히스톤 3 아세틸화(H3Ac)를 통한 De-Etiolated 1 (DET1)의 애기장대 생체시계 조절 (Regulation of Arabidopsis Circadian Clock by De-Etiolated 1 (DET1) Possibly via Histone 3 Acetylation (H3Ac))

  • 자기 현가적(self-sustaining) 조절 장치인 생체시계는 24시간 주기의 생체리듬을 조절하며 또한 생물체로 하여금 매일 변화하는 자연환경의 외부 신호를 인지할 수 있도록 해준다. 생체시계 유전자의 발현 조절은 전사/해독의 역환류 기작을 통해 이루어진다. 애기장대 LATE ELONGATED HYPOCOTYL (LHY)와 CIRCADIAN CLOCK-ASSOCIATED 1 (CCA1)는 아침에 최고조로 발현되며 해독된 LHY and CCA1는 저녁에 최고로 발현되는 TIMING OF CAB EXPRESSION1 (TOC1)의 발현을 억제한다. TOC1단백질은 LHY와 CCA1 발현을 촉진시킴으로써 생체시계의 핵심 진자(oscillator)를 형성한다. 동물에서 생체시계의 주요 전사 인자인CLOCK은 아세틸화효소 활성 기능을 가지며, 이는 생체시계의 기능 유지에 아세틸화의 중요함을 의미한다. 하지만 애기장대 생체시계에 아세틸화를 담당하는 인자에 대한 정보는 현재 보고된 바가 없다. 본 연구에서 DET1 (De-Etiolated1)는 암조건하에서 애기장대 생체시계 관련 핵심인자 중 하나인 LHY발현을 억제하는데 필요하며 이의 억제는 H3Ac 조절을 통해 이루어짐을 증명하였다. 하지만 LHY 아세틸화를 담당하는 효소의 발굴 및 이들 효소와 DET1과의 연결을 찾는 문제는 여전히 미재로 남아있다.

Drosophila CrebB is a Substrate of the Nonsense-Mediated mRNA Decay Pathway that Sustains Circadian Behaviors

  • Post-transcriptional regulation underlies the circadian control of gene expression and animal behaviors. However, the role of mRNA surveillance via the nonsense-mediated mRNA decay (NMD) pathway in circadian rhythms remains elusive. Here, we report that Drosophila NMD pathway acts in a subset of circadian pacemaker neurons to maintain robust 24 h rhythms of free-running locomotor activity. RNA interference-mediated depletion of key NMD factors in timeless-expressing clock cells decreased the amplitude of circadian locomotor behaviors. Transgenic manipulation of the NMD pathway in clock neurons expressing a neuropeptide PIGMENT-DISPERSING FACTOR (PDF) was sufficient to dampen or lengthen free-running locomotor rhythms. Confocal imaging of a transgenic NMD reporter revealed that arrhythmic Clock mutants exhibited stronger NMD activity in PDF-expressing neurons than wild-type. We further found that hypomorphic mutations in Suppressor with morphogenetic effect on genitalia 5 (Smg5) or Smg6 impaired circadian behaviors. These NMD mutants normally developed PDF-expressing clock neurons and displayed daily oscillations in the transcript levels of core clock genes. By contrast, the loss of Smg5 or Smg6 function affected the relative transcript levels of cAMP response element-binding protein B (CrebB) in an isoform-specific manner. Moreover, the overexpression of a transcriptional repressor form of CrebB rescued free-running locomotor rhythms in Smg5-depleted flies. These data demonstrate that CrebB is a rate-limiting substrate of the genetic NMD pathway important for the behavioral output of circadian clocks in Drosophila.

Effect of mPER1 on the Expression of HSP105 Gene in the Mouse SCN

  • The suprachiasmatic nucleus (SCN) of the anterior hypothalamus is the circadian pacemaker entrained to the 24-hr day by environmental time cues. Major circadian genes such as mPeriod ($mPer1{\sim}3$) and mCryptochrome ($mCry1{\sim}2$) are actively transcribed by the action of CLOCK/BMAL heterodimers, and in turn, these are being suppressed by the mPER/mCRY complex. In the study, the locomotor activity rhythms of mPer1 Knockout (KO) mice are measured, and the expression profiles of Heat Shock Protein 105kDa (HSP 105) genes in the SCN were measured by in situ hybridization. In agreement with previous reports, the locomotor activity rhythm of mPer1 KO mice was much shorter than that of wildtype. In addition, the total bout of activity of mPer1 KO was less in comparison to control mice. The expression of HSP 105 in the SCN of mPer1 KO mice was ranged from CT6 to CT22, with a peak level at CT14, implying that the gene are under the control of circadian clock. However, the expression of HSP 105 in the SCN of wildtype could not be detected in our study. Further analysis will reveal the direct or indirect regulation by mPer1 on the expression in the SCN and the role of the gene in the circadian clock.

고지방식이에 의한 말초 생체시계 유전자 발현 변화 (High-fat Intake is Associated with Alteration of Peripheral Circadian Clock Gene Expression)

  • Purpose: Recent studies demonstrated disruption of the circadian clock gene is associated with the development of obesity and metabolic syndrome. Obesity is often caused by the high calorie intake, In addition, the chronic stress tends to contribute to the increased risk for obesity. To evaluate the molecular mechanisms, we examined the expression of circadian clock genes in high fat diet-induced mice models with the chronic stress. Methods: C57BL/6J mice were fed with a 45% or 60% high fat diet for 8 weeks. Daily immobilization stress was applied to mice fed with a 45% high fat for 16 weeks. We compared body weight, food consumption, hormone levels and metabolic variables in blood. mRNA expression levels of metabolic and circadian clock genes in both fat and liver were determined by quantitative RT-PCR. Results: The higher fat content induced more severe hyperglycemia, hyperlipidemia and hyperinsulinemia, and these results correlated with their relevant gene expressions in fat and liver tissues. Chronic stress had only minimal effects on metabolic variables, but it altered the expression patterns of metabolic and circadian clock genes. Conclusion: These results suggest that the fat metabolism regulates the function of the circadian clock genes in peripheral tissues, and stress hormones may contribute to its regulation.

Actionspectra for Circadian Melatonin Rhythms in the Avian Pineal In Vitro

  • The avian pineal as well as the retina has been known to contain several types of photoreceptors with different visual pigments such as rhodopsin, iodopsin and the pineal specific opsin, pinopsin. These organs are also known to have circadian clock to regulate melatonin production. Exposure of animals to light causes a decline of the melatonin level and the phase shifts of melatonin rhythms in the pineal and retina. Therefore, the circadian clock system of these organs seem to consist of three elements, i.e., light input, oscillator and melatonin output systems. In birds, it was suggested that rhodopsin might be involved in the entrainment of pineal melatonin rhythms from the action spectrum experiment for controlling NAT activity rhythms. However, there are much more pinopsin-immunoreactive (Pino-IR) cells than rhodopsin (Rho-IR) and iodopsin (Iodo-IR) cells in the avian pineal. We found that Pino-IR cells appeared earlier embryonic stages than Rho-IR and Iodo-IR cells. So, we tried to identify the visual pigments involved in the circadian melatonin rhythms in the pineal and retina. Organ cultured pineals were exposed to monochromatic light to find out which opsin participates in regulation of melatonin rhythms. The action spectra showed a peak at 475nm, suggesting that pinopsin is the major photopigment to regulate melatonin production in birds.

  • The secretion of melatonin exhibits a circadian rhythm entrained with the sleep-wake cycle. An alteration of this secretory rhythm has been found in various psychiatric disorders. This review summarizes the regulation of melatonin and its relationship to the circadian rhythm, major depressive disorder, bipolar disorder, seasonal affective disorder, Alzheimer's disease and autism. The review also looks at the effect of melatonin and melatonin agonist on sleep and symptoms of depression, bipolar disorder and seasonal affective disorder. In Alzheimer's disease, the circadian rhythm alterations are associated with the change of melatonin levels and melatonin receptors. It has been reported that melatonin and melatonin synthetic enzyme levels decrease in autism spectrum disorder.

족부의 냉각과 가온이 행동성 체온조절에 미치는 영향 (Effect of Feet Cooling and Feet Warming on the Behavioral Temperature Regulation)

  • This study was conducted to investigate the effects of the thermally different states of human feet on temperature regulation in winter season. Five healthy female students of age 20 volunteered as subjects to participate in the study. Physiological responses such as rectal temperature and skin temperatures as well as subjective responses of thermal comfort and thermal sensation were observed. Preferred clothing and preferred temperature were also evaluated in terms of behavioral temperature regulation. The results obtained through the experiment were statistically analyzed using paired t test. Rectal temperature was decreased greater (p<.01) and mean skin temperature was maintained higher (p<.01) in feet wanning than in feet cooling. Results of preferred clothing were coincident with those of general thermal sensation. There was a higher tendency to prefer temperature in feet wanning than feet cooling in the morning. It was concluded that keeping feet skin temperature lower in the early morning and higher in the late evening would be effective in terms of regulating circadian rhythm of core temperature.
