DNA Mediated Energy Transfer from 4',6-Diamidino-2-phenylindole to tetra- and bis-cationic Porphyrins at Low Binding Densities

  • The fluorescence of 4',6-diamidino-2-phenylindole (DAPI) bound to DNA at a [DAPI]/[DNA base] ratio of 0.005 was quenched by meso-tetrakis(N-methylpyridinium-4-yl)porphyrin (TMPyP) or cis-bis(N-methylpyridinium-4-yl)porphyrin (BMPyP) when both DAPI and either porphyrin spontaneously bound to the same DNA strand. The quenching was investigated using the "one-dimensional inner sphere" and the "F$\ddot{o}$rster resonance energy transfer" (FRET) models. Total quenching occurred when DAPI and TMPyP were up to 19.3 base pairs or $66\AA$ apart. BMPyP could quench the fluorescence up to 13.9 base pairs or $47\AA$. TMPyP, which intercalated between the DNA base-pairs, appeared to be a better acceptor than BMPyP, which stacked along the DNA stem. The higher quenching and higher resonance energy transfer efficiency of TMPyP was due to the larger overlap integral between its absorption spectrum and the emission spectrum of DNA-bound DAPI.

  • Results of intrinsic and extrinsic fluorescence studies on choline oxidase revealed that the enzyme at high alkaline pH values has more accessible hydrophobic patches relative to acidic pH. Fluorescence quenching studies with acrylamide confirm these changes. The quenching constants were also determined at different pH(s) by using the Stern-Volmer equation. CD studies showed that at higher pH a transition from $\alpha$-helix to $\beta$- structure was appeared while at lower pH the content of $\alpha$-helix structure was increased. Furthermore, analysis of the spectral data using chemometric method gave evidence for existence of intermediate components at very high pH(s). Results of thermal denaturation evaluated that the enzyme has the most instability at higher pH(s). Altogether low and high pH values caused significant alteration on secondary and tertiary structures of choline oxidase via inducing of an intermediate.

Phosphorescent Azacrown Ether-appended Iridium (III) Complex for the Selective Detection of Hg2+ in Aqueous Acetonitrile

  • A new phosphorescent cyclometalated heteroleptic iridium (III) complex with an ancillary ligand of 4-azacrownpicolinate was prepared and its metal ion selective phosphorescent chemosensing behavior was investigated. The new iridium (III) complex exhibits notable phosphorescence quenching for Hg2+ in aqueous 50% acetonitrile solution with respect to the selective phosphorescent detection of various metal ions including $Li^+,Na^+,K^+,Cs^+,Mg^{2+},Ca^{2+},Ba^{2+},Fe^{2+},Ni^{2+},Cu^{2+},Zn^{2+},Ag^+,Pb^{2+},Cd^{2+},Cr^{2+},Cr^{3+}$ and $Hg^{2+}$. The phosphorescence quenching for $Hg^{2+}$ increased linearly with increasing concentration of $Hg^{2+}$ in the range of $10{\mu}M-700{\mu}M$ even in the presence of other metal ions, except for $Cu^{2+}$. Consequently, the new iridium (III) complex has the potential to be utilized for the determination of parts per million levels of $Hg^{2+}$ in aqueous acetonitrile media.

Kinetic Study on the Low-lying Excited States of Ga Atoms in Ar

  • Decay kinetics of Ga(5s), Ga(5p) and Ga(4d) atoms in Ar were studied by laser induced fluorescence technique. Theground state gallium atoms in the gas phase were generated by pulsed dc discharge of trimethyl gallium and argon mixtures. Both pulsed discharge and YAG-DYE laser system were controlled by a dual channel pulse generator and the delay time between the end of discharge and laser pulses was set 3.0-6.0 ms. The Ga(5s) and Ga(4d) atoms were generated by single photon excitation from the ground state Ga atoms and radiative lifetimes as well as the total quenching rate constants in Ar were obtained from the pressure dependence of the fluorescence decay rates. The Ga(5p) atoms were populated by a two-photon excitation method and the cascade fluorescence from Ga(5s) atoms were analyzed to extract quenching rate constant of Ga(5p) atoms by Ar in addition to radiative lifetimes of Ga(5p) state. The magnitudes of the quenching rate constants by Ar for the low-lying excited states of Ga atoms are 1.6-3$ {\times}10^{-11}cm^3$ molecul$e^{-1}s^{-1}$, which are much larger than those for alkali, alkaline earth and Group 12 metals. Based on the measured rate constants, kinetic simulations were done to assign state-to-state rate constants.

  • For the prevent of fire accident or explosion disasters from inflammable gas and vapour, flame arresters are used in chemical equipment, oil tank or other similar installation. The flame arresters have been used mainly wire gauze type. Wire gauze type flame arrestes is affected several factors. We have know that the quenching ability has a great of difference the preference in accordance with flame velocity, direction of flame propagation and wire net of mesh and number of qauze and introduce examination result data quoated from the abroad.

Optimal Method of Radon Analysis in Groundwater using Ultra Low-Level Liquid Scintillation Counter (극 저준위 액체섬광계수기를 이용한 지하수 중 라돈($^{222}Rn$) 측정법 연구)

  • Optimal method of radon analysis in groundwater was studied using ultra low-level liquid scintillation counter (ULLLSC) which is well known as an analytical instrument for analyzing the alpha and beta radionuclides in environmental materials. Optimization of pulse shape analyzer (PSA) in operating the LSC was performed with $^{241}Am\;and\;^{90}Sr/^{90}Y$ as well as $^{226}Ra$ Also, the chemical quenching of scintillation generation and the color quenching of the generated photon to photomultiplier tubes (PMT) were determined their effects not only to decrease the analytical efficiency but also to change the optimal PSA level and background due to high ion contents of groundwaters. The optimal PSA level was shown in the range of 90 to 110 with less than 5% error. The effects of high ion contents in groundwater for the analytical efficiency show within 10% error from the different ion contents. The chloroform as a quenching agent was used to determine the analytical efficiency with the different amount, showing that the efficiency decreases 20% using the 2% of chloroform.

Preparation and Fundamental Characterization of EVOH Hollow Fiber Membranes via Thermally Induced Phase Separation (TIPS) (열유도상분리법을 이용한 EVOH 중공사 분리막의 제조 및 기본 특성)

  • Poly(ethylene-co-vinylalcohol) EVOH hollow fiber membranes were successfully fabricated via a thermally induced phase separation (TIPS) method. It was observed that all membranes fabricated under different spinning conditions had interconnected and bicontinuous structures through liquid-liquid phase separation. Glycerol and poly(ethylene glycol) 200 were used as diluents for the TIPS method. Glycerol was used as a mixing component in quenching bath to control pores on the outer surface of the hollow fiber membrane. Hot quenching bath with a mixing component to generate large pores on the outer surface of the hollow fiber membrane. The effects of polymer concentration, diluent and quenching bath on the morphologies, water permeabilities, and mechanical properties of the EVOH hollow fiber membranes were systematically investigated. The relationship between water permeability, mechanical properties and spinning conditions was discussed in detail.

Numerical Calculation of Minimum Ignition Energy for Hydrogen and Methane Fuels

  • Minimum ignition energies of hydrogen/air and methane/air mixtures have been investigated numerically by solving unsteady one-dimensional conservation equations with detailed chemical kinetic mechanisms. Initial kernel size needed for numerical calculation is a sensitive function of initial pressure of a mixture and should be estimated properly to obtain quantitative agreement with experimental results. A simple macroscopic model to determine minimum ignition energy has been proposed, where the initial kernel size is correlated with the quenching distance of a mixture and evaluated from the quenching distance determined from experiment. The simulation predicts minimum ignition energies of two sample mixtures successfully which are in a good agreement with the experimental data for the ranges of pressure and equivalence ratio.

  • Red fluorescent organic nanopaticles (FONs) based on a diarylacrylonitrile derivative conjugated molecule were facilely prepared by surfactant modification. Such red FONs showed excellent water solubility and biocompatibility, making them promising for cell imaging applications.
