Study on the Sex Chromosome Dependent Segregation of the Third Chromosome in Drosophila melanogaster (초파리의 성 옆색체의 구성이 제 3옆색체의 분리에 미치는 영향에 대하여)

  • The present study was undertaken in an attempt to see if the segregation frequency of a third chromosome was changed by changing the sex chromosome which were free of SD in the second chromosome. The eight genotype males having different sex chromosome constitution each were constructed by appropriate matings and the two standard laboratory stocks of Drosophila melanogaster, e and se were used as the third chromosome recessive markers for the present experiment. The results of the present investigation are given below: 1. The k values which are the proportion of the se third chromosomes recovered among progeny flies from the mating of se/e males to e females were highly signiicantly different among the four genotypes and between the two sexes,and the interaction of genotype and sex was significantly different. Thus the setregation frequency of the se third chromosome in the male, when made heterozygous with the e third chromosome, was dependent upon the sex chromosome constitution. 2. Both of the k(Woman) and the k(man) remains roughly constant among genotypes. 3. The sex ratio o the se progeny class was highly significantly heterogeneous among the four genotypes but it was homogeneous for the e progeny class. 4. The values of the k(man) and the sex ratio of the se progeny class, on the average, were higher than that of the k(Woman) and of the e progeny class, respectively. 5. Those phenomena suggest that some sort of prezygotic selection could be operating such that the combination of the e third chromosome and the Y chromosome tends to be eliminated before fertilization. This tendency argues for a re-examination of the viability estimations of Drosophila melanogaster.

Induction of A Chromosome-doubled Persimmon (Diospyros kaki Thunb.) by in vitro Colchicine Treatment (기내 콜히친 처리에 의한 염색체 배가 감 식물체 유기)

  • This was carried out to develop a chromosome-doubled (12x) persimmon that will be used as a crossing parent to select seedless persimmon cultivars with the change of the consumption trend recently. To obtain a chromosome-doubled (12x) persimmon, colchicine was applied at the meristem of seedlings in vitro derived from cross among hexaploid persimmon (Diopyros kaki Thunb.). These were treated with 0.03%, 0.05% and 0.1% colchicine respectively for doubling chromosome, and it was most effective at the concentration of 0.05% colchicine. After colchicine treatment, we conducted tests to elucidate conditions for inducing shoot and root development. As the result, the shoots grew best when cultivated at 1/2MS media plus 10 and $30{\mu}M$ zeatin respectively, and the roots grew best when cultivated at 1/2MS media after dipping for 5 seconds at 10 mM NAA+5% DMSO. We also compared seedlings that have chromosome (6x) do not doubled and crossing parents (6x) and chromosome-doubled seedlings (12x). As the result, these chromosome-doubled seedlings (12x) showed lower stomatal density and larger stomatal size.

Study on the Chromosome Size, Number and Shape by the Centromeric Index, Arm Ratio and Relative Length in Single Comb White Leghorns (단관백색레그혼순계에 있어 중심입지수, 등완비 및 상대적길이에 의한 염색체의 형태적 특징과 수에 관한 연구)

  • Chromosome size, number and shape were studied by the centromeric index, the arm ratio and the relative length of chromosome. The chromosomes of 50 early chick embryos which were derived from a pure line of Single Comb White Leghorns were examined. Using a colchicine, hypotonic treatment, fixation and air-drying technique, the clear prometaphase figures were obtained from the whole embryo. The results of the present investigation of chromosome pairs were as follows, 1. Pair 1 and 2; metacentric and submetacentric chromosomes which could be clearly distinguished from each other by size. 2. Pair 3 and 4: acrocentric chromosomes of similar length but the 4th pair had a distinct short arm which was not present in the 3rd. 3, Pair 5; metacentric sex chromosomes, 2 chromosome had relative 5th length but the W chromosome had slightly shorter length than 7th pair of chromosomes. 4. Pair 6; acrocentric chromosomes similar in shape to pair 3 but of little more than half the size. 5, Pair 7 and 8; acrocentric chrocentric but the 7th pairs had a definite short arm. 6. Pair 9; similar length to the 7, 8 pairs but had a medially placed centromere. 7. microchromosomes of 30 pairs ; nearly all acrocentric chromosomes which appeared as paired dots. The total number of diploid was appeared to 72-78. But a number of observations presented the total diploid number in 78 (58%). The inconstancy in number observed in this study was presumably due to the minute size of the microchromosomes. Thus, the modal numbers for the diploid chromosome was at least 78.

Analysis of Chromosome aberrations by fluorescence in situ hybridization using triple chromosome-specific probes in human lymphocyte exposed to radiation (3중 DNA probe를 이용한 FISH(fluorescence in situ hybridization) 기법으로 방사선에 의한 염색체 이상 분석)

  • Fluorescence in situ hybridization with chromosome-specific probe has been shown to be a valid and rapid method for detection of chromosome rearrangements induced by radiation. This method is useful for quantifying structural aberrations, expecially for stable ones, such as translocation and insertion, which are difficult to detect with conventional method in human lymphocyte. In order to apply FISH method for high dose biological dosimetry, chromosomal abberations by radiation at doses of 1, 3, 5, and 7Gy were analysed with whole chromosome-specific probes by human chromosome 1, 2 and 4 according to PAINT system. The frequencies of stable translocation per cell equivalent were 0.04, 0.33, 1.22, 2.62, and 5.58 for the lymphocyte exposed to 0, 1, 3, 5, and 7Gy, respectively, and those of dicentric were 0.00, 0.06, 0.52, 1.19 and 2.44, respectively. Significantly more translocation of t(Ab), a translocated chromosome with a piece of painted acentric matrial 'b' attached to unpainted piece containing centromere 'A', than reciprocal chromosome t(Ba) was observed. The frequencies of all type of chromosome rearrangements increased with dose. From above result, FISH seemed to be useful for radiation biodosimetry by which the frequencies of various types of stable aberrations in human lymphocyte can be observed more easily than by conventional method and so will improve our ability to perform meaningful biodosimetry.

  • A karyotype analysis of the chromosome of Neodiplostomum seoulense, one of causative agents of human intestinal trematodiasis, was done from the gonad tissue by the squashing method. The chromosome number was n=10 and 2n=20. Chromosome length was $1.30-4.0{\;}\mu\textrm{m}$. Chromosome pairs in the complement consisted of two pairs of metacentric, five pairs of submetacentri$cs_telocentric and three pairs of telocentric chromosomes. These data were comparable with those of other intestinal trematodes.es.

  • The growth charateristics and karyotypes of Aster spathulifolius collected from 5 sites including coastal and island region on the Korean peninsula, were analysed. Several morphological characteristics of the plants such as leaf length, leaf width, top internode, medium internode, spike branching, flower diameter, number of petal, leaf color, leaf form, stem and leaf hair, viscosity, and serration of the plants were distinctly different depending on the native region from which they were collected. Karyotypic analysis showed that the chromosome number was all diploid (2n=18), with one pair of submetacentric satellite chromosomes. The chromosome composition included 7 pairs of metacentric chromosomes and 2 pairs of submetacentric chromosomes in all plants. However, chromosome order and the ranges of the chromosome lengths were a little different from plant to plant according to their native growing regions. The plants from Geoje-Do especially showed large differences in the chromosome lengths between the longest and the shortest compared to the plants from other places. This results provide important data to support the classification of the species into several sub-species.

  • The chromosomes of two similar lymnaeid snail species, Austropeplea ollula from Korea and "Lymnaea"sp. introduced to Australia, were karyologically investigated by using an air-dry method. The diploid chromosome number found in A. ollula was 32, and that of "Lymnaea" sp. was 30. The mitotic chromosome complements of A. ollula were 5 metacentric, 9 submetacentric, and 2 subtelocentric pairs. "Lymnaea" sp. had 5 metacentric, 8 submetacentric, and 2 subtelocentric pairs. Austropeplea ollula is distinguishable from "Lymnaea" sp. by their chromosome numbers.

  • Karyotype analysis and chromosomal localization of 5S and 45S rDNAs using FISH (fluorescence in situ hybridization) technique were carried out in Nicotiana plumbaginifolia. Somatic chromosome number of N. plumbaginifolia was 2n=20 and kyarotype was composed of three pairs of metacentric chromosomes (1,2 and 7) and seven pairs of submetacentric chromosomes (3, 4, 5, 6, 8, 9 and 10). Length of chromosomes was ranged from 2.29 to 4.50 ${\mu}{\textrm}{m}$. Chromosome 1 and 2 were identified as satellite chromosomes. In FISH experiment, one pair of 5S rDNA signals was detected on the centromeric region of chromosome 2 and one pair of 45S rDNA signals was detected on the terminal region of chromosome 1.

Karyotype of Lilium Miqueliannum Makino (하늘말나리의 핵형)

  • Lilium Miquelianum Makino is a species which originated in Korea. The Karyotype of the species was examined in materials collected at Mts. Kaya, Kasan, Chejung, and Kaji. The results are as follows: 1) The somatic chromosome number was found to be 2n=24. 2) The karyotype is described as: K=2Am+2Bkm+2Csst+2Dsst+2Ests+2Fst+2Gst+2Hst+2Ist+2Jsst+2Kst+2Lst m: metacentric, sm: submetacentric st: subtelocentric, s: secondary constriction 3) A single subcentric supernumerary B-chromosome was found in some bulbs from Mt. Kasan. 4) The shape of the supernumerary B-chromosome was similar to that of the E chromosome which had separated at its secondary constriction and lost its lower chromosome fragment. 5) From three to eight nucleio of varying sizes were found in the telophase or interphase nucleus of root tip cells. The maximum number of eight nucleoli corresponds to the number of chromosomes that have a secondary constriction.

Effects of gamma-irradiation on the infectivity and chromosome aberration of Clonorchis sinensis

  • Effects of gamma irradiation on the worm survival and chromosomal aberration of Clonorchis sinensis were studied. The metacercariae irradiated with various amounts of gamma radiation (ranging from 5 Gy to 50 Gy) were fed to rats, and the effects were compared with those of non-irradiated controls. Recovery rates of adult worms in irradiated groups were reduced gradually as increasing of the irradiation doses. No worm was recovered from rats which were fed with 50 Gy irradiated metacercariae. The chromosome number was 2n = 56 in all worms from all experimental groups. However, the groups irradiated with 20 Gy, 25 Gy or 30 Gy showed variations in the chromosome number, depending on different cells in the same individual. Radiation doses used in this study did not appear to induce chromosome aberrations, however, irradiation with 30 Gy showed slightly reduced chromosome size.
