Characteristics of Antifreeze Protein-1 Induced during Low Temperature Acclimation in the Protaetia brevitarsis (Coleoptera; Cetonidae) Larva

  • Change of proteins was confirmed during low temperature acclimation of overwintering larva, and some biochemical characteristics of the induced antifreeze protein-1 (AFP-1) were investigated in Protaetia brevitarsis. As the freezing point depression by the action of induced AFPs, a considerable thermal hysteresis was observed in the haemolymph and in partially purified proteins. AFP-1 was purified from the cold acclimation larvae by ammonium sulfate precipitation ion exchange chromatography, gel permeation chromatography, and electroelution. The purified AFP-1 was determined to be a glycoprotein (approximately 320 kDa, pl 5.8) composed of a single type of subunit (80 kDa). The high contents of hydrophilic amino acids (Asp, Glu, Lys, Asn, Gln, Arg, Ser, Thr) were also confirmed, showing similarity with antifreeze proteins from other insects.

A Study on the Estimation of the Structural Stability of a Container Crane According to the Change of the Boom Shape using Wind Tunnel Test

  • In this study we carried out to analyze the effect of wind load on the structural stability of a container crane according to the change of the boom shape using wind tunnel test and provided a container crane designer with data which can be used in a wind resistance design of a container crane assuming that a wind load at 75m/s wind velocity is applied on a container crane. Data acquisition conditions for this experiment were established in accordance with the similarity. The scale of a container crane dimension, wind velocity and time were chosen as 1/200, 1/13.3 and 1/15. And this experiment was implemented in an Eiffel type atmospheric boundary-layer wind tunnel with $11.52m^2$ cross-section area. Each directional drag and overturning moment coefficients were investigated and uplift forces at each supporting point due to the wind load were analyzed.

Solution Structure of Water-soluble Mutant of Crambin and Implication for Protein Solubility

  • Water-soluble mutant of intrinsically insoluble protein, crambin, was produced by mutagenesis based on the sequence analysis with homologous proteins. Thr1, Phe13, and Lys33 of crambin were substituted for Lys, Tyr, and Lys, respectively. The resultant mutant was soluble in aqueous buffer as well as in dodecylphosphocholine (DPC) micelle solution. The $^1H-^{15}N$ spectrum of the mutant crambin showed spectral similarity to that of the wild-type protein except for local regions proximal to the sites of mutation. Solution structure of water-soluble mutant crambin was determined in aqueous buffer by NMR spectroscopy. The structure was almost identical to the wild-type structure determined in non-aqueous solvent. Subtle difference in structure was very local and related to the change of the intra- and inter-protein hydrophobic interaction of crambin. The structural details for the enhanced solubility of crambin in aqueous solvent by the mutation were provided and discussed.

이종의 도로망 데이터 셋에서 면 객체 매칭 기반 변화탐지 (Automatic Change Detection Based on Areal Feature Matching in Different Network Data-sets)

  • 차량용 내비게이션의 빠른 확산과 스마트폰 등 개인 단말기의 측위 기술 발달로 사용자 중심위치기반서비스, 특히 보행자 내비게이션 서비스에 대한 관심이 증대되고 있다. 보행자 내비게이션 서비스를 위한 핵심정보인 수치지도는 대용량이고 짧은 갱신주기를 요구하는 경우가 많아 수치지도의 효율적인 갱신이 중요한 이슈가 된다. 본 연구에서는 구축시기가 상이한 이종의 도로망 데이터 셋에 형상유사도 기반 면 객체 매칭을 적용하여 변화 탐지하는 기법을 제안하였다. 변화탐지에 앞서 이종의 도로망 데이터 셋의 면 객체 매칭에서 탐지될 수 있는 갱신 유형을 정의하였다. 면 객체 매칭 기반 변화 탐지를 위하여 이종의 두 도로망 데이터 셋의 선형인 도로객체를 이들로 둘러싸인 면인 블록으로 변환하였다. 변환된 블록을 중첩하여 중첩된 블록 간의 형상유사도를 계산하고, 이 값이 0.6 이상인 것을 후보 블록 쌍으로 추출하였다. 객체 유형별로 이분 그래프 군집화와 오목다각형 특성을 적용하여 정의된 갱신유형별 블록 쌍을 탐지하고, 해당 블록 쌍을 구성하거나 내부에 있는 도로 세그먼트 간의 프레셰 거리를 계산하였다. 이때, 프레셰 거리가 50 이상인 도로명주소기본도 도로구간의 도로객체가 갱신 도로객체로 추출된다. 그 결과 0.965의 높은 탐색율을 보여 제안된 기법이 이종의 도로망 데이터 셋의 선형 객체의 변화탐지에 적용될 수 있음을 확인할 수 있었다.

  • 보은과 괴산에 위치한 GNSS상시관측소의 GNSS관측자료와 경사계자료를 이용하여, 1시간과 3시간 동안의 GNSS 차분벡터의 변위와 시간에 따른 차분 경사 벡터의 변위를 계산했다. 그리고, 시간에 따른 두 변위 사이의 관련성을 얻었다. 이렇게 계산된 관련성은 ‘높음‘보다는 낮았다. 그러나, 일사량의 일변화는 일어남이 확실하고, 측정 결과 안테나탑의 경사 변위 때문에 안테나 위상중심 위치의 회전 운동이 존재함이 확실하므로, 우리는 DGNSS에 있어서 보정 오차를 방송할 때와 가상기준국체계(VRS)에 있어서 상시관측소에서 측정된 자료를 방송할 때에 안테나 위상중심 위치의 일변화를 고려할 것을 제안한다.

Molecular Characterization and Expression of LDHA and LDHB mRNA in Testes of Japanese Quail (Coturnix japonica)

  • The LDH isozymes are key catalysts in the glycolytic pathway of energy metabolism. It is well known that the distribution of the LDH isozymes vary in accordance with the metabolic requirements of different tissues. The substrates required for energy production change noticeably at successive stages of testes development suggesting a significant flexibility in the expression of glycolytic enzymes. Therefore, expression of LHDA and LDHB mRNAs was examined in adult and prepubertal quail testis. The mRNA of both LDHA and LDHB were expressed and no significant difference was observed in prepubertal testes. The mRNA levels of LDHB significantly increased during testicular development. In the adult testis, LDHA mRNA was not expressed. Expression studies revealed the presence of different LDH isozymes during testicular development. In contrast, electrophoresis of both testicular samples revealed only single band at a position indicative of an extreme type of LDH isozyme in quail testes. Furthermore, nucleotide and amino acid sequence analysis revealed significant similarity to chicken, duck and rock pigeon. These sequence results confirmed the similarity of LDHA and LDHB subunit protein in different avian species.

  • Objective To study the effectiveness of Yuldahansotang against Allergic Contact Dermatitis, the change of cutaneous shape, histochemistry, immunohistochemistry and distribution of apoptotic cells was researched. materials and methods 4-month-old rats were divided into three groups of 10. One is a contrastive group which has applied Acetone olive oil only. Another is ACD group which has intentionally activated Allergic Contact Dermatitis by DNCB. And the other is YST group which has given medication of Yangkyuksanhawtang extract. Each group of mice were observed after 24, 48 and 72 hours. results 1. With the result of Contact hypersensitivity assay, YST group shows appreciably less ear swelling than ACD group. 2. Comparing YST and ACD groups to each other regarding general change of skin, YST group shows less hyperplasia of epidermis, less migration of inflammatory cells and less damage of epidermis than ACD group. 3. Regarding the change of collagen fiber, ACD group has appeared to be low in number of collagen fiber while YST shows similarity with the contrastive group. 4. In dermis YST group has showed lower number of mastocyte than ACD group and is granulated type. 5. In dermis YST group has showed less MAC-1,IL-1, $IL-2R-{\alpha}$, ICAM-1 and VCAM-1 than ACD group. 6. The distribution of apoptotic cells has appeared littler in YST group than in ACD 7. Among signal molecule of apoptosis Bcl-2 has distributed more in YST group than ACD group and Bax and Fas has distributed less in YST group than ACD group.

한국 제조업 지식네트워크 구조변화의 특성 (The Characteristics of Structural Change in Knowledge Network of Korean Manufacturing Industries)

  • This paper analyzes the characteristics of technological knowledge flow-structure of Korean manufacturing in dynamic perspective. In doing that, the concept of the knowledge network is introduced which is defined as a set of industries and their interaction(knowledge flow) or linkage. The analysis of the inter-industrial knowledge flows is based on the technological similarity by using R&D researchers' academic background in the year of 1984, 1987, 1990. The analysis is carried out by such methodology as network analysis, indicator analysis and simple statistical analysis. And the final results are drawn both in absolute terms(dimension effect) and in relative terms(proportion effect) respectively. The main findings are as follow. First, the Korean manufacturing knowledge network appears to strengthen existing inter-industrial knowledge linkages rather than to construct new linkages. Second, the network seems to form a dualistic structure in that some high-technology sectors(knowledge production sectors) emerge along with traditional sectors(knowledge absorbing sectors). Third, since the mid-1980s, an inter-industrial fusion is witnessed among technologically intensive sectors, indicating that some sophisticated innovation modes are emerging in Korean manufacturing system. And fourth, by using the relations of the inter-industrial knowledge-flows, we classified manufacturing industries into 3 type ; knowledge-outflow sector, knowledge-inflow sector and knowledge intermediary sector.

의왕시 오봉산 육교형 생물 이동통로 모니터링에 의한 관리 및 개선방안 (A Study on Improvement and Administration of Ecoduct through Monitoring in Uiwang's Mt. Obong)

  • This study is to introduce the improvement and management of Ecoduct located in Uiwang's Mt. Obong, Gyeonggi province. The inner planting of Ecoduct is not very related to the flora of mountain areas nearby. The difference in the index of plant crown volume(GVZ), the similarity index(S.I.), and monitored animals is significant between Ecoduct and mountain areas. Putting together all of the results, the initial selection of movement and target species is excessive in number. Target species must be selected again. They will be species that are currently found on the site. The Ecoduct space must be divided into two parts: buffer planting space and wildlife corridor. Reforestation is needed since the GVZ is remarkably lower in Ecoduct than in the nearby mountain areas. Besides the vegetation of Ecoduct is densely planted in guidance planting space to connect the southern Quercus mongolica community and the northern Quercus acutissima community. Proper upkeep after construction should be continuously conducted to maintain the procedure of natural change.

충남 광역생태네트워크 자연녹지의 훼손지 식생구조 변화 (A Study on Vegetation Structure Changes between Natural land and Damaged land in Regional Ecological Network at Chungnam Province)

  • This study was carried out to analyze vegetation structure and change of the disturbed area in forest ecosystem(FE), riparian ecosystem(RE) and coastal ecosystem(CE) related to Regional Ecological Network at Chungnam province through constancy and dominance analysis, species diversity index, similarity index and canonical correlation analysis. Data were collected from April to October in 2015. As a result of constancy and dominance analysis of forest disturbed area, non-effective species(NES) was 30 species(17.0%), exported species(ES) was 98 species(55.7%) and imported species(IS) was 48 species(27.3%) among the total 176 species, respectively. In riparian disturbed area with total 139 species, there were 16 NES(11.5%), 98 ES(70.5%) and 25 IS(18.0%) respectively. In coastal disturbed area with 140 species, there were 20 NES(14.3%), 88 ES(62.9%) and 32 IS(22.9%) respectively. In all types of disturbed areas, the ratio of ES was higher than IS. As a result of species diversity, species richness and shannon's diversity index of disturbed area decreased in all kinds of crown strata such as tree, subtree, shrub and herb layer compared to the control area. As a result of similarity index, that of each type between control site and disturbed site was 0.374 in FE, 0.329 in CE and 0.259 in RE in the order. As a result of the CCA analysis, the number of present species, vine plants ratio and exported species ratio in disturbed area of FE and RE were decreased, and the naturalized plant ratio, imported species ratio and herb ratio were increased. But environmental factors of CE were not shown any clear tendency. In conclusion, many species occupied in control site disappeared into the disturbed area, and the naturalized plants and herb species were abundantly imported in the area. Therefore, it was considered that this study could be applied to the development of long-term and short-term ecological restoration techniques in view of vegetation changes.
