Establishment and Selection of Indicator Materials for Cervi Parvum Cornu Pharmacopuncture

  • Background: Recently, Cervi Parvum Cornu pharmacopuncture has been widely used. But no studies on the indicator materials for Cervi Parvum Cornu pharmacopuncture have been conducted. The aim of this study was to select indicator materials that would aid in the uniform preparation of standardized Cervi Parvum Cornu pharmacopuncture. Methods: Three lots of Cervi Parvum Cornu pharmacopuncture were analysed. Each lot was prepared using the same methods and materials. Chondroitin sulfate, alanine, and leucine were selected as the indicator materials for Cervi Parvum Cornu. For standardization, chondroitin sulfate analysis was performed using the colorimetric method, while alanine and leucine were analyzed using liquid chromatography-mass spectrometry (LC-MS). Results: Analysis of the three lots of Cervi Parvum Cornu pharmacopuncture found chondroitin sulfate levels of $108.9{\pm}17.3ug/ml$, $118.8{\pm}5.0ug/ml$ and $112.3{\pm}11.9ug/ml$. Alanine levels were $44.9{\pm}2.8ug/ml$, $44.6{\pm}0.3ug/ml$, and $43.9{\pm}0.2ug/ml$. Leucine levels were $29.6{\pm}0.7ug/ml$, $29.0{\pm}0.1ug/ml$, and $29.4{\pm}0.1ug/ml$. Conclusion: These results suggest that chondroitin sulfate, alanine, and leucine may be useful for the standardization of Cervi Parvum Cornu pharmacopuncture.

A Study on the Biophoton Emission of Cervi Pantotrichum Cornu (녹용(鹿茸)의 Biophoton(생체광자) 방출 특성 연구)

  • Objectives : The difference of delayed luminescence-biophoton emission was investigated in Cervi Pantotrichum Cornu selected randomly. Cervi Pantotrichum Cornu was used as a tonic in Korean medicine. Methods : Randomly selected samples of Cervi Pantotrichum Cornu were radiated with 150 W metal halide lamp for 1 minute. After radiation, biophoton emissions of each sample were detected by electron multiplication(EM)-charge coupled device camera. The detected biophoton image was calculated with unit of counts per pixel. Results : The average biophoton emissions of delayed luminescence with EM ratio of ${\times}l50\;and\;{\times}250$ were distinguished significantly. The maximum biophoton emissions of delayed luminescence with EM ratio of ${\times}250$ were distinguished significantly. Conclusion : These results suggest that biophoton imaging of Cervi Pantotrichum Cornu could become the meaningful method for the study of differentiation and classification of Cervi Pantotrichum Cornu.

Healing of Bony Defects by Cervi Pantotricuhum Cornu Herbal Acupuncture (녹용약침(鹿茸藥鍼)이 골형성(骨形成)에 미치는 영향(影響))

  • Objective : Bone homeostasis is maintained by balance of bone formation and resorption. Therefore, bone related diseases arose by disturbance of this balance between osteoblast and osteoclast activities. To develop a successful screening system of the therapeutic components based on oriental medicine is essential to set out systematic approach for that purpose. Methods : This study was perforated Sprague-Dawley rat femur for bony defects(${\phi}5mm$) by the fissure bur. And experimetal group were treated with Cervi Pantotricuhum Cornu injection at both $Sh{\grave{e}}nsh{\tilde{u}}$(BL23) & $D{\grave{a}}zh{\grave{u}}(BL11)(0.2m{\ell})$. This was evaluated by radiography and histological analysis with in situ hybridization. Results : Cervi Pantotricuhum Cornu Herbal Acupuncture has weak effect on bony defect healing and this was evaluated by X-ray taking and histological analysis with in situ hybridization. Osteocalcin gene expression was not changed by Cervi Pantotricuhum Cornu Herbal Acupuncture in bony defects animal model. Conclusion : Taken together this study show that the Cervi Pantotricuhum Cornu herbal-acupuncture has a weak effect healing of bony defects, this type of approach might give a good chance to explore the favorable effects of Cervi Pantotricuhum Cornu herbal-acupuncture on bone tissue.

The Effects of distilled Cervi Pantotrichum Cornu Pharmacopuncture and Zizyphi Spinosi Semen Pharmacopuncture on the Heart Rate Variability (녹용 약침과 산조인 약침이 정상인의 심박변이도(HRV)에 미치는 영향)

  • Objective : This study was to investigate the effects of distilled Cervi Pantotrichum Cornu Pharmacopuncture and Zizyphi Spinosi Semen Pharmacopuncture on autonomic nervous system with Heart Rate Variability(HRV). Purpose of the trial was to observe what influence distilled Cervi Pantotrichum Cornu Pharmacopuncture and Zizyphi Spinosi Semen Pharmacopuncture have on the autonomic nervous system. Methods : 60 healthy male volunteers were divided into three groups which consist of two experimental groups such as Cervi Pantotrichum Cornu Pharmacopuncture and Zizyphi Spinosi Semen Pharmacopuncture, and a control group. Study design was a randomized, placebo-controlled, double-blind clinical trial. 20 subjects in experimental group were injected distilled Cervi Pantotrichum Cornu Pharmacopuncture at $GB_{21}$(Kyonjong), 20 subjects in other experimental group were injected distilled Zizyphi Spinosi Semen Pharmacopuncture and 20 subjects in control group were injected Normal Saline at $GB_{21}$(Kyonjong). One volunteer of each groups were excluded from analysis because of error during measuring HRV. At the end of the study 57 volunteers completed HRV analysis. HRV was measured by QECG-3:LXC3203(LAXTHA Inc. Korea) at P0, P1, P2, P3, P4, P5 and P6 and its time-course dependent change in each group was analyzed using paired t-test, and the difference of HRV fluctuation among two experimental group and a control was evaluated by one way ANOVA(p<0.05). Results : A. Time Domain Analysis 1. Analysis of Mean HRV, SDNN and Complexity After injection of distilled Cervi Pantotrichum Cornu Pharmacopuncture, Mean HRV was significantly decreased only at 20 minutes after injection. SDNN was significantly increased from 15 minute after injection. Complexity was significantly decreased only at 15 minute after injection. After injection of distilled Zizyphi Spinosi Semen Pharmacopuncture, Complexity was significantly decreased at 10, 15 and 30 minutes after injection. 2. Analysis of HRV index, pNN50 After injection of distilled Cervi Pantotrichum Cornu Pharmacopuncture, pNN50 was significantly decreased at 15, 20 and 25 minutes after injection. Compared with Normal Saline injection, distilled Cervi Pantotrichum Cornu Pharmacopuncture and Zizyphi Spinosi Semen Pharmacopuncture showed significant difference on HRV index and pNN50 at 5 minutes after injection. B. Frequency Domain Analysis 1. Analysis of Ln(TP), Ln(VLF) After injection of distilled Cervi Pantotrichum Cornu Pharmacopuncture, Ln(TP) was significantly increased at 15 and 30 minutes after injection. Ln(VLF) was significantly increased at 10, 15 and 30 minutes after injection. After injection of distilled Zizyphi Spinosi Semen Pharmacopuncture, Ln(VLF) was significantly increased only at 30 minutes after injection. Compared with Normal Saline injection, distilled Cervi Pantotrichum Cornu Pharmacopuncture showed significant difference on Ln(TP) after 5 minutes of injection. Conclusions : Our results suggest that Cervi Pantotrichum Cornu Pharmacopuncture and Zizyphi Spinosi Semen Pharmacopuncture in healthy adult man tend to activate the autonomic nervous system and sympathetic nervous system compared to Normal Saline within normal range.

  • Objectives : The purpose of this study is to observe the effect of Cervi Pantotrichum Cornu Pharmacopuncture at Pyesu($BL_{13}$) on Chronic Cough. Methods : 6 patients with the cough treated with Cervi Pantotrichum Cornu Pharmacopuncture at Pyesu($BL_{13}$), then self-conscious complain rates were studied. Results : There are remarkable improvement in self-conscious complain rate about cough, sputum, sore throat, runny nose, chest pain, fatigue which chronic cough patients complained. Conclusions : Cervi Pantotrichum Cornu Pharmacopuncture is effective to chronic cough.

  • Objectives : This study was to investigate the effects of Cervi Pantotrichum Cornu Pharmacopuncture and Bovis calculus Fel Ursi Pharmacopuncture on autonomic nervous system with Heart Rate Variability(HRV). Purpose of the trial was to observe what influence Cervi Pantotrichum Cornu Pharmacopuncture and Bovis calculus Fel Ursi pharmacopuncture have on the autonomic nervous system. Methods : 40 healthy male volunteers were divided into two experimental groups, such as Cervi Pantotrichum Cornu Pharmacopuncture group and Bovis calculus Fel Ursi Pharmacopuncture group. Study design was a randomized, double-blind clinical trial. 20 subjects in one experimental group were injected Cervi Pantotrichum Cornu Pharmacopuncture, 20 subjects in other experimental group were injected hydrolyzed Bovis calculus Fel Ursi Pharmacopuncture at GB21(Kyonjong). We measured HRV 7 times : baseline measurement and every 5 minutes for 30 minutes after injection. The SPSS 15.0 for Windows was used to analyze the data by the paired t-test(in group) and independent sample t-test(between the groups). Results A. Time Domain Analysis 1. Analysis of Mean HRV, SDNN and Complexity After injection of Cervi Pantotrichum Cornu Pharmacopuncture, Mean HRV was significantly decreased all the times after injection. SDNN was significantly increased at immediate and 25 minute after injection. Complexity was significantly decreased only at 15 minute after injection. After injection of Bovis calculus Fel Ursi Pharmacopuncture, Mean HRV was significantly decreased all the times after injection. Analysis of HRV index, pNN50 After injection of Cervi Pantotrichum Cornu Pharmacopuncture, HRV index was significantly increased immediately after injection. pNN50 was significantly decreased at immediately, 20 and 25 minutes after injection. After injection of Bovis calculus Fel Ursi Pharmacopuncture, HRV index was significantly increased immediately after injection. pNN50 was significantly decreased at immediate and 25 minutes after injection. B. Frequency Domain Analysis 1. Analysis of Ln(TP), Ln(HF), Ln(LF), Ln(VLF) After injection of Cervi Pantotrichum Cornu Pharmacopuncture, Ln(TP) was significantly increased at immediate and 25 after injection. Ln(HF) was significantly increased all the times after injection. Ln(LF) was significantly increased at 15, 20 and 25 minutes after injection. Ln(VLF) was significantly increased at immediate and 25 minutes after injection. After injection of Bovis calculus Fel Ursi Pharmacopuncture, Ln(TP) was significantly increased at immediately and 25 after injection. Ln(LF) was significantly increased at 20 and 25 minutes after injection. Ln(VLF) was significantly increased at immediate and 25 minutes after injection. Conclusions : We results suggest that Cervi Pantotrichum Cornu Pharmacopuncture and Bovis calculus Fel Ursi Pharmacopuncture in healthy adult men tend to activate the autonomic nervous system and sympathetic nervous system within normal range.

The Experimental Study of Electroacupuncture and Cervi Pantortichum Cornu Pharmacopuncture on Pain Decrease and Nerve Regeneration after Crush Injury of Sciatic Nerve (좌골신경 압좌 손상 후 전침 및 녹용 약침의 통증 감소와 신경 재생에 관한 실험적 연구)

  • Objectives : This study was designed to evaluate effects of electroacupuncture and Cervi Pantortichum Cornu pharmacopuncture on pain and nerve regeneration after crush injury of sciatic nerve in rats. Methods : We divided the subjects into 5 groups : control group, acupuncture treated group, electroacupuncture treated group, Cervi Pantortichum Cornu pharmacopuncture treated group, electroacupuncture and Cervi Pantortichum Cornu pharmacopuncture group. Hot plate test and degree of Substance P antibody in each group were observed at the 1st, 2nd, 3rd and 5th day. Sciatic function index and change of BDNF immunoreactivity had been estimated at the 7th, 14th, 21st day respectively. Atrophy of the gastrocnemius at the 21st day was observed. Results : All the experimental groups showed significant changes in hot plate test, degree of Substance P antibody, sciatic function index and change of BDNF immunoreactivity compared with control group. Especially, when electroacupuncture and Cervi Pantortichum Cornu pharmacopuncture used together, results were more effective than other groups. Recovery of the gastrocnemius was also the same. Conclusions : It is suggested that electroacupuncture and Cervi Pantortichum Cornu pharmacopuncture may play a significant role on pain decrease and nerve regeneration after crush injury of sciatic nerve in rats and these two treatments could be more effective when used together.

Effects of Cervi Pontotrichum Cornu on Human Uterine Leiomyoma Cell in vitro (녹용(鹿茸)이 in vitro에서 자궁근종세포(子宮筋腫細胞)에 미치는 영향)

  • Purpose: This study was conducted to investigate the effects of Cervi Pontotrichum Cornu extract solution on the cell cycle regulation and apoptosis in human leiomyoma cell. Methods: The leiomyoma cell of patients was used in the study, and we administered the extract solution of Cervi Pontotrichum Cornu concentration at 1, $10mg/m{\ell}$ to the leiomyoma cell for 48 hours. We used flow cytometry and western blotting to confirm cell cycle and apoptosis. Results: In flow cytometry, G1 phase of the $1mg/m{\ell}$ group prolonged. But G1 phase of $10mg/m{\ell}$ group was shortened and S phase was increased. Cyclin D1 expression increased in higher concentration group. And Bax expression that regulates cell apoptosis increased in $1mg/m{\ell}$ and $10mg/m{\ell}$ group than control group. Bcl-2 expression decreased in 1, $10mg/m{\ell}$ groups than control group. VEGF expression rised in higher Cervi Pontotrichum Cornu concentration group. Conclusion: This study means that Cervi Pontotrichum Cornu could induce the apoptosis of leiomyoma cell by increasing Bax and decreasing Bcl-2 expression. But Cervi Pontotrichum Cornu could increase Cyclin D1 and VEGF expression, so more detailed studies would be needed.

  • To understand immunopharmacologic effects on Cervi Pantotrichum Cornu, Morindae Officinalis Radix, Cistanches Herba, Curculginis Rhizoma, Epimedii Herba, Eucommiae Cortex, we investigated chinese experimental documents, and we could reach conclusions as follows : 1. The effects on cell-mediated immune system were as follows. 1) The effects on macrophage (1) The herbal medicines promoting to increase the number of WBC in the peripheral blood were Morindae Officinalis Radix, Epimedii Herba and that promoting to reinforce the phagocytic functions of neutrophil was Curculginis Rhizoma. (2) The herbal medicines promoting the phagocytic functions of mononuclear, macrophage were Cervi Pantotrichum Cornu, Cistanches Herba, Eucommiae Cortex. 2) The herbal medicines stimulating the activities of T lymphocytes were Cervi Pantotrichum Cornu, Curculginis Rhizoma, Epimedii Herba, Eucommiae Cortex. 2. The effects on humoral immune system were as follows. 1) The herbal medicines increasing the activity of complement receptor were Cervi Pantotrichum Cornu, Curculginis Rhizoma. 2) The herbal medicines reinforcing immunity of spleen cells were Cervi Pantotrichum Cornu, Cistanches Herba, Epimedii Herba. 3) The herbal medicines promoting proliferation of spleen cells that produce antibody after having been immunized by SRBC were Cervi Pantotrichum Cornu, Cistanches Herba, Epimedii Herba. 3. The herbal medicines, reinforcing immunity on delayed type hypersensitivity were Cervi Pantotrichum Cornu, Cistanches Herba, Eucommiae Cortex. As you know in the many bibliological documents, the studies on the effects of Drugs for Tonifying Yang were started along right lines. Recently the studies on those were accomplished more rapidly and applied many immune diseases. We thought that Drugs for Tonifying Yang could be important immunopotentiators. Therefore we can apply those herbal medicines not only to immune diseases but also inflammatory diseases, senile infirmity and all sorts of tumor.

Morphology and Life History of Stylonema cornu-cervi Reinsch (Goniotrichales, Rhodophyta) from Japan

  • The morhology and life history of Stylonema cornu-cervi Reinsch from Japan were investigated. The species had multiseriate erect thalli from a basal cell. The thalli usually branched dichotomously, occasionally trichotomously near the base, and non-branched thalli were sometimes observed. A dichotomous branch on the upper portion near the base occurred only one time on each erect branch. Cells contained a stellate chloroplast, which was composed of a central rounded part with an obscure pyrenoid and 5-8 cup-like lobes connected to the central part by a small thin stipe. The biseriate part was observed on the six-celled stage in culture, and the grown thalli were multiseriate except for base and apices. Monospores forming from the immediate transformation of vegetative cells were observed. Thalli grew at 15-25$^{\circ}C$ and died at 10 and 30$^{\circ}C$. The fastest growth and maturation were observed under 25$^{\circ}C$ and 14L:10D. Although S. alsidii (Zanardini) Drew usually had uniseriate thalli, irregularly branched multiseriate thalli had been reported in cultures. It is possible that in the previous report the thalli were confused with S. cornu-cervi. In this report, S. cornu-cervi were distinguished from S. alsidii in that the branches were few, the multiseriate portions were observed on the early stage (six-celled stage), and the grown thalli were multiseriate except at the base and apices.
