Hog Cholera 병돈(病豚)의 뇌(腦) 및 임파장기(淋巴臟器)에 관한 병리조직학적(病理組織學的) 연구(硏究) I. 임상(臨床) 및 병리해부학적(病理解剖學的) 관찰(觀察) (Histopathologic Studies on the Brain and Lymphoid Organs in Hog Cholera I. Clinical and Pathological Observation in Hog Cholera)

  • This study was taken to clarify the clinical signs and macroscopical lesions of pigs naturally infected with hog cholera. The clinical and macroscopical observation on the natural cases of hog cholera and experimental cases inoculated with ALD Virus and isolated virus strains were carried out. The results obtained are as follow; In clinical inspection of the natural cases, diarrhea (73.1%) blotching of ear (50.0%), staggering (42.3%), erythema of skin (40.0%), constipation (38.5%), conjunctivitis (32.7%) and dyspnea (30.8%) were observed. Dyspnea, constipation and erythema of skin were observed mainly in the experimental cases, however, staggering and conjunctivitis in pigs infected with ALD virus were found and convulsion and hemorrhage of skin of pigs infected with isolated virus were seen, respectively. The gross lesions of natural cases were hemorrhage of lymph node (82.5%), enteritis and hemorrhage of large intestine (65.0%), splenic infarction (57.5%), pneumonia (55.0%), gastritis and hemorrhage (52.5%), cardiac hemorrhage (40.0%) and renal petechiation (37.5%), while in the experimental cases, hemorrhage of lymph node, pneumonia, gastritis and hemorrhage, enteritis and hemorrhage of laryge intestine and splenic infarction were seen mainly.

  • Cholera toxin and dibutyryl cyclic adenosine 3', 5'-monophosphate(db-cAMP) increased the rate and number of infections units produced in the in vitro reactivation of latent herpes simplex virus, whereas adenosine diminished them. cAMP concentration in latently infected trigeminal ganglia of mice was greatly increased by cholera toxin but was not affected by adenosine.

  • Yemen has been faced with the worst cholera epidemic of modern times, with more than 1 million suspected cases and 3000 deaths at the time of writing. This problem is largely due to the longstanding civil war between pro-government forces and the Houthi armed movement, which has severely damaged already vulnerable sanitation and healthcare facilities and systems in the country. It is further compounded by a dire lack of basic amenities, chronic malnutrition, and unfavourable weather conditions. Another contributory component may be aerial transfer by cholera-infected chironomid insects. To contain the spread of cholera in Yemen, a nation-wide armistice should be negotiated, and national and local committees must be convened to coordinate efforts on the ground. Community isolation facilities with proper sanitation, reliable disposal systems, and a clean water supply should be set up to isolate and treat sick patients. The continuity of vaccination programmes should be ensured. Public health campaigns to educate local communities about good hygiene practices and nutrition are also necessary. The One Health paradigm emphasizes a multi-sectoral and transdisciplinary understanding and approach to prevent and mitigate the threat of communicable diseases. This paradigm is highly applicable to the ongoing cholera crisis in Yemen, as it demands a holistic and whole-of-society approach at the local, regional, and national levels. The key stakeholders and warring parties in Yemen must work towards a lasting ceasefire during these trying times, especially given the extra burden from the mounting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) outbreak worldwide.

A 6-Week Oral Toxicity Study of Oral Cholera Vaccine in Sprague-Dawley Rats

  • The present study was carried out to examine the toxicity and target organs of oral cholera vaccine (OCV) after repeated oral administration in Sprague-Dawley rats for 6 weeks (3 administrations, once every 2 weeks). OCV is an inactivated oral cholera vaccine that contains Vibrio cholerae and confers protection against cholera caused by V. cholera serogroups O1 (Inaba and Ogawa serotypes) and O139 (strain 4260B). The animals were orally administered either OCV placebo (negative control) or OCV at a dose equivalent to 240 times the anticipated human dose. Throughout the administration period, no significant change was detected in clinical signs, body weight, food or water consumption, urinalysis results, hematological and clinical biochemistry test results, organ weights, necropsy, or histopathological examination results. Minor changes were found in hematological and clinical biochemistry tests; however, these changes were within normal ranges. The above results suggest that oral administration of OCV in rats did not induce any toxicologically meaningful changes, and the target organs could not be determined. This study was conducted in accordance with the guidelines established by Good Laboratory Practice (2009-183, KFDA, December 22, 2009) and the OECD Principles of Good Laboratory Practice (1997).

Outbreak of chronic fowl cholera in broiler breeder in Korea

  • Fowl cholera is an infectious disease caused by .Pasteurella multocida, affecting domesticated and wild birds. It usually appears as a septicemia of sudden onset with high morbidity and mortality, but chronic conditions that characterized by localized infections often occur. 13wks broiler breeders were submitted to the Kyung-pook national university for diagnosis. Clinical signs included approximately 1% mortality, severe lameness, ruffled feathers and swollen and/or cloudy eyes. At necropsy, the outstanding lesions were seen swollen hock joint, which were suppurative or caseous exudates, inflammation of conjunctiva, severe pneumonia and epicarditis. The causative agent was isolated from the hock joint, liver, sinus and sternum of the chickens, and performed physiological and biochemical test. To identify the serotype of P. multocida, capsular serotyping was conducted by multiplex polymerase chain reaction (PCR). In the antimicrobial susceptibility test, the isolates were resistance to the aminoglycosides. In this study, we confirmed chronic fowl cholera (FC) caused by P. multocida in broiler breeders in Korea.

Fowl Cholera Outbreak in Domestic Poultry and Epidemiological Properties of Pasteurella multocida Isolate

  • Symptoms of fowl cholera including orofacial edema, swollen and edematous wattles and combs, and severe respiratory disorders were detected in domestic poultry in two broiler breeder farms: one located in Gyeong-gi Province (October, 2000) and the other in Chung-cheong-nam Province (March, 2001). Gram-negative, bipolar staining bacillus was easily found in a direct smear. The biochemical properties of isolates were examined using a standard diagnosis method, proving that they were 99.7% similar to the Pasteurella multocida (P. multocida: PM), a pathogenic and causative agent of fowl cholera (FC). As a result, an FC outbreak in domestic fowls was confirmed for the first time in Korea since 1942. Because FC was detected in broiler breeder farms for the first time in 59 years at the same time as an FC outbreak was confirmed in wild birds (October, 2000), our concern was focused on whether the PM strains that originated in wild birds were transmitted into poultry forms. The possibility was tracked down by comparing phenotypic and genetic properties between the two types of PM strains. PM strains of chicken origin showed prominent differences from the PM strains of wild bird origin in both phenotypic and genetic properties. An examination of the origin of the wild bird bacteria was conducted, but no evidence has been identified that PM strains from the wild bird were introduced into domestic poultry farms.

  • This study was taken to clarify the histopathological changes of pigs naturally infected with hog cholera. Microscopic observations of the necrotic lesion and inclusion body in the lymphoid organs were carried out in the natural cases of hog cholera and experimental cases inoculated with ALD virus and isolated virus strains. Electron microscopic findings of the intranuclear inclusion bodies in the reticular cell of spleen and lymph node were also observed in the experimental cases. The results obtained are as follow, As the histological findings necrosis of lymphoid organs was observed mainly in the lymph follicle. The necrotic lymphoid organs were found to contain 35.0% in the natural and 37.5% in the experimental cases. Intranuclear inclusion bodies were found mainly in the reticular cells of lymphoid organ, the epithelium of bronchiole and alevolus, and the vascular endothelium of brain. These inclusion bodies were seen in 40.0% of the natural cases and all of the experiment. The inclusion body was appeared to compose of activated nucleoli and chromatin granules (interchromatin and perichromatin) by electron microscopy.

Molecular Identification of Cryptosporidium viatorum Infection in a Patient Suffering from Unusual Cryptosporidiosis in West Bengal, India

  • In this study, we have collected and screened a total of 268 stool samples from diarrheal patients admitted to an Infectious disease hospital in Kolkata for the presence of Cryptosporidium spp. The initial diagnosis was carried out by microscopy followed by genus specific polymerase chain reaction assays based on 70 kDa heat shock proteins (HSP70). DNA sequencing of the amplified locus has been employed for determination of genetic diversity of the local isolates. Out of 268 collected samples, 12 (4.48%) were positive for Cryptosporidium spp. Sequences analysis of 70 kDa heat shock proteins locus in 12 Cryptosporidium local isolates revealed that 2.24% and 1.86% of samples were showing 99% to 100% identity with C. parvum and C. hominis. Along with the other 2 major species one recently described globally distributed pathogenic species Cryptosporidium viatorum has been identified. The HSP70 locus sequence of the isolate showed 100% similarity with a previously described isolate of C. viatorum (Accession No. JX978274.1, JX978273.1, and JN846706.1) present in GenBank.

  • Intestinal absorption of dietary taurine is one of the regulatory component maintaining taurine homeostasis along with renal reabsorption, bile acid conjugation and secretion, and de nobo synthesis of taurine in mammals. Recent observations of decreased enterocytic levels of taurine in response to trauma, infection and surgical insults, postulate the possibility that intestinal taurine absorption might be impaired in such stressed conditions. The aim of the present study was to evaluate changes in enterocytic taurine transporter activity using the human intestinal colon carcinoma cell line, HT-29, in various stress-induced conditions. Pretreatment of the HT-29 cells with dexamethasone, a stress hormone(0.1,1,10 or 100$\mu$M) for 3 hrs, or with E coli heat-stable enterotoxin(10, 100, or 200nM) for 30 minutes in order to induce the condition of enterotoxigenic infection did not influence taurine uptake as compared to the value found in control cells. In contrast, pretreatment of the cells with cholera toxin(10, 100, 500, or 1000ng/ml)for 3hr or 24hr significantly decreased taurine uptake by HT-29 cells to 40~50% of the value found in untreated control cells. Kinetic studies of the taurine transporter activity were conducted in control and cholera toxin treated HT-29 cells with varying taurine concentrations(2~60$\mu$M) in the uptake medium. Pretreatment of the cells with cholera toxin(100ng/ml) for 3hr did not influence the Vmax, but resulted in a 55% increase in the Michaelis-Menten constant(Km) of the taurine transporter compared to those in control cells. These results suggest that cholera toxin-induced reduction in taurine transporter activity in HT-29 cells is associated with decreased affinity of the taurine transporter without altering the amount of transporter protein. Intestinal taurine absorption appears to be reduced in the condition of water-borne diseases caused by bacteria such as V. cholerae. This might influence the taurine status of infants and young children more readily, an age group in which the prevalence of intestinal infection is high and the role of intestinal absorption is crucial for maintaining the body taurine pool. 토종닭에서 가금콜레라(Fowl Cholera) 발생 (Outbreak of Fowl Cholera in native chickens)

  • Fowl cholera(FC), which has been classified as reportable animal disease in Korea, occurred in the backyard farm located in Chungju, Chungbuk province. The farm was not ordinary domestic poultry farm but backyard farm with main cultivation of fruits. The farmer reported unusual mortality of chickens which were grazed in his own fruit farm without any poultry house. There was a lots of opportunities to contact with wild animals including wild birds freely. Clinical signs observed in the chickens were torticolis, mucous excretion from mouth and greenish diarrhea. On the necropsy we found multifocal necrosis on the surface of liver, hemorrhages in the epicardium and ovary and mucopurulent exudates in the joint. The causative agents was isolated from the liver of chickens collected from the farms and identified as Pasteurella multocida using biochemical tests of bacteria. Also, the isolated bacteria were inoculated into specific pathogen free chickens to evaluate the pathogenicity. High mortality and similar pathological lesions compared to those of chickens died in the farm were observed. We report the first case of fowl cholera of the chickens in Korea and further studies are needed to evaluate the serotype, genotype and pathogenicity of isolated Pasteurella multocida.
