Synthesis of Facial Amphiphile 3,7-Diamino-5α-cholestane Derivatives as a Molecular Receptor

  • A series of facial amphiphiles 3,7-diaminocholestane were synthesized from 3,7-diketocholestane via 2 sequential reductive aminations and anion recognition was evaluated with acetate, chloride, bromide, fluoride and phosphate anions. The stereo-selective reductive amination protocol was utilized to synthesized facial amphiphiles afforded receptors in high yields. The molecular receptor 2 showed the highest binding constant with acetate in a 1:1 ratio.

Diethylpyrocarbonate Inactivation of Aspartase from Hafnia Alvei

  • An aspartase purified from Hafnia alvei was inactivated by diethylpyrocarbonate (DEP) in a pseudo-first-order inactivation. The first-order plot was biphasic. The inactivation process was not saturable and the second order rate constant was $1.3\;M^{-1}s^{-1}$. The inactivated aspartase was reactivated with NH₂OH. The difference absorption spectrum of DEP-inactivated vs native enzyme preparations revealed a marked peak around 242 nm. The pH dependence of the inactivation rate suggests that an amino acid residue having a pK value of 7.2 was involved in the inactivation. L-aspartate, fumarate (substrates), and chloride ion (inhibitor) protected the enzyme against inactivation, indicating that histidine residues for the enzyme activity are located at the active site of this aspartase. Inspection of the presence and absence of $Cl^-$ ion demonstrated that the number of essential histidine residues is less than two. Thus, one or two histidines are in or near the aspartate binding site and participate in an essential step of the catalytic reaction.

  • It is possible for concrete using sea sand to contain chloride ion as well as cation such as Na$^{+}$and $K^{+}$ during mixing process. It is known that some cations such as Na$^{+}$and $K^{+}$ remain in pore solution without binding In this study, therefore, we intend to inspect the behavior of cations in cement paste as well as NaCl, CaCl$_2$ and KCI through analysis of pore solution extracted from cement paste with high pressure vessel. As a result, increase of alkali ions by adding sea sand and admixtures to the fresh concrete means use of the cement contained high alkali contents. In this case, alkali ions in pore solution can decrease durability of cement products causing alkali-aggregate reaction or accelerated carbonation. So it needs to be studied.studied.

Potentiometric Sensor for the Determination of Dibucaine in Pharmaceutical Preparations and Electrochemical Study of the Drug with BSA

  • Plasticized poly(vinyl chloride), PVCs, with different membrane compositions tested for use in the construction of an ion-selective sensor for the determination dibucaine. A prepared membrane with dioctyl phthalate-PVC and ion-pair of N-(1-naphthyl)ethylenediamine dihydrochloride-tetraphenyl borate had a good potential to acts as a potentiometric sensor for the analysis of dibucaine. A linear relationship was obtained between potential and logC varying between $1.0{\times}10^{-6}$ and $1.0{\times}10^{-2}$ M dibucaine with a good repeatability and reproducibility. The sensor was applied for the determination of the drug in pharmaceuticals and biological fluids such as plasma and urine samples with satisfactory results. The drug electrode has also been used to study the interaction of bovine serum albumin (BSA) with dibucaine. The saturated quantities of dibucaine binding were 13.04, 5.30 and 9.70 mol/mol in 0.01, 0.02 and 0.1% of protein, respectively.

  • New porphyrin square (1) was prepared by reaction of porphyrin containing pyridine and Re$(CO)_5Cl$ in THF/toluene solvent. 2-(Methylene15-crown-5)-nicotinoly ester(2) was synthesized by reaction of 2-(Hydroxymethyl-15-crown-5 and nicotinoly chloride in $CH_2Cl_2$. In fluorescence quenching studies luminescence was quenched by addition of the guest(2) into host(l). In the host-guest system we could obtain binding constant (K= $1.13{\times}10^{7}M^{-1}$) at decreasing concentration of (2). But the luminescence was dramatically increased after $Na^{+}$ was added into the host-guest system.

  • Purine nucleosides were chlorinated by the reaction of acyl chloride in DMF with MCPBA under mild conditions with moderate yields. And, satisfactory method for the synthesis of ribonucleoside-$3^{I},5^{I}$-cyclic phosphates and its characterization by$^{1}H$ and $^{13}C$ nmr spectroscopy is described.

  • Choline esters that are used with substrate of BChE-catalyzed hydrolyses were synthesized by two methods. First, 2-chloroethyl thiohexanoate, 2-chloroethyl thioheptanoate, and 2-chloroethyl thiooctanoate were synthesized by the treatment of hexanoyl chloride with ethylene sulfide. Hexanoyl thiocholine and octanoyl thiocholine were synthesized by using 2-chloroethyl thiohexanoate and 2-chloroethyl thiooctanoate with trimethyl amine. Second, after reaction of ethylene sulfide and dimethyl amine, followed by acylation with acid anhydride and then heptanonyl thiocholine, decanoyl thiocholine were synthesized by treatment of methyl iodide.

  • The transformation of the liquid crystal complex made by binding of anionic surfactant, sodium dodecyl sulfate (SDS), into high charge density cationic polymer, the homopolymer of diallyldimethylammonium chloride (PDADMAC) was induced by adding of nonionic surfactants and investigated by means of microscopy and FE.SEM. Among nonionic surfactants in this experiments polyethylene glycol (3 mol) ether of lauryl alcohol (laureth-3) made variation in the complex. The laureth-3 transformed the complex into spherulite vesicle with the size of ca.$100{\mu}m$. This change increased the viscosity and the turbidity of the solution phase separated originally. Microscope showed that they are spherulite particles and polarized microscope suggested they are multi.lamellar liquid crystals. FE-SEM also proved that explicitly.
