Effect of Solidago altissima L. Extract on Forage Crop Germination

  • There are 28 families and 166 species of exotic weeds on agricultural land and among these, 23 families and 80 species of exotic weeds occur on pastures. Among them, the Solidago altissima is a perennial weed belonging to the asteraceae family and it is an exotic weed that spreads to the surrounding area using methods such as high seed production, vegetative propagation using underground rhizomes and allelochemical. Accordingly, in 2009, the Ministry of Environment designated it as an ecosystem-disrupting species. This study was conducted to obtain basic data about the effects of S.altissima derived allelochemicals on forage crops. The root of S.altissima was separated, dried in the shade and then pulverized to prepare an root powder. Powder was repeatedly extracted with methanol for 3 days and concentrated under reduced pressure to obtain an root methanol extract. Dissolve the extract in distilled water, dispense it in a separate-funnel and proceed with liquid-liquid extraction by adding equal amounts of n-haxane (Hex), chloroform (CHCI3), ethyl acetate (EtoAC), and butanol (BuOH) in order of increasing polarity. A seed-bioassay was performed using fractions for each solvent, followed by separation and purification by silica gel column chromatography. As a result of the fraction germination test for each solvent, the IC50 values using the fresh weight of each fraction were 898.3 mg L-1, 676.3 mg L-1, 1160 mg L-1 and 1360 mg L-1. CA, CB, and CC fractions were obtained through primary silica gel column chromatography that used CHCI3 fraction. As a result of seed-bioassay using each fraction, the IC50 values for the fresh weight of each fraction was 537.3 mg L-1, 1280 mg L-1 and 1947 mg L-1. Based on this, 5 fractions were obtained as a result of secondary silica gel column chromatography using the CA fraction. A seed-bioassay was performed, as a result, the lowest IC50 value was calculated as 226.7 mg L-1 in the CAE fraction. Based on this, the fraction was analyzed by GC-MS. The results of this study can be used as basic research data on the effects of weeds on forage crops and allelochemicals secreted from S. altissima.

Isolation of Antioxidative Components from the Bark of Rhus verniciflua STOKES Screened from Some Chinese Medicinal Plants (한약재로부터 선발된 옻나무 수피 추출물로부터 항산화 활성물질의 분리)

  • To develop new natural antioxidants, antioxidative activity of ethanol (75%) extracts from 50 edible or medicinal plants were examined on lard and palm oil by Rancimat method ($120^{\circ}C$, 20 L/hr). The extracts from Rhus verviciflua STOKES showed comparatively strong antioxidative activity on test. Of the solvents used for extraction, chloroform extract exhibited the strongest antioxidant activity. AI (antioxidant index: induction period of oil containing extract/induction period of control oil) of chloroform extract was higher than that of commercial antioxidant, such as BHT, BHA and ${\delta}-tocopherol$. Free phenolic acid fraction (200 ppm) of the chloroform extract from 75% EtOH extract of Rhus verniciflua STOKES (RCF) showed stronger activity than that of BHT, BHA, and ${\delta}-tocopherol$ at the same concentration. RCF-11 and RCF-13 fractions separated by silicagel column chromatography from the RCF showed stronger activity than other fractions by the Rancimat method.

Effect of Phenolic Extract of Dry Leaves of Lespedeza cuneata G. Don on Antioxidant Capacity and Tyrosinase Inhibition (야관문 잎 페놀 추출물의 항산화능 및 미백 효과)

  • Lespedeza cuneata G. Don is a plant commonly grown in Asian countries, which has been widely used as an oriental medicinal herb to treat diabetes, diarrhea and various other inflammatory diseases. The phenolics of dry leaves of L. cuneata G. Don were extracted by using 80% (v/v) aqueous methanol in assistance with homogenization and sonification. The phenolic extract and its five different fractions (n-hexane, chloroform, ethyl acetate, n-butanol, and water) were used to evaluate the levels of total phenolics, total flavonoids, and antioxidant capacity as well as the inhibitory effect of tyrosinase activity. Ethyl acetate fraction (1 g) had the highest levels of total phenolics at 240.8 mg gallic acid equivalents (GAE), total flavonoids as 90.4 mg catechin equivalents (CE) as well as antioxidant capacity at 523.4 mg vitamin C equivalents (VCE) on ABTS assay and 329.5 mg VCE on DPPH assay among fractions. One g of water fraction contained total phenolics at 133.1 mg GAE, total flavonoids at 34.5 mg CE, and antioxidant capacity at 333.4 mg VCE for ABTS assay and 313.2 mg VCE for DPPH assay. Inhibition of tyrosinase activity of water fraction at 300 ${\mu}g{\cdot}mL^{-1}$ was at 47.2% and 21.1% for L-tyrosine and L-DOPA as its substrate, respectively. On the other hand, ethyl acetate fraction at 300 ${\mu}g{\cdot}mL^{-1}$ showed tyrosinase inhibition of 10.2% for L-tyrosine and 11.9% for L-DOPA. These results suggested that the phenolics from dry leaves of L. cuneata G. Don may be utilized as a potent source of antioxidants and skin whitening agents.

Potent Anticarcinogenic Action of Moutan radix for Mouse Ascites Cancer Induced by Mouse Sarcoma 180 Cells (Moutan radix의 mouse sarcoma 180 cell로 유발한 mouse ascites cancer에 대한 항암효과)

  • Anticarcinogenic activity of Moutan radix for mouse ascites cancer induced by mouse Sarcoma 180 (S-180) cells was investigated. Methanol extract of Moutan radix including other folk medicinal plants (Taxus cuspidata, Curcuma longa, Artemisia capillaris, Ligrstri fructus, and Liriope platyphylla) used to remedy or cure many chronic human diseases like cancer was fractionated into hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc), and butanol (BuOH) fractions. Anticarcinogenic activity of the fractions, exhibited a strong cytotoxicity for L1210 and S-180 cells, was examined for mouse ascites cancer induced by S-180 cells. Male ICR mice (7 mice/treatment, $5{\sim}6$ weeks of age, $23{\pm}1\;g$ were injected i.p. with S-180 cells ($1{\times}10^{7}\;cell/1\;ml$ PBS). One day later, each mouse was given 0.1 ml of 10% DMSO containing sample ($30\;{\mu}g/g$ body weight) every day for 10 consecutive days. Control mice were only given 0.1ml S-180 cells and 0.1 ml 10% DMSO. Mice treated with EtOAc fraction of Moutan radix showed 28.7 days of life, which is 167% of control mice's life. Based on the dose-dependant experiment mice treated with $30\;{\mu}g$ showed longer life relative to mice treated with ootherr doses (5, 15, $60\;{\mu}g$), and mice treated with $60\;{\mu}g$ exhibited toxic symptoms. Body weight of mice treated with Moutan radix was significantly reduced relative to that of control mice (p<0.05). GC-MS analysis in conjunction with silica-gel column chromatography revealed that the EtOAc fraction contained 2-methoxylphenol, benzoic acid, 1-(4-hydroxy-3-methoxyphenyl)ethanone, 8-methyl-2,4(1H,3H)pteridinedione and 2,5-furan-dicarboxylic dimethyl ester as regards to the anticarcinogenic property of the EtOAc fraction. These results suggest that Moutan radix might be included as an anticarcinogenic medicinal plant for treatment of ascites cancer.

Antioxidant Activity of Extracts and Fractions from Aster scaber (참취 추출물과 용매분획물의 항산화 활성)

  • As an effort to develop functional food ingredients and to discover the biological activity, the total phenolic content, total flavonoid content, DPPH and ABTS radical scavenging activity, SOD-like activity, ferric reducing antioxidant power (FRAP), and $Fe^{2+}$ chelating of Aster scaber were measured using a 70% ethanol extract and various solvent fractions. As a result, the total phenolic concent was highest in an ethyl acetate fraction of 141.9 mg GAE eq/g and the total flavonoid content was 105.6 mg QUE eq/g. The DPPH radical scavenging activity was highest in an ethyl acetate fraction of 97.1% at a concentration of 1,000 ${\mu}g/mL$ (p<0.05). The ABTS radical scavenging activity showed a 86.9% ethyl acetate fraction and a 57.9% butanol fraction at a concentration of 125 ${\mu}g/mL$, and higher than that of positive control (${\alpha}$-tocopherol and BHT) (p<0.05). The SOD-like activity showed 42.8% in an ethyl acetate at a concentration of 1,000 ${\mu}g/mL$. The ethyl acetate fraction showed the highest value of FRAP at 1051.9 ${\mu}M$ and a concentration of 1,000 ${\mu}g/mL$ (p<0.05). The $Fe^{2+}$chelating was highest in the 70.1% chloroform fraction at a concentration of 500 ${\mu}g/mL$ (p<0.05). There is the highest correlation between DPPH radical scavenging activity and FRAP (r=0.981) as compared to other antioxidant assays (p<0.01). With these results, we confirmed that the ethyl acetate fraction of Aster scaber has great antioxidant potential. So it can be expected to be developed into a specific functional food ingredient.

Studies on the Positive Inotropic Mechanism of Aconiti Tuber (부자 강심성분의 작용기전에 관한 연구)

  • Aconiti tuber butanol fraction, which is isolated from the chloroform insoluble and water soluble extract of Aconitum volubile, has been recently known to have a potent positive inotropic effect in the isolated cardiac muscle preparations of various animals. The positive inotropic mechanism of Aconiti tuber butanol fraction, in relation with the external calcium, was studied using the isolated cat papillary muscle. The positive inotropic effect was dependent on the calcium concentration in the nutrient medium, and a synergistic relation could be demonstrated between Aconiti tuber butanol fraction and the external calcium. The inotropic effect of $10^{-4}g/ml$ of Aconiti tuber butanol fraction was equivalent to that of 0.06mM of calcium in the medium. After the treatment with a calcium influx inhibitor, Verapamil$(2{\pm}10^{-7}-10^{-6}M)$, the contractile force of the papillary muscle was markedly inhibited. In these preparations, Aconiti tuber butanol fraction restored the decreased contractility in a dose-dependent manner. It was suggested that the positive inotropic effect of Aconiti tuber butanol fraction might be related with the stimulating action on the calcium influx through the slow inward calcium channels in the cardiac cell membrane. In contrast with digitalis cardiac glycoside, Aconiti tuber butanol fraction infused intravenously into the anesthetized rabbit decreased the systemic arterial blood pressure and increased the carotid blood flow, but produced no prominent changes in the heart rate.

Antioxidative Effects of Campanula takesimana Nakai Extract (섬초롱(Campanula takesimana Nakai) 용매 추출물의 항산화 효과)

  • The study analyzed the total polyphenolic contents, DPPH radical scavenging activities, ABTS radical scavenging activities, reducing power levels, SOD-like activities, and tyrosinase inhibition activities of fractions from Campanula takesimana Nakai extract. Total polyphenolic content of the ethyl acetate fraction from Campanula takesimana Nakai extract was the highest. The ethyl acetate fraction of Campanula takesimana Nakai extract showed the lowest $IC_{50}$ value (0.13 mg/mL) as well as greater than 98% ABTS radical scavenging activity, which was similar to ascorbic acid. Further, reducing power was significantly higher in the ethyl acetate fraction. The SOD-like activity of edible plant was 70.56% (n-butanol fraction) and 62.27% (ethyl acetate fraction) at 1 mg/mL. Tyrosinase inhibition activity of Campanula takesimana Nakai extract was reduced to 34.77% in the n-butanol fraction and 30.85% in the ethyl acetate fraction at 5 mg/mL. These results suggest that the ethyl acetate and n-butanol fractions of Campanula takesimana Nakai extract could be applicable as food additives or cosmeceutical ingredients.

Antimicrobial Activity of Extracts and Fractions of Green Tea Used for Coarse Tea (엽차용 녹차 추출물 및 분획물의 항균효과)

  • Antimicrobial activities of green tea extracts used for coarse tea were investigated by disc diffusion method using eight different bacteria. Among the green tea extracts, the 70% ethanol extract demonstrated the strongest antimicrobial activities against Vibrio parahemolyticus (V. parahemolyticus) and Staphylococcus aureus (S. aureus) and thus was further fractionated. Among these fractions, the ethyl acetate fraction showed the strongest antimicrobial activities against V. parahemolyticus, S. aureus, Bacillus subtilis (B. subtilis), and Streptococcus mutans (S. mutans). These activities exceeded that of all extracts and fractions tested in this study. Interestingly, although green tea extracts showed significant antimicrobial activity against Micrococcus luteus (M. luteus), once fractionated, the ethyl acetate fraction did not show any antimicrobial activity against M. luteus. MICs of the ethyl acetate fraction were $5\;\;{\mu}L$/disc against B. subtilis and $3\;{\mu}L$/disc against S. aureus, S. mutans and V. parahaemolyticus. 90% inhibition of B. subtilis was observed with 0.05% ethyl acetate fraction but S. mutans needed over 0.1% ethyl acetate fraction to exhibit the same inhibition as B. subtilis. Antimicrobial activities of ethyl acetate fractions were reduced around 10% by thermal treatment at $121^{\circ}C$ for 20 min. All the results suggest that the 70% ethanol extract as well as the ethyl acetate fraction from green tea used for coarse tea could be further developed into a natural antimicrobial agent.

Effects of Fractions of Benincasa hispida on Plasma Levels of Glucose and Lipid in Streptozotocin Induced Diabetic Rats (동과 분획물의 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당 및 혈장 지질수준에 미치는 영향)

  • This study was designed to examine the effects of fractions of ethanol extract of Benincasa hispida (wax gourd) on plasma glucose and lipid levels in streptozotocin (STZ) induced diabolic rats. sprague-Dawley rats were induced diabetes mellitus by STZ injection (45 mg/kg) into the tail vein and were divided into 5 groups: normal, STZ-control, three experimental diabetic groups. fractions of ethanol extract of Benincasa hispida were administered orally into the diabetic rats for 14 days. The food intake and body weight were monitored and plasma levels of glucose, cholesterol, HDL-cholesterol, triglyceride (TG) , free fatty acid (FFA) , and aminotransferase activity were analyzed. A significant lowering effects of plasma glucose levels were observed in the chloroform fraction and $H_2O$ fraction group compared to STZ-control group at 14 days. Administrations of each of the three fractions decreased plasma TG and FFA levels in diabetic rats. Activity of alanine aminotransferase (ALT) in $CHCl_3$ fraction and $H_2O$ fraction groups and aspartate aminotransferase (AST) in $H_2O$ fraction group significantly lower than STZ-control group. The results show that $CHCl_3$ fraction of Benincasa hispida could be effective to control the STZ-induced diabetic rats. (Korean J Nutrition 38(10): 801$\sim$806,2005)

Genotoxicity and Mutagenicity of the Extracts of Morus alba L. (뽕나무 추출물의 유전독성 및 돌연변이원성)

  • This study was carried out to investigate the genotoxicity in comet and in vitro micronucleus assay and mutagenicity in Ames test of the extracts from leaves and stem of Morus alba L. The samples showed a very weak cytotoxicity on the NIH/3T3 cells by SRB assay. The cell viability of the extracts and fractions from leaves and stems of Morus alba L. was 80% over at $500\;{\mu}g/ml$, and that of the chloroform fractions from leaves and stems showed lower than others. The genotoxicity at $250\;{\mu}g/ml$ of 100% EtOH and water extracts on the NIH/3T3 cells in comet assay was about 40% compared to positive control, and most fractions from 100% EtOH extract of the leaves showed stronger genotoxicity than that offractions from the stem. The genotoxicity with S-9 mix in vitro micronucleus assay of the 100% EtOH and water extracts form Morus alba L. did not indicate any significant difference as compared with control group. The cytokinesis-binucleated cells were showed in the hexan, chloroform, ethylacetate and butanol fractions from the extract of the leaves without S-9, and sample with S-9 showed CB cells in the chloroform fraction from the leaves. In the Ames test, the water and 100% ethanol extracts of Morus alba L. did not have a strong mutagenicity in TA98 and TA100, but the fractions of organic solvents of the ethanol extract had $10{\sim}26%$ of mutagenicity on the TA100 strain.
