Three isolates of Cucumber mosaic virus isolated from Commelina communis plants showing chlorosis and mosaic were collected in Chungju and Chuncheon, Korea. To confirm genetic variation of these three isolates (CMV-Co, CMV-Co2, and CMV-Co3), we performed PCR-RFLP and sequence analysis. Sequences of coat protein genes of CMV-C0, -Co2 and -Co3 were compared with CMV-Fny and showed 96.3%, 96.3%, and 95.9% similarities, respectively. In host reactions, three CMV-Co isolates induced systemic necrosis in Cucurbita pepo unlike CMV-Fny and CMV-Co, CMV-Co2 and CMV-Co3 observed differential symptoms responses in Physalis angulata and Nicotiana rustica. These results indicated that three isolates of CMV isolated from C. communis have genetic and biological variation.
Transmission of mulberry dwarf mycoplasma (MDM) from diseased mulberry to 5 herbaceous plants (periwinkle, white clover, Ladino clover, red clover, and Chinese milk vetch) through insect vector, Hishimonus sellatus, was confirmed by symptom expression and microscopic evidences. The earliest symptom appearance was noticed on periwinkle in which incubation period was 25-30 days, while it ranged 35-40 days in the other plant species. The common symptoms of MDM infected plants were characterized by poor plant growth with accompanying discolorations of leaves (chlorosis with vein clearing in periwinkle, reddish in red clover, brownish in white and Ladino clovers, and yellowish in Chinese milk vetch). Mycoplasmal infections were diagnosed light microscopically by Dienes' and toluidine blue staining of hand-cut and Epon-embedded sections, respectively. In Dienes' stain, all the plants infected with MDM showed specific staining reaction in phloems. In toluidine blue stain, mycoplasmal existence was noted by granular appearance in sieve tubes which were confirmed to be mycoplasma-like organisms under an electron microscope.
Weeds are widely grown in the field and are infected by many viruses. A survey was conducted to identify viruses infecting weeds in Korea. Virus-infected weed samples including Rorippa indica (L.) Hiern, R. islandica (Oed.) Bord, Crepidiastrum denticulatum (Houtt.) Pak & Kawanno, Achyranthes japonica (Miq.) Nakai, and Chrysanthemum boreale (Makino) Makino were collected in Kyonggi Province. These weeds were grown in the greenhouse and were isolated on 10 test plants. Several virus isolates were isolated fron infected tissues and were further studied by host range assay, serological test, electron microscopy (EM), reverse transcription-polymerase chain reaction (RT-PCR) and sequencing. Each isolated virus strain was mechanically transmitted to weeds and various hosts including Nicotiana spp., Brassica spp., Vigna unguiculata, Capsicum annuum, and Cucumis sativus and showed systemic mosaic, vein clearing, necrosis, mottle, malformation, chlorosis, and/or death of host plants in some cases. Each virus was then purified using infected leaves and observed by EM. From these results three viruses were isolated and identified as Turnip mosaic virus (TuMV), Broad bean wilt virus (BBWV), and Cucumber mosaic virus (CMV). RT-PCR using virus-specific oligonucleotide primers and the cloning were conducted to determine the nucleotide sequences of coat proteins of the three viruses their amino acid sequence were deduced. The amino acid sequence homologies were about 92.7 to 99.7%, 96.2 to 97.7%, and 93.9 to 98.6% to other reported TuMV, BBWV, and CMV strains, respectively. These results suggest that many weeds may serve as primary inoculum source of diseases caused by TuMV, BBWV, CMV and that the management of these viral diseases can be achieved through weed control.
A survey of the occurrence of abiotic diseases on kiwifruit (Actinidia deliciosa) trees was conducted at sixty-two kiwifruit orchards in twenty-one locations of Jeonnam and Jeju Provinces in Korea during the 2007 growing season. Various kinds of abiotic diseases were detected on the kiwifruit trees. Malformed fruits caused by incomplete pollination were commonly observed among the normally growing fruits on almost all of the kiwifruit trees examined. Fruits imbued with wire rust, fruits scratched or girdled by the wire and fruits injured by sunscald occurred in all of the examined orchards. Abnormal growth of girdled branches by the fence wire, dead trees killed by excessive soil moisture due to poor draining and leaf chlorosis by nutrient deficiencies were found in some orchards. Leaf spotting by herbicide and leaf scorch or blight by excessively high temperature were observed. Leaf blight on young shoots by late frost and bark split on trunks by freeze occurred in several open-field orchards. Flooding and strong wind damages by attack of typhoon 'Nari' were also found during the survey period. Cup-shaped leaves frequently occurred on young shoots in early spring and the incidence of the syndrome tends to increase annually in recent years, which are not etiologically defined until now.
Artificial induction of nitrogen deficiency symptoms (leaf chlorosis) with two root respiratory inhibitors(DNP and Na$_2$S) was studied and regarded to be the same characteristic to red discoloration in rice var. Tongil seedling leaves as well as adult ones. Tongil(IR 667) was shown to be more nitrogen sensitive and have more distinctive appearance of the leaf discoloration than Punggwang(a native Japonica-type varity.) Conclusions were drawn from the present data that so-called red discoloration of Tongil under the natural field conditions is brought about either by insufficient nitrogen supply in soils or certain factors which may limit at any time the root absorption of nitrogen (low temperature, toxic gases or substances, poor drainage, around roots, etc.) in soils even with ample supply of it.
This study was conducted to response the photosynthetic rate and anatomical characters in one year ginseng plant against the light transmission ratio (10%, 20% and 30% in natural light) under shading materials by polyethylene (PE) net. The chlorophyll content and the photosynthesis of ginseng according to the light transmissivity showed decreasing trend as the light transmission ratio increased. At the higher light transmission ratio condition, leaves showed the chlorosis due to the physiological disorder, stems formed hypodermis, and phloem development inside the vascular tissue shrunk remarkably. Also, roots showed the epidermis destruction and less cambium development at the higher light intensity. The root weight also decreased as the light transmission ratio increased.

We surveyed disease outbreak status that has recently become a problem in organic tomatoes and cucurbit in plastic greenhouse that were grown without spraying pesticides during the plastic greenhouse growing season of 2015 to 2019. It was found that the incidence of leaf mold, tomato spotted wilt virus, and tomato chlorosis virus disease was severe in tomato, and disease incidence of powdery mildew and zucchini yellow mosaic virus were severe in Cucurbit. The disease outbreak was found to be faster and more severe in crops grown in pesticide-free cultivation plastic greenhouses than in plastic greenhouses that are cultivated in general using pesticides. In particular, the occurrence of viral diseases mediated by thrips and aphids was found to be severely damaged. Therefore, in order to produce good organic products, it is important to effectively control pests, and in order to minimize the damage caused by disease, sanitation and physical blocking, and comprehensively utilize organic materials or microorganisms to prevent them.
Visible injury symptoms such as necrosis, chlorosis and premature senscence in the leaves of Ginkgo bloba and Pinus thunbergii treated with acid rain of pH 3.2 or below were observed. The epicuticular wax erosions were observed by SEM after exposure to acid rain of pH 2.4 and 3.2 in G. biloba and pH 4.0 below in P. thunbergii. The adaxial epidennal cells and sponge parenchyma cells were compressed, and those were distorted in the leaves of G. biloba treated with simulated acid rain of pH 3.2 or below. However, vascular tissue was intact. With increase of acidity, mesophyll cells were smaller than those of control while intercellular space in mesophyU was increased. In P. thunbergii, sponge parenchyma cells and vascular tissue except epidennis were distorted after exposure to acid rain of pH 2.4. The size change of stomata in foliar injury was not observed, but the stomatal index and size of stomatal aperture in leaves treated with acid rain increased. The stomata of injured leaf were opened in both species examined.amined.
Field and nutrient cultures of American ginseng (Panax quinquefolius L.) were used to establish foliar symptoms related to boron (B) concentration in leaves and soils, and to evaluate radish as a time-saving model system for B nutrition. Application of excess B, 8 kg/ha versus the recommended 1.5 kg/ha, to field plantings of 2-, 3-, and 4-yr-old American ginseng plants just prior to crop emergence caused, within 4 wk after crop emergence, leaf symptoms of chlorosis followed by necrosis starting at the tips and progressing along the margins. The B concentration in leaves of 2-4-yr-old plants receiving 1.5 kg/ha Bwas $30{\mu}g/g$ dry mass compared to $460{\mu}g/g$ dry mass where 8 kg/ha B was applied. Similarly, B concentration in soils receiving the lower B concentration was 1.8 mg/g dry mass and $2.2-2.8{\mu}g/g$ dry mass where the higher B concentration was applied. Application of 8 kg/ha B reduced the dry yield of 3rd-yr roots by 20% from 2745 kg/ha to 2196 kg/ha and 4th-yr roots by 26% from 4130 kg/ha to 3071 kg/ha. Ginseng seedlings and radish were grown under greenhouse conditions in nutrient culture with four B concentrations ranging from 0 mg/L to 10 mg/L. At 5 mg/L and 10 mg/L ginseng and radish developed typical leaf B toxicity symptoms similar to those described above for field-grown plants. Increasing B in the nutrient solution from 0.5 mg/L to 10 mg/L decreased, in a linear fashion, the root and leaf dry mass of ginseng, but not radish. Given the many similarities of ginseng and radish to B utilization, radish might be used as a timesaving model system for the study of B, and other micronutrients, in the slow-growing perennial ginseng.
Gibberella fujikuroi species complex (GFSC) was isolated from rice (Oryza sativa L.) seed samples from ten Asian countries and investigated for incidence of GFSC, molecular characteristics, and pathogenicity. Regardless of geographic origin, GFSC was detected with incidences ranging from 3% to 80%. Four species, Fusarium fujikuroi, F. concentricum, F. proliferatum, and F. verticillioides, were found to show an association with rice seeds, with F. fujikuroi being the predominant species. In phylogenetic analyses of DNA sequences, no relationship was found between species, isolates, and geographic sources of samples. Unidentified fragments of the ${\beta}$-tubulin gene were observed in ten isolates of F. fujikuroi and F. verticillioides. With the exception of three isolates of F. fujikuroi, F. fujikuroi, F. proliferatum, and F. verticillioides were found to have FUM1 (the fumonisin biosynthetic gene); however, FUM1 was not found in isolates of F. concentricum. Results of pathogenicity testing showed that all isolates caused reduced germination of rice seed. In addition, F. fujikuroi and F. concentricum caused typical symptoms of bakanae, leaf elongation and chlorosis, whereas F. proliferatum and F. verticillioides only caused stunting of seedlings. These findings provide insight into the characteristics of GFSC associated with rice seeds and might be helpful in development of strategies for management of bakanae.
