Induction of Apoptotic Cell Death in Human Jurkat T Cells by a Chlorophyll Derivative (Cp-D) Isolated from Actinidia arguta Planchon

  • The chloroform and methanol (2;1, v/v) extract from an edible plant, Actinidia arguta Planchon, appeared to possess antitumor activity against human leukemias Jurkat T and U937 cells through inducing apoptosis. The substance in the solvent extract was purified by silica gel column chromatography, preparative TLC, and Sephadex LH-20 column chromatography. Characteristics of the substance analyzed by UV scanning analysis, $^1H$ and $^{13}C$ NMR spectra suggested that the substance belongs to the chlorophyll derivatives-like group. The $IC_{50}$ value of the chlorophyll derivative (Cp-D) determined by MTT assay was $15\mu\textrm{g}/ml$ for Jurkat, $10\mu\textrm{g}/ml$ for U937, and $11.4\mu\textrm{g}/ml$ for HL-60m and was more toxic to these leukemias than to solid tumors or normal fibroblast. In order to elucidate cellular mechanisms underlying the cytotoxicity, the effect of the Cp-D on Jurkat T cells was investigated. When cells were treated with the Cp-D at a concentration of $15\mu\textrm{g}/ml$, [3H]thymidine incorporation declined rapidly and wa undetectable in 1h. However, no significant changes were made in the cell cycle distribution of the cells by 24h. The sub-Gl peak representing apoptotic cells began to be detectable in 36h, at which time apoptotic DNA fragmentation was also detected on agarose gel electrophoresis, demonstrating that the cytotoxic effect of the Cp-D is attributable to the induced apoptosis. Under the same conditions, although the protein level of cyclin-dependent kinases such as cdc4, csk6, cdk2, and cdc2 was not significantly changed until 24h, the kinase activity of all c안 rapidly declined and reached a minimum level within 1-6h and then recovered to the initial level by 12h and sustained until 24h. These results suggest that inactivation of cdks at an inappropriate time during the cell cycle progression in jurkat T cells following a treatment with the Cp-D leads to induction of apoptotic cell death.

Changes of Chlorophyll and their Derivative Contents during Storage of Chinese Cabbage, Leafy Radish and Leaf Mustard Kimchi (배추, 열무 및 갓김치 저장 중의 Chlorophyll 및 그 유도체의 함량변화)

  • Three kinds of kimchi using Chinese cabbage, leafy radish and mustard leaf were prepared by conventional method and stored at 5$^{\circ}C$ or 2$0^{\circ}C$ for 13 dyas. During storage at both temperatures, changes of the amounts of salt and ascorbic acid, pH and total acidity were determined, and the relationship of the decomposition of chlorophylls with the production of their derivaties was studied. At both storage temperatures, salt concentration of Chinese cabbage kimchi(3.7%), leafy radish kimchi(3.6%), mustard leaf kimchi(3.5%) was relatively constant during the entire storage period. However, pH and total acidity wre fluctuating with the remarkable changes during 3 days of storage. Ascorbic acid content was slowly decreased during the storage period and the decompositin rate of ascorbic and was greater at 2$0^{\circ}C$ than 5$^{\circ}C$. Among the kinds of kimchi tested, mustard leaf kimchi with the slow decomposition rate of ascorbic acid contained relatively high ascorbic acid content, while leafy radish kimchi contained the lowest content. At both storage temperatures, the production of pheophytin and pheophorbide from decomposition of chlorophyll was least in mustard leaf kimchi, but similar production rates in leafy radish and Chinese cabbage kimchi were observed.

  • This research determined the spectral reflectance characteristics and vegetation indices when intermixed with field crops and soil. Ground-level spectral reflectance were collected in the field experiment containing plots of soybean and other seven crops. The first and second derivative of reflectance spectra showed several peaks that were dependent in different degrees on leaf age and chlorophyll concentration in the crop leaves. This study evaluated a number of spectral indices for estimating chlorophyll concentrations at the leaf scale difference, using samples from field crops at various stages of senescence. Five vegetation indices were evaluated which had advantages over previous techniques. Experimental data recorded on field crops during the growing season are in good agreement with previous theoretical results.

  • Lincomycin, an inhibitor of plastid protein synthesis, was found to block the synthesis of apoprotein P700 with a molecular mass of 72 kDa and the assembly of the Chl a-protein of PS I. Synthesis of the polypeptides of 48, 43.5, and 32 kDa of the PS II complex is also suppressed. This process is accompanied by the disappearance of the PS Two reaction center Chl a at 683 nm, and of the PS One reaction center Chl a at 690, 696, and 705 nm on the fourth derivative of the absorption spectra at 77K. Lincomycin does not affect the synthesis of LHC subunits. It increases the content of the two main Chl forms of LHC at 648 nm (Chl b) and 676 nm (Chl a). The low-temperature fluorescence ratio F736/F685 is also increased. However, the effect of cycloheximide (an inhibitor of cytoplasmic protein synthesis) leads to the reduction of polypeptides of the light-harvesting Chl a/b-protein complex in the range of 29.5-22 kDa. Under these conditions, the relative amount of Chl b and the F736/ F685 fluorescence ratio decrease significantly. This is obviously the result of blocking the LHC I and LHC II synthesis. At the same time rifampicin and actinomycin D (inhibitors which block transcription in chloroplast and nuclear genome, respectively) inessentially affect the characteristics of these complexes.

Early Alterations of Chlorophyll Fluorescence by Light-Chilling in Cucumber (Cucumis sativus) Leaves and Their Usage as Stress Indicators (오이 잎에서 저온 광저해에 의한 형광유도과정의 초기 변이와 스트레스 지표)

  • To investigate the early symptoms of light-chilling, alterations of chlorophyll fluorescence transients were monitored in cucumber (Cucumis sativus L. cv. Ilmichungjang) leaves. During 24 h chilling, decreases in (Fv)m/Fm, qE and qQ, and an increase in Fo were observed. The chilling effects were not recovered at room temperature, and a significant increase in Fo was observed during the recovery period. After 6 h chilling, ‘dip’(D) level of the transients became obscure, and the negative slope after ‘peak’(P) disappeared. The first derivative (dFv/dt) of the fast fluorescence rise curve was used to obtain more accurate information about the changes in the transients. The maximal rate of the fluorescence increase in the D-p rise curve (Fr) has been the most frequently used chilling stress indicator. However, a correct value of Fr could not be measured when the D level became obscure. This problem was overcome by introducing a new indicator, HFr (dFv/dt at Fv = 1/2 (Fv)m), and HFr gave very similar values to Fr. To monitor the changes in curvature around D level, another new parameter, ${\Delta}S$(D-Fr), was also introduced. These three parameters decreased very sensitively during light-chilling. In addition, increases in these parameters were observed during the first 2 h chilling, but this increase in Fr was also observed in pea leaf discs dark-chilled for 15 min, suggesting that this very early change is a common response to chilling in both pea and cucumber leaves. Quenching coefficients were also very sensitive to chilling, especially qE. Discussion on the usage of these parameters as chilling stress indicators is given in the text.

The Near-Infrared Imaging Spectroscopy to Visualize the Distribution of Sugar Content in the Flesh of a Melon

  • To improve the accuracy of sweetness sensor in automated sorting operations, it is necessary to clarify unevenness of the sugar content distribution within fruits. And it is expected that the technique to evaluate the content distribution in fruits contribute to the development of the near-infrared (NIR) imaging spectroscopy. Sugiyama (1999) had succeeded to visualize the distribution of the sugar content on the surface of a half-cut green fresh melon. However, this method cannot be applied to red flesh melons because it depends on information of the absorption band of chlorophyll (676 nm), which is affected by the color of the fresh. The objective of this study was to develop the universal visualization method depends on the absorption band of sugar, which can be applied to various kinds of melons and other fruits. The relationship between the sugar contents and absorption spectra of both green and red fresh melons were investigated by using a NIR spectrometer to determine the absorption band of sugar. The combination of 2$\^$nd/ derivative absorbances at 902 nm and 874 nm was highly correlated with the sugar contents. The wavelength of 902 nm is attributed to the absorption band of sugar. A cooled charge-coupled device (CCD) imaging camera which has 16 bit (65536 steps) A/D resolution was equipped with rotating band-pass filter wheel and used to capture the spectral absorption images of the flesh of a vertically half-cut red fresh melon. The advantage of the high A/D resolution in this research is that each pixel of the CCD is expected to function as a detector of the NIR spectrometer for quantitative analysis. Images at 846 nm, 874 nm, 902 nm and 930 nm were acquired using this CCD camera. Then the 2$\^$nd/ derivative absorbances at 902 nm and 874 nm at each pixel were calculated using these four images. On the other hand, parts of the same melon were extracted for capturing the images and squeezed for the measurement of sugar content. Then the calibration curve between the combination of 2$\^$nd/ derivative absorbances at 902 nm and 874 nm and sugar content was developed. The calibration method based on NIR spectroscopy techniques was applied to each pixel of the images to convert the 2$\^$nd/ derivative absorbances into the Brix sugar content. Mapping the sugar content value of each pixel with linear color scale, the distribution of the sugar content was visualized. As a result of the visualization, it was quantitatively confirmed that the Brix sugar contents are low at the near of the skin and become higher towards the seeds. This result suggests that the visualization technique by the NIR imaging spectroscopy could become a new useful method fer quality evaluation of melons.

Studies on Photosensitized Oxidation in the Lipids of Irish moss, Laver and Oyster (진두발, 김 및 굴의 지질에 있어서 광증감 산화에 관한 연구)

  • In order to investigate the influence of photosensitized oxidation in the sun-dried irish moss (Chondrus Ocellatus), laver (Porphyra Yezoensis) and ultra violet irradiated oyster (Crassostrea gigas) the oxidation of lipid and isomers of hydroperoxides were analyzed by gas chromatography-mass spectrometry. The lipid contents of oyster, irish moss and layer were $2.7\%,\;0.1\%,\;0.1\%$ of respectively. Peroxide value, 56,7 meq/kg in the raw oyster was increased of 100.9 meq/kg by the U.V, irradiation for 4 hours. Also the peroxide values of the irish moss and laver were increased by the sun-drying. In the identification of hydroperoxides isomers by trimethylsily (TMS) derivative of photo-oxidized lipid from oyster, irish moss and laver, the proportions of positional isomer, 9-OOH and 13-OOH were dominant than those 10-OOH and 12-OOH.

Analysis of Spectral Reflectance Characteristics Using Hyperspectral Sensor at Diverse Phenological Stages of Soybeans

  • South Korea is pushing for the advancement of crop production technology to achieve food self-sufficiency and meet the demand for safe food. A medium-sized satellite for agriculture is being launched in 2023 with the aim of collecting and providing information on agriculture, not only in Korea but also in neighboring countries. The satellite is to be equipped with various sensors, though reference data for ground information are lacking. Hyperspectral remote sensing combined with 1st derivative is an efficient tool for the identification of agricultural crops. In our study, we develop a system for hyperspectral analysis of the ground-based reflectance spectrum, which is monitored seven times during the cultivation period of three soybean crops using a PSR-2500 hyperspectral sensor. In the reflection spectrum of soybean canopy, wavelength variations correspond with stages of soybean growths. The spectral reflection characteristics of soybeans can be divided according to growth into the vegetative (V)stage and the reproductive (R)stage. As a result of the first derivative analysis of the spectral reflection characteristics, it is possible to identify the characteristics of each wavelength band. Using our developed monitoring system, we observed that the near-infrared (NIR) variation was largest during the vegetative (V1-V3) stage, followed by a similar variation pattern in the order of red-edge and visible. In the reproductive stage (R1-R8), the effect of the shape and color of the soybean leaf was reflected, and the pattern is different from that in the vegetative (V) stage. At the R1 to R6 stages, the variation in NIR was the largest, and red-edge and green showed similar variation patterns, but red showed little change. In particular, the reflectance characteristics of the R1 stage provides information that could help us distinguish between the three varieties of soybean that were studied. In the R7-R8 stage, close to the harvest period, the red-edge and NIR variation patterns and the visible variation patterns changed. These results are interpreted as a result of the large effects of pigments such as chlorophyll for each of the three soybean varieties, as well as from the formation and color of the leaf and stem. The results obtained in this study provide useful information that helps us to determine the wavelength width and range of the optimal band for monitoring and acquiring vegetation information on crops using satellites and unmanned aerial vehicles (UAVs)

AN EXPERIMENTAL STUDY ON THE CHEMOPREVENTIVE EFFECT OF CHLOROPHYLLIN IN HAMSTER CHEEK POUCH TUMOR INDUCED BY 7, 12-DIMETHYLBENZ[A]ANTHRACENE (7, 12-dimethylbenz[a]anthracene(DMBA)로 유발된 햄스터 협낭암에서 chlorophylln의 암예방효과에 관한 실험적 연구)

  • Carcinogenesis is a multi-stage process that generally consists of at least three steps; initiation, promotion, and progression. If one of these carcinogenic steps were suppressed or delayed, the cancer could be prevented. Cancer chemoprevention is defined to be inhibition or reversal of the carcinogenic process by the specific chemical agents and is a novel approach to cancer management alternative to conventional chemotherapy. Chlorophylln(CHL), a water-soluble derivative of chlorophyll, containing sodium and copper, has been known to be strong antimutagen in several test systems, but its mechanism of antimutagenic action is unknown. In the present experiment, the possibility of CHL as chemopreventive drugs on 7,12-dimethylbenz[a]anthracene(DMBA)-induced hamster buccal pouch carcinogenesis was investigated by mutagenicity test, carcinogenicity test, and frequency or spectrum of H-ras mutations in the both of DMBA-induced and chlorophylln-pretreated-DMBA induced tumor by polymerase chain reaction and non-isotopic restriction fragment length polymorphism. The treatment of CHL reduced the yields and multiplicity of the 0.5% DMBA-induced tumor, 86% to 62.5% and $3.7{\pm}0.6$ to $1.4{\pm}0.3$, respectively. The occurrence of histidine revertant by $20{\mu}mole$ DMBA was inhibited 25.6 to 81.7% by 1 to $5{\mu}M$ CHL in a dose-dependent manner. The mutation rates of H-ras gene in DMBA-induced and CHL-pretreated-DMBA induced tumor were 96%, 94% of which the most mutations were in codon 12/13. These results suggest that CHL inhibits the carcinogenic action of DMBA by the formation of complex between CHL and DMBA or the inhibition of the activation of DMBA in vivo. But CHL did not affect the mutation rates or its spectrum in already formed tumor.
