Synthesis of Two Dimensional Polymer Network (이차원 구조를 갖는 고분자의 합성)

  • Cholesterol-containing surfactant was synthesized, and it was sonicated with monomer [tetradeca(ethylene glycol) diacrylate] in water to from a vesicle solution. This vesicle solution was dried to construct a membrane which had a molecular multilayer structure. The monomer which stay in this membrane was polymerized with photoinitiation, and then surfactant was extracted by organic solvent. The physical properties of the two dimensional polymer network were measured, and these physical properties are tensile strength, elongation, and swelling.

Production of Cholesterol Inhibitor, Monacolin Produced from Monasces pilosus M-15 (Monascus pilosus M-15에 의한 콜레스테롤 합성저해제 Monacolin의 생산)

  • The monacolin retard in vivo synthesis of cholestrol. This study was conducted to select the best strain of Monascus sp. for monacolin production among 21 strains. In addition, research on suitable condition for mass production was conducted. Monascus pilosus M-15 was selected as the best strain for production of monacolin. The appropriate substrate for the monacolin production seem to be as follow : glycerin 5%, glucose 2%, corn steep liquor 0.5%, sodium nitrate 0.5% and length of fermentation was at $25^{\circ}C$ for 10~12days, respectively. The monacolin obtained from this organism was compared with standard monacolin using UV, IR, and GC/MS system and it was very close to the standard monacolin.

Effects of Saengshik Supplementation on Health Improvement in Diet-Induced Hypercholesterolemic Rats (고지혈증 유발 흰쥐에 있어서 생식의 건강개선효과)

  • The study was conducted to investigate the effect of Saengshik supplementation on lipid metabolism in rats fed high cholesterol diets. Male Spraque-Dawley rats were administrated 1% cholesterol to induce hypercholesterolemia and were fed on diet containing Saengshik (30%, w/w). Serum and liver lipid profiles and fecal bile acids excretion were examined after 7 weeks of experimental diet. The feeding of diet containing 30% Saengshik significantly decreased total cholesterol (TC) contents in liver and plasma. Since cholesterol was balanced by entero-hepatic circulation through bile acid synthesis and reabsorption, inhibition of bile acid re-uptake in intestine decreases total cholesterol in liver and blood. In this point, significant increament of fecal bile acid excretion in Shaengshik group decreased TC and improved hypercholesterolemia. Also plasma high density lipoprotein-cholesterol (HDL-C) decreasing risk for coronary heart disease in Saengshik supplemented group was significantly higher than control group, whereas low-density lipoprotein-cholesterol (LDL-C) accumulation in arterial vessel wall was significantly lower than control group. In this result, we supposed that supplementation of Saengshik, uncooked natural food, may improve hypercholesterolemia through increment of cholesterol excretion.

Effect of Fermented Ice Plant Extract on the Inhibition of Triglyceride and Cholesterol Synthesis and Tyrosinase Activity (발효 아이스플랜트(Mesembryanthemum crystallinum L.) 추출물의 triglyceride, cholesterol 합성저해 및 tyrosinase 활성억제 효과)

  • This study investigated changes in triglyceride and cholesterol synthesis and tyrosinase activity induced by ice plant (Mesembryanthemum crystallinum L.) extract, which cannot be stored for long periods of time due to its high moisture content when it was fermented to improve its storage stability. The accumulation of triglyceride and cholesterol in HepG2 cells inhibited the accumulation with a relatively large magnitude in n-butanol and aqueous fractions that generally have high polarity, however, changes in inhibition potency due to the fermentation were not significant. As for the effect to inhibit tyrosinase activity, when L-tyrosine was used as a substrate, the inhibitory activity was the highest for the aqueous fraction at $60.58{\pm}4.03%$ and $63.35{\pm}4.35%$, before and after fermentation, respectively, which amounted to 72% of that of the positive control group (arbutin, $100{\mu}g/ml$). In addition, when L-3,4-dihydroxyphenylalanine (L-DOPA) was used as a substrate, the inhibitory activity was also found the highest for the aqueous fraction at $56.85{\pm}1.57%$ and $59.38{\pm}1.74%$, before and after fermentation, respectively, which amounted to at least 88% of that in the positive control (kojic acid, $100{\mu}g/ml$). Overall, the activity of the fermented ice plant extract was similar or a little higher compared to that of the one without fermentation, indicating that fermentation can be a good approach to improve the storage stability of the ice plant.

The Effect of Rice with Aspergillus terreus on Lipid Metabolism in Rats (황국곡자 투여가 지방질 대사에 미치는 영향)

  • The aim of this study was to investigate the effects of rice contatining Aspergillus terreus (Hwangkuk, HK) on lipid metabolism in male Sprague-Dawley (SD) rats fed a high-cholesterol diet (HCD) for 8 weeks. SD rats were divided into five groups: Normal, [Negative Control (HCD), Positive Control (lovastatin)], [HK 0.5 g/kg and HK 2 g/kg]. Hepatic total lipids significantly decreased following treatment with rice contatining Asp. terreus. Furthermore, this treatment led to higher expression levels of HMG-CoA reductase, LDL receptor and SREBP2 mRNA in the liver compared with the HCD group. In addition, histopathologic evaluation showed that feeding rats with rice containing Asp. terreus suppressed hepatic steatosis. These results suggest that rice containing Asp. terreus may be able to regulate of cholesterol synthesis and prevent hyperlipidemia.

  • Synthesis of a new copoly (M4Ch-ChMA), copoly ((4-methacryloyloxy) chalcone-cholestery methacrylate), with chalconyl and cholesterol moiety characteristics for photoalignment materials was investigated. Good thermal stabilities of the synthesized copolymers are confirmed by thermogravimetric analysis (TGA) measurement. The pretilt angles of the nematic liquid crystal (NLC) are reduced as UV exposure time is increased on the copolymer surfaces. A pretilt angle of 81$^{\circ}$in NLC was observed with UV exposure of 3 min on the copolymer-3 surface. The NLCs pretilt angle is attributable to increased chalcone with increasing the UV exposure time.

Synthesis of $\beta$-Sitosterol Esters with Conjugated Linoleic Acid and Medium Chain Fatty Acids by Using Lipase as Catalyst

  • Plant steryl esters have good effects on plasma cholesterol level and are used as functional food ingredient. Conjugated linoleic acid (CLA) presents mainly in animal foods and has a good benefit and medium chain fatty acids (MCFAs) are a rapid energy source for human. In this study, we produced the ${\beta}$-sitosterol esters from CLA and MCFAs using various lipases as catalysts. Among lipases, AYS (from Candida rugosa) was the most effective for synthesis of ${\beta}$-sitosterol esters in the presence of water (24.35% conversion) or hexane (25.33% conversion). The second esterification extent was obtained by lipase AK (from Pseudomonas sp), showing 10.26% conversion in water and 15.94% conversion in hexane, respectively. The reaction condition was 1:3 molar ratio (${\beta}$-sitosterol:fatty acid, 1:3) and stirred (175 rpm) at 55$^{\circ}C$ in water bath shaker for 48h.

Testicular fat deposition attenuates reproductive performance via decreased follicle-stimulating hormone level and sperm meiosis and testosterone synthesis in mouse

  • Objective: Testicular fat deposition has been reported to affect animal reproduction. However, the underlying mechanism remains poorly understood. The present study explored whether sperm meiosis and testosterone synthesis contribute to mouse testicular fat deposition-induced reproductive performance. Methods: High fat diet (HFD)-induced obesity CD1 mice (DIO) were used as a testicular fat deposition model. The serum hormone test was performed by agent kit. The quality of sperm was assessed using a Sperm Class Analyzer. Testicular tissue morphology was analyzed by histochemical methods. The expression of spermatocyte marker molecules was monitored by an immuno-fluorescence microscope during meiosis. Analysis of the synthesis of testosterone was performed by real-time polymerase chain reaction and reagent kit. Results: It was found that there was a significant increase in body weight among DIO mice, however, the food intake showed no difference compared to control mice fed a normal diet (CTR). The number of offspring in DIO mice decreased, but there was no significant difference from the CTR group. The levels of follicle-stimulating hormone were lower in DIO mice and their luteinizing hormone levels were similar. The results showed a remarkable decrease in sperm density and motility among DIO mice. We also found that fat accumulation affected the meiosis process, mainly reflected in the cross-exchange of homologous chromosomes. In addition, overweight increased fat deposition in the testis and reduced the expression of testosterone synthesis-related enzymes, thereby affecting the synthesis and secretion of testosterone by testicular Leydig cells. Conclusion: Fat accumulation in the testes causes testicular cell dysfunction, which affects testosterone hormone synthesis and ultimately affects sperm formation.
