방울토마토(Lycopersicon esculentum Mill.) 생산단계에서 Fluopicolide 및 Metrafenone의 잔류허용기준 설정 (Establishment of Pre-Harvest Residue Limits (PHRLs) of Fluopicolide and Metrafenone in Cherry Tomato (Lycopersicon esculentum Mill.))

  • 본 연구는 생산단계 방울토마토 중 fluopicolide 및 metrafenone의 경시적 잔류변화를 조사하여 반감기를 산출함으로서 생산단계 잔류허용기준을 설정하고자 수행하였다. Fluopicolide 및 metrafenone 농약을 안전사용기준에 준하여 각각 포장 1, 포장 2 지역으로 나누어 기준량 처리하였으며, 약제처리 2시간 후를 0일차로하여 0, 1, 2, 3, 5, 7, 10일차에 시료를 채취하여 분석하였다. 두 약제 모두 LC-MS/MS를 이용하여 분석하였으며, MLOQ(Method Limit of quantitation)는 0.005 mg/kg이었다. 분석결과, fluopicolide 회수율은 92.7∼94.8%이었고, metrafenone의 회수율은 82.6 ∼88.0%이었으며, 두 약제 모두 표준편차는 10% 미만이었다. 방울토마토 중 각각의 농약에 대한 반감기는 fluopicolide의 경우 15.0일(포장 1)과 12.8일(포장 2)로 나타났으며, metrafenone의 경우 18.9일(포장 1), 21.5일(포장 2)로 나타났다. fluopicolide 및 metrafenone에 대하여 방울토마토 수확 10일 전 잔류량이 각각 0.27 mg/kg, 2.29 mg/kg이하로 나타난다면 수확 시 잔류량은 MRL 수준 이하일 것으로 판단된다.

원적외선 난방시스템이 방울토마토 생육에 미치는 영향 (Growth Characteristics of Cherry Tomato in Greenhouse using Far Infrared Heating Systems)

  • This study was conducted to investigate the growth characteristics of cherry tomatoes in greenhouse using far infrared heating system. The far infrared greenhouse heating systems were installed in two ways on the greenhouse side wall and at the greenhouse ceiling. The heating characteristics of far infrared heating system were analyzed by investigating the heating load, internal temperature, energy consumption, growth characteristics and quality evaluation. The results were compared with heated air heating system using kerosene. The results showed that tomatoes grown in the greenhouse with the far infrared heating system had relatively better plant height, leaf length, leaf width, stem diameter than ones from the greenhouse with hot air heating system and both heating methods had no significant difference on Cherry tomato sugar contents. At the same time, the far infrared heating system reduced heating cost from 34.5 to 41.4% on comparing with hot air heating system.

New Antioxidant Sources; Tinged Autumnal Leaves of Maple and Cherry Trees

  • Living systems constantly suffer from atmospheric reactive oxygen species and also produce inevitably them by the course of metabolism. Therefore, antioxidants play important roles in protecting the systems from various diseased caused by them. In this study, we investigated various tinged autumnal leaves as antioxidant sources. Among the, the red leaf extracts of Acer Palmatum THUNBERG.(Aceraceae: maple tree) and Prunus Donarioum Sieb. Var. spontanea Makino(Rosaceae : cherry trees) showed the highest anti-oxidativities. The major antioxidants in their red leaves were isolated and identified as vitexin from maple leaves and isoscutellarein-4'-O-$\beta$-glucopyranoside from cherry leaves. Finally, we evaluated the efficacy of skin care products containing the extracts by human use study. As a result, the tinged leaves of maple and cherry trees were evaluated as good antioxidant sources on the bases of antioxidativities, cytotoxicities, cell proliferation effects and human use study.

RT-PCR Detection of Three Non-reported Fruit Tree Viruses Useful for Quarantine Purpose in Korea

  • A simple and reliable procedure for RT-PCR detection of Apple stem pitting virus (ASPV), Cherry rasp leaf virus (CRLV), and Cherry necrotic rusty mottle virus (CNRMV) was developed. Two virus specific primer sets for each virus were found to specifically detect each virus among fourteen sets of designed oligonucleotide primers. Total RNAs extracted from healthy and from ASPV-,CRLV- and CNRMV-infected plant tissues were used to synthesize cDNA using oligo dT primer and then amplified by virus-specific primers for each virus. Each primer specifically amplified DNA fragments of 578 bp and 306 bp products for ASPV (prAS CP-C and prAS CP-N primers, respectively); 697 bp and 429 bp products for CRLV (prCR4 and prCR5-JQ3D3 primers, respectively); and 370 bp and 257 bp products for CNRMV (prCN4 and prCN6-NEG 1 primers, respec-tively) by RT-PCR. DNA sequencing of amplified DNA fragments confirmed the nature of each amplified DNA. Altogether, these results suggest that these virus specific primer sets can specifically amplify viral sequences in infected tissues and thus indicate that they can be used for specific detection of each virus.

경골어류 잉어과 3종의 수정란 난막 미세구조 비교 (Comparative Ultrastructure of the Fertilized Egg Envelope in Three Species, Cyprinidae, Teleost)

  • The structures of the fertilizedegg envelope in threespecies of cyprinidae, leoparddanio (Brachydanio frankei), cherry barb (Barbustitteya) and white cloudmountain fish (Tanichthys albonubes) were investigated by light and electron microscopy. The fertilized eggs of all three species have colorless, transparent, spherical and non-floted type. The egg envelopes have a single micropylewhich is thought to the pathway of sperm in the area of the animal pole. Also, an outer layersof both cherry barband white cloud mountain fish areadhesive types, but in that of leoparddanio has non-adhesivetype. In leopard danio, an outer surface ofegg envelope is covered by mushroom-likestructure, that of cherry barb is arranged by button-like structure, and that of white cloud mountain fish isarranged by rod-likestructure. Inboth leopard danioand cherrybarb, the eggenvelopes consist of three distinct layers; an outer layer, a middle layer and an inner layer, and that of white cloud mountain fish hastwo layers. An innerlayer of egg envelopein leopard danio consist of 10 layers, cherry barb has 8 layers, and white cloud mountain fish has $5\sim6$ layers.

벚나무 열매의 색소 추출물에 의한 견 및 면섬유에 대한 염색성 및 견뢰도 (Dyeing and Fastness of Silk and Cotton Fabrics Dyed with Cherry Extract)

  • A natural colorants was extracted from cherry by 10wt% aqueous acetic acid solution as an extractant. Silk and cotton fabrics were dyed with the cherry extract in the temperature range of $40-80^\circ{C}$ and for the time range of 30-60min. by pre- and post-mordanting with various mordants, their dyeability and fastness were investigated. The natural cherry extract prepared in this study has a maximum absorbance at 520nm. It was found that the optimum dyeing temperature and time were $40^\circ{C}$ and 50min, respectively. The pre-mordanting method was more effective than post-mordanting. All mordants except $SnCl_2\cdot{2H}_2O\;and\;FeSO_4\cdot{7H}_2O$ were effective for silk fabrics. However, the dyeability on cotton fabrics increased in the order of $CrK(SO_4)_2\cdot{12H_2O>(CH_3COO)_2Cu\cdot{H}_2O>CuSO_4\cdot{5H}_2O>AlK(SO_4)_2\cdot{12H}_2O>FeSO_4\cdot{7H}_2O>SnCl_2\cdot{2H}_2O$ among the mordants used in this study Fastness(light, water, washing, perpspiration fastness) on the silk and cotton fabrics increased with using mordants. The post-mordanting using mordant$(CH_3COO)_2Cu\cdot{H}_2O$ among the various mordants in this study gave the best fastness.

방울토마토의 여름재배시 적정수경재배방식 구명 (The investigation of Appropriate Hydroponic System for Cherry Tomatoes in Summer Season)

  • 방울토마토에 적합한 수경재배방식에 관한 연구 결과, 담액수경에서 주경의 기부직경, 엽장, 엽폭, 화방길이 등의 생육이 가장 좋았으며, 암면재배와 NFT는 유사하였다. 개화시기와 과실의 착색일에 대하여는 각 방식마다 차이를 보이지 않았으나, 과실의 수확량은 담액수경에서 많았다. 건물율, 유기산, 경도, 당도 등은 담액수경에서 낮았고, 당산비와 vitamin C는 높아 담액수경에서 품질이 약간 우수한 것으로 나타났다. 따라서, 방울토마토의 여름재배에서는 담액수경방식이 바람직한 것으로 평가되었다.

Effect of Bacillus mesonae H20-5 on Fruit Yields and Quality in Protected Cultivation

  • A variety of microorganisms in rhizosphere affect plant health by plant growth promotion, mitigation of abiotic stresses as well as protection from pathogen attacks. In our previous study, we selected a bacterium, Bacillus mesonae H20-5, for alleviation of salinity stress in tomato plants. In this study, we verified the effect of a liquid formulation of B. mesonae H20-5 (TP-H20-5) on fruit production and phytochemical accumulation including lycopene and polyphenol in cherry tomato and strawberry fruits in on-farm tests of protected cultivation under salinity stress. When vegetables including tomato, cherry tomato, strawberry, and cucumber were treated with TP-H20-5 by irrigated systems, final marketable yields were increased by 21.4% (cherry tomato), 9.3% (ripen tomato), 120.6% (strawberry), and 14.5% (cucumber) compared to untreated control. Moreover, treatment of TP-H20-5 was showed increase of phytochemicals such as lycopene and total polyphenol compared to untreated control in cherry tomato and strawberry. Therefore, these results indicated that a formulant of B. mesonae H20-5 can be used as a potential biofertilizer for increasing fruit production and quality.

Rhamnogalacturonan I-rich fractions from cherry tomatoes stimulate phagocytosis in RAW 264.7 macrophages

  • Tomato (Lycopersicon esculentum) is widely known for its beneficial effects on human health. To investigate the beneficial effects of polysaccharides from cherry tomato, cherry tomato polysaccharides (CTP) were prepared, the component sugars were analyzed, and the immunomodulatory activities in RAW 264.7 macrophages were assessed. CTP mainly contained arabinose (Ara) and galactose (Gal), suggesting that CTP might be enriched with an arabinogalactan (AG) moiety. The Ara and Gal present in CTP are likely components of AG-II (35.4%), namely $arabino-{\beta}-(3,6)-galactan$. To investigate the immunomodulatory activity of CTP, cytokine levels and iNOS2, COX-2, and $NF-{\kappa}B$ protein levels were analyzed, and $NF-{\kappa}B$ nuclear translocation and phagocytosis were observed by immunofluorescence. CTP significantly increased the levels of $TNF-{\alpha}$, MCP-1, and IL-6. CTP also increased iNOS2 and COX-2 expression as well as $NF-{\kappa}B$ nuclear translocation in RAW 264.7 cells. CTP significantly stimulated phagocytosis activity. These results showed that CTP stimulates macrophage activity, which can boost the innate immune response. CTP with high AG-II content could be used as a prebiotic to strengthen immunity.

벚나무 추출물의 OVA 유도 천식동물모델에서 항염증 효능 (Ant-Inflammatory Effect of Prunus serrulata var. spontanea Extract in OVA-Induced Asthma Animal Model)

  • The objective of this study was to determine the efficacy of a natural product of cherry tree (Prunus serrulata var. spontanea: PS) as a test substance for improving cytokine and ovalbumin-specific IgE using an ovalbumin-induced asthma disease model of 5-week-old male BALB/c mice. Lung tissue pathology was analyzed to confirm anti-inflammatory and asthmatic effects. As a result of examining the effect on changes in inflammatory cells in bronchoalveolar lavage fluid in an ovalbumin-induced asthma disease model by administering the PS sample, total cells, eosinophil, neutrophil, lymphocyte, and monocytes were significantly decreased. Concentrations of cytokine-based TNF-alpha and IL-4 and immunoglobulin E in serum were significantly increased in the asthma-inducing negative control group than in the normal group. However, high concentrations of PS decreased them. In histopathological examination of the lung tissue, it was confirmed that inflammatory cells infiltrated around the alveoli and bronchioles were increased in ovalbumin-induced asthma disease model. After administration of cherry tree extract, bronchiolar morphological changes such as mucosal thickening were slightly improved. From the above results, it was confirmed that extract of cherry tree significantly reduced inflammation expression and tissue damage in alveolar tissues. It was also confirmed that the cherry tree extract had an excellent efficacy in improving asthma inflammation.
