Effect of Cimicifuga rhizoma extract on the odontoblastic differentiation of MDPC-23 cells (승마추출물이 MDPC-23세포의 분화에 미치는 영향)

  • Objectives: The purpose of this study was to examine the cell proliferation and expression of alkaline phosphatase (ALP) during the differentiation of murine odontoblast-like cells (MDPC-23) by Cimicifuga rhizoma extract. Cimicifuga rhizoma extract was prepared using 70% ethanol. Then, the cells were treated with 25, 50, 100, 150, and $200{\mu}g$ of Cimicifuga rhizoma extract. Methods: We determined the Cimicifuga rhizoma effects of MDPC-23 using WST-1 (water soluble tetrazolium salt-1) assay, ALP activity assay and histochemical staining. Results: $25-200{\mu}g$ of Cimicifuga rhizoma extract did not inhibit the growth of MDPC-23 cells; $100{\pm}0$, $100{\pm}3.29$, $99{\pm}4.86$, $98{\pm}3.80$, $98{\pm}1.73$, $99{\pm}5.05%$ (p<0.794). $50{\mu}g$ of Cimicifuga rhizoma extract stimulated ALP activity on MDPC-23; $5.1{\pm}0.20units/{\mu}{\ell}$ (p<0.001). Conclusions: It was proven that Cimicifuga rhizoma promoted differentiation of MDPC- 23 cells.

The Effect of Cimicifuga heracleifolia on Carben Tetrachloride-induced Hepatotoxicity in Rats (승마추출물이 흰쥐의 사염화탄소 유발 간독성에 미치는 효과)

  • The effect of Cimicifuga heracleifolia on CC1$_4$-induced hepatotoxicity was investigated. Cimicifuga heracleifolia has been used to diaphoresis, antipyretics and detoxification in oriental remedy. We examined the effect of Cimicifuga heracleifolia methanol extract by blood chemical analysis and histopathologic examination. ALT and AST were decreased by 38% and 67% in pretreatment group of Cimicifuga heracleifolia compared to CC1$_4$control group respectively. There were significant changes neither in total protein, albumin, triglyceride and cholesterol nor in BUN and creatinine. In histopathologic examination, there were severe necrosis and hemorrhage with infiltration of inflammatory cells around the central vein, zone 3 in the liver of CC1$_4$ treated rat. Ballooning degeneration of hepatocytes were frequently noted in the periphery of the hemorrhagic necrosis. In Cimicifuga heracleifolia pretreatment group, we observed mild degree of ballooning degeneration and inflammatory cell infiltration. No gross necrosis was observed. We measured malondialdehyde (MDA) formation by TBA method. It showed that the formation of MDA in Cimicifuga heracleifolia pretreatment group was decreased compared to the $CCl_4$control group. We got the result of the effect of Cimicifuga heracleifolia on CC1$_4$-induced hepatotoxicity by decreasing serum ALT and AST. It seems that the decrease of lipid peroxidation is related to the recovery effect.

  • This report describes the studies as follows through the research into the data of Cimicifuga heraclefolia Komarov blended prescriptions from Donguibogam. Cimicifuga heracleifola Komarov blended prescriptions are utilized for 39 therapeutic ranges. such as in dentistry, onclology, and 16 therapeutic ranges occupy over 84% totally, from dentistry to nasal disease. Cimicifuga heracleifola Komarov blended prescriptions use in the symptpms resulting from dentistry, intestine, internal disease, and they are used 22 disease also. The main pathologies of Cimicifuga heracleifola Komarov blended prescriptions found weakness in the digestive system, heat of stomach, wind of stomach in the splanchnology, and pathologies of wind-heat, wind-moisture, cold, excluding they are using for energy-blood, weakness, internal disease. The actual amount of Cimicifuga heracleifola Komarov blended has a wide capacity from 2 pun to 5 don, 1 don is broadlly used by 26%. Through the research into the main factors that we know Cimicifuga heracleifola Komarov blended prescriptions are listed in Yangmyungkyung, as they are regularly used as disease treatment and increasing the level of the general energy of the body, relaxing a skin, increasing stamina, detoxication, eliminating tumor, widely.

Anti Allergic Effects of Cimicifuga Racemosa on Allergic Models (승마(升麻)추출물이 RBL-2H3 Cell과 Mouse에 미치는 항알레르기 효과)

  • Cimicifuga racemosa (Black cohosh) has been used as therapeutics for pain and inflammation in Korean folk medicine. The potential effects of cimicifuga racemosa extract on mast cell dependent allergy reaction, however, have not been well elucidated yet. In the present study, we investigated the effect of cimicifuga racemosa extract on the allergy reaction using mast cell dependent in vivo and in vitro models. The oral administration of cimicifuga racemosa extract showed inhibitory potential on the compound 48/80 induced active systemic anaphylactic shock. cimicifuga racemosa extract also significantly inhibited the anti DNP IgE induced passive cutaneous anaphylaxis (PCA) reaction and acetic acid induced vascular permeability. In addition, cimicifuga racemosa extract inhibited the beta hexosaminidase release and TNF alpha and IL 4 mRNA induction by DNP HSA in rat leukemia mast cells, RBL 2H3. but cimicifuga racemosa extract didn't affected to RBL 2H3 cell viability. These results demonstrated that cimicifuga racemosa extract has an anti allergic potential and it may be due to the inhibition of histamine release and cytokine gene expression in the mast cells.

Study on the Effect of Cimicifuga heracleifolia Ethanol Extract on Hyaluronic Acid Synthesis

  • Hyaluronic acid (HA) is a mucopolysaccharide, occurring naturally in living organisms. It is one of the most hydrophilic molecules, so it has been known as being related to skin hydration and skin aging. The purpose of this study was to examine the effects of Cimicifuga heracleifolia ethanol extract on the hyaluronic acid synthesis and the inhibition of hyaluronidase activity. To determine cytotoxicity, hyaluronic acid synthase 2 (HAS2) gene expression, HA production and, hyaluronidase inhibitory effects, 3-(4,5-dimethylthiazol-2-ly)-2,5-diphenyl tetrazolium bromide (MTT) assay, real time - polymerase chain reaction (RT-PCR), hyaluronic acid enzyme linked immunosorbent assay (HA-ELISA), and hyaluronidase assay were used, respectively. When the Cimicifuga heracleifolia extract was treated in the HaCaT cells up to 500 ㎍/mL concentration, cytotoxicity was confirmed by the Cimicifuga heracleifolia extract at concentrations above 200 ㎍/mL. Therefore, the optimum concentration of all experiments used in this study was determined to be 200 ㎍/mL. HAS2 gene expression increased by Cimicifuga heracleifolia extract in a concentration-dependent manner at all treatment concentrations. The production rate of HA was tended to decrease at the highest concentration of 200 ㎍/mL. The hyaluronidase activity inhibition effect of Cimicifuga heracleifolia extract was very high compared to the control group. Based on these results, Cimicifuga heracleifolia extract was expected to have a moisturizing effect on human skin and special attention should be paid to the determination of the concentration of Cimicifuga heracleifolia when developing cosmetic materials using it.

  • Seung Ma(升摩) is a famous Chinese crude drug used for treatment of fever, piles and suppurative inflamation. With regard to the botanical origin of Seung Ma, it has been considered to be Cimicifuga spicies of Ranunculaceae, but there has no phrarmacognostical confirmation on it. To clarify the botanical origin of Seung Ma from Korea, the anatomical characteristics of Cimicfuga dahurica, Cimicifuga foetida, Cimicfuga heracleifolia and Cimicifuga simplex were studied. As a result, it was clarified that Seung Ma from Korea was the rhizomes of Cimicifuga dahurica, C, foetida and C, heracleifolia.

Anti-allergic and Anti-inflammatory Actions of Cimicifuga heracleifolia: Partial Purification of Active Components

  • Anti-allergic and anti-inflammatory actions of the water extract from Cimicifuga heracleifolia were evaluated in mice and rats. Several criteria were employed to assess the anti-allergic and anti-inflammatory actions of Cimicifuga heracleifolia, such as hyaluronidase activity, mediators-induced vascular permeability changes, 48 hour homologous passive cutaneous anaphylaxis (PCA) histamine release from mast cells, and the carrageenan-induced rat paw edema. To further characterize the active components, the water extract was either extracted with organic solvent or fractionated according to molecular weight, and each fraction was tested for some of anti-allergic parameters. Hyaluronidase activities, both in activating and in activated states, were significantly inhibited by the water extract of Cimicifuga heracleifolia and by some of its subfractions, molecular weight less than 1,000. The water extracts (50~400 mg/kg) significantly inhibited 48 hr homologous PCA and vascular permeability changes induced by chemical mediators (histamine, serotonin, and leukotriene $C_4$) in mice. In the case of histamine-induced vascular permeability changes, more extensive studies were conducted; water extract was either fractionated according to molecular weight or extracted with butanol. Anti-histamine actions were observed only from the water layer, and these active components were of the molecular weight less than 1,000. These anti-allergic actions were observed mainly from mice than from rats. On the other hand, anti-inflammatory actions of the water extract from Cimicifuga heracleifolia were significant in rats.

A Systematic Study on the Cimicifuga foefida L Complex and Related Species (Ranunculaceae) (Cimicifuga foetida L. Complex 및 근연종(미나리아재비과)에 대한 분류학적 연구)

  • The delimitation and systematic relationships of the C. foetida complex and related species were evaluated by numerical methods, including principal components analysis and cluster analysis of major morphological characters. Results of these analyses strongly support recognition of three varieties in C. foetida; these include C. foetida var. foetida, C. foetida var. micrantha, and C. foetida var. velutita. The results also indicated that C. brachycarpa, C. frigida, C. heracleifolia, and C. yunnanensis are distinct species, and C. heracleifolia comprises C. heracleifolia var. heracleifolia and C heracleifolia var. bifida. Cimicifuga mairei and C. foetida var. foliolosa, however, appeared to be morphologically indistinguishable from C. foetida var. velutina. In addition, C. foetida var. longibracteata and C. foetida var. bifula are conspecific with C. frigida.rigida.

Trichome Morphology of Cimicifuga L. (Ranunculaceae) and Its Taxonomic Significance

  • Trichomes found in the genus Cimicifuga were examined with scanning electron microscopy. Four basic types of trichomes were found in the genus; (1) filiform unicellular trichomes, (2) saccate unicellular trichomes, (3) pyriform unicellular trichomes, and (4) uniseriate multicellular trichomes. All the taxa examined develop at least two different types of trichomes, and many taxa are distinguished by differences in type, microornamentation, density, and position of trichomes. In addition, trichome features appear to be very useful in recognizing species relationships in the genus.
