Association between PCR-RFLP Polymorphism of the Fifth Intron in Lipoprotein Lipase Gene and Productive Traits in Pig Resource Family

  • The study was aimed at detecting polymorphism of the fifth intron in lipoprotein lipase (LPL) gene and analyzing association between the polymorphism and productive traits. A pair of primers was designed for amplifying the fifth intron. Sequence analysis indicated that a G1171C substitution existed in Large White breed. The mutation was detected by PCR-AfaI-RFLP. Polymorphism analysis in a pig resource family showed that there existed significant effects on carcass and meat quality traits. Thoraxwaist fat thickness of BB genotype was significantly higher (14.2%, p<0.05) than that of AA on carcass traits, while BB genotype was significantly lower (3.6% p<0.01, 4.1% p<0.01; 2.3% p<0.01, 1.9% p<0.01; 1.8% p<0.01, 1.4% p<0.05) than AA and AB genotype in pH of m. Longissimus Dorsi (LD), m. Biceps Femoris (BF), m. Semipinali Capitis (SC). The allelic frequencies were also significantly different between indigenous Chinese breeds and exotic breeds. Data analyzed revealed that the mutation locus affected production traits mostly by additive effects. Based on these results, it is necessary to do more studies on LPL gene before making the LPL locus into the application of marker-assisted selection (MAS) programs.

돼지 5품종에 있어서 mtDNA ND2 유전자의 선택적 개시코돈의 특성과 빈도 (Characteristics and Frequencies of Alternative Initiation Codon(AIC) of mtDNA ND2 in Five Pig Breeds)

  • 다양한 포유동물의 미토콘드리아 유전자들에서 선택적 개시코돈(AIC)들이 보고되었다. 본 연구는 돼지 5 품종에서 mtDNA ND2 유전자의 AIC 양상을 분석하였다. 두 종류의 AIC 서열들이 발견되었고, 품종 집단간에서 각기 다른 출현빈도를 보였다. ND2의 methionine codon으로 Large White와 Landrace는 각각 전 개체에서 ATA와 ATT 서열로 확인되었다. 다른 3 품종(Berkshire, Duroc, Hampshire)의 경우는 두 서열이 모두 발생하였으며, 91.9, 21.3, 60.0%의 빈도로 ATA를 보였다. 기존의 연구들에 의하면 중국재래돼지들이 모두 개시코돈 ATA를 갖는다고 보고되어 있다. 미토콘드리아 ND2 단백질의 합성과정에서 AIC가 어떤 영향을 미치는지는 설명할 수 없으나, 본 연구에서 나타난 AIC 다형성과 품종특이적인 분포양상은 돼지의 육종에 있어 모계추적을 위한 분자적 표지인자로서 이용될 수 있을 것을 사료된다.

초위성체 표지를 이용한 한국재래돼지 집단의 분자유전학적 고찰 (Molecular Genetic Evaluation of Korean Native Pig Populations Based on Microsatellite Markers)

  • 초위성체(MS) 표지를 이용하여 한국재래돼지 집단의 각각의 분자유전학적 특성을 조사하고 그 평가를 통해 한국재래돼지에 대한 품종 및 계통분류의 기초를 마련하고자 본 연구를 수행하였다. 또한, 각 재래돼지 집단 내 및 집단간의 유전적 변이성을 확인하고, 그 분류 및 특성평가를 위한 MS 분석체계를 마련하여 국내 가축 유전자원 관리에 활용하고자 하였다. 국내 관리기관 및 농가에서 보유하고 있는 6개 재래돼지 집단을 중국의 4개 재래돼지 집단 및 외래종 돼지 7개 집단과 함께 분석하였다. 도합 17집단 648두를 대상으로 26개 MS 표지로 분석한 결과, 한국재래돼지 집단은 외래종과 중국재래돼지로부터 분자유전학적으로 별개의 집단으로 확연히 구분되는 것을 확인하였다. 한국재래돼지 집단의 기대이형접합도($H_E$)는 0.65의 값을 보인 두 집단(B, D)을 제외한 나머지에서 0.48~0.55의 수준을 보여 전반적으로 외래종에 비해 낮았다. 한국재래돼지 집단간의 유전거리 또한 0.12~0.34 정도로 비교대상에 비하여 낮았다. 분석대상 한국재래돼지 6집단 중 세 개의 집단은 높은 유전적 균일도를 보였으나, 두 집단에서는 일부 집단의 혼입을, 나머지 하나의 집단에서는 둘 이상의 집단으로부터의 복잡한 혼입이 의심되는 매우 낮은 유전적 균일도를 확인하였다. 본 연구를 통하여 한국재래돼지 집단간의 유전적 차이 및 동질성, 그리고 집단내의 유전적 균일성을 확인하였다. 이러한 결과는 국내유전자원의 고유성을 인정할 수 있는 과학적인 근거로서, 국가수준의 가축유전자원 평가, 관리의 기초자료로 활용될 수 있을 것이다.

Tissues Expression, Polymorphisms Identification of FcRn Gene and Its Relationship with Serum Classical Swine Fever Virus Antibody Level in Pigs

  • Neonatal Fc receptor (FcRn) gene encodes a receptor that binds the Fc region of monomeric immunoglobulin G (IgG) and is responsible for IgG transport and stabilization. In this report, the 8,900 bp porcine FcRn genomic DNA structure was identified and putative FcRn protein included 356 amino acids. Alignment and phylogenetic analysis of the porcine FcRn amino acid sequences with their homologies of other species showed high identity. Tissues expression of FcRn mRNA was detected by real time quantitative polymerase chain reaction (Q-PCR), the results revealed FcRn expressed widely in ten analyzed tissues. One single nucleotide polymorphism (SNP) (HQ026019:g.8526 C>T) in exon6 region of porcine FcRn gene was demonstrated by DNA sequencing analysis. A further analysis of SNP genotypes associated with serum Classical Swine Fever Virus antibody (anti-CSFV) concentration was performed in three pig populations including Large White, Landrace and Songliao Black pig (a Chinese indigenous breed). Our results of statistical analysis showed that the SNP had a highly significant association with the level of anti-CSFV antibody (At d 20; At d 35) in serum (p = 0.008; p = 0.0001). Investigation of expression and polymorphisms of the porcine FcRn gene will help us in further understanding the molecular basis of the antibody regulation pathway in the porcine immune response. All these results indicate that FcRn gene might be regarded as a molecular marker for genetic selection of anti-CSFV antibody level in pig disease resistance breeding programmes.

Single nucleotide polymorphism-based analysis of the genetic structure of Liangshan pig population

  • Objective: To conserve and utilize the genetic resources of a traditional Chinese indigenous pig breed, Liangshan pig, we assessed the genetic diversity, genetic structure, and genetic distance in this study. Methods: We used 50K single nucleotide polymorphism (SNP) chip for SNP detection of 139 individuals in the Liangshan Pig Conservation Farm. Results: The genetically closed conserved population consisted of five overlapping generations, and the total effective content of the population (Ne) was 15. The whole population was divided into five boar families and one non-boar family. Among them, the effective size of each generation subpopulation continuously decreased. However, the proportion of polymorphic markers (PN) first decreased and then increased. The average genetic distance of these 139 Liangshan pigs was 0.2823±0.0259, and the average genetic distance of the 14 boars was 0.2723±0.0384. Thus, it can be deduced that the genetic distance changed from generation to generation. In the conserved population, 983 runs of homozygosity (ROH) were detected, and the majority of ROH (80%) were within 100 Mb. The inbreeding coefficient calculated based on ROH showed an average value of 0.026 for the whole population. In addition, the inbreeding coefficient of each generation subpopulation initially increased and then decreased. In the pedigree of the whole conserved population, the error rate of paternal information was more than 11.35% while the maternal information was more than 2.13%. Conclusion: This molecular study of the population genetic structure of Liangshan pig showed loss of genetic diversity during the closed cross-generation reproduction process. It is necessary to improve the mating plan or introduce new outside blood to ensure long-term preservation of Liangshan pig.
