A Study of Protein Ion Exchange Chromatography based on Plate Theory (단이론에 따른 단백질 이온교환 크로마토그라피의 연구)

  • Protein ion exchange chromatography was studied experimentally in order to prove the theoretical prediction from the linear model of Yamamoto, S. et al (1). Adsorption isotherms were measured as a function of ionic strength in a batch experiment. The relationship between the characteristics of chromatogram and the operating conditions of ionic strength, flow rate, length of column, concentration and amount of protein sample were studied. At the higher ionic strength, the lower flow rate and the longer column conditions, the higher number of plate was obtained. Satisfactory agreement was observed between the experimental and the calculated chromatograms except for the case of high protein concentration.

  • Red ginseng tails were extracted with ethanol solutions over a range of concentrations and temperature conditions. Investigations were carried out to study the effects of treatments on yields, soluble solids, saponin and precipitate occured in red ginseng extract beverage during storage. It was found that: (1) Higher concentration of ethanol at low temperature resulted in less yield of crude extract (2) The amount of precipitate in the non-purified extract beverage were less with decrease in ethanol concentration used (3) The treatment for purification of extracts and storage of purified extract at 37$^{\circ}C$ for 6 months had no effect on HPLC chromatogram pattern of saponins (4) The amount of purified extract decreased by purification treatment and more decrease was found as the temperature and concentration of ethanol increased. For Preparation of red ginseng extract beverage, the treatment of extracts with ethanol at low temperature was found to be more effective to minimize precipitation in tile beverage.

  • Saponins in leaf and stem of Gynostemma pentaphyllum that was collected from Jeju Island were extracted by the method for ginseng saponin. Comparison by overlapping chromatogram(HPLC) of pentaphyllum on that of ginseng and cochromatogram and ginseng although ginsenoside Rg2, Rg1 and Rf might be in common with rare possibility. It seems to be little difference in the kind of saponin glycosides between leaf and stem of pentaphyllum. Saponin content in leaf of pentaphyllum was higher than in stem, and much higher than those in ginseng. The kind of saponin glycoside in pentaphyllum appeared to be less than 22 and greater than those in ginseng. There was almost no change in saponins of pentaphyllum in methanol for 3 years at room temperature.

Detection of N,N-Dimethylaniline in Ampicillin Trihydrate by Thin Layer Chromatography and Densitometry (박층크로마토그라피 및 농도 측정법에 의한 Ampicillin Trihydrate 중의 N,N-Dimethylaniline의 검색)

  • A simple and convenient method of detecting N, N-dimethy laniline (D.M.A.) residues in ampicillin trihydrate (A.T.) was established. D.M.A. was extracted by chloroform from the chloroform presaturated N/10-ammonia water solution of A.T. and chromatographed on silica gel G thin layer. Blue spot appeared in 15minutes after spray of 2, 6-dichloroquinonechlorimide solution was compared with the blue spot of reference concomitantly processed. The developing solvent was prepared by mixing equal volume of cyclohexane and chloroform. To quantitate the amount of D.M.A. in A.T., T.L.C. was performed with the Eastmann Chromatogram sheet, then color density was measured by Cosmo Superclick densitometer. The developing solvent at this time was cyclohexane-chlorofonn (3+7) mixture. The peak areas obtained with the amount of D.M.A ranging from 0.05 to 2.0 .mu.g were linear to color density. Better sensitive results would be available with the densitometer equipped with monochromator.

Saponins of Gynostemma pentaphyllum (돌외의 Saponin 성분에 관한 연구)

  • Saponins of Gynostemma pentaphyllum that collected from Geochang region(Korea) and Dokujima(Japan) were extracted by the method for ginseng saponin. Comparison by retention time in chromatogram(HPLC) of G. pentaphyllum to that of ginseng showed that it is hardly to find out a common saponin between penta-phyllum and ginseng saponins. Saponin content extracted from G. pentaphyllum growing in Geochang region was higher than that growing in Dokujima region.

Detection of Taurine in Basidiomycetes

  • Taurine is one of the essential amino acids for humans and many of mammals. It is produced and contained in fleshes, shells, plants and algae, but has never been found in fungi. We examined six mushrooms for taurine and detected taurine in five of them. Taurine was determined by an automated amino acid analyzer using ion-exchange chromatography, being eluted between phosphoserine as a distinct peak on the chromatogram. Fruit bodies of Flammulina velutipes contained $83\mu$moles/100g fresh wt. the highest level among them, Agaricus bisporos $65\mu$moles, Lentinus edodes $49\mu$moles, Pleurotus ostreatus $9\mu$moles, and Auricularia auricula-judae $20\mu$moles. Taurine was not detected in Ganoderma lucidum. As far as fungi are concerned, this is the first report of the detection of taurine in Basidiomyces.

  • The essential oils of Elscholtzia ciliata and the cultivated tissue have been studied. The composition and contents of essential oils were identified by gas chromatography and mass spectrometry. The main component of essential oils of E. ciliata was naginata ketone. The essential oils from the flowers, leaves and stems of E. ciliata showed similar patterns of gas chromatogram. In experimental studies on the tissue culture of callus, it has been found that NAA induced higher growth rate and higher content of essential oils than 2, 4-D. The essential oils from the cultivated callus showed different composition from that of mother plants.

  • The contents of sterols in bean sprouts have been determined by mass spectrical analysis. From non-saponifiable fraction of n-Hexane extracts of the under ground parts sterol-like substance $(m.P.\;142-145^{\circ}C)$ was isolated. It was shown to be a sterol mixture on the basis of three peaks in gas liquid chromatogram. By mass spectrometrical analysis of the mixture, it was composed of 4.52mg% of Campesterol $(C_{28},\;{\Delta}^{5})$, 10.35mg% of ${\beta}-sitosterol\;(C_{29},\;{\Delta}^{5})$ and 7.75mg% of stigmasterol $(C_{29},\;{\Delta}^{5,\;22})$.

  • Melanoidins, which were higher polymers with intense brown color, were investigated on their decolorization and degradation by ozonolysis. Amino acids linked up with melanoidins were readily separated by depolymerization of melanoidins with ozonolysis. The IR spectra of ozone-untreated MRPs showed a higher peak at $1665cm^{-1}$ (C=N) and $1600cm^{-1}$ (C=C) than the corresponding peaks of ozone-treated MRPs. Ozone-treated melanoidins with molecular weight of above 900 showed the highest nitrogen composition of all melanoidins tested. Ozone-treated melanoidins with molecular weight of 900 to 1000 were separated into five peaks on recycling preparative HPLC chromatogram. Major functional groups in ozone-treated melanoidins with molecular weight of 900 to 1, 000 were -CH$_2$-CO-, -CH$_2$-O- and CH$_2$-.

  • In the present experiment the amino acid composition in the pollen of Pinus taeda and Pinus rigida and methbolic changes of the composition occurring in germinating pollen were examined by the method of paper chromatography. The results obtained in this study are as follows. 1. The mature pollen of Pinus taeda and Pinus rigida contained 12.00%, 13.19% crude protein respectively and sixteen protein-constituting amino acids and three unknowns were detected on the chromatogram. 2. Main components of the protein-constituting amino acids were leucines, valine, alanine, glutamic acid and aspartic acid. 3. The free amino acids such as leucines, valine, tyrosine, alanine and glutamic acid increased evidently during germination of the pine pollen.
