Changes in Radical Scavenging Activity and α-Glucosidase Inhibitory Activity of Dried Daraesoon (Shoot of Hardy Kiwi, Actinidia arguta) during Cooking (건조 다래순의 조리 중 라디칼 소거 활성과 알파글루코시데이스 억제 활성의 변화)

  • This study evaluated the in vitro radical scavenging and ${\alpha}$-glucosidase inhibitory activities of dried daraesoon (shoot of hardy kiwi) during cooking involving rehydration and subsequent heating at $180^{\circ}C$ with or without perilla oil. Pigments and antioxidants were quantified by HPLC and spectrophotometry. Unlike the tocopherol content, the polyphenol, flavonoid, chlorophyll, and carotenoid contents as well as the DPPH radical scavenging and ${\alpha}$-glucosidase inhibitory activities of daraesoon extract were significantly decreased by rehydration (p<0.05). Heating the rehydrated daraesoon for 10 or 20 min increased its radical scavenging activity irrespective of perilla oil addition, whereas the ${\alpha}$-glucosidase inhibitory activity increased significantly only after heating with perilla oil (p<0.05). During cooking, changes in both activities showed a similar pattern to that showed by polyphenol content changes. These results suggest that the health functionality of daraesoon can be enhanced by an appropriate cooking process that retains polyphenols.

Algae and Nutrient Removal by Vegetated Artificial Floating Island (인공식물섬에 의한 조류(Algae)및 영양염류의 제거)

  • We investigated the effect on the removal of BOD, SS, TN and TP and algal growth inhibition of Vegetated Artificial Floating Island (VAFI), by examining microorganism activity and nutrient uptake in the batch test of various conditions: (1) Blank (Control group), (2) VAFI of $0.25m^2$, (3) AFI of $0.25m^2$ which has no vegetation, (4) buoyant plate of $0.25m^2$, (5) buoyant plate of $0.25m^2$ with linear media. The proportion of BOD removal in the VAFI, AFI, buoyant plate and buoyant plate with media were 82.7, 80.8, 45.2% and 59.6% respectively. TN removal in the VAFI, AFI and buoyant plate with media were 51.2, 31.7% and 25.1% respectively. TP removal in the VAFI, AFI, buoyant plate and buoyant plate with media were 23.3, 16.7, 10.0% and 13.3% respectively. Chlorophyll-${\alpha}$ removal in the VAFI was 97.9%. The factors of chlorophyll-${\alpha}$ removal in the VAFI accounted for the shading effect of 35.1%, microorganisms activity of 61%, and plant root of 1.8%.

Cobalt complex structure of the sirohydrochlorin chelatase SirB from Bacillus subtilis subsp. spizizenii (Bacillus subtilis subsp. spizizenii의 sirohydrochlorin chelatase SirB의 코발트 복합체 구조)

  • Chelatase catalyzes the insertion of divalent metal into tetrapyrrole and plays a key role in the biosynthesis of metallated tetrapyrroles, such as cobalamin, siroheme, heme, and chlorophyll. SirB is a sirohydrochlorin (SHC) chelatase that generates cobalt-SHC or iron-SHC by inserting cobalt or iron into the center of sirohydrochlorin tetrapyrrole. To provide structural insights into the metal-binding and SHC-recognition mechanisms of SirB, we determined the crystal structure of SirB from Bacillus subtilis subsp. spizizenii (bssSirB) in complex with cobalt ions. bssSirB forms a monomeric ${\alpha}/{\beta}$ structure that consists of two domains, an N-terminal domain (NTD) and a C-terminal domain (CTD). The NTD and CTD of bssSirB adopt similar structures with a four-stranded ${\beta}-sheet$ that is decorated by ${\alpha}-helices$. bssSirB presents a highly conserved cavity that is generated between the NTD and CTD and interacts with a cobalt ion on top of the cavity using two histidine residues of the NTD. Moreover, our comparative structural analysis suggests that bssSirB would accommodate an SHC molecule into the interdomain cavity. Based on these structural findings, we propose that the cavity of bssSirB functions as the active site where cobalt insertion into SHC occurs.

Ecological studies on Togyo Reservoir in Chulwon, Korea. V. Seasonal Changes of Size-Fractionated Standing Crops and Chlorophyll a of Phytoplankton in Kyungan Stream of Paldang River-Reservoir Systems and Togyo Reservoir, Korea (철원북방 DMZ내의 중영양호 토교저수지의 생태학적 연구 V. 경안천(팔당호)과 토교저수지에서 식물플랑크톤의 크기별 현존량과 Chlorophyll $\alpha$의 계절 변동)

  • With physico-chemical environmental factors such as temperature, DO, pH, conductivity and nutrients, size fractionated of phytoplankton standing crops and chlorophyll a concentrations $(>8\mu{m},\;3-8\mu{m},\;<3\mu{m})$ and their relationships were investigated at each station of Kyungan Stream and Togyo Reservoir from April to November in 1997. The two sampling sites showed different nutrient status: Kyungan Stream was eutrophic, while Togyo Reservoir was mesotrophic. Large sizes of phytoplankton and chl. a were higher in Kyungan Stream, opposite to those of Togyo Reservoir; Standing crops of phytoplankton $(>8\mu{m)$ and chl. a $(3-8\mu{m)}$ were high in Kyung-an Stream, while phytoplankton $(3-8\mu{m)}$and chi. a $(<3\mu{m)$ were abundant in Togyo Reservoir. These results imply that phytoplankton community in the highly eutrophicated water mainly comprised the large filamentous and/or colonial algae, such as Microcystis spp. and Aphanizomenon flos-aquae, which easily enriched by nutrients loading.

Comparison of Four Different Ordination Methods for Patterning Water Quality of Agricultural Reservoirs

  • We patterned water quality of agricultural reservoirs according to the differences of six physico-chemical environmental factors (TN, TP, DO, BOD, COD, and SS) using four different ordination methods: Principal Components Analysis (PCA), Detrended Correspondence Analysis (DCA), Nonmetric Multidimensional Scaling (NMS), and Isometric Feature Mapping (Isomap). The data set was obtained from the water quality monitoring networks operated by the Ministry of Agriculture and Forestry and the Ministry of Environments. Chlorophyll-${\alpha}$ displayed the highest correlation with COD, followed by TP, BOD, SS, and TN (p<0.01), while negatively correlated with altitude and bank height of the reservoirs (p<0.01). Although four different ordination methods similarly patterned the reservoirs according to the gradient of nutrient concentration, PCA and NMS appeared to be the most efficient methods to pattern water quality of reservoirs based on the explanation power. Considering variable scores in the ordination map, the concentration of nutrients was positively correlated with Chl-${\alpha}$, while negatively correlated with altitude and bank height. These ordination methods may help to pattern agricultural reservoirs according to their water quality characteristics.

Spatio-temporal Variation of Intertidal Microphytobenthos in the Nakdong Estuary

  • The species composition and the biomass of intertidal microphytobenthos (MPB) were investigated at four sites in the Nakdong Estuary from Feb. to Dec. 2006. The chlorophyll (chl) ${\alpha}$ concentration showed a positive correlation with MPB abundance, and depth profiles showed similar patterns: high at the surface, rapidly decreasing within 4 cm from the surface, and slowly decreasing thereafter. A MANOVA analysis revealed that the chl ${\alpha}$ concentration varied significantly not only with depth, month, and site, but also with combinations of these factors. Among the four investigated sites, site D (Baekhabdeung) showed a seasonal biomass variation trend distinct from those of the other sites: higher in summer, decreased in autumn and with sustained low values until winter. As indicated, the other sites contrastingly showed low biomasses in summer, after which the biomasses continuously increased, with some variation among the sites. A cluster analysis of species composition indicated that sites near to each other and with similar sediment structures had closer similarities in the same seasons. The species of genus Amphora and Navicula were dominant at the four sites throughout the study period.

Bleaching of cold-pressed rapeseed oil using activated clay (산성백토를 이용한 저온압착 유채유의 탈색 평가)

  • Acid-activated clays (SUPER-DC, DC-A3, and P1) are used for the bleaching of cold-pressed rapeseed oil. In this study we tested the bleaching performance of cold-pressed rapeseed oil according to the different reaction time (20, 40, 60, 80 min) and temperature (40, 80, $120^{\circ}C$). Oil color (lightness, redness, yellowness), pigments (chlorophyll A and carotenoid content) and quality properties (fatty acid composition, tocopherols (${\alpha}$, ${\beta}$, ${\gamma}$, ${\delta}$), and plant sterols (${\beta}$-sitosterol, campesterol, stigmasterol) content) were analyzed. The results showed that bleaching of cold-pressed rapeseed oil with 2% acid-activated clays at $40^{\circ}C$ for 20 min, brightness (L) increased, but redness (a) and yellowness (b) decreased. Bleaching of cold-pressed rapeseed oil with 2% DC-SUPER at $40^{\circ}C$ removed chlorophyll A and carotenoids pigments significantly. In addition, about 50% of total tocopherol content in cold-pressed rapeseed oil was reduced by bleaching. Originally total tocopherol content was 46.62mg/100g in cold-pressed rapeseed oil. But after bleaching, total tocopherol content was 12.67mg/100g (20 min bleaching), 15.31mg/100g (40 min bleaching), and 13.56mg/100g (60 min bleaching). However plant sterols content in cold-pressed rapeseed oil remained unchanged by bleaching. Overall, acid-activated clays were useful for the bleaching of pigmented rapeseed oil.

Short-Term Nutrient Enrichment Bioassays and Nutrient Limitation in Daechung Reservoir (대청호에서의 단기 영양염 첨가 실험 및 제한 영양염류 분석)

  • In situ experiments of Nutrient Enrichment Bioassays (NEBs) were conducted in the field along with in the laboratory to determine which nutrient limited phytoplankton growth as a indicator of primary productivity. For the NEBs, the water was sampled using a polyethylene-lined container and dispensed into 6 L water tank in the laboratory. The control (C, no nutrient spike) and six treatments of phosphorus (P), 2-fold phosphorus (2P), 4-fold phosphorus (4P), nitrate nitrogen ($NO_3$-N), 2-fold nitrate nitrogen ($2NO_3$-N), and phosphorus and nitrate nitrogen (P+$NO_3$-N) were set up in the lacustrine zone near the dam site, Daechung Reservoir in September, 2009 and analyzed the diel changes of total nitrogen (TN), total phosphorus (TP), and chlorophyll-$\alpha$ (Chl-$\alpha$) in the cubitainers. The short-term NEBs showed that algal response in the treatments spiked phosphorus (P, 2P, and 4P) were significantly (p < 0.05) greater than the response in the control (C), and nitrogen-spike. Also, the response in 4P-treatment was greater than those in the P- and 2P-treatments. In contrast, there was no significant differences (p > 0.20) between the $NO_3$-N and $2NO_3$-N treatment. The outcomes of the NEBs suggest that phosphorus limited the phytoplankton growth and nitrogen was not limited in this system. Furthermore, in the N + P treatments, the response was minimum, compared to all other treatments and the control, indicating that even if the system is evidently P-limited system, when added the nitrogen, the response showed the inhibition. Also, > 95% of observed long-term TN:TP ratios in the ambient water showed > 17, which is the criteria of P-limitation, supporting the P-limitation in the system. Overall, these results suggest that phytoplankton biomass near the dam is a direct linear function of P-loading near the watershed, if the phosphorus pool is mainly dissolved fraction.

Effects of Seed Soaking of Kinetin with Zinc Treatment on ${\alpha}-amylase$ Activity and Free Proline Content during Germination of Rice(Oryza sative L.) (아연처리 볍씨의 발아중 ${\alpha}-Amylase$ 활성과 유리 Proline 함량 변화에 미치는 Kinetin 침종의 영향)

  • The study was carried out to elucidate the changes of ${\alpha}-amylase$ activity and free proline content for zinc toxicity in two rice cultivars(Ilpumbyeo and Namchunbyeo) during germination and early growth stages. Plant height in all kinetin treatments was promoted but zinc 120ppm treatment was decreased. Soaking treatment of kinetin $10^{-3}M$ increased germination rate of both cultivars, Ilpumbyeo and Namchunbyeo by 95% and 96% as compared with zinc 120ppm. Chlorophyll content of Ilpumbyeo was higher than that of Namchunbyeo. Activity of ${\alpha}-amylase$ in kinetin $10^{-3}M$ of both rice cultivars was most highest in the 3days after treatment of zinc 120ppm. Free proline content in all rice cultivars of zinc 120ppm treatment was sharply increased at the 3days after treatment of zinc 120ppm. As a result, the effects of kinetin treatment were recognized to promote the plant height and germination rate under zinc toxicity(120ppm) during rice seed germination and early growth stages.

Assessment of Salinity-Induced Antioxidative Defense System of Diazotrophic Cyanobacterium Nostoc muscorum

  • The present study examined the salinity-induced oxidative damage and differential response of enzymatic and nonenzymatic antioxidants of Nostoc muscorum. As compared with carotenoid content that showed induction, the chlorophyll and phycocyanin contents were inhibited after salt stress. Acceleration of lipid peroxidation and peroxide production suggested the onset of oxidative damage. The activities of all studied enzymatic antioxidants were significantly increased by salt stress, with maximum induction occuring with superoxide dismutase (154.8% at 200 mM NaCl treatment). Interestingly, under severe stress condition (250 mM NaCl), ascorbate peroxidase seemed to be more crucial than catalase for peroxide scavenging. Among the studied nonenzymatic antioxidants, ${\alpha}$-tocopherol was induced maximally (56.0%); however, ascorbate and reduced glutathione were increased by only 8.9% after 250 mM NaCl treatment as compared with control cells. Therefore, salinity was found to induce the antioxidative defense system of N. muscorum.
