Prediction of Genomic Relationship Matrices using Single Nucleotide Polymorphisms in Hanwoo (한우의 유전체 표지인자 활용 개체 혈연관계 추정)

  • The emergence of next-generation sequencing technologies has lead to application of new computational and statistical methodologies that allow incorporating genetic information from entire genomes of many individuals composing the population. For example, using single-nucleotide polymorphisms (SNP) obtained from whole genome amplification platforms such as the Ilummina BovineSNP50 chip, many researchers are actively engaged in the genetic evaluation of cattle livestock using whole genome relationship analyses. In this study, we estimated the genomic relationship matrix (GRM) and compared it with one computed using a pedigree relationship matrix (PRM) using a population of Hanwoo. This project is a preliminary study that will eventually include future work on genomic selection and prediction. Data used in this study were obtained from 187 blood samples consisting of the progeny of 20 young bulls collected after parentage testing from the Hanwoo improvement center, National Agriculture Cooperative Federation as well as 103 blood samples from the progeny of 12 proven bulls collected from farms around the Kyong-buk area in South Korea. The data set was divided into two cases for analysis. In the first case missing genotypes were included. In the second case missing genotypes were excluded. The effect of missing genotypes on the accuracy of genomic relationship estimation was investigated. Estimation of relationships using genomic information was also carried out chromosome by chromosome for whole genomic SNP markers based on the regression method using allele frequencies across loci. The average correlation coefficient and standard deviation between relationships using pedigree information and chromosomal genomic information using data which was verified using a parentage test andeliminated missing genotypes was $0.81{\pm}0.04$ and their correlation coefficient when using whole genomic information was 0.98, which was higher. Variation in relationships between non-inbred half sibs was $0.22{\pm}0.17$ on chromosomal and $0.22{\pm}0.04$ on whole genomic SNP markers. The variations were larger and unusual values were observed when non-parentage test data were included. So, relationship matrix by genomic information can be useful for genetic evaluation of animal breeding.

Analysis of SNPs in Bovine CSRP3, APOBEC2 and Caveolin Gene Family (소의 CSRP3, APOBEC2, Caveolin 유전자들의 단일염기다형 분석)

  • The cysteine and glycine rich protein 3 (CSRP3), apolipoprotein B mRNA editing enzyme catalytic polypeptide‐like 2(APOBEC2) and caveolin (CAV) gene family(CAV1, CAV2, CAV3) have been reported to play important roles for carcass and meat quality traits in pig, mouse, human and cattle. As an initial step, we investigated SNPs in these 5 genes among eight different cattle breeds. Eighteen primer pairs were designed from bovine sequence data of NCBI database to amplify the partial gene fragments. Sequencing results revealed 9 SNPs in the coding regions of three caveolin genes, 1 SNP in CSRP3 and 3 SNPs in APOBEC2 gene. All the identified SNPs were confirmed by PCR-RFLP. Also, 9 more intronic SNPs were detected in these genes. However, all identified mutations in the coding region do not change amino acid sequence. Allelic distributions were significantly different for 5 SNPs in CAV2, CAV3, CSRP3 and APOBEC2 genes among the eight different breeds. These results gave some clues about the polymorphisms of these genes among the cattle breeds and will be useful for further searches for identifying association between these SNPs and meat quality traits in cattle.

Histopathologic Changes of Articular Cartilage and Subchondral Bone in Cylindrical Biopsy Specimen from Talar Osteochondral Lesions (거골의 골연골 병변의 원주형 생검에서 관절 연골과 연골하 골의 조직병리학적 변화)

  • Purpose: This study was aimed at elucidating the pathogenesis of talar osteochondral lesion by analyzing the histopathological findings. Materials and Methods: Twenty specimens from 20 patients who underwent surgical treatment for talus osteochondral lesions were studied. Preoperative MRI images including T1, T2, and stir images were taken and cases were classified according to modification of the Anderson's classification. There were 5 cases of MRI group 1, 6 cases of group 2, 7 cases of group 3 and 2 cases of group 4. A full thickness osteochondral plug including the osteochondral lesion of the talus was harvested from each patient and reviewed histopathologic changes of osteochondral fragment using H-E staining. Mean diameter of specimens was 8.5 mm and mean depth was 10.3 mm. Pathologic changes of articular cartilage and subchondral bone were observed. Subchondral bone was divided into superficial, middle and deep zones according to depth. Cartilage formation, trabecular thickening and marrow fibrosis were observed in each zone. Results: There were detachment of the joint cartilage at the tidemark in 16 cases of 20 cases and the separated cartilages were almost necrotic on the histopathologic findings. Cartilage formation within subchondral bone was discovered beneath the tidemark in 12 cases. Trabeculae were increased and thickened in 17 cases. These pathologic changes were similar to fracture healing process and these findings were more conspicuous near the tidemark and showed transition to normal bone marrow tissue with depth. No correlation between the pathological progression and MRI stages was found. A large cyst shown on MRI's was microscopically turned out to be multiple micro-cysts accompanied by fibrovascular structure and newly formed cartilage tissue. Conclusion: The histopathologic findings of osteochondral lesions are detachment of overlying cartilage at the tidemark and subsequent changes of subchondral bone. Subchondral bone changes are summarized as cartilage formation, marrow fibrosis and trabecular thickening that mean healing process following repeated micro fractures of trabecular. These osteochondral lesions should have differed from osteochondral fractures.

Comparison of Characteristics of Local Meteorological and Particulate Matter(TSP) on the Beopjusa Temple and Seonamsa Temple (법주사와 선암사의 국지 기상 및 미세먼지 특성 비교)

  • It is crucial to measure meteorological elements in relation to the biodeterioration of building cultural heritages. The Beopjusa and Seonamsa temples located respectively in Chungbuk and Jeonnam provinces, Korea, exhibit biological damage to the building cultural heritages and cause noticeable climatic differences. To compare biodeterioration environments of the abovementioned temples, 10 meteorological elements were observed, and particulate matter (TSP) was collected at each location. Furthermore, a correlation analysis was conducted between meteorological elements, and between meteorological elements and TSP. The local meteorology at Beopjusa temple characteristically showcased high total horizontal radiation, UV radiation, evaporation, wind speed, and TSP concentration, whereas, that at Seonamsa temple showcased high temperature, humidity, dew point temperature, air pressure, precipitation and number of days with precipitation. An elemental analysis of TSP revealed the presence of sae-salts at Seonamsa temple, and compared to that of Beopjusa temple, the monthly frequencies of biogenic aerosol and Fe-containing particles were higher. The correlation analysis showed that wind speed and humidity were major meteorological factors at Beopjusa and Seonamsa temples, respectively. Subsequently, the characteristics of the local meteorology at Seonamsa temple are expected to affect the biological damage of the building cultural heritages, which is favorable for the growth of various organisms.

Physico-chemical properties and biological activity of controlled-release granular formulations for the herbicide dicamba (방출조절형 dicamba 입제의 물리화학성 및 생물효과)

  • Dicamba (3,6-dichloro-o-anisic acid) granular formulations for controlled release (DGFCRs) were prepared with biodegradable polymers, corn starch and pregelatinized starch, to minimize harmful side effects, extend weed control performance, and control the releasing rate of the active ingredient. Physico-chemical properties and biological activity of DGFCRs were studied. Six different granules were formulated by applying two processes, granulation and extrusion. Formulation efficiencies of active ingredient (A.I.) in the granules prepared by granulating and extruding were $90.0{\sim}96.3%$. Incorporation ratios of A.I. in the granules prepared by granulating and extruding showed $89.5{\sim}94.5%$ and $46.7{\sim}82.0%$, respectively. The highest swellability was DG-2 formulation prepared with corn starch. Whereas, the lowest floatability in water was DG-2 formulation, while the highest one was DG-1 formulation prepared with pregelatinized starch, Miragel 463. The degradation rates of dicamba in the granules under the elevated temperature of $50^{\circ}C$ were less than 5% for DG-1 and DG-2 formulations even after 90 days, meanwhile, those of DE-1 formulations prepared with pregelatinized starch, Mirasperse, were more than 5%. The release rates of A.I. from the granules into water under a static condition were about 100% after 2 weeks. Weeding effects of the granules on broad leaf weeds tested in greenhouse were more than 90% after 30 days.

Sensitivity of Colletotrichum gloeosporioides Isolated from Persimmon to Benzimidazoles, Mancozeb and Propineb (감나무 탄저병균 Colletotrichum gloeosporioides의 Benzimidazole계 살균제, Mancozeb 및 Propineb에 대한 감수성)

  • In 2008, a number of 110 isolates of Colletotrichum gloeosporioides were obtained from infected twigs of persimmon (Diospyros kaki) collected at Sangju and four fungicides (carbendazim, thiophanate-methyl, mancozeb, and propineb) were evaluated to determine their growth effect on fungicide-medium. Among them, the mycelial growth of 68.2 and 35.5% of isolates was inhibited over 91% in response to carbendazim ($415{\mu}g/m{\ell}$) and thiophanate-methyl ($750{\mu}g/m{\ell}$), respectively, compared to untreated control and the correlation between the two benzimidazole fungicides was higher ($r^2=0.7886$). Moreover, in responses to mancozeb ($1,500{\mu}g/m{\ell}$) and propineb ($1,500{\mu}g/m{\ell}$), the inhibition of mycelial growth of 90% and 53.6% of isolates was inhibited over 91%, respectively. But correlation between theses two fungicides was lower ($r^2=0.0174$). There is no cross-resistance response between benzimidazole funficides and mancozeb / propineb.

Survey of Manager's Perception of Slaughter and Further Processed House for the Determination of Grading of Poultry Meat Cuts (닭고기 부분육 품질 등급 설정을 위한 도계 및 추가가공장 관리자 의식 조사)

  • This study was conducted to establish a comprehensive guideline for grading system of poultry meat cuts. Slaughter and further processed houses were divided into large(>70,000 heads/day), middle($50,000\~25,000$ heads/day), and small scale(<25,000 heads/day), and the production managers were asked by the Questionnaire. Most poultry meat cuts produced by larger- and middle-slaughter house were consumed by dealers, franchisee and department store in increasing order, whereas further processed poultry meat cuts(small scale) provided meals for the students in the high amount. The packaging step was best f3r the determination of grading in the large slaughter house. This survey indicated that two or more grades were proper to determine the grading in the middle slaughter house, however only two grades were proper for the others. Freshness, bruise, wound, trimming and the inclusion of foreign materials affected to the grading. Poultry meat cuts were accepted for the grading in large-slaughter house, but whole poultry carcass was accepted for the grading in the others. Most managers agreed with that the grading system was only applied for the amounts requested. They also suggested that the grading system of poultry meat cuts were necessary in near future, even though it was not urgent to apply.

Changes of Properties and Gas Components according to Accelerated Aging Test of Vegetable Transformer Oil (식물성 절연유의 가속열화에 따른 주요 성분 및 물성 변화)

  • Mineral oil is the most widely used for electrical transformer, though some factors should be considered such as their environmentally harmfulness when it spill and low flash point. To cover these disadvantages, vegetable oil has developed because of its high biodegradability and thermal stability. However, it is necessary that many studies should conduct to reveal the detailed impacts of long-term operation as transformer oil. In this paper, we applied the accelerated aging test which simulate the real transformer circumstances using insulation paper, coil, steel at $150^{\circ}C$, which is higher than normal operation, for 2 weeks. To figure out the oxidation characteristics between mineral oil and vegetable oil test major properties and components such as total acid number, dielectric breakdown and dissolved gas components during that period. As a result of these tests, we found that vegetable oil has higher electric insulation ability than mineral oil though poor total acid number by hydrophile property. Vegetable oil also kept its thermal stability under the given circumstances.

Hydraulic Impact Scope and Dissolved Oxygen Distribution by the Micro-bubble Aeration in an Artificial Lake (인공호소에서 마이크로 버블 포기에 의한 수리학적 영향반경과 용존산소 분포)

  • This study investigated the hydraulic impact scope and dissolved oxygen (DO) concentration distribution by the micro-bubble aeration in the Juksan Lake located in Asan city in Chungcheongnam-do province. A tracing experiment for hydraulic impact scope was used which constituted a 20% rhodamine solution. A 160 m-guideline was installed in the horizontal direction of the micro-bubble jet flow and the rhodamine concentration, water temperature, and DO concentration were measured at depths of 1 m, 2 m, and 3 m at intervals of 10 m. In the Juksan Lake, the effective range of jet flow discharged by the micro-bubble generator was about 40 m, and after then the jet plume moved up to 80 m to 120 m through the advection and diffusion processes of ambient water. DO concentration in the lake was maintained at 7.4-12.6 mg/L during tracking experiment. The DO of the lake sediments improved from 0.2 mg/L to 8.0 mg/L after applying micro-bubble aeration. In conclusion, the micro-bubble aeration can be an effective technology for the management and improvement of water quality in an agricultural reservoir.

A Study on Important Problem Features of Hospitalized Senile Dementia Patients (시설에 있는 치매노인의 주요문제특성에 대한 기초 연구)

  • The purpose of this study was to extract important problem features for care of senile dementia patients. Selected cognitive ability test (Korean Mini-Mental State Examination: K-MMSE) and survey of basic & problem characteristics were conducted on 110 hospitalized senile dementia patients and 30 normal subjects. Problem features of senile dementia patients were extracted using factor analysis. The frequency difference of problem features due to the gender and dementia severities was verified using one-way ANOVA. Twenty problem features were extracted by the factor analysis. According to the gender, there are significant differences in the frequency of problem features in violent language & confabulation, collecting behavior, and repetitive behavior. According to the dementia severities, there are significant differences in the frequency of all problem features except abnormal sexual behavior and audio-visual disorder. The result of this study is expected to be used for the development of the senile dementia patients' life-care monitoring system.
