Clinical Application of Serum Tumor Associated Material (TAM) from Non-small Cell Lung Cancer Patients

  • Objective: To explore the associations of serum tumor associated material (TAM) with other common tumor markers like carcinoembryonic antigen (CEA), carbohydrate antigen 125 (CA125), carbohydrate antigen19-9 (CA19-9) and its clinical application in non-small cell lung cancer (NSCLC) patients. Methods: A total of 87 patients were enrolled into this study, all with histologically or cytologically confirmed NSCLC. With the method of chemical colorimetry, the level of TAM was determined and compared, while chemiluminescence was used to measure the levels of common tumor markers. Results: The level of TAM decreased after chemotherapy compared with before chemotherapy when CT or MRI scans showed disease control. Furthermore, it increased when disease progessed and there was no statistically significant difference in monitoring of TAM and common tumor markers (P>0.05). Conclusions: Detecting TAM in NSCLC patients has a higher sensitivity and specificity, so it can be used as an indicator for clinical monitoring of lung cancer chemotherapy.

Superoxide Formation and Cytotoxicity of RAW264.7 Macrophages Induced by Nitric Oxide

  • We have studied cytotoxicity of S-nitroso-N-acetyl- N-DL-penicillamine(SNAP), a Nitric oxide (NO)-releasing compound, in RAW264.7 macrophages. SNAP is cytotoxic to RAW264.7 cells in a concentration-dependent manner. PMA(200 nM) stimulated cells to produce superoxide anton radical($O_2^{-\cdot}$) and caused a little loss of RAW264.7 cell viability for 12 hr and diminished the cytotoxicity of SNAP. The mechanism by which PMA can protect cells against NO-mediated cytotoxicity was studied by peroxynitrite-enhanced chemiluminescence method. Observed results suggested that $O_2^{-\cdot}$ produced by PMAstimulated RAW264.7 cells may quench NO released by SNAP and reduce NO, thus attenuating NO-related damages.

  • The HCCI combustion mode poses its own set of narrow engine operating by knocking. In order to solve this, inhomogeneity method of mixture and temperature is suggested. The purpose of this research is to get fundamental knowledge about the effect of thermal stratification on HCCI combustion of PRF -Air mixture. The temperature stratification is made by buoyancy effect in combustion chamber of RCM. The analysis items are pressure, temperature of in-cylinder gas and combustion duration. In addition, the structure of flames using the two dimensional chemiluminescence's images by a framing camera are analyzed. Under stratification, the LTR starting time and the HTR starting time are advanced than that of homogeneous. Further, the LTR period of homogeneous conditions became shorter than that of the stratified conditions. With the case of homogeneous condition, the luminosity duration becomes shorter than the case of stratified condition. Additionally, under stratified condition, the brightest luminosity intensity is delayed longer than at homogeneous condition.

Optimal Conditions for Propagation in Bottom and Top Brewing Yeast Strains

  • The method of yeast propagation has an influence on yeast physiology, fermentation ability, flocculation rate, and taste stability of beer. In order to find optimal conditions for propagation, several parameters were investigated in combinations. The bottom brewing yeast grown at $10^{\circ}C$ indicated that a higher flocculation capacity during the $1^{st}$ fermentation. However, the taste stability and the aroma profile were not affected by parameters of propagation investigated. The beer quality was rather affected by storage duration. In addition, a correlation between tasting and chemiluminescence was found at the beer, which was produced using bottom brewing yeast. The propagation at $10-25^{\circ}C$ with addition of zinc ion indicated the best condition to improve fermentation ability, flocculation rate, and filterability for bottom brewing yeast, whereas the propagation at $30^{\circ}C$ with addition of zinc ion showed the best condition to increase fermentation ability for top brewing yeasts.

Biological Effects of Korean Puerariae Radix Catechins on the Liver Function in Rats Administrated with Ethanol

  • Crude catechin extracts were prepared using ethyl acetate from Korean Puerariae Radix (PR) and their biological effects on the alcoholic liver damage were investigated in male Sprague-Dawley rats (6 weeks old). Ethanol (5g ethanol/kg body weight/day) administration was performed for 8 weeks and after ethanol consuming rats were treated with one or two% catechin extracts of diet for 8 weeks. At the end of experimental period, lipid hydroperoxides in liver were analyzed using a chemiluminescence-high performance liquid chromatography (CL-HPLC) method. Compared with control animals, ethanol consumed rats showed lighter body weights, lower ratios of liver/body weight, higher activities of GOT and GPT, and increased lipid hydroperoxide amount in liver. With one or two% catechin extracts treatment, GOT and GPT activities returned to normal ranges. Lipid hydroperoxide contents in liver of on or two% PR treated rats lowered to 20% or 25% respectively, compared with the levels of those ethanol consumed animals without catechin extracts treatment. Therefore, we concluded that on or two% PR crude catechins treatment could be effective for alcoholic liver damage caused by lipid peroxidation.

  • A spectrophotometric method for the simultaneous determination of anionic and nonionic surfactant based on the application of multivariate calibration method such as principal component regression(PCR) and partial least squares(PLS) has been studied. The calibration models in PCR and PLS were obtained from the spectral data in the range of 400~700 nm for each standard of a calibration set of 26 standards, each containing different amounts of two surfactants. The relative standard error of prediction(RSEP$_{\alpha}$) was obtained to assess the model goodness in quantifying each analyte in a 5 validation samples which containing different amounts of two surfactants.

Clinical Utility of Haptoglobin in Combination with CEA, NSE and CYFRA21-1 for Diagnosis of Lung Cancer

  • Purpose: To investigate the clinical value in lung cancer of a combination of four serum tumor markers, haptoglobin (Hp), carcinoembryonic antigen (CEA), neuron specific enolase (NSE) as well as the cytokeratin 19 fragment (CYFRA21-1). Materials and Methods: Serum Hp (with immune-turbidimetric method), CEA, NSE, CYFRA21-1 (with chemiluminescence method) level were assessed in 193 patients with lung cancer, 87 patients with benign lung disease and 150 healthy controls. Differences of expression were compared among groups, and joint effects of these tumor markers for the diagnosis of lung cancer were analyzed. Results: Serum tumor marker levels in patients with lung cancer were obviously higher than those with benign lung disease and normal controls (p<0.01). The sensitivities of Hp, CEA, NSE and CYFRA21-1 were 43.5%, 40.9%, 23.3% and 41.5%, with specificities of 90.7%, 99.2%, 97.9% and 97.9%. Four tumor markers combined together could produce a positive detection rate of 85.0%, significantly higher than that of any single test. With squamous carcinomas, the positive detection rates with Hp and CYFRA21-1 were higher than that of other markers. In the adenocarcinoma case, the positive detection rate of CEA was higher than that of other markers. For small cell carcinomas, the positive detection rate of NSE was highest. The area under receiver operating characteristic curve ($AUC^{ROC}$) of Hp in squamous carcinoma (0.805) was higher than in adenocarcinoma (0.664) and small cell carcinoma (0.665). Conclusions: Hp can be used as a new serum tumor marker for lung cancer. Combination detection of Hp, CEA, NSE and CYFRA21-1 could significantly improve the sensitivity and specificity in diagnosis of lung cancer, and could be useful for pathological typing.

  • This is to investigate the methodology for the simultaneous determination of Wk, mk and TSNA using gas chromatography(GC) in combination with chemiluminescence detector, thermal energy analyzer(TEA) . The simultaneous analysis has been estimated by evaluating tobacco. The TEA was linked to GC equipped with non -polar SPB -5 fused silica capillary column which was introduced into the ceramic pyrolysis tube by the point of 16cm from the end of TEA. Quantification was carried out by internal standardization with WDPA after calibration of retention times and response factors with authentic nitrosoamines. It was demonstrated that WDPA was most preferable as internal standard for the simultaneous analysis. The recoveries of the internal standard were in the range of 83∼96% . Nitrosoamines in this method were detected with determination limit of 0.1ng and was made by a straight line in calibration curve by TEA response. The suitability of nitrosoamines extraction in tobacco leaf was investigated. It was most suitable to extract nitrosoamines from tobacco leaves with 0.01 M NaOH within a period of 8 hours. Thimerosal as an antibacterial agent was added to NaOH solution to prevent artifactual formation. The fractionation and the purification of nitrosoamines form alkaline extracts were conveniently performed using Extrelut multilayer column and dichloromethane. Reproducible and reliable results were obtained for the determination of nitrosamines in a relatively short time compared to previous known method. TSNA contents in burley were about 4 times higher as those in the fluecured tobacco.

  • Thyroxine (3,5,3',5'-L-tetraiodothyronine; T$_4$) is the most commonly measured thyroid hermono for the diagnosis of various thyroid disorders. Although radioimmunoassay (RIA) is still considered as the reference technique for the measurement of T$_4$, it is generally regarded that RIA has its primary disadventages in handling the wastes and controling the human and material resources. Therefore, establishment of enzyme-linked immunosorbent assay (ELISA) has of great significance. To verify the usefulness of our enzyme immunoassay, we have obtained the standard dose response curve of T$_4$ in patient's sera which is inversely proportional to the amount of herseradish peroxidase (HRP) conjugated monoclonal antibody of T$_4$ bound to the wells. The correlation coefficient (r) between the ELISA and chemiluminescent assay was 0.444 (n=38). Thus we have investigated the establishment of rapid and sensitive competitive ELISA assay method for detection of T$_4$ in patient's sera.

Alteration of Thyroid Function in Indian HER 2-Negative Breast Cancer Patients Undergoing Chemotherapy

  • Background: Thyroid hormones (TH) are regulated by the hypothalamic-pituitary axis, which plays an important role in cell growth, differentiation, development and other aspects of metabolism. It is believed that an active hypothalamic-pituitary axis increases the susceptibility of thyroid dysfunction during systemic chemotherapy. In order to investigate the relation between thyroid function and chemotherapy the present study was designed to investigate TH in breast cancer patients receiving at least three cycles of chemotherapy. The levels of TH were measured at the baseline and before each cycle of chemotherapy. Materials and Methods: Blood samples for estimation of TH levels were collected from 80 (pre-menopausal-40; post-menopausal-40) breast cancer patients just before they were undergoing - $1^{st}$, $2^{nd}$, $3^{rd}$ and $4^{th}$ cycle of chemotherapy. The serum was separated and $T_3$, $T_4$ and TSH levels were determined by chemiluminescence method. Results: $T_3$ and $T_4$ were found significantly decreased and TSH was found significantly increased after $1^{st}$ (p<0.001), $2^{nd}$ (p<0.0001) and $3^{rd}$ cycle of chemotherapy (p<0.0001). The variation of $T_3$ levels (decreased) and TSH levels (increased) was found more in post-menopausal (p<0.0001) women then in pre-menopausal women after $3^{rd}$ cycle of chemotherapy as compared to baseline (p<0.001). Conclusions: TH were remarkably altered after each cycle of chemotherapy leading to decline in thyroid function of breast cancer patients. Further, the results also indicated that post-menopausal women were more prone towards decline in thyroid function then pre-menopausal women. The present study proposes the monitoring of TH after each cycle of chemotherapy in breast cancer patients.
