밤 속껍질로부터 기능성 음료의 개발(III) -뇌혈류역학, 평균혈압, 흉선세포 증식율에 미치는 효과- (The Development of Functional Beverage from the Inner Skin of Chestnut Castanea crenata ( In ) Effects on the Regional Cerebral Blood Flow, Mean Arterial Blood Pressure, Proliferation of Thymocytes -)

  • The purpose of this study was to investigate effects of inner skin of chestnut on the activation of a living body's function (regional cerebral blood flow and mean arterial blood pressure in Sprague-Dawley rats, proliferation of thymocytes in normal mice and L1210 cells transplanted mice) . We used inner skin of chestnut extract(Sample A : inner skin of chestnut-panbroiled after driedextract (100$\^{C}$ ), Sample B , inner skin of chestnut-panbroiled-extract(100$\^{C}$ ) , Sample C : inner skin of chestnut -panbroiled after dried-extract(80$\^{C}$ ), Sample D : inner skin of chestnut-panbroiled-extract(80$\^{C}$)} Regional cerebral blood flow(rCBF) and Mean arterial blood pressure(MABP) were tested using Leser -Doppler Flowmetry(LDF), and the proliferation of thymcytes was tested using a colorimetric tetrazoliun assay ( MTT assay) The experimental results as follows 1. rCBF was significantly increased by Sample C in a dose-dependent manner. 2. MABP was not changed by Sample C in a 0.1mg/kg∼10.0mg/kg treated group. 3. Proliferation of thymocytes was not changed by Sample C in normal mice. 4. Proliferation of thymocytes was significantly accelerated by Sample C in L1210 cells transplanted mice.

밤 박피 시스템 개발 (Development of Chestnut Peeling System)

  • The chestnut is a well-known and important forest product in Korea. The annual production of chestnut is about 100, 000tons and its cultivating area is 80, 000ha. However, the peeling process of outer and inner skins of chestnut is very difficult due to hardness and adhesiveness of chestnut skin. The peeling process of chestnut was operated by manual work and the performance of chestnut peeling machine is very low. The purpose of this study is to develope the prototype of new chestnut peeling system. The hardness of chestnuts was tested with six different drying conditions and its range was from 949$g/mm^2$ to 2, 149$g/mm^2$. The hardness of chestnuts was decresed gradually during the drying process. The chestnut peeling Process includes sorting, storage, drying, outer skin cutting, flame peeling, continuous frictional skin peeling, and inner skin cutting operation. The developed chestnut peeling system consists of outer skin cutter, flame peeler, continuous frictional skin peeler and inner skin cutter. The system can peel domestic chestnuts at 150$kg/hr$ with peeling rate of 78%.

  • 본 연구는 밤가공 과정에서 폐기물로 나오는 밤껍질에서 분리한 밤 가루의 이화학적 특성을 규명하고, 식품 자원으로의 이용가능성을 검토해보고자 빵, 국수, 묵 등 식품을 제조하여 기호성과 관능검사를 실시하였다. 밤 가루의 물결합능력은 순수한 밤에서 분리한 것보다 껍질에서 분리한 것이 약 40%정도 높았으며 요드반응은 유사한 상태를 보였으며 amylose 분해한도는 밤 껍질에서 분리한 밤가루가 약 8.0%높았다. 밤 가루로 만든 빵, 국수, 묵 등은 냄새, 경도, 탄력성, 응집성, 부착성, 씹힘성, 검성 등에서 밀가루나 도토리로 만든 것과 유의적 차이가 없었다.

Characterization of Antioxidant Activities from Chestnut Inner Skin Extracts

  • Total phenolics and antioxidant activities of water and 80% methanol extract of chestnut inner skin were investigated. The antioxidant properties of both extracts of chestnut inner skin were evaluated using different antioxidant tests, including 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activities, reducing power, ferric reducing ability of plasma (FRAP), and inhibitory effects on ${\beta}$-carotene bleaching. The 80% methanol extract of inner skin exhibited the higher DPPH, ABTS radical scavenging activities, reducing power, and FRAP than water extract of inner skin and did also in a concentration-dependent manner. However, inhibitory effects on ${\beta}$-carotene bleaching of 80% methanol extract was similar to those of water extract, 35.09 and 39.07% at 1 mg/mL, respectively. The total phenolic contents of water and 80% methanol extract from chestnut inner skin were 5,801.42 and 9,735.56 mg/100 g, respectively. High performance liquid chromatography (HPLC) analysis showed that gallic acid was the predominant phenolic compound in water and 80% methanol extract from inner skin. These water and 80% methanol extracts of chestnut inner skin can be utilized as an effective and safe source of antioxidants.

Effects of the Chestnut Inner Shell Extract on the Expression of Adhesion Molecules, Fibronectin and Vitronectin, of Skin Fibroblasts in Culture

  • The inner shell of the chestnut (Castanea crenata S. et Z., Fagaceae) has been used as an anti-wrinkle/skin firming agent in East Asia, and preliminary experiments have found that a 70% ethanol extract from this plant material can prevent cell detachment of skin fibroblasts from culture plates. In order to examine the molecular mechanisms underlying this phenomenon, its effects on the expression of adhesion molecules, such as fibronectin and vitronectin, were investigated using the mouse skin fibroblast cell line, NIH/3T3. Using fixed-cell ELISA, Western blotting and immunofluorescence cell staining, it was clearly demonstrated that the chestnut inner shell extract enhanced the expression of the cell-associated fibronectin and vitronectin. Scoparone (6,7-dimethoxycoumarin), isolated from the extract, also possessed similar properties. These findings suggest that the enhanced expression of the adhesion molecules may be one of the molecular mechanisms for how the chestnut inner shell extract preventing cell detachment and may be also responsible for its anti-wrinkle/skin firming effect.

우피의 울금 염색시 사용된 율피의 매염 효과 (Mordanting effect of Chestnut's inner skin on the cow leather dyed with Turmeric powder)

  • This research was carried out to find the mordant effect of Chestnut's inner skin on the cow leather dyed by Turmeric powder. The best proper mordanting conditions were examined by changing mordant method, concentration, temperature, bath ratio, time, and repetition. Also dyeability and surface color changes were evaluated by various mordanting methods. The optimum mordanting conditions of chestnut's inner skin extract on the cow leather were pre mordant, 80%, $50^{\circ}C$, 50:1, 40minutes, 4 repetition. The K/S values as a mordant were higher in pre mordant than post mordanting condition. Although the ${\Delta}E$ was slightly higher in post mordant than pre mordant, it was too small to find any means. The surface colors of all dyed cow leathers were yellow. Among light, dry cleaning, and abrasion fastnesses, only light fastness was increased one degree, others were same degrees.

  • 결명자차, 녹차 및 밤차가 인체의 생체기능활성화에 미치는 영향을 알아보기 위하여 국소뇌혈류량 및 혈압, in vitro상에서의 면역세포의 활성화, 그리고 암세포가 이식된 동물에서의 면역세포 활성화와 암세포의 증식억제 효과를 관찰한 결과 다음과 같은 결론을 얻었다. 1. 단당류는 현미녹차와 결명자차에는 glucose, ga-lactose등이 들어 있으나 밤차에는 glucose, gal-actose, mannose가 들어 있다. 2. 아미노산은 현미녹차, 밤차, 결명자차 순으로 들어 있다. 3. 카페인은 현미녹차에는 들어 있으나 밤차와 결명자차에는 들어 있지 않았다. 4. 결명자 차는 국소뇌혈류량을 증가시킨 반면 밤차는 감소시켰다. 5. 녹차는 혈압을 증가시켰다. 6. 밤차는 in vitro 상에서 흉선세포와 비장세포의 증식을 유의성있게 증가시켰다. 7. 결명자차와 녹차는 in bitro 상에서 흉선세포의 증식을 감소시켰다. 8. 결명자차와 녹차는 L1210세포가 이식된 동물의 흉선세포 증식을 감소시켰다. 9. 밤차는 L1210세포가 이식된 동물의 비장세포 증식을 촉진시켰다. 10. 밤차와 결명자차는 L1210세포가 이식된 동물의 암세포 증식을 억제하였다.

율피.솔잎.호프의 수성 아세톤 추출물에 의한 Melanin 생성 억제 효과 (Inhibitory Effects of Water-Acetone Extracts of Chestnut inner shell, Pine needle and Hop on The Melanin Biosynthesis)

  • The skin whitening effects of pine needle extract, hop extract and chestnut inner shell extract were evaluated both in vitro and in B 16 mouse melanoma cell lines. Each extracts significantly inhibited tyrosinase activity, dopa auto-oxidation and melanin biosynthesis in vitro and in B 16 cell lines. In vitro, hop extract inhibited melanin biosynthesis 15 times stronger than kojic acid at $10{\;}\mu\textrm{g}/ml$ concentration. Each extracts were stronger inhibitors of melanin biosynthesis than kojic acid in B 16 mouse melanoma cell at less than $4{\;}\mu\textrm{g}/ml$ concentration. These results show that extracts fo pine needle, hop and chestnut inner shell could be developed as skin whitening component of cosmetics.

항산화 활성을 갖는 율피 추출물의 향장 소재 특성 (Characterization as Cosmetic Substances of Chestnut Inner Skin Extracts with Antioxidant Activity)

  • In vitro 항산화 활성을 갖는 율피 추출물의 향장 소재 특성을 조사하였다. 율피로부터의 다양한 추출물 중 총 페놀성 화합물 함량은 60% 메탄올 추출물 및 에틸아세테이트 분획물이 각각 164.82 및 191.14 mg/g으로 가장 높은 것으로 나타났다. 또한, 상기 두 분획물에 의한 in vitro 항산화 활성(DPPH radical scavenging activity, lipid peroxidation 저해)은 농도 의존적으로 증가한다는 것을 확인하였다. 상기 두 분획물은 UV-B(290-320 nm) 영역에서 강한 UV 흡수도를 보여주었다. 특히, 율피 유래 두 분획물은 $1000{\mu}g/mL$에서 42.88 및 33.02%의 elastase 저해 활성을 나타내어 우수한 주름 개선 특성을 가지는 것을 보여주었다. 아울러, 율피로부터의 60% 메탄올 추출물 및 에틸아세테이트 분획물은 피부 상재균에 대하여서도 항균 활성을 지니는 것으로 나타났다. 이러한 결과들은 율피에서 추출될 수 있는 물질이 향장 산업소재로서 활용될 수 있는 가능성을 제시하는 것으로 판단된다.
