Exogenous Bio-Based 2,3-Butanediols Enhanced Abiotic Stress Tolerance of Tomato and Turfgrass under Drought or Chilling Stress

  • Among abiotic stresses in plants, drought and chilling stresses reduce the supply of moisture to plant tissues, inhibit photosynthesis, and severely reduce plant growth and yield. Thus, the application of water stress-tolerant agents can be a useful strategy to maintain plant growth under abiotic stresses. This study assessed the effect of exogenous bio-based 2,3-butanediol (BDO) application on drought and chilling response in tomato and turfgrass, and expression levels of several plant signaling pathway-related gene transcripts. Bio-based 2,3-BDOs were formulated to levo-2,3-BDO 0.9% soluble concentrate (levo 0.9% SL) and meso-2,3-BDO 9% SL (meso 9% SL). Under drought and chilling stress conditions, the application of levo 0.9% SL in creeping bentgrass and meso 9% SL in tomato plants significantly reduced the deleterious effects of abiotic stresses. Interestingly, pretreatment with levo-2,3-BDO in creeping bentgrass and meso-2,3-BDO in tomato plants enhanced JA and SA signaling pathway-related gene transcript expression levels in different ways. In addition, all tomato plants treated with acibenzolar-S-methyl (as a positive control) withered completely under chilling stress, whereas 2,3-BDO-treated tomato plants exhibited excellent cold tolerance. According to our findings, bio-based 2,3-BDO isomers as sustainable water stress-tolerant agents, levo- and meso-2,3-BDOs, could enhance tolerance to drought and/or chilling stresses in various plants through somewhat different molecular activities without any side effects.

UV-A LED을 이용한 고추 묘의 저온 내성 유도 (Elicitation of Chilling Tolerance of Pepper Seedlings Using UV-A LED)

  • BACKGROUND: After transplanting, the recent abnormal low temperature caused physiological disorders of pepper seedlings. This study was conducted to evaluate the effects of UV-A LED, a physical elicitor, on the chilling tolerance of pepper seedlings. METHODS AND RESULTS: Seedlings were continuously irradiated with 370 and 385 nm UV-A LEDs with 30 W·m-2 for 6 d. After that, seedlings were exposed to 4℃ for 6 h and then recovered under the normal growing condition for 2 d. There were no significant differences in growth characteristics of UV-A treatments compared to the control. Fv/Fm values of two UV-A treatments were below 0.8. Electrolyte leakage in the control was increased by chilling stress, while 385 nm UV-A had the significantly lowest value. Total phenolic content and antioxidant capacity of two UV-A treatments significantly increased due to UV-A radiation. However, total phenolic content and antioxidant capacity of the control increased due to chilling stress and tended to decrease again during the recovery time. CONCLUSION: We confirmed that UV-A light was effective to induce the chilling tolerance of pepper seedling, and the supplemental radiation of 385 nm UV-A LED before transplanting could be used as a cultivation technique to produce high quality pepper seedlings.

Resistance of Cucumber Grafting Rootstock Pumpkin Cultivars to Chilling and Salinity Stresses

  • Grafting using a pumpkin (Cucurbita sp.) rootstock is an effective way to improve cucumber (Cucumis sativus) resistance to a combination of chilling and salinity stresses. We evaluated the tolerance of 15 pumpkin cultivars to chilling, salinity, and combined stresses at the germination and seedling stages. Selected plant characteristics, including germination rate, germination potential, germination index, plant height, stem thickness, fresh weight, and dry weight, were analyzed. We used the unweighted pair group method with arithmetic mean for cluster analyses to determine the stress tolerance levels of the pumpkin cultivars. The 15 cultivars were divided into three clusters: tolerant, moderately tolerant, and susceptible to stress treatments. The stress tolerances of all cultivars were variable in the germination and seedling stages, and most cultivars were not tolerant to individual treatments of chilling or salinity stresses at both stages. These results suggest that identifying suitable cultivars for use as rootstock during cucumber grafting should involve the evaluation of stress tolerance during different growth stages. Additionally, cultivars tolerant to chilling stress may not be tolerant to salinity stress; therefore, the choice of pumpkin rootstock should depend on where the grafted plant will be grown. Cultivars tolerant to a combination of chilling and salinity stresses may be useful as rootstock for cucumber grafting. Our findings may serve as reference material for choosing appropriate pumpkin rootstocks for cucumber grafting.

Cross-Tolerance and Responses of Antioxidative Enzymes of Rice to Various Environmental Stresse

  • In order to examine the cross-tolerance of two chilling-tolerant cultivars (Donganbyeo and Heukhyangbyeo) and two chilling-susceptible cultivars (Hyangmibyeo and Taekbaekbyeo) to salt, paraquat, and drought, changes of physiological response and antioxidant enzymes were investigated. The seedlings were grown in a growth chamber until the 4-leaf stage. The seedlings were exposed to chilling at $5^{\circ}C$ for 3 days. For drought treatment, the seedlings were subjected to drought by withholding water from plants for 5 days. For paraquat study, plants were sprayed with $300{\mu}M$ paraquat. For the salt stress, the seedlings were transferred to the Hoagland's nutrient solution containing 0.6% (w/v) NaCl for 4 days. Chilling-tolerant cultivars showed cross-tolerant to other stresses, salt, paraquat, and drought in physiological parameters, such as leaf injury, chlorophyll a fluorescence, and lipid peroxidation. The baseline levels of antioxidative enzyme activities, catalase (CAT) and peroxidase (POX) activities in chilling-tolerant cultivars were higher than in the chilling-susceptible cultivars. However, there were no differences in ascorbate peroxidase (APX) and glutathione reductase (GR) activities between chilling-tolerant and -susceptible cultivars in untreated control. CAT activity in chilling-tolerant cultivars was higher than that in chilling-susceptible cultivars during chilling, salt, and drought treatments, but not during paraquat treatment. However, other antioxidative enzymes, APX, POX, and GR activities showed no significant differences between chilling-tolerant and -susceptible cultivars during chilling, salt, paraquat, and drought treatments. Thus, it was assumed that CAT contribute to cross-tolerance mechanism of chilling, salt, and drought in rice plants.

Chilling Tolerance of Photosynthesis in Plants is Dependent on the Capacity to Enhance the Levels of the Xanthophyll Cycle Pigments in Response to Cold Stress

  • Plants possess the ability to dissipate the excitation energy for the protection of photosynthetic apparatus from absorbed excess light. Heat dissipation is regulated by xanthophyll cycle in thylakoid membranes of chloroplasts. We investigated the mechanistic aspects of xanthophyll cycle-dependent photoprotection against low-temperature photoinhibition in plants. Using barley and rice as chilling-resistant species and sensitive ones, respectively, chilling-induced chlorophyll fluorescence quenching, composition of xanthophyll cycle pigments and mRNA expression of the zeaxanthin epoxidase were examined. Chilled barley plants exhibited little changes in chlorophyll fluorescence quenching either of photochemical or non-photochemical nature and in the photosynthetic electron transport, indicating low reduction state of PS II primary electron acceptor. In contrast to the barley, chilled rice showed a marked decline in those parameters mentioned above, indicating the increased reduction state of PS II primary electron acceptor. In addition, barley plants were shown to have a higher capacity to elevate the pool size of xanthophyll cycle pigments in response to cold stress compared to rice plants. Such species-dependent regulation of xanthophyll cycle activity was correlated with the gene expression level of cold-induced zeaxanthin epoxidase. Chilled rice plants depressed the gene expression of zeaxanthin epoxidase, whereas barley increased its expression in response to cold stress. We suggest that chilling-induced alterations in the pool size of xanthophyll cycle pigments related to its capacity would play an important role in regulating plant's sensitivity to chilling stress.

CHLOROPHYLL FLUORESCENCE IN CUCUMBER (Cucumis safivus L.) AND PEA (Pisum sativum L.) LEAVES UNDER CHILLING STRESS IN THE LIGHT AND DURING THE SUBSEQUENT RECOVERY PERIOD

  • To investigate the chilling sensitivity related injuries in the photosynthetic apparatus of cucumber leaves, the light-chilling induced alterations of chlorophyll fluorescence transients in cucumber leaves were compared with those in pea leaves. As an early effect of light-chilling, an increase in Fp/Fm$^*$ was observed in both pea and cucumber leaves, which was saturated by about 6 h chilling. However, the saturated value of Fp/Fm was almost 1.0 in cucumber, in contrast to about 0.8 in pea. During the recovery period after 24 h chilling, the light-chilling induced changes in pea seemed to be reversed, but those in cucumber leaves were thought to be irreversible, because Fo was increased significantly. Light-chilling caused significant decreases in qQ and qE in cucumber leaves, but qR was increased until 6 h, and decreased thereafter. In both pea and cucumber leaves, Fm was increased by 2 h dark treatment. The Fm from the predarkened pea leaf discs was higher than the value from the preilluminated ones during the whole period of light-chilling (500 $\mu$mol m$^{-2}$s$^{-1}$ PAR). However, the predarkened cucumber leaf discs showed a reduction in Fm and an increase in Fo during the 2 h chilling in the light. These results indicate that the causes of chilling sensitivities in photosynthetic apparatus of cucumber leaves are possibly related with the damage in PSI reaction center and the ability of acidification of lumen by PSII.

Mechanisms of Chilling Tolerance in Relation to Antioxidative Enzymes in Rice

  • In order to examine the mechanistic basis for differential sensitivities to chilling and subsequent recovery between two rice (Oryza sativa L.) cutivars, a chilling-tolerant japonica type (Ilpumbyeo) and a chilling-susceptible indica type (Taebaekbyeo), changes of physiological responses and antioxidant enzymes were investigated. Both cultivars at 3 leaf stage were exposed at a low temperature of $5^{\circ}C$ for 3 days and subsequently recovered in a growth chamber at a $25^{\circ}C$ for 5 days with 250 mmol $m^{-2}$ $s^{-1}$. Physiological parameters such as leaf fresh weight, relative water content, cellular leakage, lipid peroxidation, and chlorophyll a fluorescence showed that the chilling tolerant cultivar had a high tolerance during chilling. However, the chilling-susceptible cultivar revealed severe chilling damages. The chilling-tolerant cultivar was also faster in recovery than the chilling-susceptible cultivar in all parameters examined. We analyzed the activity and isozyme profiles of four antioxidant enzymes which are: superoxide dismutase (SOD), caltalase (CAT), ascorbate peroxidase (APX), and glutation reductase (GR). We observed that chilling-tolerance was due to a result of the induced or higher antioxidant enzyme system, CAT and APX in leaves and SOD, CAT, APX, and GR in roots. Especially, we observed the most significant differences between the chilling-tolerant cultivar and -susceptible cultivar in CAT and APX activity. Also in isozyme profiles, CAT and APX band intensity in the chilling-tolerant cultivar was distinctively higher than in the chilling-susceptible cultivars during chilling and recovery. Thus, the cold stability of CAT and APX are expected to contribute to a tolerance mechanism of chilling in rice plants. In addition, the antioxidative enzymes activity in roots may be more important than in that of leaves to protect chilling damage on rice plants.

Effect of Salicylic Acid on Growth and Chilling Tolerance of Cucumber Seedlings

  • The present study was undertaken to investigate the effect of low temperature and salicylic acid(SA) on the chilling tolerance of acclimated and nonacclimated cucumber(Cucurmis sativus L.) seedlings. The acclimation phenomenon was characterized in chilling-sensitive cucumber seedlings and found to have a significant effect on the survival and shoot dry weights. The injuries experienced by the acclimated seedlings in the third leaf stage were on average smaller by half than those experienced by the nonacclimated seedlings. Chilling also caused a large increase in the free proline levels, regardless of the acclimation status. Exogenous treatment with SA(0.5mM) resulted in improved growth and survival of the nonacclimated chilled seedlings, indicating that SA induced chilling tolerance and SA and acclimation had common effects. The application of cycloheximide in the presence of SA restored the acclimation-induced chilling tolerance. The elevated proline level observed in the cold-treated and SA-treated plants was more pronounced in the light than in the dark at a chilled temperature, indicating that endogenous proline may play a role in chilling tolerance by stabilizing the water status in response to chilling. From these results it is suggested that SA provided protection against low-temperature stress by increasing the proline accumulation, and pre-treatment with SA may induce antioxidant enzymes leading to increased chilling tolerance.

외생 살리실산과 일산화질소 처리가 고추묘의 저온 내성 및 병 저항성에 미치는 영향 (Effect of Exogenous Application of Salicylic Acid or Nitric Oxide on Chilling Tolerance and Disease Resistant in Pepper Seedlings)

  • 저온은 식물 생장을 저해하는 주된 요인이며 병원균에 대한 감수성을 증가시킨다. 그러므로 식물체에서 스트레스 내성을 증대시키는 것은 불리한 환경 조건에서 살아남기 위한 중요한 전략이다. 본 실험의 목적은 고추 묘에서 저온 내성과 식물병 발생에 대한 외생 살리실산(SA)과 일산화질소(NO) 처리의 효과를 밝히는 것이다. 정식 후 23일 동안 고추 묘(Capsicum annuum L. '기대만발')는 온도 $20/25^{\circ}C$(낮/밤), 광주기 15시간, 광도 $145{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$의 정상적인 생육환경에서 자랐다. 1주일에 2번 계면활성제 0.1%를 포함한 SA와 NO 3mL을 고추 묘에 각각 분사해주었다. 처리 후 고추 묘는 암 상태에서 6시간 동안 $4^{\circ}C$ 저온에 노출시킨 후 정상적인 생육환경에서 2일 동안 회복시켜주었다. 저온 스트레스에 대한 식물 내성을 평가하기 위해 저온 처리 후 생육특성, 엽록소 형광 값, 세포막 투과성을 측정하였다. 총 페놀릭 농도와 항산화도는 실험하는 동안 측정하였다. 또한, 고추의 점무늬병과 풋마름병 발생 정도도 조사하였다. 저온 처리 전 후를 비교하여 대조구 고추묘에서는 저온에 의해 상대적으로 많은 수분을 손실하여 건물율이 높지만 SA와 NO 처리 된 고추 묘는 비슷한 건물율을 유지하였다. 저온 처리 후 대조구에 비해 SA와 NO 처리구의 전해질 유출 값은 더 낮았다. 저온 처리 동안 SA와 NO 처리구의 엽록소 형광값은 약 0.8 수준으로 유지하였지만 대조구는 빠르게 감소하였다. 화학적 처리 동안 SA 처리구의 총 페놀릭 농도와 항산화도는 NO 처리구보다 높았다. 또한 저온 처리 후 대조구와 NO 처리구의 총 페놀릭 농도는 증가하였다. 고추에서 풋마름병에 대한 저항성은 SA가 보다 효과적이었다. 본 실험의 결과는 SA와 NO의 외생처리는 고추 묘의 저온 내성을 증대시켰고 병 발생 정도를 감소시키는 데 효과적이었음을 보여준다.
