Protection against Whole Body γ-Irradiation Induced Oxidative Stress and Clastogenic Damage in Mice by Ginger Essential Oil

  • Radioprotective effects of ginger essential oil (GEO) on mortality, body weight alteration, hematological parameters, antioxidant status and chromosomal damage were studied in irradiated mice. Regression analysis of survival data in mice exposed to radiation yielded LD50/30 as 7.12 and 10.14 Gy for control (irradiation alone) and experimental (GEO-treated irradiated) mice, respectively, with a dose reduction factor (DRF) of 1.42. In mice exposed to whole-body gamma-irradiation (6 Gy), GEO pre-treatment at 100 and 500 mg/kg b.wt (orally) significantly ameliorated decreased hematological and immunological parameters. Radiation induced reduction in intestinal tissue antioxidant enzyme levels such as superoxide dismutase, catalase, glutathione peroxidase and glutathione was also reversed following administration of GEO. Tissue architecture of small intestine which was damaged following irradiation was improved upon administration of GEO. Anticlastogenic effects of GEO were studied by micronuclei assay, chromosomal aberration and alkaline gel electrophoresis assay. GEO significantly decreased the formation of micronuclei, increased the P/N ratio, inhibited the formation of chromosomal aberrations and protected agaisnt cellular DNA damage in bone marrow cells as revealed by comet assay. These results are supportive of use of GEO as a potential radioprotective compound.

Galangin 및 양강추출물의 $KBrO_3$ 유도 DNA 및 염색체 손상에 대한 보호효과 (Protective Effect of the 70% Ethanolic Extract of Alpinia officinarum and Galangin Against $KBrO_3$-induced DNA and Chromosomal Damage in Mice)

  • The aim of this study was to evaluate the in vivo effect of galangin and the 70% ethanolic extract of Alpinia officinarum (AO) toward $KBrO_3$-induced DNA and chromosomal damage in mice. Galangin and AO inhibited the formation of 8-hydroxy-2'-deoxyguanosine (8-OH2'dG) as an indicator of DNA oxidative damage in the liver cell. Galangin and AO showed the inhibitory effect on the formation of DNA single strand break in the splenocyte by single cell gel electrophoresis (SCGE) assay and also inhibited micronucleated reticulocyte (MNRET) formation of peripheral blood in tail blood of mice. Vit-E revealed antigenotoxic effects in DNA and chromosome levels, but galangin was more potent active compound compare to vit-E under our experimental conditions. The results suggest that the extract of Alpinia officinarum containing galangin can modify the oxidative DNA and chromosomal damage and may act as chemopreventive agent against oxidative stress in vivo.

Glucoamylase 유전자 STA의 염색체내 삽입에 의한 Saccharomyces diastaticus의 glucoamylase 생성능 향상 (Improvement of Glucoamylase Productivity of Saccharomyces diastaticus by Intergration of Glucoamylase Gene, STA, into Chromosomal DHA)

  • Saccharomyces distaticus 의 glucoamylase 생성능을 증진시킬 목적으로 STA1 유전자를 YIp vector 를 이용하여 염색체에 도입해 주고자 하였다. STA1 유전자 5.8-Kb 를 YIp vector 에 재조합하여 YIp-STA 를 재작하고, S. diastaticus GMT-11 (a, ura 3, STA1) 을 숙주균주로 하여 염색체의 STA1 유전자 부위에 homologous recombination 되어 삽입하도록 형질전환을 실시하였다. 이렇게 하여 glucoamylase 생성능이 모균주에 비해 최대 6배까지 증대된 다양한 형질전환체들을 얻을 수 있었다. 그리고 glycoamylase 생성능이 증대된 형질전환체들의 염색체 DNA 를 분리하여 Southern hybridization 을 실시한 결과 YIp-STA 가 multi-copy integration 되었음을 확인하였고, 또한 도입해 준 YIp-STA 는 세포분열인 30세대기간 동안 계속되었어도 안정하게 유지되었음을 알았다.

한국산 벼멸구 생태형의 염색체 변이 (Chromosomal Variation among Brown Planthopper, Nilaparvata lugens (Stal), Biotypes in Korea)

  • 벼멸구 수컷 정소에서 염색체의 변이정도를 생태형 별로 관찰하였다. 세포 분열 지수는 생태형 3에서 가장 높았고 그 다음으로 생태형 1, 생태형 2이었다. Agmatoploidy, aneuploidy, 성염색체의 loose pairings과 같은 염색체의 구조적 변화는 생태형 1, 생태형 2, 생태형 3순으로 높았다. 감수분열 제1분열기 중기는 성염색체가 상염색체로부터 현격하게 분리되어 있는 세포의 수는 생태형 2에서 가장 높았다.

비뇨기과 환자에서의 염색체 이상에 관한 연구 (A Study of Chromosomal Abnormality in Urological Patients)

  • A chromosomal study was performed in a total of 162 urological patients during past 2$2{\frac{1}{2}}$ years (Feb. 1984 - Aug. 1986). Of these 78(48%) patients had abnormal chromosome complements. Among all patients with chromosome abnormalities, 88% (69/78) had aberrations of chromosome number, 8% (6/78) had aberrations of chromosome structure and 4% (3/78) had aberrations of both. 90% (65/72) of numerical abnormality was Klinefelter's syndrome and the structural abnormality rate (5.6%, 9/162) was less than that (6.99%) of general population. The chromosomal study was mandatory for the detection of intersex in small testes or hypospadias with cryptorchism or clitoromegaly or bilateral cryptorchism. But unilateral cryptochism or hypospadias with normal scrotal testes was not thought to be indication of the chromosomal study if the external genitalia are otherwise quite normal.

Binding of IciA protein to the dnaA promoter region

  • IciA protein has been shown as an inhibitor for the initiation of E. coli chromosomal DNA replication at oriC. IciA protein binds the AT-rich region in oriC and then blocks the initiation of chromosomal DNA replication. Two binding sites for IciA protein were identified in dnaA gene, encoding the initiator for the E. coli chromosomal replication, promoter region by gel-shift assay and DNase I footprinting, One, named as IciA site I, is located upstream of the dnaA promoter 1P. The other, named as IciA site II, is located downstream of the dnaA promoter 2P. The sequence comparison of the regions protected from the DNase I cleavage did not result in a clear consensus sequence for the binding of IciA protein, suggesting that IciA protein may be a member of multimeric complex dsDNA binding proteins. This study provided information about the binding mode of IciA protein. Even though the IciA site II and IciA binding site in oriC seem to be composed of two IciA binding units, one binding unit is likely enough to cause the binding of IciA protein to the IciA site I. The binding of IciA protein to the dna4 promoter implies that IciA protein may involve not only the control of the initiation of chromosomal DNA replication but also the control of the dna4 gene expression.

Evaluation of Genotoxicity of SU-Eohyeol Pharmacopuncture Using an In Vitro Chromosome Aberration Test in Chinese Hamster Lung Cell

  • Objectives: This study was conducted to evaluate the safety of SU-Eohyeol pharmacopuncture (SUEP) by assessing its potential to cause chromosomal abnormalities in Chinese hamster lung cells (CHL/IC). Methods: A dose-curve was conducted to determine the highest dose of SUEP. Doses of 10, 5, 2.5, 1.25, 0.625, and 0.313% were used, and no cytotoxicity or SUEP precipitation was observed. SUEP doses of 10, 5, and 2.5%, with positive and negative controls, were used in a chromosome aberration test. Results: In this study, the frequency of abnormal chromosomal cells in the SUEP group did not show a statistically significant difference from that of the negative control group in short-term treatments with and without metabolic activation and the continuous treatment without metabolic activation. Compared with the negative control group, the positive control group had a significantly higher frequency of cells with structural chromosomal abnormalities. This test's results satisfied all conditions for determining the results. Conclusion: SUEP did not induce chromosomal aberrations under the conditions of this study. Other toxicity evaluations, safety studies in humans, and various clinical trials are required to evaluate the safety and efficacy of SUEP.

Toxicity Assessment of a No-Pain Pharmacopuncture Extract Using a Standard Battery of In Vitro Chromosome Aberration Tests

  • Objectives: Genotoxicity is evaluated through a chromosomal aberration test using cultured mammalian cells to determine the toxicity of no-pain pharmacopuncture (NPP), which has recently been used to treat musculoskeletal pain disorders in Korean medical clinical practice. Methods: An initial test was performed to determine the dosage range of the NPP, followed by the main test. In this study, NPP doses of 10.0, 5.0, and 2.5%, and negative and positive controls were tested. An in vitro chromosome aberration test was performed using Chinese hamster lung cells under short-term treatment with or without metabolic activation and under continuous treatment without metabolic activation. Results: Compared with the saline negative control group, NPP did not significantly increase the frequency of chromosomal abnormalities in Chinese hamster lung cells, regardless of the presence or absence of metabolic activation. Additionally, the number of cells with structural chromosomal abnormalities was significantly higher in the positive control group than that in the negative control group that received saline. Conclusion: Based on the above results, the chromosomal abnormality-producing effect of NPP was determined to be negative under these test conditions.

Chitosan Oligosaccharide Inhibits $^{203}HgCl_2-Induced$ Genotoxicity in Mice: Micronuclei Occurrence and Chromosomal Aberration

  • The purpose of this study was to investigate the safety of chitosan oligosaccharide and the effects of chitosan oligosaccharide on mercury induced genotoxicity in mice using the micronuclei and chromosome aberration. The micronuclei test was performed by microscopic examination $(\times1,000,\;stained\;using\;a\;May-Grunwald\;solution)$ after administering 0.01, 0.1, and $1\%(10\;mg/mL)$ chitosan oligosaccharide for 7, 60, and 180 days ad libitum in mice. Total micronuclei of 1,000 polychromatic erythrocytes were recorded for each group. There was no difference between the untreated and experimental groups. The intake periods and concentrations of chitosan oligosaccharide did not affect the occurrence of micronuclei in bone marrow cells (P>0.05). The chromosomal aberration test was performed by microscopic examination $({\times}1,000,\;stained\;using\;a\;4\%\;Giemsa\;solution)$ after administering the same concentration of chitosan oligosaccharide to mice, in $F_1,\;F_2,\;F_3$ generations and parents. The frequency of chromosomal aberrations was defined as [Ydr=(D+R)/total number of counted lymphocytes]. Similar to the micronuclei test, there was no difference between the untreated and treated groups. These results showed that the intake periods and concentrations of chitosan oligosaccharide did not affect chromosomal aberrations in bone marrow cells (P>0.05). To investigate the effect of chitosan oligosaccharide on mercury-induced chromosome aberration, mice in each condition were supplied with $^{203}HgCl_2$ and chitosan oligosaccharide ad libitum. Chitosan oligosaccharide significantly inhibited $^{203}HgCl_2-induced$ chromosome aberration in mice. Based on the results of this study, it may be concluded that the chitosan oligosaccharide is a nontoxic material that could be used as a suppressor of heavy metal-induced genotoxicity.

In vitro에서 1-β-D-arabinofuranosyl-cytosine의 염색체 파열 유도 (1-β-D-Arabinofuranosyl-cytosine Induces Chromosomal Breaks in vitro)

  • 목 적 : 염색체 취약부위는 세포를 특정의 화학물질에 노출시키거나 특수한 배양조건에서 배양 할 때 쉽게 파열되는 염색체상의 특정구역이다. 취약부위는 유전성질환 및 악성종양과 관련이 있으며, 현재에는 분자생물학적 실험기법의 개발로 분자수준에서 이해가 되고 있다. 새로운 취약부위 및 취약부위의 발현을 높이거나 쉽게 관찰할 수 있는 실험실적 조건을 알아보기 위하여 항암제로 사용되는 Ara-C를 이용하여 염색체파열을 조사하여 보았다. 한편 염색체 취약부위가 종양형성과 관련이 있다는 사실에 기초하여 Ara-C에 의해 발열되는 취약부위와 암유전자가 위치하는 부위 및 종양에서 일정하게 염색체변이가 관찰되는 특정의 염색체 부위와의 상관관계도 알아보고자 하였다. 방 법 : 정상 성인 남녀 각각 3명의 말초혈액 내 T-림프구를 세포배양 후 Ara-C를 첨가하고 다시 caffeine을 처리한 뒤 종전에 시행했던 방법과 동일하게 검체 처리하여 염색체파열 부위를 관찰하였다. 염색체 취약부위는 염색체상의 일정부위에서 100개의 염색체파열 당 2회 이상의 염색체파열이 6명 중 4명 이상에서 관찰되는 경우로 정의하였다. 결 과 : 1) T-림프구를 엽산이 부족한 MEN-FA 배지에서 배양 시 Ara-C는 100개의 분열세포 당 252.1개의 염색체파열을 유도하였으며, Ara-C를 처리하지 않은 대조군에서 25.2개가 관찰되어 Ara-C를 처리한 경우에 염색체파열이 의미 있게 많았다(P<0.05). 2) Ara-C에 의한 염색체파열은 엽산이 부족한 MEM-FA 배지에서 엽산이 충분한 RPMI 1640 배지에서 보다 많이 되었다(P<0.05). 한편, 2.0 mM 농도의 caffeine은 엽산만 부족한 배양 배지에서는 염색체파열을 상승시키지 못했으나, Ara-C와 병행사용 시 상승시켰다. 3) Ara-C에 의해 가장 많이 파열된 부위는 3p14.2.이었으며, 발현된 취약부위는 20부위였다. 4) 발현된 취약부위 중 7 부위는 JUN, SKI, REL, N-MYC, FHIT, MET, ETS-1, FOS의 암유전자가 위치하는 부위와 일치하였으며, 15부위는 급성림프구성백혈병, 급성골수성백혈병, 만성림프구성백혈병, 골수이형성성증, 악성흑색종, 신경모세포종, 소세포성폐암, 난소암, 유전성 신장암, 혼합성 지방육종시 염색체상에 이상이 있는 부위와 일치하였다. 결 론 : S기 특이성 항암제인 Ara-C는 정상성인의 T-림프구를 엽산이 부족한 MEM-FA배지에서 배양시 염색체 파열을 대조군에 비해 의미 있게 유도하였다. 한편으로 Ara-C 특이성 염색체 취약부위는 암유전자가 위치하는 부위 및 특정 종양에서 관찰되는 특이한 염색체변이가 있는 부위와 상당 예에서 일치하여 아직 알려지지 않은 암유전자를 찾거나 분석하는데 기초적인 자료를 제공할 뿐 아니라 염색체변화와 종양형성과정의 상관관계를 이해하는데 도움이 될 것으로 사료된다.
