Ichthyotoxicity of a Harmful Dinoflagellate Cochlodinium polykrikoides: Aspect of Hematological Responses of Fish Exposed to Algal Blooms

  • To clarify the ichthyotoxic mechanisms of a harmful dinoflagellate Cochlodinium polykrikoides, hematological responses of the flounder Paralichthys olivaceus and red sea bream Pagrus major exposed to this algal bloom were investigated. The mortality of red sea bream was considerably larger than that of flounder, and the threshold lethal density of C. polykrikoides to the test fish was approximately 3,000 cells/ml. Blood $PO_2$declined in proportion to the increasing density of algal cells. The blood $PO_2$ of moribund fish was about $40-60\% of control test fish. Particularly, the fishes began to be killed when the blood $PO_2$ fell below 30-40 mmHg. However, the blood pH dropped almost 1.0 unit just before fish kill. Hemoglobin and hematocrit levels of fish exposed to C. polykrikoides of 5,000 cells/ml for 24 h and of moribund fish did not show great difference. The concentrations of plasma $Na^+$, $K^+$ and $Cl^-$ were slightly elevated to different magnitudes except $Ca^{2+}$ and plasma osmolality was also increased in Cochlodinium-exposed fish. In the plasma cortisol level, these values of moribund flounder and red sea bream were 4- 5 times higher than those of control fish. These results suggest that the drop of blood $PO_2$ was may be one of the principal causes of fish kill by C. polykrikoides, and the changes of other hematological parameters were secondary responses elicited by the decrease in blood $PO_2$.

The Effects of Alternating Set Pressure Changes on Dynamic Tissue Perfusion Characteristics (교대부양 설정압력 변화가 인체조직의 동적인 관류특성에 미치는 영향)

  • The quantitative effectiveness of powered support surfaces such as APAM in preventing and treating pressure ulcers has not been sufficiently evaluated because of uncertainty of alternating pressure load input and lack of interpretation of dynamic perfusion characteristics of soft tissue. The aim was to verify the dynamic loading effects to sacral tissue perfusion characteristics from alternating set pressure changes. We developed integrated experiment system to supply alternating load to supinely positioned sacrum and concurrently measured $TcPO_2$, $TcPCO_2$ and air cell pressure. Ten aged subjects (5 female, 5 male) were tested with alternating set pressure 20, 30, 40, 50 and 60mmHg. From the dynamic perfusion response eight characteristic parameters were proposed such as average, minimum, maximum and perfusion range regarding to $TcPO_2$ and $TcPCO_2$. A one-way ANOVA was carried out to determine whether the manipulation of alternating set pressure had any effect on $TcPO_2$ and $TcPCO_2$. From the dynamic tissue perfusion response we found mean $TcPO_2$ decreased exponentially as alternating pressure load increased and perfusion range varied mainly because of minimum level change of $TcPO_2$. And perfusion range of $TcPCO_2$ affected by increase of maximum value of $TcPCO_2$. From the results we can get more strict insights about actual physiological dynamic tissue perfusion mechanism under alternating pressure load.

Electrical Conductivity of S$m_2O_3-ZrO_2$ Systems (S$m_2O_3-ZrO_2$계의 전기전도성)

  • Electrical conductivities of $ZrO_2-Sm_2O_3$ systems containing 10, 20, 30, 40, and 50 mol% of $ZrO_2$ have been measured as a function of temperature and of oxygen partial pressure at temperature from 500 to 1000${\circ}C$ and oxygen partial pressures from 1 ${\times}10^{-5}to 1{\times}10^{-1}$ atm. Plots of log conductivity vs. 1/T are found to be linear with an inflection point at around 650$^{\circ}C$ and the temperature dependence of conductivity shows two different defect structures. The conductivities are increased with increasing pressure, slowing a p-type character. The electrical conductivity dependences on $Po_2$ are found to be ${\sigma}{\propto}Po_2^{1/5.3}$ at 650∼1000$^{\circ}C$ and ${\sigma}{\propto}Po_2^{1/6}$ at 500∼650$^{\circ}C$, respectively, The defect structures are Oi" at 650-1000$^{\circ}C$ and $Vs_m$"' at 500-650$^{\circ}C$. The electron hole is main carrier type, however ionic contribution is found at low temperature portion. Ionic contributions increased with the increasing amount of $ZrO_2$ dopant. In 60mol% $ZrO_2-Sm_2O_3$ system, the conductivity is increased with decreasing oxygen pressure.

  • The characteristics of the deposited thin films of the zinc oxide (ZnO) at different oxygen pressures will be elucidated in this work. The resistivity of ZnO thin films were dominated by the carrier concentration under high oxygen pressure conditions while controlled by the carrier mobility at low oxygen ambiences. In addition, we will show the characteristics of the transparent ZnO based thin film transistor (TFT) fabricated at a full room temperature process with gate dielectric of gadolinium oxide ($Gd_2O_3$) thin films.

  • ${CO_3}^{2-}$ containing whisker-like hydroxyapatite powders were synthesized byhomogeneous precipitation method using urea, Dicalcium phosphate anhdrate[DCPA; $CaHPO_4$] and octacalcium phosphate [OCP; $Ca_8H_2(PO_4_)6\cdot5H_20$]were obtained as precursors and they transformed to high crystalline hydroxyapatites at pH 5.62, and 6.54 respectively. According to the condition of the final pH in the solutions for the solution products and urea contents OCP was remained. When the solution product of $Ca^{2-}$ and ${PO_4}^{3-}$ was $1.5\times 10^4$[$mM^2$] and the content of urea was 0.25 mol.$dm^{-3}$ well crystallized whisker-like hydroxyapatite tens of micrometer in length was obtained. By heat treatment DCPA and OCP were decomposed into $\beta$-tricalcium phosphate [$\beta$-TCP ; $\beta$-$Ca_3{PO_4}_2$] and $\beta$-dicalcium phosphate [$\beta$-DCP ;$\beta$-$Ca_2P_2O_4}_2$]. And well-crystallized hydroxyapatite was partially decomposed into $\beta$-TCP at $800^{\circ}C$.

Mutation Spectrum of Manganese (II) Peroxidase Gene in the Pleurotus ostreatus Mutants Induced by Gamma Radiation

  • The mutational spectra in the manganese (II) peroxidase gene (mnp) of the Pleurotus ostreatus mutants induced by gamma radiation (Co$\^$60/) give evidence to prove the effect of gamma radiation on the gene. mnp of each mutant was cloned, sequenced and analyzed. Among the 1941 base pairs of the sequenced region of the mnP genes of 4 mutants (PO-5,-6,-15 and -16), nine mutational hotspots on which the same base was mutated simultaneously were found, additionally 6 mutations were also found at different positions in the mnp gene. These mutation-spectra were predominantly A:T\longrightarrowG:C transitions (50.1%). By the analysis of putative amino acid sequences, PO-5 and PO-16 mutants have 3 and 1 mutated residues, respectively. Since the mutational spectra reported herein are specific to the mnp gene, we propose that the mutational hotspots for the gamma radiation could be in the gene(5) within cells.

Gender-dependent difference in serum paraoxonase 1 levels of Hanwoo, Korean native cattle, and a positive association with meat quality

  • Objective: Paraoxonase 1 (PON1), a calcium-dependent serum enzyme, has been shown to be involved in lipid metabolism. In this study, we examined the putative correlation of the serum PON1 level of Hanwoo, Korean native cattle, with gender and meat quality grade. Methods: PON1 levels were estimated by determining the arylesterase and paraoxonase activities (AE and PO, respectively) in serum samples from Hanwoo individuals (n = 56). Serum PON1 levels were analyzed in different gender groups (female [n = 21], castrated male [n = 17], and male [n = 18]), and meat quality grades (${\geq}1$ [n = 23], 2 [n = 21], and 3 [n = 12]). Results: Serum PON1 levels were similar in female ($AE=120{\pm}55U/mL$, $PO=84{\pm}43mU/mL$) and castrated male ($123{\pm}44U/mL$, $PO=89{\pm}30mU/mL$), while male showed a significantly lower level ($AE=65{\pm}43U/mL$, $PO=44{\pm}34mU/mL$). Furthermore, analysis of serum PON1 levels in three different grades of meat quality showed similar levels in the grades ${\geq}1$ ($AE=118{\pm}49U/mL$, $PO=84{\pm}37mU/mL$) and 2 ($AE=116{\pm}54U/mL$, $PO=82{\pm}43mU/mL$), while the level was significantly lower in the grade 3 ($AE=58{\pm}35U/mL$, $PO=39{\pm}27mU/mL$) of lower meat quality. Conclusion: We discovered the gender-dependent differences in serum PON1 levels of Hanwoo and a positive association of the serum PON1 level with meat quality. Results in this study suggest that PON1 would be a useful serum marker for preliminary screening of Hanwoo individuals with high-quality meat and applicable for genetic improvement.

Molecular Cloning and Characterization of a Novel Exo-β-1,3-Galactanase from Penicillium oxalicum sp. 68

  • Arabinogalactans have diverse biological properties and can be used as pharmaceutical agents. Most arabinogalactans are composed of β-(1→3)-galactan, so it is particularly important to identify β-1,3-galactanases that can selectively degrade them. In this study, a novel exo-β-1,3-galactanase, named PoGal3, was screened from Penicillium oxalicum sp. 68, and hetero-expressed in P. pastoris GS115 as a soluble protein. PoGal3 belongs to glycoside hydrolase family 43 (GH43) and has a 1,356-bp gene length that encodes 451 amino acids residues. To study the enzymatic properties and substrate selectivity of PoGal3, β-1,3-galactan (AG-P-I) from larch wood arabinogalactan (LWAG) was prepared and characterized by HPLC and NMR. Using AG-P-I as substrate, purified PoGal3 exhibited an optimal pH of 5.0 and temperature of 40℃. We also discovered that Zn2+ had the strongest promoting effect on enzyme activity, increasing it by 28.6%. Substrate specificity suggests that PoGal3 functions as an exo-β-1,3-galactanase, with its greatest catalytic activity observed on AG-P-I. Hydrolytic products of AG-P-I are mainly composed of galactose and β-1,6-galactobiose. In addition, PoGal3 can catalyze hydrolysis of LWAG to produce galacto-oligomers. PoGal3 is the first enzyme identified as an exo-β-1,3-galactanase that can be used in building glycan blocks of crucial glycoconjugates to assess their biological functions.
