뽕나무(Morus alba)와 꾸지뽕나무(Cudrania tricuspidata)의 수용성 추출물에 의한 항산화 활성 (Antioxidative Activities by Water-Soluble Extracts of Morus alba and Cudrania tricuspidata)

  • 뽕나무(Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata)의 잎과 줄기 껍질의 수용성 추출물에 대하여 in vitro 실험계에서 항산화 활성을 비교 검토하였다. 성장기 마우스의 간장 microsome을 이용한 생체막 지질 과산화 억제정도는 꾸지뽕나무 줄기 껍질(53%)>뽕나무 줄기 껍질(43%)>꾸지뽕나무 잎(38%)>뽕나무 및(34%) 순으로 나타났다. Linoleic acid산화 실험계에서는 꾸지뽕나무 줄기 껍질과 뽕나무 잎의 추출물이 강한 항산화 활성을 나타내었으며, 꾸지뽕나무 줄기 껍질의 추출물은 반응 8일째까지 강한 활성을 보였다. TBA method에서는 뽕나무 줄기 껍질이 다른 것에 비해 약간 강한 활성을 보였으며, 또한 DPPH에 의한 수소공여능은 각 시료첨가 0.05%에서 뽕나무 잎>꾸지뽕나무 잎>통나무 줄기 껍질>꾸지뽕나무 줄기 껍질 순으로 나타나 및 추출물에서 비교적 수소공여 작용이 강하였다. 이들 추출물의 총 폴리페놀 화합물의 함량을 측정한 결과 뽕나무 잎(1.32%), 뽕나무 줄기 껍질(1.28%), 꾸지뽕나무 잎(1.34%), 꾸지뽕나무 줄기 껍질(1.30%)로 큰 차이가 없었다.

감나무 잎으로 부터 분리한 tyrosinase 억제물질의 응용 (Application of Isolated Tyrosinase Inhibitory Compounds from Persimmon Leaves)

  • 감잎으로부터 tyrosinase 억제물질의 추출을 위하여 60% ethanol을 사용하여 추출한 경우 추출물의 phenol성물질 함량이 21.91 mg/g로 가장 높았고, tyrosinase 억제효과도 37%의 억제율로 가장 높게 나타났다. 감잎 추출물로부터 tyrosinase 저해 활성 물질을 정제하기 위하여 Sephadex LH-20 column과 MCI-gel CHP-20 column을 이용하여 gradient로 용출한 결과 tyrosinase 저해 활성을 가지는 2가지 순수한 compound를 정제하였으며, 정제된 2가지 compound를 구조 동정한 결과, (+)-gallocatechin과 prodelphinidin B-3이었다. 정제물의 tyrosinase 저해활성을 측정한 결과 (+)-gallocatechin과 prodelphinidin B-3는 29.5, 40.2%의 저해활성을 나타내었으며, melanoma 세포에서의 멜라닌 생합성 저해효과를 측정한 결과 (+)-gallocatechin과 prodelphinidin B-3는 각각 32.5, 46.7%의 저해활성을 나타내어 멜라닌생성억제효과가 매우 뛰어남을 알 수 있었다. 정제된 미백물질을 이용하여 제조한 essence 제품의 안정성을 살펴본 결과, 미백 essence제품의 pH는 10,000 ppm의 농도에서 4.90~4.95였으며, 점도는 23,000~26,000 cP로 60일 동안의 저장기간 동안 큰 변화를 나타내지 않았다. 일반적 보존 시험인 온도에 따른 안정성과 광안정성(인공광, 자연광) 검사에서도 안정하였으며, 특수, 가혹 보존 시험인 온도순환에 따른 안정성 검사에서도 상의 분리와 변색, 변취 없이 모두 안정함을 확인할 수 있었다.

Effects of Ultraviolet-B Radiation on Growth and Photosynthesis in Cucumber Primary Leaves

  • In the present study we studied the growth, photosynthetic traits and protective mechanisms against oxidative stress in the primary loaves of cucumber (Cucumis sativus L.) seedlings with or without UV-B treatment. Cucumber seedings were irradiated with UV-B for 10 days in environment-controlled growth chambers. The primary leaves irradiated with UV-B showed reduction in leaf length and decreased biomass production. The reduced biomass production seemed to be due to a negative effect of UV-B radiation on the photosynthetic process. Changes in chemical properties of leaf, such as chi a/b ratio affected photosynthesis. UV-B significantly affected chl b content compared with chi a in the light harvesting complex resulting reduced photosynthetic activity Fv/Fm decreased with an UV-B stress, suggesting that the photosynthetic apparatus, and particularly, PS II was damaged under UV-B stress. Malondialdehyde(MDA) concentration which represents the state of membrane lipid peroxidation Increased significantly under UV-B stress confirming an oxidative stress. UV-B exposure with SA solution(0.1-1.0 mM) can partially ameliorated some of the detrimental effects of UV-B stress. Leaf injuries including loss of chlorophyll and decreased ratio of Fv/Fm were reduced with combined application of UV-B and SA. ABA and JA showed similar mode of action in physiological effects on photosynthetic activities though the levels were lower than those from SA treated plants. Chloroplast ultrastructure was also affected by UV-B exposure. The thickness of leaf tissue components decreased and the number of grana and thylakoids was reduced in chloroplast applied UV-B or SA alone. At combined stress granal and stromal thylakoids were less affected. The leaves under combined stress acquired a significant tolerance to oxidative stress. From these results, it can be suggested that SA may have involved a protective role against UV-B induced oxidative damage.

수집약쑥의 유효성분 함량변이 (Chemical Component of Mugwort (Artemisia princeps Pampanini) leaves Collected in Korea)

  • 우리 나라를 비롯한 동남아시아에서 주로 약용으로 이용 되어온 약쑥의 품질평가를 위한 기초자료를 얻고자 본 연구를 수행하였다. 우리 나라에서 특수한 용도로 이용되어 온 약쑥을 48개 지역에서 2004년부터 2006년까지 3년에 걸쳐 210점을 수집하여 잎의 유효성분을 조사한 바 그 결과는 다음과 같다. 잎 100 9에 함유된 eupatilin 함량은 $0{\sim}228.0mg$(평균 43.8 mg), jaceosidin 함량은 $0{\sim}73.8mg$(평균 12.8 mg)이 었으며, sesamin 함량은 $0{\sim}6.78mg$(평균 3.58 mg)의 변이를 보였다. 이들 화학성분 중 수집지역별로 함량변이가 큰 eupatilin 성분을 비교한 결과 강화도, 백령도를 중심으로 한 서해안 지역에서 수집된 것에서 높고, 동해안과 남해안지역에서 수집된 약쑥에는 eupatilin이 거의 함유되지 않았다.

Flavonoids from the Leaves of Ailanthus altissima Swingle and their Antioxidant Activity

  • Phytochemical studies on the leaves of Ailanthus altissima (Simaroubaceae) have not been reported previously. Thus, the authors isolated and identified secondary metabolites from A. altissima. Dried and powdered leaves were extracted with 80% aqueous methanol, and the concentrated extract was successively partitioned with ethyl acetate, n-butanol, and water. Four flavonoids were isolated from the ethyl acetate fraction through repeated silica gel and octadecyl silica gel column chromatography. Spectroscopic data including NMR, MS, and IR allowed for identification of the chemical structures as quercetin (1), afzelin (2), quercitrin (3), and isoquercitrin (4). This is the first report of the isolation of these compounds from A. altissima. The four isolated flavonoids 1-4 as well as solvent fractions (ethyl acetate, n-butanol, and water), were evaluated for DPPH radical scavenging activity.

Chloride Meter(CL-2Z형)를 이용한 담배잎 중 chloride의 신속한 검정방법 (Rapid Quantification of Chloride in Tobacco Leaves by a Chloride Meter (Model: CL-2Z))

  • The aim of this study is to develop a simple and rapid field method for the determination of chloride content in tobacco green or cured leaves. To set up a practical process, several factors such as the proper sampling position of each leaf, the size of leaf lamina pieces for blending, the substitution of deionized water fur the extracting solution, 0.1N $HNO_3$, the submerging durations before blending, and the standing times before measuring were studied using flue-cured and burley tobacco with a stable and reproducible Chloride Meter, KRK CL-2Z. From the results, chloride contents of tobacco leaves could be analyzed with the Chloride Meter(CL-2Z) using the suspension of 14 mm diameter leaf disks after soaking in distilled water for 30 min., blending with a mini-blender for 30 seconds. The chloride contents measured in this way showed highly significant correlations with ones by the conventional potentiometric method using an ion meter-chloride electrode and $AgNO_3$ titration. In our method, the procedures such as drying, powdering and weighing samples, and using chemical reagents for extraction (0.1N $HNO_3$) and titration(0.028N $AgNO_3$) could be eliminated. These results suggest that the newly constructed method using the Chloride Meter is more efficient to determine the chloride content of tobacco leave in comparison with the one by the Ion $Meter:AgNO_3$ titration.

오갈피나무의 정유성분 GC-MS 분석 (GC-MS Analysis of Volatile Constituents from Acanthopanax sessiliflorus)

  • The chemical composition of the volatile constituents from the leaves, stems, and roots of Acanthopanax sessiliforus growing wild in Mt. Deok-Yu was determined by GC and GC-MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were ${\delta}-3-carene$ (7.24%), limonene (8.10%), ${\beta}-thujene$ (17.85%), trans-bicyclic hydrocarbon (13.35%) and ${\delta}-cadinene$ (4.28%) in the leaves from one year-grown plants, tricyclene (7.21%), ${\beta}-myrcene$ (7.62%), limonene (10.23%), ${\beta}-thujene$ (15.61%) and dihydroedulan I (6.12%) in the leaves from three years-grown plants, ${\delta}-3-carene$ (4.96%), limonene (5.93%), ${\beta}-phellandrene$ (17.31%) and naphthalene (7.79%) in the stems from one year-grown plants, ${\alpha}-pinene$ (5.21%), limonene (5.12%) and ${\beta}-phellandrene$ (9.82%) in the stems from three years-grown plants, ${\alpha}-pinene$ (12.73%), ${\beta}-pinene$ (11.16%), ${\delta}-3-carene$ (6.16%) and ${\gamma}-cadinene$ (23.39%) in the roots from one year-grown plants, and ${\alpha}-pinene$ (17.25%), ${\beta}-pinene$ (9.35%), ${\delta}-3-carene$ (7.26%) and ${\gamma}-cadinene$ (17.95%) in the roots from three years-grown plants.

Induction of Systemic Resistance against Cucumber mosaic virus in Arabidopsis thaliana by Trichoderma asperellum SKT-1

  • Trichoderma asperellum SKT-1 is a microbial pesticide that is very effective against various diseases. Our study was undertaken to evaluate T. asperellum SKT-1 for induction of resistance against yellow strain of Cucumber mosaic virus (CMV-Y) in Arabidopsis plants. Disease severity was rated at 2 weeks post inoculation (WPI). CMV titre in Arabidopsis leaves was determined by indirect enzyme-linked immunosorbent assay (ELISA) at 2 WPI. Our results demonstrated that among all Arabidopsis plants treated with barley grain inoculum (BGI) of SKT-1 NahG and npr1 plants showed no significant reduction in disease severity and CMV titre as compared with control plants. In contrast, disease severity and CMV titre were significantly reduced in all Arabidopsis plants treated with culture filtrate (CF) of SKT-1 as compared with control plants. RT-PCR results showed increased expression levels of SA-inducible genes, but not JA/ET-inducible genes, in leaves of BGI treated plants. Moreover, expression levels of SA- and JA/ET-inducible genes were increased in leaves of CF treated plants. In conclusion, BGI treatment induced systemic resistance against CMV through SA signaling cascade in Arabidopsis plants. While, treatment with CF of SKT-1 mediated the expression of a majority of the various pathogen related genes, which led to the increased defense mechanism against CMV infection.

생쪽잎분말의 염색성 및 저장성 (I) - 동결건조방법 - (Dyeing Properties and Storage Stability of Leaf Powder Prepared from Dyer's Knotweed (I) - by Freeze Drying method -)

  • The objective of this study is to investigate the efficacy of leaf powder colorants as substitutes for traditional fresh juice extract dyeing. Three kinds of leaf powder colorants were prepared by freeze drying method with or without deep freezing as pre-treatment: one powder colorant from fresh leaf juice with deep freezing; two kinds of powder colorant from fresh leaves with and without deep freezing. Their dyeing properties and storage stabilities were studied and compared with the traditional fresh juice extract dyeing. The presence of indigo in the powder colorants was confirmed by UV/Visible absorption spectra. They showed absorption peak at 602nm which was same with indigo absorption peak. Dyeing was done at low temperature around 6$^{\circ}C$. All three powder colorants produced B colors on silk fabrics, showing similar color to the one dyed traditionally with fresh juice extract. The powder colorants from leaves gave higher color strength than the powder from leaf juice. The powder colorant prepared from leaves with deep freezing was the most stable for long term storage as its color and color strength were not changed after 360 days. So, this was used for further dyeing to study the effects of concentration and repeat dyeing on color strength and colorfastness. Fastnesses to dry cleaning and rubbing were fairly good above 4 rating. Further study is needed to improve light fastness. It was concluded that the leaf powder colorant with deep freezing could be used as a substitute for traditional juice extract dyeing at all seasons.

구증구포(九蒸九)에 의한 녹차 제조 I. 일반성분 분석 및 화학적 조성 (Korean Green Tea by Ku Jeung Ku Po′s I. Analysis of General Compositions and Chemical Compositions)

  • This study was carried out to prepare green teas by traditional roasting manufacturing process, Ku Geung Ku Po and to determine the quality charateristics of the green teas by examining the change of their main components induced during this roasting process. The content of total sugars in unroasted tea leaves was 7.82%. Its content of roasted teas was decreased by increasing the number of roasting and there was significantly differences between samples. The content of total sugars in 9th roasted green tea was 3.98%. The total nitrogen contents of green teas produced by this process were 4.96∼6.38%. The more the number of roasting and the less content of tannin in green teas. And the ratio of total nitrogen and tannin in tea leaves 21.97 but its ratio in 9th roasted tea was increased to 45.54. The ascorbic acid in tea leaves consisted in 1,820.3 mg/100g but its content was decreased by increasing the number of roasting process. Whereas the content of caffeine in green teas was not reduced significantly by this traditional method. Of all amino acids, green tea produced by Ku Jeung Ku Po was rich glutamic acid, lysine and aspartic acid. And especially, phenylalanine, which was rarely found in other green teas, was abundant in these green teas. The compositions of glutamic acid and methionine were increased as increasing the number of roasting process but those of aspartic acid and arginine decreased by these processing. The main fatty acids of Ku Jeung Ku Po green tea were linolenic acid, linoleic acid and palmitic acid and the contents of unsaturated fatty acid in green teas were composed over 70% of total fatty acid. And the fatty acid contents in green teas were not affected during Ku Jeung Ku Po's process. The contents of minerals in these teas was rich in the oder of potassuim. magnessuim and calciumim, and these contents were not observed the prominant change during the process.
