Phylogenetic Characterization of Tomato chlorosis virus Population in Korea: Evidence of Reassortment between Isolates from Different Origins

  • Tomato chlorosis virus (ToCV) is a whitefly-transmitted and phloem-limited crinivirus. In 2013, severe interveinal chlorosis and bronzing on tomato leaves, known symptoms of ToCV infection, were observed in greenhouses in Korea. To identify ToCV infection in symptomatic tomato plants, RT-PCR with ToCV-specific primers was performed on leaf samples collected from 11 tomato cultivating areas where ToCV-like symptoms were observed in 2013 and 2014. About half of samples (45.18%) were confirmed as ToCV-infected, and the complete genome of 10 different isolates were characterized. This is the first report of ToCV occurring in Korea. The phylogenetic relationship and genetic variation among ToCV isolates from Korea and other countries were also analysed. When RNA1 and RNA2 are analysed separately, ToCV isolates were clustered into three groups in phylogenetic trees, and ToCV Korean isolates were confirmed to belong to two groups, which were geographically separated. These results suggested that Korean ToCV isolates originated from two independent origins. However, the RNA1 and RNA2 sequences of the Yeonggwang isolate were confirmed to belong to different groups, which indicated that ToCV RNA1 and RNA2 originated from two different origins and were reassorted in Yeonggwang, which is the intermediate point of two geographically separated groups.

쿠퍼 수화제 살포시 마늘에 미치는 인공산성비의 영향 (Effects of simulated acid rain on garlic in spraying copper hydroxide solution)

  • 본시험은 쿠퍼 77% 수화제 살포에 따른 산성비가 마늘에 미치는 영향을 조사하였다. 인공 산성비만을 마늘에 처리한 결과 경미한 백화 현상이 발생되었고, 잎이 구부러지고 표피세포가 손상을 받았으며 기공이 열려 있었다. 이러한 현상은 인공산성비를 처리한 후 쿠퍼 수화제를 처리할 경우 더욱 심했지만, 마늘수량 감소에 미치는 영향은 없었다. 마늘에 대한 구리 이온의 영향을 조사하기 위하여 황산구리 희석액을 살포한 결과 구리 이온으로 100 ppm 이상의 농도에서 백화증상이 발생되었다. 쿠퍼 수화제 희석액에 산을 첨가하여 pH를 낮추면 구리 이온의 용출량이 수용해도 이상으로 크게 증가됨을 확인할 수 있었다. 인공산성비가 처리된 후 쿠퍼 수화제를 살포한 마늘잎에서 인공산성비 무처리구에 비해 구리이온의 잔류량이 증가되었다. 인공산성비에 의해 피해를 입은 마늘에 동제를 안전하게 사용하기 위해 탄산칼슘을 쿠퍼 수화제와 혼용하여 살포하면 백화증상을 줄일 수 있었다.

식물체에 미치는 연, 아연 ( Pb, Zn ) 의 영향 (Ecological Effects of Zinc and Lead on Plants)

  • This study was carried out to investigate the effects of Zn and Pb concentration on seed germination and plant growth in water and soil culture, and the frequency of chlorosis invegetation and the relationship between plants and soil in the Sambo mine. The inhibition of germination were observed in 1000ppm of Zn, 10ppm of Pb and 5000ppm of Zn + Pb, but germination was more stimulated in 10ppm of Zn than control. The symptoms of chlorosis and abnormality were occurred in plant leaves grown to the soils treated with more than 1000ppm of Pb. Reasons of chlorosis were considered as an antagonistic effect of other metals towards uptake of iron by the plant in Zn treatment. The contents of Zn and Pb in fruits were lower than those of leaves, and that was remarked in case of Pb. With increasing rate of Zn and Pb treatment, chemical components of soils in pot culture were accompanied by slight decrease in pH, total nitorgen and exchangeable K. Chlorotic individuals of 10 species were shown in the areas of the Sambo mine. Chlorotic symptoms were especially extensive and severe in Sophora angustifolia, Populus alba, Spiraea prunifolia, Amorpha fruticota, Lespedeza bicolor and Salix dependens. Plants in the investigated areas grew in soils containing Zn of 311ppm and Pb of 151ppm on an average, and accumulated Zn of 2084ppm and Pb of 49ppm.

Efficient Transmission and Propagation of Tomato Chlorosis Virus by Simple Single-Leaflet Grafting

  • Tomato chlorosis virus (ToCV), a member of the genus Crinivirus, has caused an epidemic disease in tomato worldwide. ToCV is phloem-limited and transmitted by whiteflies in a semi-persistent manner, but not by mechanical inoculation. Experimental propagation of ToCV has been performed primarily by using whitefly-mediated inoculation. To develop a simple and convenient method for transmission of ToCV, we investigated grafting single-leaflets from tomato plants infected with ToCV to recipient tomato seedlings. Forty-one of 46 tomato seedlings tested were grafted successfully with single-leaflets infected with ToCV. Among them, 36 seedlings (87.8%) were systemically infected with ToCV and developed typical symptoms. Our results demonstrated that single-leaflet grafting could provide a sufficient amount of inoculum for the transmission of ToCV to the grafted seedlings.

배나무 엽 황화증상 발생 과원의 토양 및 엽 특성 분석 (Analysis of Soil and Leaf Characteristics of Pear Orchards with Lime-Induced Chlorosis Leaves)

  • 본 연구에서는 황화증상이 발생한 전국 4개 지역의 배 과원에서 토양 특성과 잎의 특성을 분석하여 황화증상의 원인을 구명하고자 하였다. 황화증상이 발생한 잎의 색은 육안 또는 SPAD와 Hunter 값을 통해 진단할 수 있었다. 황화증상이 발생한 과원의 토양은 정상 과원에 비해 토양의 pH가 유의하게 높았다. 유효토심은 황화증상과 큰 관계가 없었으나, 토양에 석회 등이 과다 시비되는 조건과 결합되어 식물체의 철 흡수를 저해할 가능성이 있다. 황화증상이 발생한 잎에서는 정상엽에 비해 철과 칼슘 함량이 유의하게 낮았고, 마그네슘 함량이 유의하게 높았다. 따라서 고온다습한 6-7월경 2차 신초 발달이 일어나는 시기에 집중적으로 황화증상이 발생하는 것은 철과 칼슘의 흡수 저해와 밀접한 관계가 있는 것으로 판단된다. 이를 해결하기 위해서는 석회의 과다 시비를 피하고, 철 성분을 함유한 비료를 엽면시비하거나 수관부 토양에 관주하는 방식이 적합하다.

  • From 1975 to 1981, the survey was carried out to find out radioecological effects of uranium ore deposits on natural vegetation in Koisan, Korea. The symptoms of spotty and mosaic chlorosis, and necrosis were observed in flowering plants in the areas of uranium ore deposits at Deok-Peung-Ri A, B, and C in Koisan. Although 13 species were found to be chlorosis and necrosis, foliages observed are small and very rare. The features of these symptoms closely resemble those occured by the effects of heavy meetals. The amount of transparent radiation throughout the depth of soils from uranium radiation sources decreases exponentially. The mean contents in leaves of spotty and mosaik chlorotic plants, and soils were 1.36~1.53 and 5.3~7.4 ppm, respectively.

  • During the period of 1975-76, a survey was carried out to find out zinc indicators in the natural vegetation in Korea. The symptoms fo chlorosis were observed in flowering plants in the areas of zinc outcrop of Wolgok-A, Seokgok-9, and Sowolgok. Although 28 species were found to be chlorotic, the total quantity of chlorotic foliage observed was small. Reasons for chlorosis in the areas of zinc are deposits is considered as effects of zinc, lead, copper and calcium ions. Sedum sp. and Dianthus sinensis were confined to soil containing more than exchangeable zinc of 30 ppm and to accumulation in the plants contained at least 1,300-14,000 ppm of zinc. Therefore Sedum sp. and Dianthus sinensis might be used as zinc indicators in Korea.

Little Leaf and Yellowing Symptoms on Castanea crenata are Associated with Phytoplasma in Korea

  • For unknown reasons, a few trees in a private chestnut orchard in Icheon si, Gyunggi-do suffered leaf chlorosis and growth decline. Based on symptoms, phytoplasma was a probable cause. Leaf samples were collected from two symptomatic and non-symptomatic trees in the orchard for phytoplasma detection. An amplicon of about 1.2 bp size was obtained from both symptomatic trees by PCR with the universal 16S rDNA primers. Sequences of these amplicons were found to have 99% nucleotide sequence identity to the corresponding genomic region of 16SrIII (X-disease group). More than 100 phytoplasma isolates, such as Candidatus phytoplasma pruni, Milkweed yellows phytoplasma, Goldenrod yellows phytoplasma, Tsuwabuki witches'-broom phytoplasma, Candidatus Phytoplasma trifolii, etc. were involved in the list. Phylogenetic analysis revealed that the sequence obtained in this study closely clustered with Candidatus phytoplasma groups. While one of the amplicons shared 91% identity with the Candidatus phytoplasma castaneae, the other shared only 47%. It needs further analysis and investigation to determine the exact taxonomy. Meanwhile, based on the analysis of the sequences, chlorosis, and small leaves were associated with phytoplasma.

Chlorosis of Ogura-CMS Brassica rapa is due to down-regulation of genes for chloroplast proteins

  • Cytoplasmic male sterility (CMS) is a maternally inherited trait leading to loss of the ability to produce fertile pollen and is extensively used in hybrid crop breeding. Ogura-CMS was originally generated by insertion of orf138 upstream of atp8 in the radish mitochondrial genome and transferred to Brassica crops for hybrid breeding. Gene expression changes by dysfunctional mitochondria in Ogura-CMS result in pollen developmental defects, but little is known about gene expression patterns in vegetative tissue. To examine the interaction between nuclear and organellar regulation of gene expression, microarray and subsequent gene expression experiments were conducted with leaves of $F_1$ hybrid Chinese cabbage derived from self-incompatible (SI) or Ogura-CMS parents (Brassica rapa ssp. pekinensis). Out of 24,000 genes deposited on a KBGP24K microarray, 66 genes were up-regulated and 26 genes were down-regulated by over 2.5 fold in the CMS leaves. Up-regulated genes included stress-response genes and mitochondrial protein genes, while genes for ascorbic acid biosynthesis and thylakoid proteins were down-regulated. Most of the major component genes for light reactions of photosynthesis were highly expressed in leaves of both SI and CMS plants, but most of the corresponding proteins were found to be greatly reduced in leaves of CMS plants, indicating posttranscriptional regulation. Reduction in thylakoid proteins and chlorophylls led to reduction in photosynthetic efficiency and chlorosis of Ogura-CMS at low temperatures. This research provides a foundation for studying chloroplast function regulated by mitochondrial signal and for using organelle genome introgression in molecular breeding.

Ultrastructural Changes During Programmed Cell Death of Tobacco Leaf Tissues Infected with Tobacco mosaic virus

  • Tobacco (Nicotiana tabacum cvs.Xanthi-nc and NC 82) plants infected with Tobacco mosaic virus (TMV) were examined ultrastructurally. Local lesions produced by TMV were sunken and withered. The plants were subjected to temperature shift (TS), a method to produce programmed cell death (PCD), by placing the infected plants initially at high temperature (35$^{\circ}C$) for 2 days and then shifting them to greenhouse temperature (22-27$^{\circ}C$). As a result, expanded lesions around the original necrotic lesions were produced. The expanded area initially had no symptoms, but it withered and became necrotic 15 h after TS. No ultrastructural changes related to PCD were noted at 0 h after TS in Xanthi-nc tobacco tissues as well as in healthy and susceptible tobacco tissues infected with TMV, At 6 h after TS, chloroplasts were convoluted and cytoplasm began to be depleted; however no necrotic cells were found. At 17 h after TS, ground cytoplasm of affected cells was completely depleted and chloroplasts were stacked together with bent cell wall or dispersed in the intracellular space. Necrotic cells were also observed, containing virus particles in the necrotic cytoplasm. There were initially two types of symptoms in the expanded lesions: chlorosis and non-chlorosis (green). Abundant TMV particles and X-bodies were only found in the chlorotic tissue areas. These results suggest that PCD by TMV infection may start with the wilting of cells and tissues before necrotic lesion formation.
