Chromosomal Inversions in a Natural Population of Drosophila melanogaster (초파리 集團의 染色體 多型現象)

  • 1. A total of 969 flies were tested and 19 different inversions were found from the present samples collected three times at yearly intervals in an area. 2. Of the 19 polymorphisms, except for the six common paracentric types of cosmopolitan inversions, 13 were endemic. Among this latter group, one was X-chromosomal, 4 were on second chromosome and 8 on third chromosome. 3. Beside two overlapping inversions all the others were identified as single ones. And the inversion frequency on an average in every three samples tested was 38.9%. 4. The high distribution of the inversions apeared to the second chromosome suggests that a differential adaptive force may be present between the second and third chromosome.

  • Karyotype of the ricessh Orygias latipes (Temminck et Schlegel) was investigated using 151 specimens of eleven populations in the southern Korea. Based on the karyotype analysis, two distinct chromosomal groups were found. One group, occurring in the Kum River, the Mankyong River, the Dongjin River, the Ysngsan River and the SEmjin River had a diploid chromosome number of 46 and arm number of 70, which contained one pair of "large" metacentric chromosomes. The other group, occurring in the Tamjin River, the Hyongsan River andthe all four island populations observed had a diploid chromosome number of 48 and arm number of 68. These results showed that 0. latipes of 6 inland populations was identical with the Chinese population in diploid number, arm number and presence of "large" metacentric chromosome, while the present species of island populations ana populations of the Tamjn R. and Hyongsan R. was similar to the Japanese population in chromosome number and arm number. The karyotype variation in populations indicated that karyotypes will probably provide useful information in studying zoogeography of this species.phy of this species.

  • Male pseudohermaphrodites is one of abnormalities of phenotypic sex and have an XY chromosome constitution and testes, but the internal or external genitalia are to some degree those of a female. We surveyed one case which were referred in Veterinary Medical Teaching Hospital of Seoul National University. When patient was purchased, its phenotype was entirely female that have female external genitalia. However, from the age of 6 months, clitoris was gradually increased to Os clitoris, and patients showed male behavior. Critical clinical sign was trauma of protruded Os clitoris. In radiographic and ultrasonographic examination, he had normal vagina and urinary bladder. In chromosome examination, he had an XY chromosome constitution. This case was performed removal of Os clitoris and ovariohysterectomy.

  • To investigate the mutant induction of wild ginseng culture extract, we performed chromosomal aberration assay with chinese hamster lung cell in vitro. The test concentration of the extract was decided for the standard with the 50% suppression of cell propagation in the cell. The concentrations for the chromosome test were 1,250, 2,500 and 5,000 ㎍/ml with metabolic activation (+S, 6 hours treatment), 1,100, 2,200 and 4,400 ㎍/ml without metabolic activation (-S, 6 hours treatment) 800, 1,600 and 3,200 ㎍/ml without metabolic activation (-S, 24 hours treatment). No significant increase in chromosome aberrations was observed at any of these concentrations both in the absence and presence of metabolic activation system. Cyclophosphamide monohydrate (CPA) and ethylmethanesulfonate (EMS) caused a significant increase in chromosome aberration. These results may be concluded that wild ginseng culture extract is not capable of inducing chromosome aberration in cultured chinese hamster lung cell regardless of metabolic activation and genotoxicity of that is negative under the present experimental condition.

  • Reports of constitutional ring chromosome 22, r(22) are rare. Individuals with r(22) present similar features as those with the 22q13 deletion syndrome. The instability in the ring chromosome contributes to the development of variable phenotypes. Central nervous system (CNS) atypical teratoid rhabdoid tumors (ATRTs) are rare, highly malignant tumors, primarily occurring in young children below 3 years of age. The majority of ATRT cases display genetic alterations of SMARCB1 (INI1/hSNF5 ), a tumor suppressor gene located on 22q11.2. The coexistence of a CNS ATRT in a child with a r(22) is rare. We present a case of a 4-month-old boy with 46,XY,r(22)(p13q13.3), generalized hypotonia and delayed development. High-resolution microarray analysis revealed a 3.5-Mb deletion at 22q13.31q13.33. At 11 months, the patient had an ATRT ($5.6cm{\times}5.0cm{\times}7.6cm$) in the cerebellar vermis, which was detected in the brain via magnetic resonance imaging.

  • The vegetative nuclear divisions in hyphae and the chromosome of Fusarium were observed by use of HCI-Giemsa technique and light microscope. The chromosome of nuclear in F. moniliforme both 7150 and 7219 were eight. F. subglutinans 1082 was n=8 and n=7 in F. sub­glutinans 1083. F. nygamai 5668 was n=7 and n=5 in F. nygamai 7132. F. beomiforme 9758 and 9760 were n=7. F. coccidicola ATCC 24138 and F. acuminatum ATCC 16560 were n=6. From these results and other reports, the basic chromosomal number of these fungi might be speculated to be four.

  • The cellular toxicity and antitumor effects of bleomycin are thought to be occurred by formation of O$_2$-Fe$^{2+}$-bleomycin complexes that degrade DNA and release O$_2^-$ and $^{\cdot}$OH radicals. Hydroxyl radicals derived from hydrogen peroxide seem most likely to be involved in the various stages of carcinogenesis, and transition metals such as iron play a central role in activation of bleomycin and in formation of hydroxyl radicals. This study was performed to investigate whether treatment with ferrous sulfate increase chromosome aberration induced by bleomycin and hydrogen peroxide, and whether there is different repair mechanism for DNA damage induced by those chemicals. Treatment with 3AB, Ara C, during G$_1$ and post-treatment with caffeine, and Hu during G$_2$ increased the frequency of chromosome aberration induced by bleomycin but post-treatment with caffeine only did function that way when hydrogen peroxide was treated. When 6.6X 10$^{-7}$ M of bleomycin or 5.0X10$^{-5}$M of hydrogen peroxide were treated simultaneously with iron, the frequency of chromosome aberration was reduced, if compared with the results by bleomycin or hydrogen per oxide alone.

  • To determine the structure of chromosome and to identify the sex chromosome of Ginkgo biloba and G. biloba var. fastigiata, the samples were obtained from root tips of trees growing in seven different provinences. The results are as follows. The basic number of somatic chromosomes was 2n=24. The range of a relative length of long chromosome was between $14.88{\sim}11.18{\mu}m$ and that of short chromosome was $8.11{\sim}6.24{\mu}m$. The chromosome sets were composed with one long pairs of m type and 11 short pairs of sm or st type. These short pairs showed the continuous descending in length. There was a satellite on the short arm of the Longest chromosome pair, and were satellites of the one or both long arm of 7th or 8th chromosome pair which were sm or st type, or the shortest st type chromosome pair. Sometimes, a satellite on the short arm of the longest chromosome pairs of Ginkgo biloba was double satellite, but that of G. biloba var. fastigiata was not. Karyotype was $2n=24=2^{2s}A^m+2B^{st\;or\;sm}+2C^{st}+2D^{st}+2E^{st}+2F^{st\;or\;sm}+2G^{sm}+2^{2s}H^{sm}\;or\;(^{1s}H^{sm}+H^{sm})+2I^{st}+2J^{st}+2K^{st}+2^{2s}L^{st}\;or\;(^{1s}L^{st}+L^{st})$. The male and female trees were not apparently distinguished by the chromsome structures. However the differences between the satellites could be used to identify the male and females. The male tree has double satellite on short arm of a longest chromosome pairs and females' has not. Also female trees have a satellite on a short chromosome more frequently than male trees.

  • The spermatogenesis and chromosome number were investigated in the pupal testes of Helicouerpa assulta Guenee by light microscopy. During the spermatogenesis, each bundle of P8(256) sperms developed by 6 mitotic and 2 meiotic spermatogonial divisions. From the early stage of spermatogenesis, it was distinguishable between two kinds of sperm differentiation, eupyrene and apyrene spermatogenesis, which are characteristic in Lepidoptera, by the differences in nuclear shape and cell distribution in immature spermatocyst. Through the followed spermiogenesis, the spermatocysts were developed into two kinds of mature cyst, a streamline-shaped eupyrene cyst with nucleated sperms of thready head or a long spindle-shaped apyrene cyst with anucleated sperms of cylindrical head. As the results off chromosomal analysis at metaphase of the spermatogonial mitosis and spermatocytic meiosis, the chromosome number were 2n=6a/n=31, respectively, and no variation between individuals.

  • The alkaline elastase is an extracellular serine protease of the alkalophilic Bacillus strain Ya-B. To increase the gene copy number and the production level of the alkaline elastase Ya-B, we designed, on the B. subtilis chromosome, a gene amplification of the 10.6 kb repeating unit containing amyE, aleE (alkaline elastase Ya-B gene) and tmrB. The aleE was inserted between amyE and tmrB, and B. subtilis APT119 strain was transformed with this amyE-aleE-tmrB-junction region fragment. As a result, we succeeded in obtaining tunicamycin-resistant (Tm$^{r}$) transformants (Tf-1, Tf-2) in which the designed gene amplification of 10.6 kb occurred in chromosome. The transformants showed high productivity of $\alpha $-amylase and alkaline elastase Ya-B. The copy number of the repeating unit (amyE-aleE-tmrB) was estimated to be 25, but plasmid vector (pUC19) was not integrated. The amplified aleE of chromosome was more stable than that of plasmid in absence of antibiotics.
