The Plasma Fatty Acid Composition and Cholesterol Levels of Rates Fed Different Sources of $\omega$3 Fatty Acid and Excess DHA during Gestation, Lactation, and Growth

  • This study was designed to investigate the effect of different sources of $\omega$3 fatty acid in the diet with a similar polyunsaturated/saturated (P/S) fatty acid ratio and $\omega$6/$\omega$3 fatty acid ratio as well as excess DHA on the plasma fatty acid composition and cholesterol level of rats. Three experimental diets contained 10% (w/w) dietary lipids. The control diet and one treatment diet were corn oil-based diets with different $\omega$-3 fatty acid sources: perilla (CO) or fish oil (CF), respectively. In order to examine the effect of excess DHA, the other treatment diet (FO) was a fish oil-based diet with corn oil to supply essential fatty acids at the level of 1.8% (w/w) of the diet. Female Sprague Dawley rats were fed the experimental diets for 2 weeks prior to mating and throughout gestation and lactation. Pups were weaned to the same diet of dams at 21 days of age. Plasma fatty acid compositions and cholesterol contents were analyzed for pups at 3th, 7th and 10th week after birth. Plasma DHA concentrations increased significantly as the level of fish oil supplementation increased. Three-, seven- and ten-week old rats fed on CO diet which contained only $\alpha$-lino1enic acid as a $\omega$-3 fatty acid Source had Plasma DHA levels of 4.85%, 3.15% ana 2.47%, respectively, suggesting that rats at this period of development can convert $\alpha$-linolenic acid to DHA. But the ability to form DHA might be limited, since dietary DHA showed to be more effective in raising the plasma level of DHA. There was a significant negative correlation between DHA and cholesterol concentration of the rat plasma at 7th week (r=0.34, p<0.05) and l0th week after birth (r=036, p<0.05), proving the hypocholesterolemic effect of DHA.

Seamustard (Undaria pinnatifida) Improves Growth, Immunity, Fatty Acid Profile and Reduces Cholesterol in Hanwoo Steers

  • The study was designed to evaluate the effect of 2% seamustard (Undaria pinnatifida) by-product (SW) on growth performance, immunity, carcass characteristics, cholesterol content and fatty acid profile in Hanwoo steers. A total of 20 Hanwoo steers (ave. 22 months old; 619 kg body weight) were randomly assigned to control (basal diet) and 2% SW supplemented diet. Dietary SW supplementation significantly (p<0.05) improved average daily gain and gain:feed ratio as well as serum immunoglobulin G concentration. Chemical composition and quality grade of meat and carcass yield grades evaluated at the end of the trial were found to be unaffected by SW supplementation. Dietary SW significantly reduced meat cholesterol concentration (p<0.05). Dietary SW supplementation significantly reduced the myristic acid (C14:0) and palmitoleic acid (C16:ln-7) concentration, while SW increased the concentration of stearic acid (C18:0) and linolenic acid (C18:3n-3) compared to control (p<0.05). Dietary SW supplementation had no effect on saturated fatty acids (SFA), unsaturated fatty acids, poly unsaturated fatty acid (PUFA) or mono unsaturated fatty acid content in muscles. A reduced ratio of PUFA/SFA and n-6/n-3 were found in SW supplemented group (p<0.05). In conclusion, 2% SW supplementation was found to improve growth, immunity and fatty acid profile with significantly reduced cholesterol of beef.

Age-Related Changes of Lipid Metabolism and Thrombogenic Capacity in Rats (노화과정에 따른 흰쥐의 지방대사와 혈전능의 변화)

  • This study was performed to investigate the age-related changes of the lipid metabolism and thrombogenic capacity in Sprague-Dawley (SD) rats at the ages of 4, 8, 12, 16, 20 and 24 months old. Total lipid, triglyceride (TG) and total cholesterol in plasma and liver, HDL-cholesterol concentration, and eicosanoid contents in plasma were measured. Lipid peroxides were determined by the levels of thiobarbituric acid reactive substance (TBARS) in LDL fraction. Body weight was increased continuous until 16 months and decreased after 20 months. Epididymal fat pad (EFP) weight was increased continuously until 20 months and decreased at 24 months. Total lipid and TG concentrations in plasma were increased until 20 months and then rapidly decreased at 24 months but plasma cholesterol was increased continuously with aging. HDL-cholesterol level was increased continuously until 12 months, but decreased at 16 months and maintained there after. The TBARS levels in LDL fraction were the highest level at 24 months. Liver total lipid, TG, and total cholesterol concentrations were shown a tendency to increase with aging, and especially TG concentration was increased rapidly from 12 months to 16 months. Plasma thromboxane B$_2$ (TXB$_2$) and 6-keto-prostaglandin F$_1$ (6-keto-PGF$_1$) contents did not change with aging, but the ratio of TXB$_2$/6-keto-PGF$_1$ was increased with aging, especially from 8 to 12 months. These results showed that lipid levels in plasma and liver, TBARS levels in LDL fraction, and TXB$_2$/6-keto-PGF$_1$ ratio were increased with aging.

The Effect of the Long-Term Ginseng Intakes on Serum Lipids Profile and Hemostatic Factors in Human (장기적인 인삼섭취가 인체의 혈전 관련 요인에 미치는 영향)

  • This study was designed to investigate the effectiveness of ginseng intakes in modifying serum lipid profiles and plasma clotting factors. The participants in this study were 47 normal healthy volunteers(men 24, women 23) with an age range of 35-49 years and a mean age of 41 years residing in Taejon. Based on the diet intakes, subjects were classed into one of three groups : control, vegetarian, and ginseng consumed over 3-4 years. There was no significant difference in their physical characteristics. Dietary calorie intakes were not significantly different in subjects. The ratio of energy intake in the control and ginseng consumed group was 63-64% : 20-21% : 15-16%(Cho : Fat : Pro), but 70-73% : 13-14% :14-15%(Cho : Fat : Pro) in the vegetarians. The intakes of animal food in the vegetarian was significantly lower than the control and ginseng consumed group in men. The ratio of P/S(1.27) was the highest in the vegetarians. Venous blood samples were taken for serum lipid profiling, plasma clotting assay and platelet function. The concentration of serum triglyceride in the men ginseng group is significantly lower than those of the men control group. Serum lipid profiles values of the men ginseng group, such as total cholesterol and phospholipid were lower those of the men control group, but higher those of the men vegetarian group. the serum lipid profile in the women were not significant, but total cholesterol, triglyceride and LDL cholesterol levels in the ginseng groups were low. The concentration of HDL cholesterol was not significantly different. Platelet cell count and platelet aggregation were low in the ginseng groups. APTT(Activated Partial thromboplastin time) was significantly elongated in ginseng groups in the normal range. In seems that the major beneficial effects of ginseng intakes in especially men were on the blood concentrations of triglyceride, total cholesterol and elongation of plasma clotting time.

Effect of Cheonggukjang Made from Sword Bean on Serum Lipid Profile and Tissue Change in Rats

  • This study was to investigate serum triglyceride (TG), phospholipid (PL), and cholesterol concentrations after feeding the combined diets of prepared traditional Cheonggukjang (TC) or Cheonggukjang made from sword bean powder (CS), the feed, and high-fat cholesterols at certain ratio for 8 weeks in rats, and then confirmed the effects of the diets based on the changes in the tissues of livers, kidneys, and spleens. The serum TG concentrations were lower in CS group compared to that of in TC group. The increase of PL concentrations was greater in CS group than that of TC group. FChCS6 group (92% feed+2% cholesterol+6% CS) showed the lowest total-cholesterol (T-C), $60.1{\pm}8.5mg/dL$, indicating that CS was effective to lower the T-C. Both TG/PL and T-C/PL ratio tended to be decreased. As CS intake was effective in the reduction of low density lipoprotein-cholesterol level as well as the increase of high density lipoprotein-cholesterol level, it would be effective in the prevention of adult diseases and circulatory diseases. It was confirmed that there were no changes in organ appearance and tissues in the rats with CS intake. Therefore, CS intake showed superior functionalities than that of TC intake.

Effects of Herbal Acupuncture(Raphanus Sativus) at $BL_{20}$ on the Hyperlipemia Rats Induced by High Fat Diet (비유에 대한 래복자 약침(藥鍼)이 고지방 식이로 유발된 비만(肥滿) 백서(白鼠)에 미치는 영향)

  • Objectives : This research was performed to investigate the effect of herbal acupuncture(Raphanus sativus) at Bisu($BL_{20}$) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into high fat diet group(Control), high fat diet and saline in $BL_{20}$ treated group(Saline), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-1, 0.058mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-2, 0.029mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-3, 0.011mg/ml/g), Herbal acupuncture was bilaterally treated at 0.2cc per 4days, total 14 times in 56days. Results : Body weight and food efficiency were decreased in saline, RHA-1, RHA-2. Feed intake was increased in saline, RHA-1, RHA- 2, RHA- 3. The level of serum total cholesterol was decreased in saline, RHA-1. The level of serum HDL-cholesterol, LDL-cholesterol were not significant. The level of serum triglyceride, Total Bilirubin were decreased in saline, RHA-1, RHA-2, RHA-3.The level of serum AST was decreased in RHA-1. But the level of serum ALT was increased in RHA-1. The level of atherogenic index was decreased in saline, RHA-1. The level of HTR was increased in saline, RHA-1. Conclusions : Raphanus sativus-herbal acupuncture in Bisu($BL_{20}$) can control body weight, feed intake and feed efficency ratio, total cholesterol, total bilirubin, and prevention cardiovascular risk.

Effects of the Korean Mistletoe Hot-Water Extract on the Lipid Components and Blood Pressure Level in Spontaneously Hypertensive Rats (한국산 겨우살이 열수추출액이 본태성 고혈압쥐의 지질성분 및 혈압 저하에 미치는 영향)

  • This study was designed to observe the effects of Korean mistletoe(Viscum album L. var. coloratum Ohwi) hot-water extract administration on the improvement on the lipid components, apolipoprotein and blood pressure level in spontaneously hypertensive rats (SHR, Wistar strain, male) fed the experimental diets for 33 days. Concentrations of total cholesterol, triglyceride (TG), LDL-cholesterol, free cholesterol and atheroscieotic index in serum were significantly lower in the Korean mistletoe extract administration group [group KM(Korean mistletoe 10.0 g% hot-water extract)] than those in the control group. In the ratio of HDL-cholesterol concentration Korean mistletoe 10.0% hot-water extract administration group were higher percentage than in the control group. However, concentrations of total cholesterol and TG in liver and brain were significantly lower in the group KM than those in the control group. But the components of apolipoprotein (Apo) A-I and Apo A-II in serum were significantly higher in the KM group than in the control group. However, components of Apo C-II, Apo C-III, Apo E and ratio of Apo B to Apo A-I in serum were fairly reduced in the group KM than in the control group. The levels of systolic and diastolic blood pressure were significantly lower in KM cup than control group. From these results, Korean mistletoe hot-water extract administration were effective on the improvement of the lipid components, coronary heart disease and hypertension in spontaneously hypertensive rats. And particularly, Korean mistletoe extract administration were more effective as a therapeutic regimen for the control of blood pressure in hypertension.

Effects of Amino Acid Composition and Average Hydrophobicity of Soybean Peptides on the Concentration of Serum Cholesterol in Rats (대두 펩타이드의 아미노산 조성 및 평균소수도가 흰쥐의 혈청 콜레스레롤 농도에 미치는 영향)

  • Effects of amino acid composition and average hydrophobicity of soybean peptides on serum cholesterol in rats were investigated. Soybean protein(ISP), casein(SCN), their peptic hydrolyzates fractionated by acid precipitations(SHT, SH8, SH6, SH4, CHT), and amino acid mixtures of the same composition as the proteins(SAA, CAA) were prepared to feed to rats. The amino acid composition of the peptides was analyzed by HPLC and the concentration of serum cholesterol in the rats was measured. By data analysis, it was found that there was no relationship between ratio of Lys/Arg or molar ratio of hydrophobic amino acids and serum cholesterol level. And also there was no relationship between the concentration and average hydrophobicity calculated by the method of Tanford, Manavalan, for Meirovitch, only except by the method of Krigbaum(r=-0.736); the higher the average hydrophobicity of Krigbaum was, the lower the concentration of serum cholestrol became.

Effects of Invasive Low Level Laser Acupuncture Therapy(LLLAT) at LU7, LR5, LU7+LR5 on the Hyperlipemia Rats Induced by High Fat Diet (열결(列缺), 여구, 열결배여구에 침습적(侵襲的)으로 조사(照射)된 레이저침료법(鍼療法)이 고지혈증(高脂血症) 백서(白鼠)에 미치는 영향(影響))

  • Objectives: This research was performed to investigate the effect of invasive low level laser acupuncture therapy(LLLAT) at Yolgyol(LU7), Yogu(LR5) and Yolgyol+Yogu(LU7+LR5) on weight gain, food intake, food efficiency, lipid metabolism, atherogenic index, HTR(HDL-cholesterol to total cholesterol ratio) and liver function in hyperlipidemia rats. Methods : Experimental groups were divided into high fat diet group(Control group), high fat diet and LLLAT at LU7(LU7 group), high fat diet and LLLAT at LR5(LR5 group), LLLAT at LU7 and LR5(LU7+LR5 group). Animals was treated by the LLLAT at 30mW-5min once a 2day during 5 weeks. Results: Body weight was decreased significantly in LU7+LR5 group when compared with control group. Food intake was increased significantly in LU7, LR5, LU7+LR5 group when compared with control group. Food efficiency was decreased significantly in LU7, LR5, LU7+LR5 group when compared with control group. In the lipid metabolism, total cholesterol and HDL-cholesterol was decreased significantly in LU7+LR5 group, LDL-cholesterol and phospholipids were decreased significantly in LR5, LU7+LR5 group, triglyceride and fee fatty acid were decreased significantly in LU7 group when compared with control group. Atherogenic index was decreased significantly in LU7, LR5, LU7+LR5 group when compared with control group. HTR was increased significantly in LU7 group when compared with conool group. In the liver function, the significance was not showed in AST and ALT, ALP was decreased significantly in LU7+LR5 group when compared with control group. Conclusions: LLLAT at LU7 and LR5 maybe can manage hyperlipidemia by controlling body weight, food intake, food efficiency ratio and lipid metabolism.

Effects of Different Forms of Chromium Supplements on Serum Glucose, Insulin and Lipids in Rats

  • This study evaluated the effects of different forms of chromium supplements on serum glucose, insulin and lipid concentrations in rats. Sprague-Dawley male rats were randomly assigned to one of three dietary groups and fed AIN-76 semi-purified basal diets supplemented with 300 ppb Cr from Cr methionine (CrMet) and Cr chloride $(CrCI_3)$ or without Cr (control). By the end of the $4^{th}$ week, all rats were decapitated, blood collected, and serum glucose, insulin and lipid concentrations were determined. The CrMet and $CrCl_3$ supplementation did not affect weight gain and feed efficiency ratio. However, feed intake was significantly higher in CrMet groups than control (p < 0.05). CrMet-supplemented rats had markedly increased insulin levels (p < 0.05) compared with controls. Serum lipids were not significantly different between the control and the CrMet groups. $CrCl_3$ supplementation decreased total cholesterol and triglyceride, but the decreases were only significant for the control group. $CrCl_3$ supplementation was associated with significant decreases in total cholesterol compared with CrMet supplementation. These results indicate that CrMet supplementation is effective for increasing serum insulin, and $CrCl_32$ may improve lipid concentrations, because we observed decreased serum total cholesterol and an improved total cholesterol/HDL-cholesterol ratio (THR).
