Chemical and Absorption Characteristics of Water-soluble Organic Carbon and Humic-like Substances in Size-segregated Particles from Biomass Burning Emissions

  • In this study, measurements of size-segregated particulate matter (PM) emitted from the combustion of rice straw, pine needles, and sesame stem were conducted in a laboratory chamber. The collected samples were used to analyze amounts of organic and elemental carbon (OC and EC), water-soluble organic carbon (WSOC), humic-like substances (HULIS), and ionic species. The light absorption properties of size-resolved water extracts were measured using ultraviolet-visible spectroscopy. A solid-phase extraction method was first used to separate the size-resolved HULIS fraction, which was then quantified by a total organic carbon analyzer. The results show that regardless of particle cut sizes, the contributions of size-resolved HULIS ($=1.94{\times}HULIS-C$) to PM size fractions ($PM_{0.32}$, $PM_{0.55}$, $PM_{1.0}$, and $PM_{1.8}$) were similar, accounting for 25.2-27.6, 15.2-22.4 and 28.2-28.7% for rice straw, pine needle, and sesame stem smoke samples, respectively. The $PM_{1.8}$ fraction revealed WSOC/OC and HULIS-C/WSOC ratios of 0.51 and 0.60, 0.44 and 0.40, and 0.50 and 0.60 for the rice straw, pine needle, and sesame stem burning emissions, respectively. Strong absorption with decreasing wavelength was found by the water extracts from size-resolved biomass burning aerosols. The absorption ${\AA}ngstr{\ddot{o}}m $ exponent values of the size-resolved water extracts fitted between 300 and 400 nm wavelengths for particle sizes of $0.32-1.0{\mu}m$ were 6.6-7.7 for the rice straw burning samples, and 7.5-8.0 for the sesame stem burning samples. The average mass absorption efficiencies of size-resolved WSOC and HULIS-C at 365 nm were 1.09 (range: 0.89-1.61) and 1.82 (range: 1.33-2.06) $m^2/g{\cdot}C$ for rice straw smoke aerosols, and 1.13 (range: 0.85-1.52) and 1.83 (range: 1.44-2.05) $m^2/g{\cdot}C$ for sesame stem smoke aerosols, respectively. The light absorption of size-resolved water extracts measured at 365 nm showed strong correlations with WSOC and HULIS-C concentrations ($R^2=0.89-0.93$), indicating significant contribution of HULIS component from biomass burning emissions to the light absorption of ambient aerosols.

  • As natural plant-based industry has been expanded, the use of oriental medicinal plants as cosmeceutical material received a lot of attentions in the cosmetic industry. Among various medicinal plants, Potentilla chinensis have drawn interests for its biological effects. Although several attempts were tried to study its biological effect as medicinal plant, only limited results were reported to evaluate its biological effect as cosmeceutical material. In this study, we examined the possibility of root extract from Potentilla chinensis as a cosmeceutical material because the root part has been reported to have several kinds of health promoting effects. After extraction of roots, biological evaluation including anti-inflammation, anti-wrinkle, whitening effect and moisturizing effect was performed. As a result, the root extract showed remarkable biological activities through stimulating synthesis of elastin and aquaporin-3, and inhibiting melanin synthesis, cyclooxygenase-2 expression and expression of metallopeptidase-1.

Extraction and Quality Characteristics of Porphyran from Laver(Porphyra yezoensis) Waste (김 부산물로부터 Porphyran의 추출 및 품질특성)

  • Chemical compositions such as total sugar, sulfate/total sugar, protein and 3,6-anhydrogalactose content of porphyran extracted from commercial laver, discolored laver and laver waste were measured. The total sugar contents of commercial laver, discolored laver and laver waste showed 54.5%, 45.7% and 56.2%, respectively. The total sugar of the commercial laver and laver waste were showed almost similar portion although that of discolored laver showed relatively lower level. The sulfate/total sugar contents showed 17.6%, 13.9% and 17.3%, and 3,6-anhydrogalactose contents showed 13.8%, 9.1% and 13.9% in commercial laver, discoloredlaver and laver waste respectively.

  • Methane, as a greenhouse gas, is some 21~25 times more detrimental to the environmental than carbon dioxide. Landfills generally constitute the most important anthropogenic source, and methane emission from landfill was estimated as 35~73 Tg per year. Biological approaches using biocover (open system) and biofilter (closed system) can be a promising solution for older and/or smaller landfills where the methane production is too low for energy recovery or flaring and installation of a gas extraction system is inefficient. Methanotrophic bacteria, utilizing methane as a sole carbon and energy source, are responsible for the aerobic degradation (oxidation) of methane in the biological systems. Many bench-scale studies have demonstrated a high oxidation capacity in diverse filter bed materials such as soil, compost, earthworm cast and etc. Compost had been most often employed in the biological systems, and the methane oxidation rates in compost biocovers/boifilters ranged from 50 to $700\;g-CH_4\;m^{-2}\;d^{-1}$. Some preliminary field trials have showed the suitability of biocovers/biofilters for practical application and their satisfactory performance in mitigation methane emissions. Since the reduction of landfill methane emissions has been linked to carbon credits and trading schemes, the verified quantification of mitigated emissions through biocovers/biofilters is very important. Therefore, the assessment of in situ biocovers/biofilters performance should be standardized, and the reliable quantification methods of methane reduction is necessary.

Synthesis and Small Animal Brain PET Study of a Serotonin Receptor Radiotracer, 18F-Mefway (세로토닌 5-HT1A수용체 방사성 추적자 18F-Mefway의 합성과 소동물 뇌 PET 연구)

  • $^{18}F$-mefway has been developed as radioligand for serotonin receptor 5-$HT_{1A}$. The object of this study was to obtain the mefway precursor with the higher yield than previous method and to identify whether $^{18}F$-mefway can bind to 5-$HT_{1A}$ or not. from microPET imaging of small animal brain. Precursor was prepared by a modification of the reported procedure then [$^{18}F$] labeling was performed by adding $^{18}F$ ion at $130^{\circ}C$ in the hot cell for 30min. After purification of reaction mixture using alumina Sep-pak and HPLC, microPET images of small animal brain were determined. The chemical yield of precursor was increased from 9% to 34% using oxalyl chloride and LAH/diethylether. We synthesized a precursor which was successfully labeled with no-carrier-added $^{18}F$-by new synthetic route. This research suggest that $^{18}F$-mefway will be used a radiopharmaceutical for evaluation of central nerve system disorder as imaging a gent for 5-$HT_{1A}$ receptor.

Characteristics of Bacterial-Koji and Doenjang(soybean paste) Made by using Bacillus subtilis DJI (Bacillus subtilis DJI을 이용하여 제조된 세균형 코지와 속성된장의 특성)

  • One bacterium with high proteinase production and spore-forming ability was isolated from korean traditional soybean paste(doenjang). The isolated strain was identified as Bacillus subtilis, based on gram-staining, biochemical properties and l6S rRNA gene sequencing, and designated as B. subtilis DJI. Its growth rate was very fast, and it reached its stationary phase within 9 h, and then started to form spores. Bacterial-koji and doenjang were prepared using B. subtilis DJI. Chemical components of the doenjang were determined after 2 months of aging period: amino nitrogen 507 mg%, crude protein 14.3%, crude fat 4.8% and water 54.9%. The composition of total and free amino acids and their ratios of doenjang were changed during the aging period. Among total amino acids in DJI doenjang, glutamic acid, aspartic acid, leucine and arginine were the major amino acids. The fibrinolytic activities of DJI doenjang and traditional doenjangs were 909.7 units/ml and $363.3{\sim}618.6\;units/ml$, respectively. Flavor compounds of DJI doenjang and traditional doenjang were extracted by SDE(simultaneous steam distillation and extraction), and analyzed by GC/MS; DJI doenjang possessed the typically favorable flavor compounds in traditional korean doenjang, with reduced off-flavor compounds.

Discrimination of the Origin of Ephedrine and Pseudoephedrine for Pharmaceutical Powder Materials by Carbon and Nitrogen Stable Isotope Ratio Analysis ($^{13}C$$^{15}N$ 안정성 동위원소 비율 분석에 의한 원료의약품에서 에페드린 및 슈도에페드린의 기원 판별)

  • The abuse of methamphetamine (MA) is one of the most serious drug abuses in Asia. And, the prevention of precursor production for abuse drug is one of the most effective drug control system. Isotope ratio analysis at natural abundance levels have been used to establish the environmental source or the geographic origin of various biological and nonbiological materials. Ephedrine, the precursor of MA, is produced by one of three methods; extraction from Ephedra plants, full chemical synthesis or via a semi-synthetic process involving the fermentation of sugar, followed by amination. We investigated the origin of ephedrine and pseudoephedrine based on the carbon and nitrogen values for nineteen pharmaceutical powder materials (PPMs) obtained from pharmaceutical company in Korea by stable isotope ratio mass spectrometry coupled to an elemental analyser (EA-IRMS). The carbon delta values for the ephedrine and pseudoephedrine were -24.21~-22.72 (mean=-23.72) $^{\cir}/_{\circ\circ}$ and -23.79~-22.71 (mean=-23.48) $^{\cir}/_{\circ\circ}$. The nitrogen delta values were 3.51~5.55 (4.43) $^{\cir}/_{\circ\circ}$ and 2.24~8.22 (5.42) $^{\cir}/_{\circ\circ}$. These results indicate that PPMs are semi-synthetic products. Therefore the origins of ephedrine(natural, semi-synthetic or synthetic) could be discriminated by using carbon and nitrogen stable isotope ratios. we are sure tat this stable isotope ratio analysis can discriminate the origins of precursors of methamphetamine.

Laboratory Study on the Removal of Heavy Metals Using Apatite for Stabilization of Tailings at the Ulsan Abandoned Iron Mine (울산폐철광산 광미 안정화를 위한 인회석의 중금속 제거 실내실험)

  • The purpose of this study is to evaluate laboratory experiments on arsenic and cadmium removal from tailings using apatite at the Ulsan Abandoned Iron Mine, and to develop a stabilization technique. The results of this study show that the permeability is decreased proportionally to the amount of apatite when it is added below 8%, while this is almost constant when the amount of apatite is added above 10%. The water extraction test from tailings using deionized water for several days shows that pH (7.4-8.4) is almost constant or slightly increased when apatite is added below 8%, while it is slightly decreased when apatite is added above 10%. According to TCLP test, reduction of concentrations of heavy metals in leachate is proportional to amount of apatite added. It seems that precipitates generated from leachate-apatite chemical reaction are not redissolved. As a result, cadmium and arsenic in leachate is mostly removed when apatite is added above 10%, and it is suggested that a proper technique should be selected for field application because either mixed or layered method shows almost same removal efficiencies of cadmium and arsenic in tailings.

Comparison of Volatile Compounds in Plant Parts of Angelica gigas Nakai and A. acutiloba Kitagawa (참당귀와 일당귀의 부위별 휘발성 정유성분 비교)

  • Volatile flavor compounds Angelica gigas Nakai and Angelica acutiloba Kitagawa were extracted by SDE (simultaneous steam distillation & extraction) using the mixture of n-pentane and diethylether (1:1, v/v) as an extract solvent and analyzed by GC-FID and GC-MS. The amount of essential oils of top part and root in Angelica gigas were obtained in 0.063% (v/w) and 0.389% (v/w) yields as a fresh weight base, respectively. The main compounds in top parts and the root were identified as nonane (7.51% and 24.49%, respectively), ${\alpha}-pinene$ (14.64% and 31.75%), limonene+${\beta}-phellandrene$ (14.01% and 9.66%), ${\gamma}-terpinene$ (7.85% and 1.20%), germacrene-d (5.85% and 0.22%), (E,E)-${\alpha}-farnesene$ (6.05% and 1.40%), ${\beta}-eudesmol$ (5.26% and 1.84%). Although these compounds were present in both parts. The results showed large differences in. the concentrations of them much varied. The amount of essential oils stem and leaf obtained (0.068% and 0.127% in A. gigas) and (0.153% and 0.243% in A. acutiloba) yields as a fresh weight base, respectively. More than 18 and 32 components in stem and leaf have been identified, which of main components in A. gigas were ${\alpha}-pinene$, myrcene, limonene, germacrene-d, eudesmol and butylphthalide, but germacrene-d and butylphthalide contents were also different in stem and leaf. And more than 21 and 32 components in A. acutiloba were ${\gamma}-terpinene$ and butylphthalide. Volatile compounds were very different in both species.

Isolation and Characterization of Paramyosins of Marine Gastropods (해산복족류(海産腹足類)의 Paramyosin의 분리(分離) 및 그 특성(特性)에 관(關)한 연구(硏究))

  • The muscle of abalone, Notohaliotis discus (REEVE), and top-shell, Turbo cornutus Solander, were examined for protein composition. Then paramyosins which are known as one of the important structural protein of the muscle fibrils were isolated from the both muscle and their physico-chemical properties such as solubility, salting-out behaviour, intrinsic viscosity, ATPase activity, etc. involving amino acid composition and N-terminal amino acid residues were investigated to elucidate phylogenie characteristics more intensively from the viewpoint of comparative biochemistry. The analysis of protein composition resulted in the following estimations: abalone muscle; water-soluble protein of 22 %, salt-soluble protein, 34%, alkali-soluble protein, 20%, and stroma protein, 24%, and top-shell muscle; water-soluble protein of 16%, salt-soluble protein, 30%, alkali-soluble protein, 29%, and stroma protein, 25%, respectively. It is demonstrated in sedimentation analysis that paramyosin and myosin-actomyosin account for approximately 65% and 35% of the salt-soluble protein of abalone, and that the composition of both sediments in top-shell was approximately 70% and 30%, respectively. The ultracentrifugally homogenous paramyosins isolated essentially according to Bailey's ethanol-dried method from both of the muscle showed a $S^{\circ}_{20,w}$ of 3. 14s for abalone and a $S^{\circ}_{20,w}$ of 3.50s for top-shell. The both paramyosins were commonly rich in arginine, aspartic acid, and glutamic acid, while scarcely contained proline and tryptophan, in rough accord with the other paramyosins thus far reported. It is clear that these gastropod paramyosins showed of having the characteristic N-terminal amino acid residues such as N-aspartic acid, N-valine, N-serine, and N-threonine in common. The abalone paramyosin completely salted in with KCl beyond $0.35{\mu}$ and the top-shell paramyosin beyond $0.30{\mu}$. The abalone paramyosin was salted-out between 18% and 30% saturation of ammonium sulphate and the top-shell paramyosin between 22% and 29% saturation. The intrinsic viscosities at abalone and top-shell paramyosins at $25^{\circ}C$ were estimated respectively to be 3.1 dl/g and 2.6 dl/g showing somewhat higher than the values for some other paramyosins from lamellibranchs. In regard with the ATPase activity, the para myosin specimens did not exhibit any significant activity over through the pH conditions of 5 to 9.5. irrespective of the presence of $Ca^{++}$ or $Mg^{++}$. So was the case with the abalone paramyosin prepared by a slightly modified Bailey's wet-extraction method.
