본 연구에서는 유전자 재조합 기술에 의해 제조된 ${\beta}-carotene$ 15,15'-dioxygenase의 반응특성 및 효소 kinetics를 규명하였다. ${\beta}-carotene$ 15,15'-dioxygenase 효소반응을 위한 최적 온도 및 pH를 측정한 결과, 최적 온도는 $40^{\circ}C$로 판명되었으며 최적 pH 는 9.0이었다. 저장 pH 6.0-9.0 범위에서 안정하였으며, pH 11에서도 80% 이상의 활성을 보이는 호알칼리성 효소임을 확인하였다. 온도 저장성을 확인한 결과, $35^{\circ}C$에서의 효소활성 반감기가 40분으로서 열에 민감한 것으로 판단되었다. 한편, ferrous ion-chelating agent와 sulfhydryl-binding agent를 사용하여 ${\beta}-carotene$ 15,15'-dioxygenase에 미치는 영향을 살펴 보았다. Ferrousion-chelating agent인 ${\alpha},{\alpha}'-dipyridyl$과 1,10-phenanthroline은 $1{\times}10^{-4}$ M에서 최소 저해농도를 형성하였으며, sulfhydryl-binding agent인 iodoacetamide와 PCMB는 $1{\times}10^{-3}$ M에서 N-ethylmaleimide은 $1{\times}10^{-4}$} M에서 최소저해농도를 형성함을 확인할 수 있었다. 본 연구에서의 효소반응은 Michaelis-Menten 곡선을 따름을 확인하였으며, Hanes-Woolf 작도법에 따른 결과, ${\beta}-carotene$ 15,15'-dioxygenase 효소의 $K_{m}$ 및 $V_{max}$ 값은 각각 $3.39{\times}10^{-6}$ 및 1.2 pmol/mg protein/min인 것으로 산출되었다.
목 적:철은 세포 성장과 분화, 전자 전달 반응, 산소 전달, 해독작용 등 여러 가지 중요한 생체 반응에 반드시 필요한 요소로서 종양세포의 성장과 증식에도 절대적으로 필요하다. 최근에 철킬레이트제인 deferoxamine이 악성 구강 각질세포의 성장을 억제하고 세포자멸사를 유도하며, 난소암세포의 증식을 억제하고 세포자멸사를 유도하여 난소암의 성장을 억제하였다고 보고되었다. 그러므로 반복적인 수혈에 의하여 헤모시데린침착증이 발생한 소아 종양 환아에서 deferoxamine이 철을 제거 할 뿐만 아니라 암세포의 세포자멸사를 유도하는지에 대하여 알아보고 세포자멸사를 유도한다면 그 경로에 대하여 알아보고자 하였다. 방 법:골육종세포인 Saos-2에서 deferoxamine에 대한 효과를 알아보기 위하여 크리스탈 바이올렛과 트리판 블루 염색으로 세포의 성장 및 증식을 측정하였고, DNA 분획, 핵 응축, 세포주기분석으로 세포자멸사를 분석하였고, 세포자멸사와 관련된 분자들의 발현을 Western hybridization으로 분석하였다. 결 과:Deferoxamine은 Saos-2 세포에 대하여 시간과 농도에 의존적으로 세포 증식 억제 효과를 나타내었다. 이러한 세포 증식 억제 효과는 DNA 단편화, 핵 응축, 세포주기 분석에서의 $A_{0}$ 기의 증가, PARP의 활성도의 증가 등 세포자멸사가 유도되었음을 알 수 있었다. 또한 Saos-2 세포에서 deferoxamine 처리 후 Akt/PKB의 활성화가 억제되어 caspase 9의 활성화, 그 하류의 caspase 3의 활성화로 이어지는 미토콘드리아 매개되는 경로로 세포자멸사가 유도되었다. 결 론:결론적으로 철킬레이트제인 deferoxamine이 세포증식을 억제시키고 세포자멸사를 유도시킴으로써 골육종 세포의 증식을 억제하는 것을 보여주었다. 따라서 반복적 대량 수혈에 의한 철 과부하에 따른 장기손상이 우려되는 각종 소아 종양환자들에서 deferoxamine은 체내 축적된 철을 제거할 뿐만 아니라 종양 환자의 치료에 있어서 항암제 치료의 효과를 증가시킬 수 있는 새로운 치료법으로 개발될 수 있을 것이다.
실리콘계 태양전지 제조과정에서 발생하는 불량품에서 실리콘웨이퍼를 회수하는 연구를 수행하였다. 상온($25^{\circ}C$)에서 인산용액 농도, 산성불화암모늄 농도, 킬레이트제 종류 및 농도를 변화시키면서 폐태양전지의 반사방지막 및 N층의 제거 효율을 조사하였다. 10 wt% 인산, 2.0 wt% 산성불화암모늄, 1.5 wt% Hydantoin 사용 시 제거 효율이 가장 우수 하였다. 인산농도가 증가할수록 미세입자의 표면전위가 (+)로 변하여 정전기적 인력에 의해 실리콘웨이퍼 표면에 재흡착하여 표면처리 전보다 두께가 두꺼워졌으며, 표면의 오염도도 증가하였다. 인산-산성불화암모늄-킬레이트제 용액에 의한 표면처리방법은 모든 공정이 상온에서 수행되며, 공정이 단순하고, 폐수 발생량이 적고, 표면제거 효율이 우수한 방법으로 폐 태양전지의 재활용 및 기존 RCA 세정법의 대안으로 가능성이 매우 클 것으로 판단되었다.
전형적인 음이온 교환수지인 Dowex 1-X8 (50∼100메쉬)에 술폰기를 갖는 킬레이트 시약인 8-hydroxyquinoline-5-sulfonic acid (HQS)와 7-nitroso-8-hydroxyquinoline-5-sulfonic acid (NHQS)를 흡착시킨 킬레이트 수지를 사용하여 수용액에 용해되어 있는 Fe(Ⅲ), Cu(Ⅱ), Pb(Ⅱ) 및 Zn(Ⅱ)을 흡착시켜 다른 이온들로부터 분리시키는 연구를 시도한다. 킬레이트 수지의 안정성은 수용액에 공존하는 음이온의 종류, 그 농도 및 액성에 따라 영향을 받으므로 수지사용의 최적조건을 결정하였으며, 수지에 대한 두 킬레이트 시약의 흡착성도 정성적으로 해석하였다. 금속이온의 흡착은 킬레이트 수지에 있는 HWS 및 NHQS와 각 금속이온과의 킬레이트 형성에 의한 것으로 그 흡착력은 각 킬레이트의 안정도상수값과 비례한다. 액성이 PH3∼4 범위에서 HQS-수지의 금속이온의 흡착능은 Fe(Ⅲ):0.63, Cu(Ⅱ):0.82 및 Pb(Ⅱ):0.79 mmol/g이며, NHQS-수지의 경우는 Cu(Ⅱ):1.07 및 Pb(Ⅱ):0.96mmol/g이다. 몇가지 금속이온의 혼합액을 시료로 하여 분리를 시도했다.
본 연구에서는 modified Fenton 공정에서 Fe(II)과 Fe(III)에 대한 유 무기 착제의 효율성 평가를 위해 BTEX(benzene, toluene, ethylbenzene, xylene) 분해효율을 관찰하였다. 적용된 유 무기착제의 종류는 구연산과 피로인산을 선정하였다. Fe(III)과 구연산이 적용되어진 modified Fenton 공정에서, 구연산의 농도가 증가할수록 BTEX의 분해율이 감소하였지만, 피로인산의 경우에는 농도가 증가할수록 BTEX의 분해율이 증가하였다. 또한 Fe(III)이 적용되어진 modified Fenton 공정에서 무기착제인 피로인산이 적용되어진 경우, 유기착제인 구연산이 적용되어진 경우보다 상대적으로 높은 BTEX의 분해율을 나타내었다. Fe(II)에 유 무기 착제가 적용되어진 modified Fenton 공정을 비교한 결과, Fe(II)과 구연산의 몰비가 1:1일 때 pH 변화를 최소화시킴과 동시에 BTEX의 높은 분해율을 보였다. 결과적으로 과산화수소의 효율성, 철 침전물 생성여부, 오염물질 분해율 등이 고려되어질 때, 100 ppm의 벤젠분해를 위한 최적 Fe(II), 구연산 그리고 과산화수소의 농도 조건은 7 mM/7 mM/500 mM였다.
식물조직배양기술을 이용한 Ge 함유 인삼 부정근 생산 시 Ge의 생산성을 향상시키고자, 다양한 킬레이트제가 인삼 부정근의 Ge 축적과 사포닌 형성 및 생육에 미치는 영향을 조사하였다. $GeO_2$ 50ppm과 함께 citric acid, oxalic acid, phosphoric acid, EDTA 및 EGTA와 같은 킬레이트제를 인삼 부정근 배양시 첨가하여 Ge 흡수에 미치는 영향을 조사한 결과, phosphoric acid, EDTA 및 EGTA가 Ge 흡수를 촉진시킨다는 사실을 알 수 있었다. 그러나, EDTA와 EGTA는 인삼부정근의 생육을 억제시킬 뿐만 아니라 Ge 흡수 촉진 효과도 phosphoric acid에 비해 낮으므로 인삼 부정근의 Ge 흡수를 촉진하는데 가장 효과적인 킬레이트제는 phosphoric acid인 것으로 확인되었다. 인삼 부정근의 Ge 흡수 촉진을 위한 phosphoric acid의 최적 농도는 1.0 mM이었고, 이때의 Ge 함량은$22.7{\pm}0.3$ mg%로 대조구$(16.8{\pm}0.7$ mg%)의 1.4배에 해당하는 양이다. 한편, 인삼 부정근의 생장률과 총 사포닌 함량은 phosphoric acid의 농도가 증가할수록 감소하는 경향을 나타내었다.
This study aims to investigate the impacts of chemical cleaning on the performance of a reverse osmosis membrane. Chemicals used for simulating membrane cleaning include a surfactant (sodium dodecyl sulfate, SDS), a chelating agent (ethylenediaminetetraacetic acid, EDTA), and two proprietary cleaning formulations namely MC3 and MC11. The impact of sequential exposure to multiple membrane cleaning solutions was also examined. Water permeability and the rejection of boron and sodium were investigated under various water fluxes, temperatures and feedwater pH. Changes in the membrane performance were systematically explained based on the changes in the charge density, hydrophobicity and chemical structure of the membrane surface. The experimental results show that membrane cleaning can significantly alter the hydrophobicity and water permeability of the membrane; however, its impacts on the rejections of boron and sodium are marginal. Although the presence of surfactant or chelating agent may cause decreases in the rejection, solution pH is the key factor responsible for the loss of membrane separation and changes in the surface properties. The impact of solution pH on the water permeability can be reversed by applying a subsequent cleaning with the opposite pH condition. Nevertheless, the impacts of solution pH on boron and sodium rejections are irreversible in most cases.
Ferroelectric L $a_2$$Ti_2$$O_{7}$(LTO) thin films were prepared by chemical solution deposition processes. Acetylacetone was used as chelating agent and nitric acid was added in the stock solution to control hydrolysis and condensation reaction rate. The LTO thin films were spin-coated on Pt/Ti/ $SiO_2$/(100)Si and Pt/Zr $O_2$/ $SiO_2$/(100)Si substrates. After multiple coating, dried thin films were heat-treated for decomposition of residual organics and crystallization. The role of acetylacetone in Ti iso-propoxide stabilization by possibly substituting $O^{i}$Pr ligand was studied by H-NMR. B site-rich impurity phase, i.e. L $a_4$$Ti_{9}$$O_{24}$, was found after annealing, where its appearance was dependent on process temperature indicating the possible reaction with substrate. Dielectric and other relevant electrical properties were measured and the results were compared between modified sol-gel and MOD processes.s.s.
Stable PZT coating sol was prepared using chelating agent, ethylacetoacetate(EAcAc) by sol-gel processing under ambient atmosphere. Through FT-IR spectrum analysis on solution of each reaction step, formation of metal complex was confirmed and prepared PZT sol was stable over several months. Through TG-DTA, XRD, FT-IR spetrum analysis of PZT gel powder, it was understood that the addition of EAcAc could reduce the transition temperature to ferroelectric phase, due to the increased homogeneity by matching the hydrolysis and condensation rates by chelation. Single perovskite phase was obtained by the heat-treatment at 54$0^{\circ}C$ for 30 min. The film was coated on ITO-coated glass substrate by dip coating method. After heat-treatment, PZT thin film had thickness in the range of 20~130 nm. The maximum dielectric constant of its thin film at room temperature and 1 kHz was 128.
Solvent extraction using a Schiff-base, salen$(NEt_2)_2$, as a chelating agent has been conducted on several water samples to study the determination of trace Cu(II), Mn(II) and Zn(II). Experimental conditions for the formation and extraction of metal complexes were optimized with an aqueous solution similar in composition to the samples. The matrix difference between the sample and standard solutions was approximately matched, and the pH of each sample solution was adjusted to 9.5 with $NaHCO_3/NaOH$ buffer. The concentration of salen$(NEt_2)_2$ was $7.3\;{\times}\;10^{-3}$ mol/L, and the complexes were extracted into MIBK solvent followed by the measurement of AAS absorbance. The potential interference of concomitant ions was investigated, but no interference from alkaline and alkali earth ions was shown in this procedure. The given procedure is precise, as judged from the relative standard deviation of less than 5% for five measured data. The recovery of 93-103% shows that this method is quantitative for such trace metal analysis.
