Effects of aqueous extract isolated from Platycodon grandiflorum against oxidative stress in rat primary hepatocytes

  • Herbal medicines are increasingly being utilized to treat a wide variety of disease processes. The aim of this study was to evaluate the ability of aqueous extract from the roots of Platycodon grandiflorum A. DC (Campanulaceae). Changkil (CK). to affect cellular response in primary cultures of rat hepatocytes to t-butyl hydroperoxide (t-BHP) induced oxidative stress and hepatotoxicity. CK-treated cells showed an increased resistance to oxidative challenge. as revealed by a higher percent of survival capacity in respect to control cells. CK added prior or simultaneously with I-BHP reduced enganced lipid peroxidation measured as production of malondialdehyde and enhnaced intracellular reduced glutathinoe depletion by t-BHP. Furhtermore. CK protected from the t-BHP-induced intracellular generation of reactive oxygen species assessde by montioting dichlorodihydrofluorescein fluorescence. it can be concluded that CK exerts an antioxidant action insice the cell. responsible for the abserved modulation of the cellular response to oxidative challenge. and CK have a marked anitioxdative and hepatoprotective potency.

Effects of Boshimgeonbi-tang on Gene Expression in Hypothalamus of Immobilization-stressed Mouse (보심건비탕(補心健脾湯) 투여가 Stress 유발 Mouse의 Hypothalamus 유전자 발현에 미치는 영향)

  • The genetic effects of restraint stress challenge on HPA axis and the therapeutic effect of Boshimgeonbi-tang on the stress were studied with cDNA microarray analyses, RT-PCR on hypothalamus using an immobilization-stress mice as an animal model. Male CD-1 mice were restrained in a tightly fitted and ventilated vinyl holder for 2hrs once a day, and this challenge was repeated for seven· consecutive days. In the change of body weight it showed that the Boshimgeonbi-tang is effected recovery on weight loss caused by the immobilization-stress. Seven days later, total RNA was extracted from the organs of the mouse, body-labeled with $CyDye^{TM}$ fluorescence dyes and then hybridized to CDNA microarray chip. Scanning and analyzing the array slides were carried out using GenePix4000 series scanner and GenePix $Pro^{TM}$ analyzing program, respectively. The expression profiles of 109 genes out of 6000 genes on the chip were significantly modulated in hypothalamus by the immobilization stress. Energy metabolism-, lipid metabolism-, apoptosis-, stress protein, transcriptional factor, and signal transduction-related genes were transcriptionally activated whereas DNA repair-, protein biosysthesis-, and structure integrity-related genes were down-regulated in hypothalamus. The 58 genes were up-regulated by the mRNA expression folds of 1.5 to 7.9. and the 51 genes were down-regulated by 1.5 - 5.5 fold. The 11 genes among them were selected to confirm the expression profiles by RT-PCR. The mRNA expression levels of Tnfrsf1a (apoptosis), Calm2 (cell cycle), Bag3 (apoptosis), Ogg1 (DNA repair), Aatk (apoptosis), Dffa (apoptosis), Fkbp5 (protein folding) were restored to the normal one by the treatment of Boshimgeonbi-tang.

  • In previous studies, stress in the Information and Communication Technology (ICT) work environment was focused on lowering work performance by negatively affecting the emotions of organizational members. In the current corona pandemic era, as the ratio of organizational members working from home in a smart environment increases, the need for research on the effects of positive and negative stress on work performance is being raised. Accordingly, by applying the Affective Event Theory to the emotional effects of stress on employees' emotional responses, the effects on work performance were analyzed by dividing them into positive and negative emotions. Also, the effect of smart work on work performance was analyzed. This survey was commissioned by a research company and analyzed using Smart PLS 3.0 tool. Therefore, based on the research results, it was suggested that stress does not only negatively affect the emotions of employees, but that the challenge stress that appears in the mind to achieve a goal induces positive emotions in the employees' emotions and helps to improve work performance.

Effects of dietary lysozyme supplementation on growth performance, nutrient digestibility, intestinal microbiota, and blood profiles of weanling pigs challenged with Escherichia coli

  • The aim of this was evaluate the efficacy of lysozyme on growth performance, nutrient digestibility, excreta microflora population, and blood profiles of weanling pigs under Escherichia coli (E. coli) challenge. A total of 30 piglets weaned at 25 days, 7.46 kg body weight, were assigned to three dietary treatments, composed of five replications, two piglets per replication, for 7 days. The dietary treatment groups were negative control (NC; without antibiotics and lysozyme), positive control (PC; NC + antibiotics), lysozyme (NC + 0.1% lysozyme). All piglets were challenged orally with 6 ml suspension, containing E. coli K88 (2 × 109 CFU/mL). Dietary supplementation with lysozyme and PC resulted in no significant differences in average daily gain and gain to feed efficiency. Weanling pigs fed with E. coli challenge with lysozyme and PC treatments had significantly enhanced nutrient retentions of dry matter and energy (p < 0.05); however, there was a tendency to increase nitrogen digestibility. Furthermore, dietary inclusion of lysozyme and antibiotics treatment groups had a beneficial effect on excreta, ileal, and cecal of the fecal microbial population as decreased E. coli (p < 0.05) counts, without effects on lactobacillus counts. A significant effect were observed on a white blood cells, epinephrine and cortisol concentrations were reduced in piglets fed diets containing E. coli challenge with lysozyme and antibiotics supplementation comparison with the NC group. Therefore, the present data indicate that lysozyme in diet could ameliorate the experimental stress response induced by E. coli in piglets by decreasing intestinal E. coli, white blood cells and stress hormones and improving nutrient digestibility.

  • Oxidative stress is considered to be associated with many diseases, such as inflammatory and cardiovascular diseases, aging and cancer. An important etiological mechanism of these diseases may be a causal relationship between the presence of oxidants and the generation of lipid hydroperoxides derived from enzymatic reactions or xenobiotic metabolism. The hydroperoxides can be decomposed to alkoxy- (ROㆍ) and peroxy- (ROOㆍ) free radicals that can oxidize other cell components, resulting in changes in enzyme activity or the generation of mediators, which can cause further cell damage. The aim of this study was to evaluate the ability of aqueous extract from the roots of Platycodon grandiflorum A. DC (Campanulaceae), Changkil (CK), to affect cellular response in primary cultures of rat hepatocytes to t-butyl hydroperoxide (t-BHP) induced oxidative stress and hepatotoxicity.   • Purpose: This study was performed to investigate the influence of children's hospital nurses' job characteristics and job stress on turnover intention. Method: The subjects were 148 nurses who have been working in 6 children's hospitals located in G city. Measurement tools used for this study included job characteristics were created by Lee, and modified by Hong, and job stress tool developed Kim & Ku and modified by Choi. And the tools used for assessing turnover intention were created by Mobley and Becker and modified by Kim. The collected data was analyzed using descriptive statistics, the independent t-test, one-way ANOVA, Pearson' Correlation and regression on SPSS 21.0 program. Result: Job challenge and job fitness among the sub-areas of job characteristics, job stress and salary were identified as factors influencing turnover intention. These factors explained 36.5% of variance of turnover intention. Conclusion: In conclusion, this results can be used to help the children's hospitals creates an environment where nurses can enhance Job challenge and job fitness also, reduce job stress which lead to turnover rate and turnover intention.

A study on the Duty of Arbitrator's Disclosure - Laying stress on the precedent of Korean supreme court - (중재인의 고지의무에 관한 고찰 - 한국 대법원판례를 중심으로 -)

  • An arbitrator is an impartial person chosen to decide the issue between parties engaged in a dispute. But the arbitrator appointed by a party or arbitration institution shall be impartial or independent and should disclose to the administrator any circumstances likely to give rise to justifiable doubts as to his impartiality or independence. If, at any stage during the arbitration, new circumstances arise that may give rise to such doubts, the arbitrator shall promptly disclose such circumstances to the parties and to the administrator. Upon receipt of such information from an arbitrator or a party, an party must challenge any arbitrator whenever circumstances exist that give rise to justifiable doubts as to arbitrator's impartiality or independence. Under these circumstance, there were two cases declared by the Korean Supreme Court in relation to the cancellation of the arbitration award. One arbitral case was cancelled for the reason of the having been arbitral procedure without disclosure arbitrator's impartiality, and the other case was refused to cancel the ward for the reason of the having been arbitral procedure without challenge an arbitrator. There are not, however, the standard to decide what is definitely the arbitrator's impartiality or independence and the difference on qualification between arbitrator chosen by an party and neutral arbitrator in korean arbitration law and rules. Nevertheless, korean court require arbitrator to be impartial and independent and the arbitration parties to challenge arbitrator' impartiality or independence.

Microarray Analysis of Gene Expression by Ginseng Water Extracts in a Mouse Adrenal Cortex after Immobilization Stress

  • To investigate the effects of repeated immobilization-stress challenge on the the hypothalamic-pituitary-adrenal axis, the genomic transcriptome in the adrenal cortex of immobilization-stressed mouse was analyzed by using a cDNA microarray. Mice were subjected to immobilization stress for 2 h per day for 5 consecutive d. With a 4.0-fold cutoff of arbitrary criteria, the expression levels of 168 out of 41,174 genes were significantly modulated in the adrenal cortex by stress when comparing the control and experimental groups. These genes were related to apoptosis, cell cycle, immune response, inflammatory responses, and signal transduction, and thus may be used as potential targets for the development of therapeutics for chronic stress or depression. Six significant genes among these were selected for real time polymerase chain reaction analysis to confirm the change of their expression levels. The gene for phospho 1 was also further investigated because its expression showed the greatest fold-change.

Gastric Fluid and Heat Stress Response of Listeria monocytogenes Inoculated on Frankfurters Formulated with 10%, 20%, and 30% Fat Content

  • This study evaluated the effects of frankfurter fat content on Listeria monocytogenes resistance to heat stress and gastric fluid, and the Caco-2 cell invasion efficiency of the pathogen. A 10-strain mixture of L. monocytogenes was inoculated on frankfurters formulated with 10%, 20%, and 30% fat content (10%: F10, 20%: F20, 30%: F30) and stored at $10^{\circ}C$ for 30 d. The samples were analyzed for L. monocytogenes resistance to heat stress and a simulated gastric fluid challenge. The total bacteria and L. monocytogenes survival rates were measured on tryptic soy agar plus 0.6% yeast extract and Palcam agar, respectively. L. monocytogenes colonies inoculated on F10, F20, and F30 samples were used for a Caco-2 cell invasion assay. In general, no obvious differences were observed between the survival rates of total bacteria and L. monocytogenes grown on different fat contents under heat stress and gastric fluid challenge. However, L. monocytogenes obtained from the F30 samples had a significantly higher Caco-2 cell invasion efficiency than those in the F10 and F20 samples (p<0.05). These results indicate that although high fat content in food may not be related to L. monocytogenes resistance to heat stress and gastric fluid, it may increase the Caco-2 cell invasion efficiency of the pathogen.

Unveiling the Mediating Role of Personality: Exploring the Nexus between Transformational Leadership and Work Stress in Public Organizations

  • This study investigates the interplay between transformational leadership, personality, and work stress in public organizations, focusing on public servants in Putrajaya, Kuala Lumpur. Data from 702 public servants in Putrajaya, Kuala Lumpur, out of 800 distributed questionnaires, were analyzed. Rigorous analysis employed a structural equation model (SEM) with partial least squares (PLS-SEM) methodology. Our empirical analysis challenges the conventional belief of a positive correlation between transformational leadership and work stress in public organizations, revealing no direct link between transformational leadership and increased work stress in this context. Our study highlights a significant positive correlation between personality and work stress among public servants, emphasizing the role of individual personality characteristics in shaping work-related stress levels. Finally, personality emerged as a crucial mediator in the transformational leadership-work stress relationship, indicating that transformational leadership indirectly influences work stress through its impact on personality. This underscores the importance of considering personality as a mediating factor in understanding the transformational leadership-work stress dynamic in public organizations. In summary, our research provides insights into the relationship between transformational leadership, personality, and work stress in public organizations. These findings challenge conventional assumptions, emphasize individual differences in stress levels, and underscore the mediating role of personality in addressing work stress among public servants.
