Change of Lipoxygenase Activity in Chinese Cabbages Submerged in Brines (배추의 절임 중 Lipoxygenase의 활성변화)

  • Lipoxygenase activity in Chinese cabbage was measured at various concentrations of brines. Lipoxygenase activity on linoleic acid substrate was determined by changing the rate of dissolved oxygen consumption. The inactivation of lipoxygenase by salting was increased when concentration of sodium chloride and soaking time were increased. About 60% of enzyme activity was reduced after submerging in 13% brine solution for 5 hr. The addition of calcium chloride (0.7%) reduced about $10{\sim}15%$ of lipoxygenase activity rather than without. Residual activity of lipoxygenase in Chinese cabbage submerged in 13% brine was 20% and about 60% of lipoxygenase was also inhibited by addition of garlic extract.

Relationship between Exposure Concentrations to Methylene Chloride in Air and Carboxyhemoglobin Levels in Blood of Workers Engaged in Blowing, and Cleaning Operations (발포 및 세척 공정 근로자의 공기중 Methylene Chloride 노출 농도와 혈중 Carboxyhemoglobin 수준간의 상관관계에 관한 연구)

  • The objectives of this study were to evaluate exposures to airborne methylene chloride and postshift carboxyhemoglobin (COHb) in blood of workers engaged in processes using blowing or cleaning agents, and to investigate correlation between methylene chloride concentrations and the blood COHb levels of workers. The geometric mean (GM) of workers' exposures (8 hour-time weighted averages, TWA) to airborne methylene chloride during cleaning molds using rags wetted with the solvent in the manufacture of flexible polyurethane foam (GM = 61.4 ppm), during operating the dip tank for cleaning molds in the manufacture of lens (GM = 61.0 ppm), and during cleaning the blowing nozzles by spraying the solvent in the manufacture of shoes (GM = 117.2 ppm) were exceeded the American Conference of Governmental Industrial Hygienists (ACGIH) Threshold Limit Value ($TLV^{(R)}$)-Time Weighted Average (TWA) (50 ppm). The COHb levels were significantly different among groups (p<0.05). The average COHh levels in blood of non-smoking workers were 2.0% in. low-level (<50 ppm) exposure group, and 3.9 % in high-level (>50ppm) exposure group. The average COHb levels in smoking workers were 3.1% in low-level exposure group, and 4.8% in high-level exposure group. The blood COHb levels of no-exposed workers to methylene chloride were 1.8% in non-smoking group, and 2.8% in smoking group. It was found that the COHb level dependeds on the methylene chloride concentration and smoking habit, and was highly correlated with methylene chloride concentration in air. The correlation coefficient was 0.81 among non-smoking workers. The estimated COHb level (3.6%) and 95% upper confidence limit (4.0%) corresponding to TLV-TWA of methylene chloride exceeded the current ACGIH Biological Exposure Index (COHb 3.5%) for carbon monoxide. The estimated COHb level (5.4%) at 100 ppm exceeded the standard (5%) recommended by National Institute for Occupational Safety and Health (NIOSH) for preventing adverse cardiovascular effect. The estimated COHb value and 95% upper confidence limit at 25 ppm of the Occupational Safety and Health. Adminstration (OSHA) Permissible Exposure Limit-TWA (PEL-TWA) were 2.6% and 3.0%, respectively. It is suggested that COHb in blood be kept below 3.0% to comply with OSHA PEL-TWA.

  • Artifacts due to inter-particle and gas-particle interactions during PM2.5 sampling were quantified by comparing the measurement results between the annular denuder-filterpack system and the filterpack system without denuder. Measurements were carried in Seoul for 10 days in each season; Nov. 2004, Jan. 2005, Mar. 2005, and Jul. 2005, respectively. In each day, two 12-h samples were obtained. The concentrations of nitrate and chloride showed seasonal variations mainly due to the availability of ammonium to neutralize nitrate or chloride. Nitrates and chloride losses were prominent in summer. Since most of ammonia was used to neutralize sulfuric acid and formed ammonium sulfate in summer, nitrate and chloride could not exist in particles and ammonium loss was smaller than other seasons.

Studies on the Adsorptive Properties of Korean Kaolin(III) Adsorption at Solid-Liquid Interface (국산카올린의 흡착성에 관한 연구(III) 고체-액체 계면 흡착)

  • The adsorption of quinine, atropine and methylrozaniline chloride from aqueous phase by different kaolins was studied to innovated utilization of Korean kaolins as pharmaceutical agents. The adsorption isotherms were determined at $27{\pm}1^{\circ}C$ and the results were plotted according to the Langmuir equation. The Langmuir constants were calculated from adsorption isotherms of quinine and methylrozaniline chloride; a=1.46, 1.34 b=5.7, 9.3 and slope=0.175, 0.108, respectively. The kaolins gave the same type of curves with the two alkaloids and methylrozaniline chloride. The white colored premium grade kaolins were better adsorbent for the alkaloids and methylrozaniline chloride than the lower grade ones. The results indicate that the premium grade kaolins could be utilized as an ingredients in intestinal preparations. The condition of activation for the better adsorption was under the cases with the higher temperature and the lower pressure. The smaller particle size, the greater was adsorption power and the activated kaolins had superior adsorptive properties at higher pH value than at higher hydrogen-concentrations.

A Fatal Case of Methylene Chloride Poisoning (염화메틸렌 중독에 의한 사망 1례)

  • A 52-year-old man was found dead in the driver's seat of his tank lorry. The tank lorry was used to transport industrial wastewater to a disposal plant; the material was transferred into a storage tank with the help of compressed air. The wastewater contained methylene chloride and 2-chloropyridine. No respiratory protective equipment was used while working under these conditions. The autopsy report showed extensive edema and congestion of the brain, lung, and intraperitoneal organs. The concentrations of methylene chloride in lung and brain were reported at 398 and 67 mg/kg, respectively.

  • Ganoderma resinaceum tolerated sodium chloride salt stress within a range of 0 mM till 300 mM. It responded to salt stress with fluctuation in proline formation at different NaCl concentrations. However, the mycelial dry weight, total protein contents and exopolysaccharides did not changed considerably. Increasing sodium chloride concentration led to morphological alteration in fungal mycelia with disappearance of fungal cell wall, plasmolysis, and vacuolation as indicated with electron microscopic examination of the fungal growth.

  • This study was performed to assets the operating characteristics of food waste composting and co-composting (food waste + sewage sludge) at a compelling facility. The facility was being operated successfully without being affected by kind of composting feed materials. Partial anaerobic condition was detected during food waste composting and co-com-posting, but these two composting systems were proven to be operated successfully under aerobic condition from the monitoring results of $O_2$, volatile solids reduction rate, temperature, and other parameters. The conductivity and chloride concentrations of compost were gradually increased during two composting periods, but the conductivity and chloride concentrations of co-compelling indicated lower values than those of food waste composting at final point(40 m). As a result, co-composting was turned out to be more desirable than food waste composting, considering salt problem. High correlations ($R^2$= 0.9265 for food waste composting and $R^2$= 0.9685 for co-composting) between CEC and volatile organic matter were found. Quality of composts produced from two composting process satisfied Korean heavy metal standard.

Effects of Addition of Electrolyte and Ascorbic Acid in Feed during Heat Stress in Buffaloes

  • The ameliorative effect of salts and ascorbic acid polyphosphate supplementation on heat stress was studied in buffaloes. Adult buffaloes of either sex were randomly divided into 2 groups of 4 animals each. Group I served as control and Group II was supplemented with sodium bicarbonate, potassium carbonate and ascorbic acid polyphosphate. All the animals were exposed to two conditions of temperature and humidity: hot-dry and hot-humid in a psychrometric chamber for 4 h daily for 10 days. Blood was collected on day 1, 5 and 10 of treatment. The activities of catalase and superoxide dismutase (SOD), concentrations of serum glutathione (GSH), cortisol, sodium, potassium, and chloride and lipid peroxidation were estimated in serum. Lymphocyte proliferation was assessed in blood. The activities of catalase and SOD, serum concentration of GSH, sodium, potassium and chloride decreased while lipid peroxidation and serum cortisol increased in both groups when subjected to heat stress. Dietary supplementation resulted in further decreasing of the enzyme activities but increasing of the serum concentrations of GSH, sodium, potassium and chloride. Lipid peroxidation and serum cortisol increased in the supplemented group in both types of stress. Dietary supplementation caused an increase in lymphoproliferative response to con A. Thus, supplementation of ascorbate in addition to electrolytes relieves the animals of oxidative stress and boosts cell mediated immunity.

Evaluation of alginate microspheres prepared by emulsion method for protein delivery system

  • The purpose of the study is to research the proper conditions to prepare the calcium-alginate microspheres using a emulsion method. The calcium-alginate microspheres were prepared at distinct concentrations (alginate; 1%, 1.5%, 2% (w/v), calcium chloride; 2%, 4%, 8%, 10%(w/v)). The shape of the microspheres prepared was spherical. With increasing alginate and calcium chloride concentration the mean size of the microspheres decreased gradually. In release test, the amount of ovalbumin released from the calcium-alginate mcirospheres was decreased by the increasing of alginate and calcium chloride concentration. In this study the best result was obtained at a 2% of alginate concentration and 10% of calcium chloride concentration.

  • Three synthetic chemicals, benzoyl chloride, 2-propyn-l-ol, and 2-phenoxy ethanol were selected for genotoxicity testing, based on production quantity and available genotoxic data. In our previous report, benzoyl chloride induced chromosomal aberrations in Chinese hamster lung (CHL) fibroblast in vitro with and without metabolic activation, while 2-propyn-l-ol and 2-phenoxy ethanol induced only with metabolic activation. To compare the genotoxicity of chromosome aberration assay, the single cell gel electrophoresis (comet) assay subjected using CHL cells. As a result, statistically significant differences of tail moment values of benzoyl chloride, 2-propyn-1-ol, and 2-phenoxy ethanol were observed compared with control values on almost all concentrations with S9 or without S9 metabolic activation system. This results suggest that genotoxic results of the comet assay and the chromosome aberration assay show correlationship of genotoxicity in the CHL fibroblast. In summary, the positive result of chromosome aberration of benzoyl chloride, 2-propyn-l-ol, and 2-phenoxy ethanol was also induced DNA damages in comet assay with same cell line. Consequently, comet assay will be useful and more accurate tool to detect and to confirm the genotoxicity especially DNA damages in CHL fibroblast.
