Agrobacterium tumefaciens에 대한 약용식물의 탐색 I (The Screening of Medicinal Plant Extracts against Agrobacterium tumefaciens I)

  • 본 연구는 약용식물로부터 항균활성 물질을 찾기 위하여 Glycyrrhiza uralensis, Dryopteris crassirhizoma, Dictamnus albus, Paeonia lactiflora, Angelica dahurica, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum falcatum, Magnolia kobus, Artemisia princeps, Arctium lappa, Aster tataricus, Hovenia dulcis, Citrus unshiu, Asparagus cochinchinensis, Gardenia jasminoides, Smilax china, Hovenia dulcis, Prunus sargentii, Scutellaria baicalensis등의 약용식물을 사용하였다. 총 20종류의 약용식물로부터 에탄올 추출물을 분리하여 Agrobacterium tumefaciens에 대한 항균활성을 조사하였다. Glycyrrhiza uralensis, Dryopteris crassirhizoma, Cimicifuga heracleifolia, Bupleurum falcatum,의 추출물이 Agrobacterium tumefaciens에 대한 항균활성을 나타냈다.

승마 추출물의 항알레르기 효과 (Antiallergic Effect of Cimicifuga heracleifolia Extract)

  • Effects of the aqueous extract of Cimicifuga heracleifolia (CHAE) on the allergic reactions were investigated. CHAE inhibited systemic anaphylaxis induced by compound 48/ 80 in mice dose-dependently. Especially, CHAE inhibited compound 48/80-induced systemic anaphylaxis 100% with a dose of 0.5mg/g body weight. CHAE significantly inhibited serum histamine levels induced by compound 48/80. CHAE inhibited histamine release from the rat peritonea] mast cells activated by compound 48/80 or anti-DNP IgE. Our studies provide evidence that CHAE will be beneficial in the treatment of anaphylias.

數種의 韓藥 抽出物이 抗알레르기 反應에 미치는 影響 (The study on the anti-allergic effect of a number of herb-extract.)

  • This experimental study was done to research effects of a number of extract on the anti-allergic effect. The results were obtained as follows: 1. In effect of herb-extract on compound $\frac{48}{80}$-mediated histamine release from Evans blue skin assay, Isatis Japonica Miquel, Dictamnus dasycarpus Turcz, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum chinense, Magnolia liliflora, Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(MtOH), Trichosanthes kirilowii, Phellodendron amurense Rupr, Schizonepeta tenuifolia Var, Betula platyphylla show considerable visible anti-allergic effect. In the result of quantification of histamine induced compound $\frac{48}{80}$, Spirodela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively. 2. In effect of Herb-Extract on compound $\frac{48}{80}$-mediated histamine release from RPMC, Spirodcla polyrhiza, Cimicifuga heracleifolia inhibit histamine release effectively. 3. In effect of Herb-Extract on anti-DNP IgE-mediated histamine release from Evansblue skin assay. Spirodela polyrhiza, Cimicifuga heracleifolia(0.1mg/ml). Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(0.1mg/ml), Trichosanthes kirilowii(0.1mg/ml) show considerable visible anti-allergic effect. In the result of quantification of histamine induced anti-DNP IgE, Spiradela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively 4. In the result of genetic manifestative inhibition about the human mast cell-1(HMC-1), Cimicifuga heracleifolia has considerable effect in IL-4 in IL-5, and Tussilage farfara L. has in IL-4. According to the above results, it is suggested that several Herb-Extract have anti-allergic effect.

Antimicrobial Activity of Medicinal Plant Extracts against Streptococcus mutans II

  • This study was carried out to research antimicrobial agents from medicinal plants, Pueraria thunbergiana, Sophora flavescens, Lonicera japonica, Pinellia ternata, Dictamnus dasycarpus, Paeonia lactiflora, Angelica dahurica, Spirodela polyrhiza, Cryptotympana pustulata, Pinus densiflora, Cimicifuga heracleifolia, Bupleurum falcatum, Magnolia kobus, Artemisia princeps, Houttuynia cordata, Forsythia koreana, Arctium lappa, Castanea crenata, Aster tataricus, Citrus unshiu, Gardenia jasminoides, Ledebouriella seseloides, Hovenia dulcis, Prunus sargentii. The ethanol extracts of 24 medicinal plants were tested for the antimicrobial activity against Streptococcus mutans. The extracts of Sophora flavescens, Pinus densiflora, Cimicifuga heracleifolia, Gardenia jasminoides and Ledebouriella seseloides showed antimicrobial activity against Streptococcus mutans. The extracts of Pinus densiflora and Sophora flavescens among these medical plants showed significant antimicrobial activity against Streptcoccus mutans. These results suggested that the extracts from Pinus densiflora and Sophora flavescens could be the potential source of antimicrobial agents against S. mutans.

  • Inflammation is a biological response caused by overactivation of the immune system and is controlled by immune cells via a variety of cytokines. The overproduction of pro-inflammatory cytokines enhances abnormal host immunity, resulting in diseases such as rheumatoid arthritis, cardiovascular disease, Alzheimer's disease, and cancer. Inhibiting the production of pro-inflammatory cytokines such as interleukin (IL)-12p40, IL-6, and tumor necrosis factor $(TNF)-{\alpha}$ might be one way to treat these conditions. Here, we investigated the anti-inflammatory activity of compounds isolated from Cimicifuga dahurica (Turcz.) Maxim., which is traditionally used as an antipyretic and analgesic in Korea. In primary cell culture assays, 12 compounds were found to inhibit the production of pro-inflammatory cytokines (IL-12p40, IL-6, and $TNF-{\alpha}$) in vitro in bone marrow-derived dendritic cells stimulated with LPS.

Cimicifuga heracleifolia Extract Induces iNOS Expression via a Nuclear Factor-${\kappa}B$-dependent Pathway in Mouse Peritoneal Macrophages

  • Cimicifuga heracleifolia extract (CHE) was investigated for its effects on the release of nitric oxide (NO) and at the level of inducible nitric oxide synthase (iNOS) gene expression in mouse macrophages. We found that C. heracleifolia elicited a dose-dependent increase in NO production and the level of iNOS mRNA. Since, iNOS transcription has been shown to be under the control of the transcription factor $NF-{\kappa}B$, the effects of CHE on $NF-{\kappa}B$ activation were examined. Transient expression assays with $NF-{\kappa}B$ binding sites linked to the luciferase gene revealed that the increased level of iNOS mRNA, induced by CHE, was mediated by the $NF-{\kappa}B$ transcription factor complex. By using DNA fragments containing the $NF-{\kappa}B$ binding sequence, CHE was shown to activate the protein/DNA binding of $NF-{\kappa}B$ to its cognate site, as measured by electrophoretic mobility shift assay. These results demonstrate that C. heracleifolia stimulates NO production and is able to up-regulate iNOS expression through $NF-{\kappa}B$ transactivation.

대표적 근경류 생약의 방사선 방호효과 (The Radioprotective Effects of Rhizomata Herbs)

  • 한의학에서 근경류(rhizomata)에 분류되어 있으며 각종 보익탕제에 주로 사용되는 대표적 생약인 백출(Baizhu, Atractylodes japonica), 천궁(Chuanxiong, Cnidium officinale), 산약(Shanyao, Discorea japonica) 및 승마(Shengma, Cimicifuga heracleifolia)의 방사선 방호효과를 확인하기 위해서 고선량 및 저선량의 방사선을 마우스에 조사하고 소장움 생존, 내재성 비장집락형성 및 apoptosis 유발 등을 관찰하였다. 방사선조사전 승마 투여 군에서 소장움세포의 생존율을 증가시켰으며 내재성 비장집락 형성 시험에서는 천궁 투여 군에서 평균치의 증가가 관찰되었고 천궁 및 승마는 저선량 방사선에 의한 apoptosis 형성을 억제시켰다. 이상의 결과에서 승마 및 천궁의 방사선 방호효과를 조혈세포의 생존과 회복, 소장움세포 생존을 통하여 확인하였으며 이는 독성이 적은 천연물이라는 관점에서 방사선 방호 식품으로서 적용이 가능할 것으로 사료된다.

Structure-Guided Identification of Novel Phenolic and Phenolic Amide Allosides from the Rhizomes of Cimicifuga heracleifolia

  • Two phenolic allopyranosides and two phenolic amide allopyranosides, along with eight known phenolic compounds, including cimicifugic acids, shomaside B, fukiic acid, isoferulic acid, and piscidic acid, were isolated from the n-butanolic extract of rhizomes of Cimicifuga heracleifolia. On-line spectroscopic data for UV, NMR, and MS from a combination of LC-NMR and LC-MS techniques directly and rapidly provided sufficient structural information to identify and confirm all the structures of major phenolic compounds in the extract, in addition to their HPLC profiles. This combined analytic information was then used as a dereplication tool for structure-guided screening in order to isolate unknown phenolic compounds in the extract. Successive fractionation and purification using semi-preparative HPLC acquired four unknown allopyranosides, and their structures were identified as cis-ferulic acid 4-O-${\beta}$-D-allopyranoside, trans-ferulic acid 4-O-${\beta}$-D-allopyranoside, trans-feruloyltyramine 4-O-${\beta}$-D-allopyranoside, and trans-feruloyl-(3-O-methyl)dopamine 4-O-${\beta}$-D-allopyranoside, based on a subsequent spectroscopic interpretation.

추출조건에 따른 진피(陳皮) 및 승마(升麻) 추출물의 미백 활성효과 (Inhibitory Effect of Citrus Unshiu Peel and Cimicifuga Dahurica Extracts on Melanogenesis accroding to the Various Extraction Methods)

  • Objectives: Excessive melanin production leads to skin pigmentation, which causes various cosmetic and health problems. Citrus unshiu Markovich peel (CS) and Cimicifuga dahurica (CD) have long been widely used as a oriental medicinal plant because of their pharmacological properties including anti-ulcer, anti-oxidant, and anti-inflammatory properties. Methods: In this study, we investigated the inhibitory effects of CS, CD extracts or CS:CD=1:2 mixture on melanogenesis according to the various extraction methods. CS and CD extracted were prepared by ethanol extraction (EE), ultrasonification extraction (USE), Supercritical extraction (SCE), reflux extraction (RE), respectively. Results: DPPH radical scavenging and tyrosinase inhibitory activity of CD extracts or CS:CD=1:2 mixture were increased in dose-dependent manners. In addition, we evaluated the effect of CS, CD extracts or CS:CD=1:2 mixture on tyrosinase activity and melanogenesis in α-melanocyte stimulating hormone-induced B16-F10 melanoma cells. CS, CD extracts or CS:CD=1:2 mixture significantly inhibited tyrosinase activity and melanogenesis at 10-200 ㎍/mL. Conclusions: Therefore, our study suggests that CS and CD extracts have potential as a safe treatment for excessive pigmentation or as a natural ingredient in cosmetics.

Phylogenetic Contributions of Partial 26S rDNA Sequences to the Tribe Helleboreae (Ranunculaceae)

  • Monophyly and intergeneric relationships of the tribe Helleboreae, sensu Tamura, and related genera were studied using a 1,100-bp segment at the 5'end of the 26S ribosomal RNA gene. Forty-one OTUs, including eight species of the Helleboreae, were either directly sequenced or obtained from previous publications. Data were analyzed using distance and discrete character methods to infer phylogenetic relationships among the included taxa. The inferred phylogeny did not support monophyly of either Helleboreae or Cimicifugeae whose members were intermixed in our inferred phylogeny. This result is congruent with our previous study, which recommended against finely subdividing, suprageneric higher taxa within the R-chromosome group (subfamily Ranuncluloideae, sensu lato) until more molecular data were accumulated. Our data convincingly suggest the presence of the following three monophyletic groups: the Cimicifuga group (the clade of Actaea, Cimicifuga, Souliea, Eranthis, Anemonopsis, and Beesia), the Trollius group (the clade of Trollius, Megaleranthis, Adonis), and a clade including Anemonopsis and Beesia. Our data also suggest that Trollius and Megaleranthis might be congeners and Eranthis a paraphyletic group.
