Effect of Pycnogenol on Skin Wound Healing

  • This study was carried out to investigate the effects of pycnogenol (PYC) on the cutaneous wound healing of the mice. The wounds were extracted on days 1, 3, 5, and 7 post-injury for histomorphometrical analysis including wound area, infiltrating inflammatory cells, wound contracture including collagen deposition. As the result, the wound area of PYC-treated group was larger than the control group on days 1 to 7. Inflammatory cells in the PYC-treated wounds were decreased at day 1 compared to the control wound tissue. From day 3 to 7, there was no significant difference between the control and the PYC-treated skin wounds. Though the degree of contraction in the PYC-treated group was lower than that of the control group from days 1 to 5, but appeared significantly higher on day 7. Compared to the control group, collagen accumulation in the PYC-treated group was higher than that of the control group from days 5 to 7. From this result, it may support the possibility that PYC would be useful agent for early inflammatory response and matrix remodeling phase of the skin wounds.

Recording Support System for Off-Line Conference using Face and Speaker Recognition (얼굴 인식 및 화자 정보를 이용한 오프라인 회의 기록 지원 시스템)

  • Recent multimedia technology has supported various application services based on the development of effective movie compression and network techniques. On-line video conference system is a typical example that use theses two technologies effectively. On-line video conference system can be characterized into an effective conferencing method for long-distanced on-line conference members. But, unfortunately, off-line conference with face-to-face meeting is more frequent than on-line conference and their support systems have not been sufficiently considered. In this paper, we propose a recording support system for off-Line conference using face and speaker recognition. This system finds the speaker in the conference by using three microphones and three webcam cameras. And analysis is done with face region information that gathered by currently active webcam camera, and recognizes the identity of face. Finally, the system tracks speaker and records conference with extract speaker information.

  • 국내 해양관련 HNS 데이터베이스의 정보를 구축하는 중에 확인된 문제점들은 크게 두 가지로 요약된다. 첫 번째는 물질정보가 없는 경우로서, 많은 경우 자연물질로 다양한 성분을 함유하고 있으며, 이외에도 이성질체 및 동족체 화합물로서 확인된다. 두 번째로는 국내 HNS DB의 출처가 다양하여, 정제되지 않은 경우인데 물질정보의 환경조건이 상이하거나 같은 조건에서도 서로 다른 정보가 혼재하는 경우이다. 이러한 문제점을 해결하기 위해, 유럽의 GESAMP에서 제시한 보고서와 미국 CAMEO를 참조하여 누락정보를 확보하였으며 두 데이터베이스의 명명법의 차이에서 오는 누락 정보를 두 데이터베이스의 유사명 자료비교와 화학구조를 이용해 확인하였다. 자연물질과 그룹화합물들도 주요성분의 자료를 활용하였으며, 이성질체의 경우 단일성분의 자료를 기입하거나, 위험성이 높은 성분의 자료를 선택해서 기입하였다. 물질정보 중 IMDG 등급의 경우 주위험성외에 부위험성의 중요성을 고려하여 HNS DB에 새로 기입하였으며, 부록으로 HS 번호 부여 및 해양환경거동분류를 첨부하였다. 향후 아직 남아있는 누락정보의 확보를 위해서는 GESAMP circular report를 확보하도록 할 필요성이 있다고 판단된다.

Development and Optimization of a Rapid Colorimetric Membrane Immunoassay for Porphyromonas gingivalis

  • Porphyromonas gingivalis (P. gingivalis) is a major bacterial pathogen that causes periodontitis, a chronic inflammatory disease of tissues around the teeth. Periodontitis is known to be related to other diseases, such as oral cancer, Alzheimer's disease, and rheumatism. Thus, a precise and sensitive test to detect P. gingivalis is necessary for the early diagnosis of periodontitis. The objective of this study was to optimize a rapid visual detection system for P. gingivalis. First, we performed a visual membrane immunoassay using 3,3',5,5'-tetramethylbenzidine (TMB; blue) and coating and detection antibodies that could bind to the host laboratory strain, ATCC 33277. Antibodies against the P. gingivalis surface adhesion molecules RgpB (arginine proteinase) and Kgp (lysine proteinase) were determined to be the most specific coating and detection antibodies, respectively. Using these two selected antibodies, the streptavidin-horseradish peroxidase (HRP) reaction was performed using a nitrocellulose membrane and visualized with a detection range of 103-105 bacterial cells/ml following incubation for 15 min. These selected conditions were applied to test other oral bacteria, and the results showed that P. gingivalis could be detected without cross-reactivity to other bacteria, including Streptococcus mutans and Escherichia fergusonii. Furthermore, three clinical strains of P. gingivalis, KCOM 2880, KCOM 2803, and KCOM 3190, were also recognized using this optimized enzyme immunoassay (EIA) system. To conclude, we established optimized conditions for P. gingivalis detection with specificity, accuracy, and sensitivity. These results could be utilized to manufacture economical and rapid detection kits for P. gingivalis.

A Case Report of Post-Stroke Delirium Patient Using Complex Korean Medicine Treatment Including Scalp Electroacupuncture (뇌졸중 후 섬망의 두부 침전기자극술을 포함한 한의복합치료에 대한 증례 보고 1례)

  • Objectives: This case study aimed to provide evidence of the combined use of Korean Medicine treatments, including electroacupuncture (EA) on scalp acupoints, as a useful intervention for post-stroke delirium. Methods: The patient in this study was diagnosed with Cerebral Infarction in Right Corpus Callosum and post-stroke delirium. The patient received eight sessions of EA on scalp acupoints, rehabilitation medicine treatment, and other Korean Medicine treatments such as acupuncture and herbal medicine. Evaluation of treatment effectiveness was done mainly through the Memorial Delirium Assessment Scale Korean (MDAS-K) and Mini-Mental State Examination Korean (MMSE-K). Results: MDAS-K score dropped from 20 to 9 and MMSE-K score increased from 15 to 21 during hospitalization, showing improvement in symptoms. Conclusions: Combined use of Korean medicine treatments including EA on scalp acupoints may be an effective treatment for post-stroke delirium.

  • In this paper the performance of a array invariant method is evaluated for source-range estimation in horizontally stratified shallow water ocean waveguide. The method has advantage of little computationally effort over existing source-localization methods. such as matched field processing or the waveguide invariant and array gain is fully exploited. And. no knowledge of the environment is required except that the received field should not be dominated by purely interference This simple and instantaneous method is applied to simulated acoustic propagation filed for testing range estimation performance. The result of range estimation according to the SNR for the underwater impulsive source with broadband spectrum is demonstrated. The spatial smoothing method is applied to suppress the effect of mutipath propagation by high frequency signal. The result of performance test for range estimation shows that the error rate is within 20% at the SNR above 10dB.

A Study on Underwater Source Localization Using the Wideband Interference Pattern Matching (수중에서 광대역 간섭 패턴 정합을 이용한 음원의 위치 추정 연구)

  • This paper proposes a method of underwater source localization using the wideband interference patterns matching. By matching two interference patterns in the spectrogram, it is estimated a ratio of the range from source to sensor5, and then this ratio is applied to the Apollonius circle. The Apollonius circle is defined as the locus of all points whose distances from two fixed points are in a constant value so that it is possible to represent the locus of potential source location. The Apollonius circle alone, however still keeps the ambiguity against the correct source location. Therefore another equation is necessary to estimate the unique locus of the source location. By estimating time differences of signal arrivals between source and sensors, the hyperbola equation is used to get the cross point of the two equations, where the point being assumed to be the source position. Simulations are performed to get performances of the proposed algorithm. Also, comparisons with real sea experiment data are made to prove applicability of the algorithm in real environment. The results show that the proposed algorithm successfully estimates the source position within an error bound of 10%.

Purification, Characterization, and Cloning of Fibrinolytic Metalloprotease from Pleurotus ostreatus Mycelia

  • A fibrinolytic protease (PoFE) was purified from the cultured mycelia of the edible oyster mushroom Pleurotus ostreatus, using a combination of various chromatographies. The purification protocol resulted in an 876-fold purification of the enzyme, with a final yield of 6.5%. The apparent molecular mass of the purified enzyme was estimated to be 32 kDa by SDS-PAGE, fibrin-zymography, and size exclusion using FPLC. The optimal reaction pH value and temperature were pH 6.5 and $35^{\circ}C$, respectively. PoFE effectively hydrolyzed fibrinogen, preferentially digesting the $A{\alpha}$-chain and the $B{\beta}$-chain over the ${\gamma}$-chain. Enzyme activity was enhanced by the addition of $Ca^{2+},\;Zn^{2+},\;and\;Mg^{2+}$ ions. Furthermore, PoFE activity was potently inhibited by EDTA, and it was found to exhibit a higher specificity for the chromogenic substrate S-2586 for chymotrypsin, indicating that the enzyme is a chymotrypsin-like metalloprotease. The first 19 amino acid residues of the N-terminal sequence were ALRKGGAAALNIYSVGFTS, which is extremely similar to the metalloprotease purified from the fruiting body of P. ostreatus. In addition, we cloned the PoFE protein, encoding gene, and its nucleotide sequence was determined. The cDNA of cloned PoFE is 867 nucleotides long and consists of an open reading frame encoding 288 amino acid residues. Its cDNA showed a high degree of homology with PoMEP from P. ostreatus fruiting body. The mycelia of P. ostreatus may thus represent a potential source of new therapeutic agents to treat thrombosis.

A Fibrinolytic Enzyme from the Medicinal Mushroom Cordyceps militaris

  • In this study we purified a fibrinolytic enzyme from Cordyceps militaris using a combination of ion-exchange chromatography on a DEAE Sephadex A-50 column, gel filtration chromatography on a Sephadex G-75 column, and FPLC on a HiLoad 16/60 Superdex 75 column. This purification protocol resulted in a 191.8-fold purification of the enzyme and a final yield of 12.9 %. The molecular mass of the purified enzyme was estimated to be 52 kDa by SDS-PAGE, fibrin-zymography, and gel filtration chromatography. The first 19 amino acid residues of the N-terminal sequence were ALTTQSNV THGLATISLRQ, which is similar to the subtilisin-like serine protease PR1J from Metarhizium anisopliae var. anisopliase. This enzyme is a neutral protease with an optimal reaction pH and temperature of 7.4 and $37^{\circ}C$, respectively. Results for the fibrinolysis pattern showed that the enzyme rapidly hydrolyzed the fibrin $\alpha$-chain followed by the $\gamma$-$\gamma$ chains. It also hydrolyzed the $\beta$-chain, but more slowly. The A$\alpha$, B$\beta$, and $\gamma$ chains of fibrinogen were also cleaved very rapidly. We found that enzyme activity was inhibited by $Cu^{2+}$ and $Co^{2+}$, but enhanced by the additions of $Ca^{2+}$ and $Mg^{2+}$ ions. Furthermore, fibrinolytic enzyme activity was potently inhibited by PMSF and APMSF. This enzyme exhibited a high specificity for the chymotrypsin substrate S-2586 indicating it's a chymotrypsin-like serine protease. The data we present suggest that the fibrinolytic enzyme derived from the edible and medicinal mushroom Cordyceps militaris has fibrin binding activity, which allows for the local activation of the fibrin degradation pathway.

  • A form of polycystic ovary (PCO) resembling some aspects of the human PCO syndrome (PCOS) can be induced in rats by a single injection of estradiol valerate (EV). An increase in sympathetic outflow to the ovary precedes, by several weeks, the appearance of cysts, suggesting the involvement of a neurogenic component in the pathology of this ovarian dysfunction. To test the hypotheses that the change in sympathetic tone is related to an augmented production of hippocampal and/or ovarian nerve growth factor (NGF), and that this abnormally elevated production of NGF contributes to the induction of PCOS induced by EV. The animals were sacrificed after PCOS induction and the ovaries and hippocampus were sectioned and compared to the normal control. The expression of NGF was measured by immunohistochemical staining and Western blot analysis in the ovaries and hippocampus. EV-induced PCOS showed significant increase of ovarian NGF expression. Immunohistochemical expression of NGF was confined to the follicular cells and interstitial cells. Hippocampal NGF expression was not significantly changed. In conclusion EV-induced PCOS was related to the ovarian sympathetic activation which was mediated by NGF.
