주요 한약재인 목단피의 저장에 관한 기초 연구 (A Basic Study on the Storage of Major Chinese Herbal Medicine Materials , Moutan Radicis Cortex)

  • This study was conducted to investigate desirable storage method to major chinese herbal medicine materials, Moutan Radicis Cortex in different packing materials and sealing degree. The loss in weight of Moutan Radicis Cortex as influenced by packing materials was the lowest at complete sealing treatment with transparent or black polyethylene film. The rot ratio of Moutan Radicis Cortex during the storage period was not difference between packing materials, transparent and black polyethylene film, but rot ratio of complete sealing condition was lower than punch ($\phi$J5mm) on polyethylene film.

The Effect of Chinese Herbs on Acne Pathogens

  • Chinese herbs have been used for a long period of time and less side effects than synthesized chemical drugs. Therefore, using Chinese herbs as natural additives in cosmetics becomes popular in recent years. The methanol extracts of Scutellariae Radix, Lithospermi Radix, Lonicerae Flos, Andrographitis. Herba, Angelicae Dahuricae Radix, Ligustici Rhizoma et Radix, Hedyotis Diffusae Herba, Isatidis Folium, Magnoliae Liliflorae Flos, Forsythiae Fructus, Anmarrhenae Rhizoma, Spirodelae Herba, Gardeniae Fructus, Sophorae Flavescentis Radix, Coptidis Rhizoma, Prunellae Spica, Equiseti Hiemalis Herba, Gentianae Radix, Moutan Radicis Cortex, Fraxini Cortex, Lycii Radicis Cortex, Violae Herba, Lophatheri Herba, Matricariae chamomillae Flos, Taraxaci Herba and Scutellariae Barbatae Herba are used to test the efficiency of inhibiting acne pathogens. Twenty-six Chinese herbs are extracted by methanol, and then condensed to dried powder. These extracts are divided into water-soluble part and DMSO soluble part. These two type solutions are tested for the effect on acne pathogens by paper disc diffusion method. The results show that the substances of water soluble part which are Coptidis Rhizoma, Moutan Radicis Cortex, Scutellariae Barbatae Herba have medium to high activity of inhibiting acne pathogents, and the substances of DMSO soluble part which are Coptidis Rhizoma, Ligustici Rhizoma et Radix, Sophorae Flavescentis Radix, Moutan Radicis Cortex, Scutellariae Radix, Scutellariae Barbatae Herba also have medium to high activity of inhibiting acne pathogens. Using Chinese herbs as natural additives in cosmetics is convenience and valuable application in cosmetceutical research and development. Therefore, it is worth that re-investigation and find out the potential of Chinese herbs being use in cosmetics.

중국 목단재배종의 채취시기 및 부위별 Paeonol과 Paeoniflorin의 함량변화 (Changes of Paeonol and Paeoniflorin Contents in Chinese Moutan (Paeonia suffruticosa Andrews) Cultivars with Different Harvesting Times and their Parts)

  • 중국에서 재배되는 목단의 품종 중 화형의 연화정도가 다른 대표적인 6개 품종의 뿌리에 대해 채취시기에 따른 paeonol과 paeoniflorin의 함량을 조사하였다. 6개 품종의 뿌리에서 paeonol의 양은 계절에 관계없이 거의 일정하여 변화가 없었다. 반면 paeoniflorin은 개화기인 5월에 가장 높은 값을 보이다가 6월에 감소, 7월에는 증가, 8월에 다시 감소하다가 9월에는 증가하는 경향을 보였다. 비교한 6개의 품종중 화형의 형태가 단순한 종류가 복잡한 종류에 비해 약리성분을 많이 함유하고 있었다. 단판형화관인 Dan Feng과 만첩형 화관의 Wu Long Feng Sheng의 채취시기에 따른 부위별 약리성분의 분포정도를 비교하였다. Paeonol은 두 품종 모두 뿌리를 제외한 잎과 줄기에 미량으로 존재하며 채취시기가 달라져도 변화가 없었다. Paeoniflorin은 뿌리와 잎 두 부분에 다량으로 존재하여 뿌리에서의 분포양이 계절적으로 증감을 반복하는 반면, 잎은 성장이 가장 왕성한 5월에 가장 높게 분포하고 시기가 지날수록 감소하였다.

Hypocholestrolemic Effect of CJ90002 in Hamsters: A Potent Inhibitor for Squalene Synthase from Paeonia moutan

  • Squalene synthase catalyzes the reductive dimerization of two molecules of farnesyl diphosphate to form squalene at the final branch point of the cholesterol biosynthetic pathway. Due to the unique position of this enzyme in the pathway, its inhibitors may have advantages as antihypercholesterolemic agents. Therefore, selective inhibitors of squalene synthase do not prevent the formation of the essential branch products of the isoprene pathway, such as dolichol, coenzyme-Q, and prenylated proteins, as might be expected for inhibitors of enzymes earlier in the pathway; for example, lovastatin and mevalotin. The current study reports that CJ90002, a pentagalloylglucose isolated from Paeonia moutan SIM (Paeoniaceae), which is an important Chinese crude drug used in many traditional prescriptions, was a potent inhibitor of rat microsomal squalene synthase, and also a potent inhibitor of cholesterol biosynthesis in vitro. In addition, the intraperitoneal and oral administration of CJ90002 had a significant lowering effect on plasma cholesterol levels in hamsters.

Evaluating Pharmacological Effects of Two Major Components of Shuangdan Oral Liquid: Role of Danshensu and Paeonol in Diabetic Nephropathy Rat

  • Shuangdan oral liquid (SDO) containing radix Salviae miltiorrhizae (Chinese name Danshen) and cortex moutan (Chinese name Mudanpi) is a traditional Chinese medicine using for treating vascular diseases. Danshensu (DSS) is a main effective monomer composition derived from radix Salviae miltiorrhizae and paeonol (Pae) from cortex moutan. Although the two herbs are widely used in traditional Chinese medicine, the pharmacological functions of their active compositions were not reported. Therefore, the research of DSS and Pae in mechanisms and pharmacodynamics interaction can provide scientific evidence to support clinical application. The diabetic nephropathy (DN) rats which were induced by streptozotocin (STZ) were treated with SDO, DSS, Pae, and DSS+Pae for eight weeks. The positive effects on DN animal models were investigated by detection of physiological and biochemical indexes and oxidative stress markers, within five treatments: SDO, DSS, Pae, DSS+Pae and insulin group. Compared with the model group, the DSS+Pae group improved the renal function, blood lipid metabolism and blood viscosity, increased the vitality of T-SOD or T-AOC and decreased the level of MDA or NO after the treatment. The study was successfully showed that the DSS+Pae group could delay the process of DN, especially in the renal injury part of histopathology changes. Our results suggest that the co-administration of DSS and Pae significantly may play a protective role in DN rats through decreasing the oxidative stress and improving the blood lipid metabolism mechanisms.

  • Keokhachukeu-tang is the one of Chinese traditional prescription used for the treatment of liver disease. This prescription consists of Carthami Flos (6g), Persicae Semen (9g), Pteropi Faeces (9g), Corydalis Tuber (9g), Moutan Radicis Cortex (6g), Paeoniae Radix rubra (9g), Angelicae gigantis Radix (9g), Cnidii Rhizoma (9g), Linderae Radix (12g), Cyperi Rhizoma (12g), Aurantii Fructus (9g), and Glycyrrhizae Radix (3g). Water extract of Keokhachukeu-tang showed a moderate hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. (omitted)

목단피가 세포고사의 억제에 미치는 영향에 관한 연구 (Studies on the Anti-apoptotic Effect of the Mudanpi)

  • Mudanpi (Cortex Moutan Radicis; the root cortex of Paeonia suffruticosa Andrews) is an important Chinese crude drug used in many oriental prescriptions. 1,2,3,4,6-Penta-O-galloyl-beta-D-glucose (PGG), a major component of this crude drug, has been shown to possess potent antioxidant, anti-mutagenic and anti-proliferative effects. In this study, I examined whether PGG could protect Neuro 2A cells, a kind of neuronal cell lines, from oxidative damage through the induction of HO-1 expression and HO activity. Exposure of Neuro 2A cells to PGG (10-50μM) resulted in a concentration- and time-dependent induction of HO-1 mRNA, and protein expressions and heme oxygenase activity. PGG protected the cells from hydrogen peroxide-induced cell death. The protective effect of PGG on hydrogen peroxide-induced cell death was abrogated by zinc protoporphyrin IX (ZnPP IX), a HO inhibitor. These results indicate that PGG is a potent inducer of HO-1 and HO-1 induction is responsible for the PGG-mediated cytoprotection against oxidative damage.

Antitumor and Apoptosis Induction Effects of Paeonol on Mice Bearing EMT6 Breast Carcinoma

  • Paeonol is a major phenolic micromolecular component of Moutan cortex Radicis, a traditional Chinese Medicine. It has shown antitumor effects in previous studies; however, the underlying mechanisms remain unknown. This study investigated the mechanism by giving treatments of placebo, cyclophosphamide, paeonol of 150 and 300 mg/kg to 4 groups of mice bearing EMT6 breast cancer. Apoptosis in tumor cells were confirmed by morphology analysis, including hematoxylin, eosin staining and TUNEL staining. The results showed that the weight of EMT6 breast tumor was significantly reduced in the groups treated with both 150 and 300 mg/kg of paeonol. Immunohistochemical and Western blot results showed that the expression of Bcl-2 was down-regulated while the expression of Bax, caspase 8 and caspase 3 was up-regulated respectively. These results suggest that paeonol exhibits antitumor effects and the mechanism of the inhibition is via induction of apoptosis, regulation of Bcl-2 and Bax expression, and activation of caspase 8 and caspase 3.

육미지황탕(환)의 약리와 독성에 대한 문헌고찰 (Pharmacological and Toxicological review of Yukmijihwang-tang(Hwan))

  • Objectives : Yukmijihwang-tang(Wan), a well-known formula for invigorating yin-particular kidney yin, was first recorded in "Xiao er Yao Zheng Zhi Jue", consisting of Radix Rehmanniae Preparata, Fructus Macrocarpii, Rhizoma Dioscoreae Oppositae, Poria, Rhizoma Alismatis and Cortex Moutan Radicis with dose proportion of 8:4:4:3:3:3. Although clinical trials have been lacking, various pharmacological actions for Yukmijihwang-tang has been identified newly using animal models. In addition, it was reported that Yukmijihwang-tang increases structural chromosome aberrations significantly in Chinese hamster lung cells. In this article, it is purposed that new studies for pharmacology and toxicology of Yukmijihwang-tang are reviewed. Insight into new studies of Yukmijihwang-tang at the cellular and animal levels will enhance our understanding of Yukmijihwang-tang against various diseases will provide new tools to diagnose and treat patients. Methods : Recent researches for Yukmijihwang-tang were reviewed and summarized in terms of pharmacological action and toxicity. All sources for review were based on recent studies loaded on data base of web sites such as Science Direct and National Center for Biotechnology Information. Results and Conclusions : Recently, reports showed that YMJ had antiaging effects, antioxidant and free radical scavenging activities, anti-renal hypertension and prevented tumors, and diabetes mellitus. However, there is little information on its safety except general toxicity, acute and sub-chronic oral toxicity, or genotoxicity. In addition, clinical trial for Yukmijihwang-tang was limited even though Yukmijihwang-tang has been used extensively in Korean traditional medicine. Thus, further studies are necessary to focus on safety evaluation and clinical trial for Yukmijihwang-tang.

한약이 임신중 태아에 미치는 영향(II) -한약이 돌연변이원성과 염색체이상에 미치는 효과- (Studies on the Effects of Herbal medicines on the Fetus during Pregnancy (II) - Mutagenesis and chromosomal aberration of herbal medicines -)

  • Oriental herbal medicines were examined for mutagenicity in the reverse mutation test on Salmonella typimurium T A98/100 and chromosomal aberration test on cultured mammalian cells (Chinese hamster cell lines). The reverse mutation test was performed by a plate incorporation method with and without a metabolic activation system (S9 mix). The tested herbal medicines did not significantly increase revertible colonies on any of the test strains with and without a metabolic activation system (S9 mix) at concentrations of 1 mg/ml. In the chromosomal aberration test, most tested herbal medicines did not significantly increase the number of aberrant cells on any of the test strains with a metabolic activation system (S9 mix) at concentrations of 1 mg/ml, compared with the vehicle control. However. Ansu Semen significantly increased the number of aberrant cells without a metabolic activation system (S9 mix). Paeoniae Radix. Hoelen, Aurantii nobilis Pericarpium, Cnidii Rhizoma, Angeliacae gigantis Radix, Perillae Herba and Moutan Cortex Radicis slightly increased revertible colonies on any of the test strains with a metabolic activation system (S9 mix), These results indicate that most herbal medicines might be carefully used in obstetrics and gynecology, although they do not have the potent mutagenic potential under the present test conditions.
