Sex Linked Developmental Rate Differences in Murrah Buffalo (Bubalus bubalis) Embryos Fertilized and Cultured In Vitro

  • The aim of the present study was to determine the effect of paternal sex chromosome on early development of buffalo embryos fertilized and cultured in vitro. Embryos were produced in vitro from abattoir derived buffalo oocytes. The cleaved embryos were cocultured with buffalo oviductal epithelial cells and evaluated on day 7 under the phase contrast microscope to classify development. The embryos which reached the morula/blastocyst stage were fast developing, the embryos which were at 16-32 cell stage were medium developing and the embryos below 16 cell stage were slow developing. The embryos which showed some fragmentation in the blastomeres or degenerated blastomeres, were degenerating. Sex of emberyos (n=159) was determined using PCR for amplification of a male specific BRY. 1 (301 bp) and a buffalo specific satellite DNA (216 bp) fragments. The results thus obtained show that 1) X and Y chromosome bearing sperms fertilize oocytes to give almost equal numbers of cleaved XX and XY embryos, 2) male embryos develop faster than female embryos to reach advanced stage and 3) degeneration of buffalo embryos is not linked with the paternal sex chromosome. We suggest that faster development of males is due to differential processing of X and Y chromosome within the zygote for its activation and / or differential expression of genes on paternal sex chromosome sex chromosome during development of buffalo embryos fertilized and cultured in vitro which may be attributed to a combination of genetic and environmental factors.

  • The chromosome analyses of blood culture were made of 11 Korean native and 53 crossbred males between the Korean native cattles(K) and Charolais(C), which consisted of $K\times$K, $C\times$K, $C\times$CK, CK$\times$CCK and Charolais synthetic males(CK$\times$CCK or CCK$\times$CK). 1. The diploid(2n=60, XY) Charolais synthetic male has the 29 pairs of acrocentric autosomes, a single large submetacentric X and a small metacentric Y chromosome. 2. The numbers of G-band of karyotype in these males were a few differences in the 8 pairs of autosomes(chromosome 2, 4, 5, 6, 9, 11, 19 and 26) compared to those of purebred Korean native ones. G-banding qualities were not matched in chromosome 16, 19 and 29 with the Korean native males and also in chromosome 14, 20 and 22 with other domestic cattles. 3. The G-banding pattern between chromosome 4-6-7 and 24-25-27 was alomost similar together and the possibilityof misidentification was greater in the G-banded preparations. 4. 1/29 Robertsonian translocation and other abnormalities were not observed among 11 Korean native and 53 crossbred males. This result is considered in relation to limited data and further investigation based on larger samples may be necessary for definite conclusion.

  • Present investigation was carried out in order to make clear the fertility, morphological characters, and chromosome numbers of interspecific hybrids of Chrysanthemum Zauadskii, C. indicum, and C. lavandulaefolium. 1. Hybrids were not self-pollinated. When sib-crossed the fertility was 1.3~19.3%. 2. F2 individuals were variable in their morphological Characters. 3. Chromosome numbers of three putative parents were different from those reported previously: C. zawadskii 2n=36, C. indicum 2n=20, C. lavandulaefolium 2n=16, $C. indicum{\times}C. zawadskii 2n=28, C. zawadskii{\times}C. indicum-1 2n=28, C. zawadskii{\times}C. indicum-2 2n=28, C. zawadskii{\times}C. indicum-3 2n=28, C. zawadskii{\times}C. indicum-4 2n=28, C. zawadskii{\times}C. indicum-5 2n=28, C. zawadskii {\times}C. lavandulaefolium-1 2n=26, C. zawadskii{\times}C. lavandulaefolium-2 2n=26.$

  • The somatic chromosome numbers were counted to be 2n = 16 of Codonopsis lanceolata. Chromosomes 2 and 7 of Sokrisan and Koheung II has a hetero satellite on short arm. Chromosome 5 of Bakwoonsan showed homo satllite on short arms. Short arms of chromosomes 1 and 5 has homo satellite and short arms of chromosomes 2 and 7 has hetero satellite of Jilin.

Karyotypes of Korean Endemic Land Snail, Koreanohadra koreana (Gastropoda: Bradybaenidae)

  • The karyotypes of Korean endemic land snail, Koreanohadra koreana, using air-drying method wereinvestigated. Somatic cells of this species had 2n = 58. Karyotypes were also analysed with 16 metacentric, 12 submetacentric and one subtelocentric chromosome pairs. Observed chromosomes ranged from 2.6 to $8.9{\mu}m$ and the total length was $122.3{\mu}m$. This is the second report on the chromosome numbers and the karyotype of K. koreana.

  • This study was carried out to obtain the basic information on morphological and chromosomal charateristics in the three species of Chinese carps (grass carp; Ctenopharyngodon idellua, bighead carp; Aristichthys nobilis, and silver carp; Hypophthalmichthys molitrix) introduced to Korea from China. C. idellua was differ from A. nobilis and H. molitrix by the number of gill rakers, scales, fin rays, body proportion. A. nobilis and H. molitrix were similar in having ventral keel and many scale number, but H. molitrix was differ from A. nobilis by the connected gill rakers and body color pattern. Diploid chromosome and arm number (fundamental number, NF) of the three species were all the same to 2n=48 and NF=84. Diploid chromosome numbers in the three species are consisted of 10 pairs of metacentric chromosome, 8 pairs of submetacentric chromosome and 6 pairs of acro and/ or telocentric chromosome. Morphological and karyological relationship of the three Chinese carps are discussed.

  • It was revaluated about taxonomic position of Bupleurum latissimum Nakai (Apiaceae) by the cluster analysis, the principal component analysis, and the discriminant analysis. Its diagnostic characteristics include bracteole length longer than umbellules, 16-21 pedicel numbers, plant with evergreen leaves, chromosome number 2n = 16 and cauline leaves with auriculate base. It is morphologically similar to B. euphorbioides in having ovate bracteoles, 16-21 pedicel numbers, chromosome number 2n = 16, and cauline leaves with auriculate base, but distinguished from B. longiradiatum with having linear bracteoles, 7-15 pedicel numbers, and chromosome number 2n = 12. Morphological characters support that Bupleurum latissimum Nakai is a Korean endemic species.

  • The chromosome numbers and karyotypes were investigated in four Korean native species of the genus Hosta. The chromosome complements were diploid of 2n=60 in H. japonica var. lancifolia Nakai and H. capitata Nakai, aneuploid of 2n=59 in H. minor (Bak.) Nakai, and modified triploid of 2n=92 in H. longipes (Fr. et Sav.) Matsumura. All the species carried four sets of distinctly large chromosomes of which the chromosome types were telocentrics or subtelocentrics with $4.4{\sim}7.2\;{\mu}m$ in length. The other chromosomes were meta-, submeta, subtelo-, or telocentric types and showed gradual length degradation in the range of $1.0{\sim}3.0\;{\mu}m$. The satellites appeared vestigially in a pair or a triplet set of chromosomes which depends on the species. New chromosome number and karyotype in H. longipes were the first report in this species. The structural rearrangement was suggested to explain the modified triploid composition of 2n=92.

A systematic study of Abeliophyllum distichum (Oleaceae) based on cytological characters (세포학적 형질에 기초한 미선나무의 계통분류학적 연구)

  • Cytological characteristics of Abeliophyllum distichum (Oleaceae), endemic to Korea, was examined. Somatic chromosome numbers was 2n = 28 which corresponds to diploid based on x=14. Chromosome length was varied continuously from $1.00{\mu}m$ to $2.03{\mu}m$. Karyotype of Abeliophyllum distichum was investigated in this study for the first time. The cytological characteristics including basic chromosome number, continuous variation of chromosome length, diploid and karyotype were similar to those of the genus Forsythia, which indicated the close relationship between Abeliophyllum and Forsythia, and consequently the two genera seemed to be included to same tribe.

  • The somatic chromosome number of four Korean species of Leontopodium were investigated. The chromosome number of L. leiolepis (2n = 24) is reported here as for the first time. The chromosome number of L. japonicum (2n = 28) is not varied among the 3 populations on the Korean Peninsula, but that condition is different from the previous reports for Korea (2n = 26) and Japan (2n = 21, 26). L. hallaisanense and L. japonicum, both of which are in sect. Nobilia and similar to each other in gross morphology, have the same chromosome number of 2n = 28. On the other hand the chromosome number of Korean L. leontopodioides (2n = 24) is different from that in Russian reports (2n = 26). The chromosome numbers of all Korean species of the genus Leontopodium could be inferred as tetraploid or aneuploid.
