모과 에탄올 추출물의 아세틸콜린에스테라제 저해활성과 신경세포에서 아밀로이드 전구단백질의 대사에 미치는 영향 (The Acetylcholinesterase Inhibitory Activity of the EtOH Extract of Chaenomelis Fructus and its effects on the Metabolism of Amyloid Precursor Protein in Neuroblastoma Cells)

  • Alzheimer's disease (AD) is a progressive neurodegenerative disorder symptomatically characterized by impairment in memory and cognitive abilities. AD is characterized pathologically by the deposition of ${\beta}$-amyloid ($A{\beta}$) peptides of 40-42 residues, which are generated by processing of amyloid precursor protein (APP). $A{\beta}$ has been believed to be neurotoxic and now is also considered to have a role on the mechanism of memory dysfunction. In this study, we tested that EtOH extract of the fruits of Chaenomeles sinensis Koehne (CSE) affects on the processing of APP from the APPswe over-expressing Neuro2a cell line. We found that CSE increased over 2 folds of the $sAPP{\alpha}$ secretion level, a metabolite of ${\alpha}$-secretase. We showed that CSE reduced the secretion level of $A{\beta}42$ and $A{\beta}40$ by down regulation of ${\beta}$-secretase (BACE) without cytotoxicity. Furthermore, we found that CSE inhibited BACE and acetylcholinesterase activity in vitro. We suggest that Chaenomelis Fructus may be an useful source to develop a herbal medicine for AD.

모과(木瓜)가 장운동(腸運動)에 미치는 영향 연구 (Study of the Intestinal Motility Effects of Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus)

  • Objectives : This study was conducted to compare Chaenomelis Sinensis Fructus with Chaenomelis Lagenariae Fructus, and to examine Fructus chaenomelis having an influence on Intestinal Motility. Methods : We compared Chaenomelis Sinensis Fructus with Chaenomelis Lagenariae Fructus, by observing their effects on Intestinal Motility. Oral administration of water extracts of Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus into albino rats was followed by dealing with carbachol or loperamide, injecting charcoal meal and measuring the moving length in the intestine. Results : By Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus, the Intestinal Motility of normal albino rats did not change significantly. Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus controled the accelerated Intestinal Motility of albino rats. Chaenomelis Sinensis Fructus was not different from the extracting method, Chaenomelis Lagenariae Fructus had effects of control on extracting by methanol significantly. When Chaenomelis Sinensis Fructus was compared with Chaenomelis Lagenariae Fructus, the former was proved to have control effects on Intestinal Motility in decoction extracts and extracts by ethyl ether higher than the latter. By both Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus, the declined Intestinal Motility was not changed significantly. Conclusion : Chaenomelis Fructus suppressed the exasperated Intestinal Motility by carbachol, but did not influence the dropped Intestinal Motility by loperamide. In addition, Chaenomelis Sinensis Fructus was more excellent than Chaenomelis Lagenariae Fructus in its control effects of Intestinal Motility. inhibitory effects by DKT represent a potential therapeutic approach to the treatment of inflammatory diseases.

모과 부탄올 분획의 예쁜꼬마선충 내의 항산화 효과 (Antioxidant Activity of n-Butanol Fraction of Chaenomeles sinensis Fruit in Caenorhabditis elegans)

  • Chaenomeles sinensis (Thouin) Koehne fruit (Rosaceae) has been used as a traditional medicine in Korea, Japan and China to treat sore throat, diarrhea and inflammation. The ethanol extract of C. sinensis fruit was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. Among those fractions, the n-butanol fraction showed the most potent DPPH radical scavenging and superoxide quenching activities. To verify antioxidant activities, the n-butanol fraction was checked the activities of superoxide dismutase (SOD) and catalase activities, and intracellular ROS levels and oxidative stress tolerance in Caenorhabditis elegans. Furthermore, to see if increased stress tolerance of worms by treating of the n-butanol fraction was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain. Consequently, the n-butanol fraction elevated SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Moreover, the n-butanol fraction-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.

Inhibitory effect of Chaenomelis Fructus ethanol extract on receptor activator of nuclear factor-kappa B ligand-mediated osteoclastogenesis

  • The fruit of Chaenomeles sinensis (Thouin) Koehne (Chaenomelis Fructus) known as "Mo-Gua" in Korea has been commonly used in traditional medicine to treat inflammatory diseases, such as sore throat. However, its effect on bone metabolism has not been elucidated yet. Here, we examined the effect of Chaenomelis Fructus ethanol extract (CF-E) on receptor activator of nuclear factor (NF)-κB ligand (RANKL)-mediated osteoclast differentiation and formation. CF-E considerably inhibited osteoclast differentiation and tartrate-resistant acid phosphatase-positive multinuclear cell formation from bone marrow-derived macrophages and osteoclast precursor cells in a dose-dependent manner. In addition, the formation of actin rings and resorption pits were significantly suppressed in CF-E-treated osteoclasts as compared with the findings in non-treated control cells. Consistent with these phenotypic inhibitory results, the expressions of osteoclast differentiation marker genes (Acp5, Atp6v0d2, Oscar, CtsK, and Tm7sf4) and Nfatc1, a pivotal transcription factor for osteoclastogenesis, were markedly decreased by CF-E treatment. The inhibitory effect of CF-E on RANKL-induced osteoclastogenesis was associated with the suppression of NFATc1 expression, not by regulation of mitogen-activated protein kinases and NF-κB activation but by the inactivation of phospholipase C gamma 1 and 2. These results indicate that CF-E has an inhibitory effect on osteoclast differentiation and formation, and they suggest the possibility of CF-E as a traditional therapeutic agent against bone-resorptive diseases, such as osteoporosis, rheumatoid arthritis, and periodontitis.
