한국산 뾰족쨈물우렁이 ( Oxyloma hirasei ) 의 염색체 연구 (Chromosome Study on the Oxyloma hirasei ( Gastropoda : Succineidae ) in Korea)

  • The mitotic and meiotic chromosomes of Succineidae snail one species, Oxyloma hirasei(Pilsbry), were investigated by mians of air-drying mithod. The diploid chromosome numbers were n=18, 2n=36. Chromosome complements of this species consist of seven pairs of metacentrics and 11 pairs of submetacentric chromosomes. Spermatogonial metaphase chromosomes range in length from 4.80${\mu}{\textrm}{m}$ for the largest pair to 1.44${\mu}{\textrm}{m}$ for the smallest pair.

  • The chromosome of Anodota woodiana in the Lake Uiam was analysed as using air drying technique of spermatogonial tissue to obtain mitotic and meiotic chromosomes. The chromosome chcle did mot differ, in general, from that found in other bivalves. Chromosome of this species consisted of metacentrics and submetacentrics. The longest chromosome was 2.1 ${\mu}{\textrm}{m}$ and the shortest was about 1.4 ${\mu}{\textrm}{m}$ in length.

  • The chromosome numbers and karyotypes were investigated in four Korean native species of the genus Hosta. The chromosome complements were diploid of 2n=60 in H. japonica var. lancifolia Nakai and H. capitata Nakai, aneuploid of 2n=59 in H. minor (Bak.) Nakai, and modified triploid of 2n=92 in H. longipes (Fr. et Sav.) Matsumura. All the species carried four sets of distinctly large chromosomes of which the chromosome types were telocentrics or subtelocentrics with $4.4{\sim}7.2\;{\mu}m$ in length. The other chromosomes were meta-, submeta, subtelo-, or telocentric types and showed gradual length degradation in the range of $1.0{\sim}3.0\;{\mu}m$. The satellites appeared vestigially in a pair or a triplet set of chromosomes which depends on the species. New chromosome number and karyotype in H. longipes were the first report in this species. The structural rearrangement was suggested to explain the modified triploid composition of 2n=92.

  • 한국 특산식물인 미선나무 1종을 대상으로 세포학적 조사를 수행한 결과 2n=28로 x=14의 2배수성 식물로 조사되었다. 염색체 크기는 $1.00{\mu}m$에서 $2.03{\mu}m$의 크기를 보였고, 그 크기는 연속적인 상태를 나타내었다. 본 연구에서 밝혀진 세포학적 형질인 기본염색체수, 크기의 연속성, 배수성 및 핵형에 있어 개나리속 식물과 거의 유사한 특징을 갖는 것으로 밝혀져 이들 속간에 세포학적으로 유연관계가 밀접한 것으로 판단되며, 미선나무속과 개나리속은 동일족에 포함되는 것이 타당할 것으로 사료된다.

  • This study was carried out to compare chromosomal characteristics between Atractylodes japonica and A macrocephala. Cytogenetic analysis was conducted based on karyotype analysis and physical mapping using fluorescence in situ hybridization. As a result of karyotype analysis by feulgen staining, somatic chromosome numbers of A. japonica and A. macrocephala were 2n=24. The length. of the mitotic metaphase chromosomes of A. japonica ranged from $0.70\;to\;1.60{\mu}m$ with a total length. of $12.11{\mu}m$ and the homologous chromosome complement comprised six metacentrics, five submetacentrics and one subtelocentrics. On the other hand, the length of the mitotic metaphase chromosomes of A. macrocephala ranged from $0.90\;to\;2.35{\mu}m$ with a total length of $16.58{\mu}m$ and the homologous chromosome complement comprised seven metacentrics and five submetacentrics. The total length of A. japonica chromosomes was shorter than that of A. macrocephala, but A. japonica had one subtelocentrics (chromosomes 4) different from A. macrocepha1a. chromosomes. The F1SH technique using 17S and 5S rDNA was applied to metaphase chromosomes. The signals for 17S rDNA were detected on the telomeric regions of chromosomes 4 and 5 in both A japonica and A. macrocephala. The 5S rDNA signal was found in the short arm of chromosome 1.

  • Meiotic metaphase chromosomes of the testis of two storage insects, tobacco moth (Ephestia elutella H.) and cigarette beetle (Lasioderma serricorne F.) were examined to study their karyotypes. The number of haploid chromosomes of the tobacco moth was 31 and the karyotype was characterized by 29 metacentric and 2 submetacentric chromosomes. The metaphase chromosome length of tobacco moth ranged ca. 1.44-2.11${\mu}{\textrm}{m}$ and the average length was 1, 78${\mu}{\textrm}{m}$. The number of haploid chromosomes of the cigarette beetle at metaphase was 10 and the karyotype was characterized by 8 metacentric and 2 submetacentric chromosomes. The range of metaphase chromosome length of the cigarette beetle was ca. 1.79- 2.39${\mu}{\textrm}{m}$ and the average length 2.09${\mu}{\textrm}{m}$.

Detection of Main-effect QTLs, Epistatic QTLs and QE Interactions for Grain Appearance of Brown Rice(Oryza sativa L.)

  • The objective of this study is to identify main-effect QTLs, epistatic QTLs, and the interactions between QTL and the environment associated with grain appearances of brown rice. A genetic linkage map was created with 172 DNA markers spanning 12 rice chromosomes based on 120 DH lines, which derived from a cross between 'Samgang'(Tongil) and 'Nagdong'(Japonica). One thousandgrain weight, length, width, length-to-width ratio, and thickness were evaluated regarding the DH population. Twenty independent QTLs and fourteen epistatic QTLs were identified in using CIM by two programs, known as WinQTLcart2.5 and QTLMAPPER. The QTLs of qgw9.1 in an interval of RM434-RM242 on chromosome 9 and qgw11.1 at a peak marker of RM287 on chromosome 11 for one thousand-grain weight, qgwi2.2 for grain width at a peak marker of RM450, qlw2.1 for length-to-width ratio flanked by RM492 and RM324, and qgt2.1 for thickness flanked by 2009 and RM492 on chromosome 2 were detected over two years, which can be considered as stable QTLs. The epistatic effect might be an important component for genetic basis of one thousand-grain weight and width. The main-effect QTLs of grain width and length to width ratio were easily influenced by environments.

Cytological Study of Euphaedusa fusaniana (Stylommataphora: Clausiliidae) of Korea

  • The chromosome numbers and karyotype of Euphaedusa fusaniana, a kind of Korean land snail are investigated here. From E. fusaniana, chromosome numbers of 28 (n) and 56 (2n) were counted. Euphaedusa fusaniana has 15 pairs of metacentric chromosomes and 13 pairs of submetacentric chromosomes. The mean total length of all chromosomes was $128.7\;{\pm}\;9.93\;{\mu}m$. The maximum length of chromosomes No. 1 was $9.6\;{\pm}\;0.21\;{\mu}m$.

  • In this paper, we classify between the chromosome and blood cell, and find the location of chromosome. First, the gray level images be the binary images using the threshold method. Then, the spot noises are removed by the morphological filtering. Features are obtained using the updated Run length(RL) coding and are classified using the Bayes decision rule. The performances of classification are 83.3% in chromosome and 93.3% in blood cell. Because each sub-images ($256{\times}256$) is obtained from the full image($512{\times}512$), we realize the location of chromosome if we get the corrected chromosome classifications.

벼의 이상적인 초형에 관여하는 QTL 분석 (QTL Analysis of Concerned on Ideal Plant Form in Rice)

  • 본 실험은 청청과 낙동 조합을 약배양하여 육성한 120 계통을 이용하여 이상적인 초형에 관련된 간장, 수장, 개체당 이삭수에 대한 QTLs를 분석하고 탐색된 QTLs은 다음과 같은 결과를 얻었다. 모부본인 청청과 낙동의 간장의 평균은 $75.3{\pm}6.72cm$, 수장의 평균은 $20.6{\pm}2.08cm$이었으며 개체당 이삭수의 평균은 $16.0{\pm}2.37$개로 나타났다. 120 계통의 CNDH의 간장의 평균은 $71.6{\pm}17.38cm$였으며, 수장의 평균은 $20.3{\pm}2.24cm$였으며 개체당 이삭수의 평균은 $16.1{\pm}7.17$개로 나타났다. 이에 대한 CNDH 계통의 빈도분포표의 곡선은 정규분포에 가까운 연속변이를 나타내었다. 간장, 수장, 개체당 이삭수 QTL 분석결과 간장에서 1번 염색체의 qPlL1-1, qPlL1-2, 5번 염색체의 qPlL5, 수장에서 2번 염색체의 qPL2, 3번 염색체의 qPL3, 10번 염색체의 qPL10, 개체당 이삭수에서 1번 염색체의 qPN1-1, qPN1-2, 9번 염색체의 qPN9이 탐색되었다. 간장에 대한 QTL에서 5번 염색체의 LOD 값은 3.81, 상가적 값은 5.49였으며, 수장에 대한 QTL에서 2번 염색체의 LOD 값은 7.13, 상가적 값은 -2.58 이었으며, 3번 염색체의 LOD 값은 3.20, 상가적 값은 0.88로 나타났다. 개체당 이삭수에 대한 QTL에서 9번 염색체의 LOD 값은 4.27, 상가적 값은 -1.60으로 나타났다. 초형 관련 분석 결과에서 탐색된 9개 마커를 토대로 간장에 대한 RM5311, 수장에 대한 RM555, 개체당 이삭수에 대한 RM8111을 선발하여 모부본인 청청, 낙동을 기준으로 자포니카형 22 품종, 인디카형 12 품종에 다형성을 분석하였다. 그림4와 같이 청청과 낙동과 같은 밴드양상을 나타나거나 다른크기상태의 밴드양상을 나타내었다. 간장, 수장, 개체당 이삭수의 일치율은 각각 44.11%, 41.17%, 44.11%로 나타났다.
