산벚나무 가지 추출물 및 용매 분획물의 Nitric Oxide 생성 억제 효과와 여드름 원인균에 대한 항균활성 (Antibacterial and Nitric Oxide Production Inhibitory Activities of Prunus sargentii Branches Extract and Its Fractions against Pathogens of Acne)

  • Background: In this study, we investigated the antibacterial and nitric oxide (NO) production inhibitory activities of 75% ethanol extract of Prunus sargentii branches and its fractions against acne pathogens. Methods and Results: The antibacterial activity against acne causing pathogens was determined using the disc diffusion assay. The ethyl acetate fraction showed higher activities against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis than those shown by other fractions. In the DPPH radical and NO scavenging assays, the butanol fraction showed strong DPPH radical and NO scavenging abilities. These activities were related to the total polyphenol and flavonoid contents of butanol fraction. On the other hand, the chloroform and ethyl acetate fractions exhibited the highest NO production inhibitory activity in Lipopolysaccharide (LPS)-stimulated Raw 264.7 cells compared to those exhibited by other fractions. Conclusions: The extract and its ethyl acetate fraction from the branches of P. sargentii exhibited antibacterial activity and could be used as functional materials in antimicrobial related fields. Moreover, the chloroform and ethyl acetate fractions are potential antiinflammatory agents and butanol fraction acts as an effective radical scavenger.

결명자 분획물이 당뇨 유발 흰쥐의 혈당에 미치는 영향 (Hypoglycemic Effect of Fractions of Cassia tora Extract in Streptozotocin-Induced Diabetic Rats)

  • The hypoglycemic effect of fractions of methanol extract of Cassia tora was investigated in diabetic rats. Sprague-Dawley male rats weighing 180∼230 g were divided into 6 groups. Diabetes was induced in the male rats by intravenous injection of streptozotocin at a dose of 45 mg/kg dissolved in citrate buffer (pH 4.5). The diabetic animals then had plasma glucose concentration of above 300 mg/㎗. Fractions of methanol extract of Cassia tora were administered orally into the diabetic rats for 14 days after streptozotocin injection. The food intake and body weight gain were monitored and plasma levels of glucose, triglyceride, free fatty acid, HDL-cholesterol, cholesterol and ALT activity were determined. Levels of glycogen, cholesterol and triglyceride in liver were analysed. Weights of liver and kidney were lighter in all the fraction-administered groups than streptozotocin-control group. Plasma glucose level was significantly decreased by the administration of butanol fraction at 14 days. Plasma triglyceride levels were lower in chloroform, butanol and H$_2$O fraction-administered groups compared to streptozotocin-control group. Plasma cholesterol and HDL-cholesterol levels were not significantly different in all groups. Administrations of each of the four fractions have decreased plasma free fatty acid level, ALT activity and liver triglyceride levels in the diabetic rats. Liver glycogen levels in chloroform, butanol and H$_2$O fraction-administered groups were higher than streptozotocin-control group. It is suggested from the results that butanol fraction of methanol extract of Cassia tora may contain the antihyperglycemic compounds.

오배자, 우슬, 가자 및 감초를 포함하는 혼합추출물의 항산화 및 항균활성 (Antioxidant and Antimicrobial Activities of Combined Extracts of Galla rhois, Achyranthes japonica Nakai, Terminalia chebula Retz and Glycyrrhiza uralensis)

  • In this study, antioxidant and antimicrobial activities of ethanolic crude extract and its five different solvent subfractions (namely, ethyl acetate fraction, n-butanol fraction, chloroform fraction, n-hexane fraction and the aqueous fraction) from a mixture of four different medicinal herbs (Galla rhois, Achyranthes japonica Nakai, Terminalia chebula Retz and Glycyrrhiza uralensis) were investigated. Among all the tested mixture combination of four medicinal herbs, 5:3:1:1 ratio of Galla:Achyranthes : Terminalia : Glycyrrhiza had the best antimicrobial effects against four strains of microorganisms (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa and Escherichia coli) and exhibited the highest DPPH radical-scavenging activity. Further sub-fractions with solvents were screened for antioxidant and antimicrobial activities. Antioxidant activity in order was ethyl acetate fraction > n-butanol fraction > chloroform fraction > nhexane fraction > aqueous fraction. The n-butanol extracted fraction showed the highest level of antimicrobial activity in com- parison to other fractions. In addition, all those fractions did not show any cytotoxicity against human skin cell CCD-986sk. These results suggest that 5:3:1:1 combination extracts of medicinal herbs (Galla : Achyranthes : Terminalia : Glycyrrhiza) may be potentially used as a safe natural antimicrobial preservative.

숙지황, 건지황 및 생지황 중 숙지황의 특이성분 검색 (Characteristic Component of Rehmanniae Radix Preparata Compared to Rehmanniae Radix and Rehmanniae Radix Crudus)

  • 생지황을 구증구폭하여 제조한 숙지황은 품질관리를 위한 지표물질이 아직 명확히 밝혀져 있지 않다. 생지황, 건지황과 숙지황을 methanol로 추출하고 ether, butanol, 수층으로 용매 분획하고 각 분획별로 TLC로 성분분석한 결과 ether층에서 숙지황의 특이성분이 나타났고, ether층을 증발농축하고 chloroform/methanol을 이동상으로 한 silica gel column으로 분리하고 다시 chloroform/methanol/물(7:1:1)의 preparative TLC로 단리하여 이 성분이 5-(hydroxymethyl)-2-furancarboxaldehyde(5-HMF)임이 규명되었다. 5-HMF는 생지황과 건지황에서는 숙지황에 비해 극히 미량이 존재하였다.

참나물 에탄올 추출물의 항산화 효과 (Antioxidative Effect of Pimpinella brachycarpa Ethanol Extract.)

  • 참나물의 생리활성 효과를 구명하기 위하여 in vitro에서 용매별 분획물의 항산화효능을 검토하였다. 참나물 에탄올추출물의 추출 수율은 12.01%이었으며, 이를 n-hexane, chloroform, ethylacetate, n-butanol, water로 계통 분획한 수율은 water 분획이 1.90%로 가장 높았으며, n-hexane, n-butanol, chloroform, ethylacetate 순이었다. In vitro에서 참나물 에탄올 추출물을 계통 분획하여 DPPH radical에 대한 자유기 소거능을 측정한 결과 n-hexane 분획이 $41\;{\mu}g/ml$로 가장 강한 항산화 활성을 나타내었다. 또한 Rancimat로 측정한 항산화지수도 n-hexane 분획이 1.98로 가장 높게 나타났으며, 이는 기존의 항산화제인 BHT가 1.97로 유사한 활성을 나타내었다. 아질산염 소거능에서도 모든 pH에서 n-hexane 분획이 가장 높게 나타났으며, pH 1.2에서 가장 우수하였다. 지질과산화물 생성 억제효과는 n-hexane 분획이 88.40%로 가장 높았으나, 양성대조군으로 사용한 BHT의 억제율보다도 낮았다. Linoleic acid에 대한 항산화효과를 지질과산화물가로 측정한 결과도 분획 중 n-hexane의 항산화력이 가장 우수하였고, ethylacetate, chloroform 순이었다. 이상의 결과 참나물은 식품으로의 이용가치뿐만 아니라 참나물 추출물의 in vitro 에서 항산화력의 우수함을 통해 천연 항산화제로서의 효과를 기대해 본다.

항진균성 물질을 생산하는 지의류 내생 곰팡이의 탐색 (Detection of Endolichenic Fungi Producing Antifungal Compound)

  • 순천대학교 지의류생물 소재은행(KOLABIC)에서 100종의 지의류 내생 곰팡이를 분양 받아 C. albicans에 대한 항진균 활성을 조사하였다. 고체배지 및 액체배지에 배양시 Usnea rigidula, Parmotrema pseudotinctorum 그리고 Myelochroa sp.가 높은 항진균 활성을 나타내는 물질을 생성함을 알 수 있었다. 이들 3종 지의류 내생 곰팡이의 배양액을 chloroform, ethyl acetate 용매 순으로 추출하고 disc diffusion method를 이용하여 각 추출물의 C. albicans에 대한 항진균 활성도를 측정한 결과 모두 chloroform 추출물에서 높은 활성이 가장 높았으며, 다음으로 ethyl acetate 추출물이 높은 활성을 나타냈다. 항진균성 물질이 기존에 알려진 물질인지에 대한 정보를 알기위해 6 일 동안 배양한 Usnea rigidula와 4일 동안 배양한 Parmotrema pseudotinctorum의 chloroform 추출액에 대하여 HPLC, $C_{18}$ column으로 분리하여 Usnea rigidula (2326)의 6일과 Parmotrema pseudotinctorum (2202)의 4일 배양액에서 각각 2 개의 peak 분획물을 얻었다. 각 분획물을 농축한 후 96 well plate를 이용하여 C. albicans와 C. glabrata에 대한 항진균 활성을 실험한 결과, Parmotrema pseudotinctorum의 주요peak는 control에 비해 항진균 활성이 높게 나타났으며 활성물질은 usnic acid계열의 화합물로 추정되었다.

딱지꽃 (Potentilla chinensis) 추출물의 항염증 효과 (Anti-Inflammatory Effect of Chloroform Extract from Potentilla chinensis)

  • In this study, we investigated the anti-inflammation effect of Potentilla chinensis (PC) on Raw264.7 macrophage cells. Ethanol extract of PC decreased the production of nitric oxide (NO) in LPS-stimulated RAW264.7 cells. Ethanol extract was fractioned by n-hexane, chloroform, ethyl acetate, n-butanol, water and each fraction was tested for inhibitory effects on inflammation. Among the sequential solvent fractions, PC chloroform extracts (50, 100, 300, and 500 ${\mu}g/mL$) significantly suppressed LPS-stimulated production of NO. During the entire experimental period, 200 and 300 ${\mu}g/mL$ of PC chloroform extracts had no cytotoxicity. LPS-induced NO and prostaglandin $E_2$ ($PGE_2$) production were inhibited by PC chloroform extracts up to 50% and 90% of these productions, respectively. PC chloroform extracts reduced the expression of iNOS and COX-2 gene. These results suggest that PC chloroform extracts exhibit strong effects of anti-inflammation and can be a potential candidate in the treatment of acute and chronic inflammatory diseases.

db/db 마우스 동물모델에서 Vigna nakashimae 클로로포름층 분획물의 항당뇨 효능 (Antidiabetic Activity of a Chloroform Fraction of Vigna nakashimae in db/db mice)

  • 최근 식생활의 변화에 따라 당뇨병, 비만 등의 만성대사질환의 발병률이 증가함에 따라 예방과 치료를 위한 물질연구가 많이 진행되고 있다. 본 연구에서는 선행 연구에서 항당뇨 효능이 확인된 Vigna nakashimae (VN) 메탄올 추출물로부터 VN 클로로포름층 분획물과 물층 분획물을 얻어 각 분획물의 항당뇨 효능과 분자적 기전을 살펴보았다. 각각의 VN 분획물을 제 2형 당뇨병질환모델인 db/db 마우스에 경구투여하여 VN 분획물의 혈당 및 지질 대사 변화 등을 측정하였다. VN 클로로포름층 분획물은 공복 혈당과 당화혈색소를 물층 분획물보다 더 효과적으로 감소시켰다. 또한 내당능도 개선하였으며, 혈중 지방산과 중성지방도 감소시켰다. VN 클로로포름층 분획물은 HepG2와 C2C12세포에서 인산화된 AMPK와 AMPK 하위 유전자인 CPT-1의 유전자 발현을 증가시켰다. VN 클로로포름층 분획물은 AMPK의 활성화와 일치하게 HepG2에서 당 생성 효소인 PEPCK와 G-6Pase의 발현을 감소시켰으며, C2C12에서는 당 유입을 증가시켰다. 이상의 결과는 VN 클로로포름층 분획물은 AMPK의 활성화를 통해 공복혈당을 감소시켰으며, 이러한 효과는 물층 분획물보다 더 효과적이었다.

각종 변이원에 대한 쇠비름 추출물의 돌연변이 억제 효과 (Inhibitory Effects of Poturaca oleracea L. Extract on the Mutagenicity of Various Mutagen)

  • This study was performed to determine the effects of antimutagenicity of Porturaca oleracea L. in Korea. In Ames test, the ethanol extract of Poturaca oleracea L. inhibited mutagenic activity of N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), , 4-nitroquinoline-1-oxide(4NQO), benzo($\alpha$)pyrene (B($\alpha$)P) and 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indole (Trp-P-1) in Salmonella typhimurium TA98 and TA100. But hot-water extract Poturaca oleracea L. only Inhibited mutagenic activity of MNNG in Salmonella typhimurium TA100, On 4NQO, the ethanol extract 100-1,600$\mu\textrm{g}$/plate of Porturaca oleracea L. showed a slight inhibitory effect of 13-48%, 4-47% in TA98 and TA100, respectively, but on MNNG, it showed higher inhibitory effect of 6-86% in TA100, And the treatment of 1,600$\mu\textrm{g}$/plate of ethanol extract of Porturacea L. had strong antimutagenicity with 74-87% inhibition against TA98 and TA100 induced by B(a)P and with 85-93% inhibition against TA98 and TA100 induced by Trp-P-1. The ethanol extract was fractionated with ether. chloroform, ethylacetate, butanol and water. Among them, most of the fraction except water fraction showed strong antimutagenicity effects against mutation induced by 4-NQO, MNNG, B(a)P and Trp-P-1. Chloroform fraction had strong antimutagenicity with 91% inhibition against TA100 induced by MNNG, diethyl etherfraction had strong antimutagenicity with 92%, 98% inhibition against TA98 and TA100 induced by 4NQO, Chloroform fraction had strong antimutagenicity with 97% inhibition against TA100 induced by B (a)P and diethyl etherfraction had strong antimutagenicity with 98% inhibition against both strain Induced by Trp-P-1, respectively.

귤엽산변방(橘葉散變方)이 유방암 세포주 MCF-7 생장 억제에 미치는 영향 (Anti-proliferation effect of Gyulyupsanbyonbang extracts on MCF-7 cells)

  • Purpose : This investigation was undertaken to evaluate the anti-proliferation, in hexane, chloroform, ethyl acetate, butanol and water fraction from extract of Gyulyupsanbyonbang(GYSB) using MCF-7 human breast cancer cells. Methods : GYSB was added to distilled water(1500ml) and was boiled then filtered. The residue was suspended in distilled water and extracted with hexane, chloroform, ethyl acetate, butanol and water. MCF-7 cells were cultured in RPMI1640 complex badge, NIH3T3 was cultered in 37$^{\circ}$C, 5% moisture incubator of carbon dioxide with Dulbecco's Modified Eagle Medium(DMEM) supplemented with 10% fetal bovine serum and antibiotics. Cell cytotoxicity test about cancer cell was measured used MTT assay. Results: When it synthesizes a result, hexane and butanol fraction had shown anti-proliferation effect and safety together, and those anti-proliferation effect operating selectively appeared. Ethyl acetate fraction had anti-proliferation effect however, it was not selective. The Chloroform and water soluble fraction did not almost appear anti-proliferation effect. Conclusion : I can conclude that GYSB have anti-proliferation effect and safety together on MCF-7 cells. It suggest that GYSB may be useful for brest cancer patients.
