애기똥풀 ( 양귀비과 ) 꽃에서 일어나는 성적자원 분배의 계절적 변화 (Seasonal Changes in Sexual Allocation within Flowers of Chelidonium majus ( Papaveraceae ))

  • Seasonal variation in size and number of floral structures was examined in two massachusetts populations of chelidonium major, a self-compatible herb. All floral charcters except for anther number per flower declind significantly during a 3 week period. However, temporal patterns were not identical among characters or between popolations. The result indicate that floral characters varied in conjunction with flower diameter,but that the pattern of changes in floral characters in response to environments may not be easy to predict. Principal components analysis was conducted to environments may not be easy to predict. principal compenents analysis was conducted tl identify the functional relationship among floral male function, and female function, respectively, perhaps reflecting the functional distinction of floral characters. Based on this pattern, the relative allocation to sexual structures within flowers was examined: male allocation was relatively greater than female allocation eary or in the middle of flowering season, depending upon populations. Temporalvariation in relative allocation within flowers was not independent of seed tield components:; different combinations of the size and number of floral characters were correlated with different seed yield components, for example, either seed size or number per fruit, during a season. in particular, allocations to attractive and male structures were highly correlated with mean seed weight only earlier in the season. These result provide some evidence that flowering phenolgy is an important comportant to be considered in the study of sexual allocation.

  • Free radicals cause many diseases and aging. In our body coexist useful and harmful bacteria. Some useful intestinal bacteria such as L. plantarum, B. adolescentis, B .bifidum, B. infantis, C. butyricum were presented positive growth in culture medium bearing natural products. L. plantarum was more effectively grown by mixed natural products of CORNI FRUCTUS+THEAE FOLIUM+COPTIDIS RHIZOMA+PAEONIA JAPONICA, B. adolescentis was BEIAMCANDA CHINENSIS+ANGELICAE GIGANTIS RADIX+EPIMEDII HERBA+PAEONIA JAPONICA, B. bifidum was NELUMBO NUCLFERA GAERTNER+THEAE FOLIUM+BEIAMCANDA CHINENSIS+CORNI FRUCTUS, B. infantis was THEAE FOLIUM+CHELIDONIUM MAJUS L.+ASTRAGALI RADIX+EPIMEDII HERBA, C. butyricum was EPIMEDII HERBA+PUERARIAE RADIX+NELUMBO NUCLFERA GAERTNER+PAEONIA JAPONICA,etc. These natural products will support an important role in bio-industry.

단삼 등 천연물의 항암작용 (Anticancer Activity of Natural Products including Salvia miltiorrhiza)

  • The cellular growth inhibition of 20 natural products was screened using SRB (sulforhodamine B) assay against 4 human cancer cell lines(SNU-1, SNU-C$_{4}$, Hep3B, Kato III). Ethanol extracts of Salvia miltiorrhiza, Saussurea lappa and Chelidonium majus showed potent anticancer activity among them, and further, it was fractionated into methylene chloride, hexane and methanol. Methylene chloride and methanol fraction of Salvia radix showed significant inhibitory activity against 4 human cancer cell lines. The effect of Salvia miltiorrhiza on anticancer activity in vitro models was evaluated with methylene chloride fraction of Salvia miltiorrhiza. Life span of ICR mice implanted with sarcoma-180 was increased by 40-61% and BDF$^{1}$ mice implanted with L1210 was increased by 66-89% upon intraperitoneal administration with methylene chloride fraction of Salvia miltiorrhiza. Based on these result, we suggested that Salvia miltiorrhiza showed anticancer activity on the in vivo and in vitro models

유지 불검화물과 식물체 추출물이 식용유의 $180^{\circ}C$ 가열 산화에 미치는 효과 (Effects of Oil Unsaponifiables and Plant Extracts on the Thermal Oxidation of Oils at $180^{\circ}C$)

  • 여러 종류(8종)의 유지로 부터 추출한 불검화물과, 강력한 자동산화억제력이 있다고 알려진 53종의 식물체(농산물 및 생약) 메탄을 추출물을 유지에 첨가한후 $180^{\circ}C$에서 14 혹은 16시간 연속 가열하는 동안 유지의 지방산조성을 측정하여 이들 추출물들이 유지의 고열 가열 산화에 미치는 영향을 연구하였다. 추출된 유지의 불검화물들은 모두 $180^{\circ}C$의 고온 가열산화 조건에서 항산화력을 나타내지 못하였다. 그러나 53종의 농산물 및 생약중에서는 일부 종들은 강력한 가열산화 억제력을 나타내었다. 특히, 대황, 백굴체, 정향 등의 메탄올 추출물은 강력한 가열산화 억제력을 나타내었다. 또한 이들 대황, 백굴체 및 정향의 메탄올 추출물들이, 면실유를 $180^{\circ}C$에서 1일 7시간씩 8일간 가열 처리하는 과정에서 면실유의 linoleic acid 파괴 및 중합체 생성에 미치는 영향을 연구하였다. 이 결과 정향 및 백굴체추출물은 $180^{\circ}C$에서 장기간의 가열조건에서도 linoleic acid 파괴 억제 및 중합체 생성을 억제하는 효과가 크게 나타낸 반면, 대황의 경우에는 장기 가열산화 조건에서 지방산의 파괴에 대한 억제력은 미미하였으나, 중합체 생성억제력은 비교적 양호하였다.

애기똥풀 종자의 휴면타파를 위한 층적과 GAs의 효과 (Effects of Stratification and Gibberellins on Germination of Chelidonium majus L. subsp. asiaticum H.Hara Seed)

  • 애기똥풀(Chelidonium majus L. subsp. asiaticum H.Hara)의 지상부는 백굴채라고 불리며 항암, 항균 등의 효능을 가지고 있어 예로부터 약용으로 이용되었다. 그러나 천연 약용소재로 개발가치가 높음에도 불구하고 실생묘의 대량 확보를 위한 종자번식 관련 연구가 전무한 실정이다. 본 연구에서는 애기똥풀의 발아 및 휴면 특성을 분석함으로써 추후 종자번식을 위한 기초자료를 제공하기 위해 수행되었다. 실험재료로 사용된 애기똥풀 종자는 2019년 6월 경상북도 봉화군에서 채종하였으며 정선 후 건조상태로 실온(25℃)에 보관하였다. 종자의 1000립중은 0.57±0.01g이며, 배종비는 0.21±0.12로 조사되어 형태적으로 미성숙한 배를 가지고 있다. 종자의 활력은 테트라졸륨 검정을 통해 79%로 확인되었다. 종자의 기본 함수율은 7.64±0.25%였으며, 침지 2일 차에 최대함수율(32.6±0.34%)에 도달하였다. 초기 발아검정은 25/15℃ (light/dark, 16/8h)챔버에서 명조건과 암조건으로 구분하여 30일간 진행하였으며, 암조건은 알루미늄 호일로 광을 완전히 차단하고 최종 발아율만 조사하였다. 발아율을 조사한 결과, 명조건은 0.75±0.80%, 암조건은 5.50±1.55%로 조사되어 형태적인 휴면과 생리적인 휴면을 모두 갖는 것으로 판단하였다. 애기똥풀의 생태적인 환경을 고려하여 12주의 Summer temperature(S12, 25/15℃, 16/8h)로 층적하고 연속하여 Autumn temperature(A, 15/10℃, 16/8h)를 4주(A4)와 8주(A8)로 달리하여 종자의 휴면타파를 유도하였다. 층적 후 발아율은 모두 증가되었으며, S12-A4(54%)는 S12-A8(24.5%)에 비해서 휴면타파에 효과적이었다. 휴면타파 효과의 증진을 위해 층적처리한 종자를 500ppm의 GA3와 GA4+7에 각 24시간동안 4℃에서 침지시킨 뒤 동일한 조건에 파종하였다. GA3가 처리된 S12-A4는 발아율이 82.0%, S12-A8는 30.8%로 유의적으로 증가하였으나 GA4+7 처리는 S12-A4(48.5%)와 S12-A8(21.3%)에서 발아율 향상 효과가 없었다. 결론적으로 애기똥풀의 휴면타파를 위해서 12주의 Summer temperature와 4주의 Autumn temperature 변온층적과 추가 GA3 처리가 휴면타파를 위한 효과적인 처리방법으로 판단된다.

소염약침액이 Collagen II 유발 관절염 mouse의 TNF-α, IFN-γ 생성 및 비장세포 증식에 미치는 영향 (Inhibitory Effects of Soyeum Pharmacopuncture (SPP) on Rheumatoid Arthritis in Collagen II-induced Arthritis (CIA) Mice)

  • Objective : The aim is to examine the effect of Soyeum Pharmacopuncture (SPP) on collagen-induced arthritis (CIA) in DBA/1OlaHsd mice. Methods : To determine the effect of SPP on chronic IFNlammatory joint disease, we induced CIA in DBA/1OlaHsd mice by immunization with bovine type II collagen. Animals were treated with intraperitoneal injection doses of 2 mg/kg of SPP, beginning 3 days before the expected onset of disease symptoms. Inhibitory Effects of SPP were observed by serum levels of TNF-${\alpha}$, and IFN-${\gamma}$,,, or cell proliferation in the spleen cell culture and histological examination of knee joint. Results : In the CIA Mice, serum levels of TNF-${\alpha}$, and IFN-${\gamma}$,, production in the spleen cell culture were reduced. At the histopathological examination of knee joint, chondropathy of cartilage in the synovial joint in the SP group was repaired while compared with control group. Conclusion : These results suggest that the SPP may be effective for the prevention and treatment of rheumatoid arthritis disease.

Chelidonine blocks hKv 1.5 channel current

  • Voltage-gated $K^{+}$ (Kv) channels represent a structurally and functionally diverse group of membrane proteins. These channels play an important role in determining the length of the cardiac action potential and are the targets for antiarrhythmic drugs. Many $K^{+}$ channel genes have been cloned from human myocardium and functionally contribute to its electrical activity. One of these channels, Kv1.5, is one of the more cardiovascular-specific $K^{+}$ channel isoforms identified to date and forms the molecular basis for an ultra-rapid delayed rectifier $K^{+}$ current found in human atrium. Thus, the blocker of hKv1.5 is expected to be an ideal antiarrhythmic drug for atrial fibrillation. Chelidonine was isolated from Chelidonium majus L. We examined the effect of chelidonine on the hKv1.5 current expressed in Ltk-cells using whole cell mode of patch clamp techniques. Chelidonine selectively inhibited the hKv1.5 current expressed in Ltk-cells in a concentration-dependent manner, whereas did not affect the HERG current expressed in HEK-293 cells. Additionally, chelidonine reduced the tail current amplitude recorded at -50 mV after 250 ms depolarizing pulses to +60 mV, and slowed the deactivation time course resulting in a 'crossover' phenomenon when the tail currents recorded under control conditions and in the presence of chelidonine were superimposed. We found that chelidonine also inhibited the $K^{+}$ current in isolated human atrial myocytes where hKv1.5 channels were predominantly expressed. Furthermore, we examined the effects of chelidonine on the action potentials in rabbit hearts using conventional microelectrode technique. Chelidonine prolonged the action potential durations (APD) of atrial, ventricular myocytes and Purkinje fibers in a dose-dependent manner. However, the effect of chelidonine on atrial APD was frequency-dependent whereas the effect of chelidonine on the APDs of ventricular myocytes and Purkinje fibers was not frequency- dependent. Also, the selective action of chelidonine on heart was more potent than dofetilide, $K^{+}$ channel blocker.

더덕에 발생하는 당근뿌리혹선충의 증식억제 식물 탐색 (Screening of Medicinal Plants to Suppress Population of Meloidogyne hapla in Codonopsis lanceolata Trautv)

  • 비름과 쇠무릎등 총 36과 81속 90종의 약용식물을 대상으로 당근뿌리혹선충의 감염 여부를 조사한 결과, 쇠무릎 등 70종의 식물에서 당근뿌리혹선충 감염을 확인하지 못했고, 이들 70종 중 42종의 식물을 당근뿌리혹선충에 의한 더덕의 피해가 많았던 포장에 심고, 감염여부를 조사한 결과, 쇠무릎 등 26종은 당근뿌리혹선충에 거의 감염되지 않았고, 지치 등 16종은 감염되었음을 확인하였다. 또한 42종의 식물중 30종의 식물을 Pot 재배를 통하여 당근뿌리혹선충 접종밀도별 뿌리혹수와 토양 내 유충밀도를 조사한 결과 패랭이, 원추천인국, 기린초, 아주까리, 지모, 해변아욱, 애기똥풀, 흑참깨, 짚신나물, 큰뱀무, 오이풀, 현삼 등 12종은 당근뿌리혹선충에 거의 감염되지 않았음을 확인하였다. 반면에 더덕은 당근뿌리혹선충 접종밀도가 높을수록 뿌리혹수가 많아지고, 토양 내 유충밀도 역시 높아졌으며, 이에 따라 더덕의 생육은 접종밀도가 높을수록 나빠지는 경향이었다.

박동관류 심방모델에서 강심효과를 나타내는 단미 한약재 검색 (Screening of Positive Inotropic Effect from Herbal Extracts in Beating Rabbit Atria)

  • Many medicinal plants have been used for the treatment of edema, jaundice, and gonorrhea in traditional Oriental medicine. This screening study was designed to search the positive inotropic effects of herbal extracts in beating rabbit atria. Aquarius extracts of twenty six herbs were examined in atrial mechanical dynamics such as pulse pressure and stroke volume and atrial natriuretic peptide (ANP), one of the main hormones involved in the regulation of the body fluid and blood pressure homeostasis in perfused beating rabbit atria. Sophora flavescens Ait., Rheum officinale Baill., Acorus gramineus Sol., Chelidonium majus L., Pulsatilla koreana Nakai., Reynoutria japonica Houtt., Euphorbia lathyris L., Pyrrosia lingua (Thunb.) Farwell, Poncirus trifoliata Rafin., Anemarrhena asphodeloides Bunge, Kochia scoparia Schrad. significantly increased stroke volume and pulse pressure. However, those herbal extracts were not induced ANP secretion. We clarified the eleven herbal extracts for the positive inotropic effect independent of ANP secretion in beating rabbit atria. Thus these results provide a beneficial data for the treatment of the impairment of body fluid and blood pressure in traditional Oriental medicine.

최근 독성 문제가 제기된 한약재에 대한 고찰 (A Review of Herbal Medicinal Products Associated with Toxic Events in Korea)

  • Background : There has been renewed interest in complementary alternative medicine around the world far a decade. On the other hand, the issue of safety of herbal medicine in Korea has been continuously questioned. Issues raised have included not only chemical residue or heavy metals caused by pollution, but also the toxicity of herbal medicine itself. Objectives : The purpose of this study was to review some herbal medicines subject to controversy of toxicity and figure out what kind of problems they evidence. Methods : In this review, PubMed was used to search medical journals. Keywords such as 'herb, or herbal medicine', 'safety, or adverse events, or adverse effect, or toxicity, or liver injury' were applied. Journals were selected by four inclusion criteria, as objectives should be human, researchers should be medical doctors, literature written in the English language, and types of journals should be meta-analysis or practice guideline or reviews. Finally, 9 cases from 60 journals were included in this review. Results : We reviewed 13 herbal medicines for controversy of toxicity such as ephedra(Mahuang), Arisolochia fangchi, Aristolochia manshuriensis, licorice, skullcap , germander, Syo-saiko-to(Xiao-chai-hu-tang), Chelidonium majus, mistletoe, Dictamnus dasycarpus and madder. Conclusions : It suggested that 'who it is prescribed for' is one of the most important factors to identify the causes of toxicity in herbal medicine and dietary supplements, under specific conditions in Korea.
