p-Aminosalicylic acid를 포함하고 있는 Chitin, Chitosan-Microsphere의 용출특성 (Dissolution properties of Chitin or Chitosan Microsphere Containing p-Aminosalicylic Acid)

  • The Applicability of chitin or chitosan microsphere as means to achieve sustained release of p-aminosalicylic acid(PAS) has been examined. The microsphere of chitin or chitosan containing PAS were prepared by coacervation in acidic aqueous system in range of pH 2.0-4.0. The dissolution test of PAS from polymeric drug system was carried out in vitro test. The dissolution rate of PAS from the microsphere with chitin was significanthly lower than that from the microsphere with chitosan.The dissolution rate of PAS from the microsphere was decreased with increasing of concentration of chitin and chitosan. The sustained release of PAS from the microsphere was more effective at pH 1.2 than pH 6.8.

Binding Capacity of Chitin and Chitosan to Anthocyanin Pigment Isolated from Purple Perilla Leaves

  • The binding capacity of chitin and chitosan to anthocyanin pigmentss isolated from purple perilla leaves was inves-tigated. The pigment binding capacity increased with increasing pigment concentrations and decreasing pH without being affected by reaction temperature and particle sizes. Regression analysis revealed significantly high corre-lations between pigment binding capacity of chitin and chitosan and pigment concentration at ranges of 25-100 mg of pigment/g of sample, After 1 hr settling, release of pigment from pigmented chitin and chitosan increased with increasing pH, up to 24.9% and 17.4%, respectively, at pH 9. In general, pigment binding capacity of chitosan was higher than that of chitin. There results suggest that chitosan may be useful as a potential adsorbent capable of stabilizing anthocyanin pigment.

  • 키틴의 글루코오스 단위에 메탄술폰산 존재하에서 아세트산 무수물, 프로피온산 무수물과 n-부틸산 무수물을 반응시켜 아세틸 키틴(AC), 프로피오닐 키틴(PC) 그리고 n-부티릴 키틴(BC) 등의 에스테르 형태를 갖는 개질화 키틴을 합성하였고 이들의 구조를 FT-IR과 solid state CP/MAS $^{13}C-NMR$ spectra로 확인하였다. 에스테르 형태의 키틴유도체는 대체로 키틴의 글루코오스 단위의 6번 탄소위치에 치환되었다. 유도체를 유기용매와 수용성 약산 유기용매에서 용해성을 검토하여 포름산에서 세 가지 유도체 모두 좋은 용해성을 나타냈고, 수용성 약산에서는 대부분의 유도체는 팽윤되는 것을 고찰할 수 있었다.

  • Toha-jeot, salt-fermented Toha shirmp(caridina denticulata denticulata DeHAAN) is a traditional fermented food in Korea. Samples of Toha-jeot used in the present study were a low-salt group of 15% sodium chloride(L), a high-salt group of 23% sodium chloride(H), a 50% conventional soybean sauce group(S), a low-salt group contraining 2% wheat bran (W2%-L) , a high -salt group containing 2% wheat bran(W2%-H), a low-salt group containing 4% wheat bran(W4%-L)and a high-salt group containing 4% wheat bran (W4%-H). These seven groups were refrigerated at 4 $\pm$1$^{\circ}C$ and then taken out for analysis at theree month intervals. We investigated the functional components of Toha-jeot during fermentation . Long fermentation of Toha-jeot lowered the viscosity of chitin and tended to reduce the distribution of molecular weight. THe formation of chitin oligosaccharides on the other hand, increased significantly. After nine months of fermenttion, chitin oligosaccharides(M.W. 823~1789) constituting 24.75% of Toha chitin were produced in the sample of W2%-H. During the same period, chitin oligosaccharides(M.W.1436-1879) constituting 66.30% of Toha chitin were produced in the samples of S. However, chitin oligosaccharides were not produced in Jeotsaeu-jeot made of sea-water shrimp when fermented for six months.

근적외선 분광분석법을 이용한 Chitin 및 Chitosan의 탈단백 및 탈아세틸화도 측정 (Measurement of Deproteinization and Deacetylation of Chitin and Chitosan by Near Infrared Spectroscopy)

  • NIR spectroscopic analysis was used for the measurement of deproteinization and deacetylation to apply the merits of NIR spectroscopic analysis to the quality management in the process of chitin and chitosan production. In measuring squid pen and red snow crab shell, which are raw materials of chitin and chitosan by NIR there were typical peaks in 1200 nm, 1510 nm, 2050 nm and 2180 nm. Squid pen had somewhat higher peak than red snow crab shell. In producing chitin, amount of protein was decreased. Measuring it by NIR, reduction of protein caused by deproteinization was identified in producing chitin. Chitosan is a derivative material made from chitin by processing the deacetylation. During this processing, acetyl groups were removed and amide bends were appeared. From NIR spectra, peaks at 1530 nm and 2030 nm indicated amide II peak of chitosan, and these peaks were used for identifying the differences of structure between chitin and chitosan. The error in measurement of nonidentified sample was below $1\%$ and the error in the standard curve was below 0.006. These errors were very low and the accuracy of NIR was considered to be superior to the existing methods.

Chitin의 분자량과 Phosphorylation 정도가 Microcrystalline Chitin의 유화안정 능력에 미치는 영향 (The Effect of Molecular Size and Degree of Phosphorylation on the Emulsion Stability of Microcrystalline Chitin)

  • 게껍질로부터 얻은 분자량이 다른 chitin을 phosphorylation하여 얻은 MCC의 유화 안정능력은 중간 분자량의 chitin에 40% phosphorylation시킨 것(M-40-MCC)이 가장 좋았다. 그리고 MCC의 지방 결합 능력은 사용한 chitin의 분자량 크기에 별 영향을 받지 않았으나, phosphorylation 정도가 높은 MCC는 지방 결합 능력 이 커지는 경향을 보였다. 액상 커피 creamer에 사용되는 유화제의 50-70%를 MCC로 대체하여 제품을 만들어 품질을 평가한 결과 유화 안정성이 만족스러웠고, MCC를 첨가하면 creamer의 점도가 감소하는 경향을 나타내었다. 그리고 creamer의 백탁능력을 커피에 평가한 결과 MCC 첨가한 creamer의 L 값이 대조구보다 우수하였다. 그리고 creamer제품의 관능검사 결과 시료간에 여러가지 관능적 특성에 차이가 없었으나, MCC를 70% 첨가하면 creamer의 구수한 맛이 대조구보다 떨어지고 전체적인 선호도도 감소하는 것으로 나타났다 이들 결과를 종합해 보면 액상 커피 creamer에 50%정도의 MCC를 첨가하는 것이 적당한 것으로 평가되었다.

키틴 기반 흡착제 PEI-chitin을 이용한 반응성염료의 제거 (Removal of Reactive Dyes using Chitin-based Adsorbent PEI-chitin)

  • 산업폐수 속의 대표적인 색도유발물질인 염료를 효과적으로 제거하기 위한 생체흡착제로 폴리에틸렌이민을 키틴에 가교결합한 PEI-chitin을 개발하였다. 대표적인 반응성염료인 Reactive Orange 16 (RO16)을 모델염료로 사용하였고, RO16에 대한 PEI-chitin의 흡/탈착 능력을 평가하기 위해 pH의 영향, 등온흡착, 흡착속도론, 탈착 실험을 수행하였다. 그 결과, Langmuir 식에 의해 산출된 최대흡착량은 pH 2에서 266.3 mg/g이었고, 흡착평형에 도달하는 시간은 50 mg/L에서는 약 20분, 100 mg/L에서는 약 60분 그리고 200 mg/L에서는 약 240분으로 평가되었다. 탈착실험은 암모니아/에탄올 혼합용액, NaOH, $NaHCO_3$, $Na_2CO_3$를 용리액으로 이용하여 평가하였으며, 암모니아/에탄올 혼합용액에서 75.24%로 가장 높은 탈착율을 보였다.

Isolation of Chitin-utilizing Bacterium and Production of Its Extracellular Chitinase

  • A bacterial strain, designated as WY22, producing extracellular chitinase was isolated from the soil around the Youngduck area, after enrichment culture in a medium containing $1{\%}$ (w/v) wet colloidal chitin as a sole carbon source. The isolate was identified as a strain of Bacillus sp. based on its morphological and physiological characteristics. It was observed that Bacillus sp. WY22 could inhibit the growth of Fusarium oxysporum with hyphal extention-inhibition assay on potato dextrose agar plate supplemented with $1{\%}$ collidal chitin. Optimum culture conditions of Bacillus sp. WY22 were examined for chitinase production in a chitin medium. High level production of chitinase was observed not only in the chitin medium but in a medium supplemented with $1{\%}$ N-glucosamine or lactose instead of chitin. The optimum concentrations of colloidal chitin and yeast extract were 3.0 and $0.5{\%}$, and the optimum culture conditions for initial pH of medium and temperature were 7.0 and $30^{\circ}C$, respectively, for the production of chitinase.

Suppression of Clubroot Formation in Chinese Cabbage by the Chitin Compost and Broth

  • Chitin compost and broth were used to suppress club root. Individual cabbage seedlings were transplanted into pots(3500 ml) containing a mixture of 3% chitin compost and 50 ml of chitin broth (T1) or the same quantity control compost and control compost broth(T2). The media in each pot was then infected with Plasmodiophora brassicae. Samples were taken at 6, 7 and 8 weeks after transplanting. The population of chitinase producing bacteria in T1 was consistently larger than that observed in T2. Chitinase activity in the T1 rhizosphere was two-fold greater than that of T2 at each time point observed. Shoot dry weight, leaf number and leaf area in T1 were enhanced 20%, 10% and 12% relative to those seen in T2, respectively. The disease index and root mortality at 8 weeks after transplanting were reduced by 50% and 25% in T1 compared to T2, respectively. Results presented in this study are strongly indicative that chitin compost and broth suppress clubroot in Chinese cabbage.

Chitin, CHitosan 및 수용성 Chitosan 유도체가 흰쥐의 Cadimium 중독과 지방대사에 미치는 영향 (Effect of Dietary CHitin, Chitosan and NOCC on Cadmium Toxicity and Lipid Metabolism in Rats)

  • This study was to investigate the effects of chitin, chitosan , NOCC and cellulose on cadmium toxicity and lipid metabolism in rats. Fifty male rats of Sprague-Dawley strain weighing 155$\pm$17g were divided into 10 groups according to body weight, and were raised for 5 weeks. Levels of 0 and 400 ppm of cadmium chloride were in the diets. And chitin, chitosan , NOCC and cellulose were given at the level of 0 and 4%(w/w) of the diets. Chitosan and NOCC decreased Cd concentration of liver and kidney, and increased the fecal excretion of Cd. Although cellulose was less effective, cellulose also decreased Cd concentration of liver and increased fecal Cd excretion . However chitin had little effect on alleviating Cd toxicity. In addition, chitosan and NOCC lowered total lipid, cholesterol and triglyceride contents in serum and liver by decreasing the lipid absorption. The lowering of cholesterol especially had a remarkable effect. Although it was less effective than chitosan and NOCC, chitin decreased the lipid absorption ratio and lowered cholesterol contents in serum and liver. In conclusion , chitosan and NOCC showed effects of decreasing the absorption of Cd and lipid.
