Development of Protein Chip by Random Fluidic Self-Assembly Interaction (무작위 액중 상호 작용에 의한 단백질칩의 개발)

  • In this paper, we have been proposed a new method of multichannel biosensor using random fluidic self-assembly. A metal particle and an array was fabricated. Biomaterials were immobilized on the metal particle. The array and the particles were mixed in a buffer solution, and were arranged by self-assembly. A quarter of total Ni dots were covered by the particles. The binding direction of the particles was controllable, and condition of particles was almost with Au surface on top. The particles were successfully arranged on the array. The biomaterial activities were detected by chemiluminescence.

  • The purpose of this research was to investigate the effect of Codonopsis lanceolatae Radix water extract(CLE) on immunocytes. The effect of CLE on the proliferation of thymocytes was estimated by MTT colorimetric assay, sub-population of thymocytes was estimated by laser flow cytometry, production of nitric oxide from macrophages was estimated by Griess method, and phagocytosis of human polymorphonuclear cells was estimated by lucigenin chemiluminescence. CLE increased the proliferation of thymocytes in vivo, but did not affect the proliferation of thymocytes in vitro. CLE accelerated the activation of heper T cells in thymocytes. CLE inhibited the production of nitric oxide from peritoneal macrophages, and increased the phagocytosis of human polymorphonuclear cells. These results suggest that CLE have immuno-regulatory action in vivo.

  • Nitric oxide of gas phase in cigarette smoke was conveniently determined by Chemiluminescence NO analyzer with 20 ports rotary type smoking machine Data are presented to support this observation. On the basis of this method the amount of NO in the smoke from nonblended and commercial cigarettes were estimated. Gas phase NO concentration in main stream smoke was correlated well with the nitrate levels in tobacco leaves, but there was no correlation with the tar yields. In consequence, it would be considered that the methodology in this study provided simple and rapid quantification for NO in cigarette smoke.

Biomaterials were immobilized on the metal particle. The array and the particles were mixed in a buffer solution, and were arranged by magnetic force interaction and random fluidic self-assembly. A quarter of total Ni dots were covered by the particles. The binding direction of the particles was controllable, and condition of particles was almost with Au surface on top. The particles were successfully arranged on the array. The biomaterial activities were detected by chemiluminescence and electrochemical methods.

  • An experimental study of a dual swirl burner was conducted to analyze NOx emission in the lean conditions. The dual swirl burner is composed of a combination of swirling jet premixed(main section) and diffusion flames(pilot section). It was operated with a co-swirling configuration and overall equivalence ratios between 0.6 and 0.8. The purpose of this study is to analyze experimentally the characterization of flame temperature and NOx concentration in reacting zone and to supply the useful experimental data for numerical simulations. The measurements of temperature and NOx concentration were captured using a thin digitally-compensated thermocouple and a sampling quartz probe with quenching effect of sudden expansion, and were measured by the NOx analyzer of chemiluminescence method. We could analyse the NOx emission characteristics comparing the temperature distributions in the lean equivalence ratios.
