Establishing porcine jejunum-derived intestinal organoids to study the function of intestinal epithelium as an alternative for animal testing

  • Background: The small intestine plays a crucial role in animals in maintaining homeostasis as well as a series of physiological events such as nutrient uptake and immune function to improve productivity. Research on intestinal organoids has recently garnered interest, aiming to study various functions of the intestinal epithelium as a potential alternative to an in vivo system. These technologies have created new possibilities and opportunities for substituting animals for testing with an in vitro model. Methods: Here, we report the establishment and characterisation of intestinal organoids derived from jejunum tissues of adult pigs. Intestinal crypts, including intestinal stem cells from the jejunum tissue of adult pigs (10 months old), were sequentially isolated and cultivated over several passages without losing their proliferation and differentiation using the scaffold-based and three-dimensional method, which indicated the recapitulating capacity. Results: Porcine jejunum-derived intestinal organoids showed the specific expression of several genes related to intestinal stem cells and the epithelium. Furthermore, they showed high permeability when exposed to FITC-dextran 4 kDa, representing a barrier function similar to that of in vivo tissues. Collectively, these results demonstrate the efficient cultivation and characteristics of porcine jejunum-derived intestinal organoids. Conclusions: In this study, using a 3D culture system, we successfully established porcine jejunum-derived intestinal organoids. They show potential for various applications, such as for nutrient absorption as an in vitro model of the intestinal epithelium fused with organ-on-a-chip technology to improve productivity in animal biotechnology in future studies.

Mechanical behaviour of a syntactic foam/glass fibre composite sandwich: experimental results

  • This note presents the main results of an experimental investigation into the mechanical behaviour of a composite sandwich conceived as a lightweight material for naval engineering applications. The sandwich structure is formed by a three-dimensional glass fibre/polymer matrix fabric with transverse piles interconnecting the skins; the core is filled with a polymer matrix/glass microspheres syntactic foam; additional Glass Fibre Reinforced Plastics extra-skins are laminated on the external facings of the filled fabric. The main features of the experimental tests on syntactic foam, skins and sandwich panels are presented and discussed, with focus on both in-plane and out-of-plane responses. This work is part of a broader research investigation aimed at a complete characterisation, both experimental and numerical, of the complex mechanical behaviour of this composite sandwich.

Development of a Prototype System for the Optical-Video-Detection and Characterisation of Meteors/Fireballs in South Korea

  • During a six-month period (autumn 2014 within the framework of a research & education project) we have constructed a professional double-station video-meteor detection network at the SOAO and BOAO mountain summits. Meteor detection is achieved by pixel-to-pixel motion-detection trigger. Each station is nearly autonomous and has three cameras with fixed viewing angles monitoring part of the night-sky over Korea. Various field of views are in use for testing purpose and captured video-meteor data is automatically transferred to a central FTP server on a nightly basis. Data is publicly available. The network has been operational since September 2014 and could serve as a prototype system for a more extended national network for meteor/fireball monitoring and detection in Korean airspace.   • [ $V_{0.9}Sb_{0.1}O_x$ ]systems, bulk and deposited on different supports (five types of ${\gamma}$-aluminas, ${\alpha}$-alumina, silica-alumina, silica gel, magnesium oxide), have been tested in the oxidative dehydrogenation (ODH) of iso-butane. This statement is derived from the data obtained by a set of characterisation techniques(specific surface area measurements, X-ray diffraction, X-ray photoelectron spectroscopy, laser Raman spectroscopy, in situ differential scanning calorimetry and in situ diffuse reflectance-absorption infrared Fourier transform spectroscopy).

Characterisation of Ramie Yarn Treated with Sodium Hydroxide and Crosslinked by 1,2,3,4-Butanetetracarboxylic Acid

  • The microstructure and tensile properties of ramie treated with NaOH and crosslinked with 1, 2, 3, 4-butanetetracarboxylic acid were characterised. X-ray diffraction and FTIR were used to study the crystalline structure o f the resultant ramie yarns. Results showed that the maximum change in the structure of the modified ramie took place at 16% NaOH, which would completely transform Cellulose I to Cellulose II. Also, the crystallinity index and fibre orientation decreased to the minimum value while the absorption properties were enhanced. The average degree of polymerization of the treated ramie yams slightly decreased after NaOH treatment.

  • SiC는 wide bandgap 물질로서 그 material properties로 인하여, high tmperature, high power, high frequency영역으로의 사용이 기대되는 물질이다. 따라서 SiC에 대한 기본적인 연구와 더불어, 그 소자 제작 및 응용에의 연구가 절실한 시점이다. 이에, SiC 기본적인 소자중 하나인 Schottky diode에 대해 연구하였다. 본 논문은 Schottky contact 물질로써 현재까지 연구가 미비한 Ru을 사용하였다. Ru은 Pt 계열물질로써, 다른 metal에 비하여 열역학적으로 안정하며, 또한 그의 산소 화합물인 RuO2는 다른 oxide에 비하여 전도성 이 높은 장점을 가지고 있다. 따라서 Ru-SiC diode는 이러한 측면에서 연구할 만한 가치가 있다.

  • The peroxo titanic acid solution was successfully prepared using titanium trichloride as a precursor. The basic properties of the TiO2 film prepared by the solution were investigated in view of phase change, bandgap energy, crystalline size etc. The film displayed amorphous TiO$_2$ at room temperature, anatase above 281$^{\circ}C$ and a mixture of anatase and rutile at 99$0^{\circ}C$, The crystalline size increases with annealing temperatures, while the bandgap energies decrease due to the quantum size effect and the formation of rutile phase which has low bandgap energy. As a result of TG-DTA, it was found that annealing treatment at 99$0^{\circ}C$ for 2h formed a mixtures of anatase and rutile through three steps: (1) the removal of physically adsorbed water (2) the decomposition of peroxo group (3) amorphous-anatase or anatase-rutile phase transformation.

Overproduction of Lactic Bacterial Enzymes and Bioactive Components

  • Recent developments in the application of molecular biology to food grade lactic acid bacteria (LAB) have shown that it could be feasible to engineer metabolic pathways to either enhance specific metabolic fluxes or to divert metabolites for the production of different or new end products. This engineering requires detailed knowledge of enzymes involved in metabolism and regulation within the targeted organism but little works have been done in this area. During biochemical and molecular characterisation of lactic bacterial enzymes, some of probiotic Lactobacillus and Bifidobacterium species were found to be very useful for food, nutraceutical and pharmaceutical industries. The enzymes are usually intracellular and the yields are very low to be useful for industrial applications. Among many enzymes and proteins of lactic bacteria studied, some of our gene cloning achievements have contributed to overproduction of lactic bacterial enzymes such as peptidases, esterases, lactases, bile salt hydrolases and linoleate isomerases for foods and nutraceuticals.

  • There are presented certain results on extending continuous linear operators defined on spaces of E-valued continuous functions (defined on a compact Hausdorff space X) to linear operators defined on spaces of E-valued measurable functions in a way such that uniformly bounded sequences of functions that converge pointwise in the weak (or norm) topology of E are sent to sequences that converge in the weak, norm or weak* topology of the target space. As an application, a new description of uniform closures of convex subsets of C(X, E) is given. Also new and strong results on integral representations of continuous linear operators defined on C(X, E) are presented. A new classes of vector measures are introduced and various bounded convergence theorems for them are proved.
