Characterizations of Yields and Seed Components of Sesame (Sesamum indicum L.) as Affected by Soil Moisture from Paddy Field Cultivation

  • Accurate and optimal water supply to cereal crop is critical in growing stalks and producing maximum yields. Recently, upland crops are cultivated in paddy field soils to reduce overproduced rice in Korea. In order to increase productivity of cereal crops in paddy fields which have poor percolation and drainage properties, it is necessary to fully understand crop response to excessive soil water condition and management of soil drainage system in paddy field. The objectives of this study were to investigate effects of excessive soil water to sesame growth and to quantify stress response using groundwater levels. Two cultivars of sesame were selected to investigate; Gunbak and Areum. These sesames were planted in paddy fields located in Miryang, Gyeongnam with different soil drainage levels and drainage systems. The experiment site was divided into two plots by drainage class; very poorly and somewhat poorly drained. Two different drainage systems were applied to alleviate excessive soil water in each plot: open ditch and pipe drainage system. Soil water contents and groundwater levels were measured every hour during growing season. Pipe drainage system was significantly effective to alleviate wet injury for sesame in paddy fields. Pipe drainage system decreased average soil moisture content and groundwater level during sesame cultivation. This resulted in greater yield and lignan contetns in sesame seeds than ones from open ditch system. Comparison between two cultivars, Gunbak had greater decrease in growth and yield by excessive soil water and high groundwater level than Areum. Seed components (lignan) showed decrease in seeds as soil water increased. When soil moisture content was greater than 40%, lignan content tended to decrease than ones from less soil moisture content. Based on these results, pipe drainage system would be more effective to reduce wet injury to sesame and increase lignan component in paddy field cultivation.

Reverse Transcription Recombinase Polymerase Amplification Assay for Rapid and Sensitive Detection of Barley Yellow Dwarf Virus in Oat

  • Barley yellow dwarf virus (BYDV) is an economically important plant pathogen that causes stunted growth, delayed heading, leaf yellowing, and purple leaf tip, thereby reducing the yields of cereal crops worldwide. In the present study, a reverse transcription recombinase polymerase amplification (RT-RPA) assay was developed for the detection of BYDV in oat leaf samples. The RT-RPA assay involved incubation at an isothermal temperature (42℃) and could be performed rapidly in 5 min. In addition, no cross-reactivity was observed to occur with other cereal-infecting viruses, and the method was 100 times more sensitive than conventional reverse transcription polymerase chain reaction. Furthermore, the assay was validated for the detection of BYDV in both field-collected oat leaves and viruliferous aphids. Thus, the RT-RPA assay developed in the present study represents a simple, rapid, sensitive, and reliable method for detecting BYDV in oats.

Rapid and Visual Detection of Barley Yellow Dwarf Virus by Reverse Transcription Recombinase Polymerase Amplification with Lateral Flow Strips

  • Barley yellow dwarf virus (BYDV) has been a major viral pathogen causing significant losses of cereal crops including oats worldwide. It spreads naturally through aphids, and a rapid, specific, and reliable diagnostic method is imperative for disease monitoring and management. Here, we established a rapid and reliable method for isothermal reverse transcription recombinase polymerase amplification (RT-RPA) combined with a lateral flow strips (LFS) assay for the detection of BYDV-infected oat samples based on the conserved sequences of the BYDV coat protein gene. Specific primers and a probe for RT-RPA reacted and optimally incubated at 42℃ for 10 min, and the end-labeled amplification products were visualized on LFS within 10 min. The RT-RPA-LFS assay showed no cross-reactivity with other major cereal viruses, including barley mild mosaic virus, barley yellow mosaic virus, and rice black streaked dwarf virus, indicating high specificity of the assay. The sensitivity of the RT-RPA-LFS assay was similar to that of reverse transcription polymerase chain reaction, and it was successfully validated to detect BYDV in oat samples from six different regions and in individual aphids. These results confirm the outstanding potential of the RT-RPA-LFS assay for rapid detection of BYDV.

Effect of light, ultrasonication and liquid smoke on germination of proso millet (Panicum miliaceum L.) seeds

  • High quality seed of proso millet that has high germination percentage, germination speed, and uniformity demanded to increases rates of mechanization in cereal crop cultivation. In order to improve germination characteristics, proso millet seeds were treated with red light, ultrasonication and liquid smoke (LS) solution that generated from hickory wood. All treatments were performed in seed priming solution with 100mM $NH_4NO_3$ at $15^{\circ}C$ for 24hrs under aeration condition. Seeds were exposed under light intensity of 2000 lux for 15m, 30m, 60m, and 120m in priming solution. Ultrasonic treatment was performed at 60%, 80%, and 100% intensity of 21.6 KHz for 5m, 10m, and 20m in priming solution. For LS treatment seed were soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted solution with $dH_2O$ or 100mM $NH_4NO_3$ solution. The effect of seed treatment was evaluated with germination percentage (GP), mean germination time (MGT), germination index (GI), germination rate (GR), Germination uniformity (GU) and heath seed percentage (HS). Our results demonstrate that red light (15min) or ultrasonication (21.6kHz, 5min) treatment improved MGT, GI, GR, and GU comparing to untreated control. Importantly, we show that LS treatments have significant effect on the health seedling and germination characteristics. Proso millet seeds that treated with 5% LS solution for 24hrs improves HS up to 97% that similar results obtained in 100mM $NH_4NO_3$ priming for 24hrs. The combined treatment with LS solution and 100mM $NH_4NO_3$ priming were not effective in all treatments. Our results demonstrate that treating seeds with LS or 100mM $NH_4NO_3$ priming or ultrasonication improves germination characteristics. The methods described here will help advance research using this species by increasing the germination performance at which successive seed pellet process.

Inhibitory Effects of Resveratrol and Piceid against Pathogens of Rice Plant, and Disease Resistance Assay of Transgenic Rice Plant Transformed with Stilbene Synthase Gene

  • Resvestrol has been known to inhibit bacterial and fungal growth in vitro, and can be accumulated in plant to concentrations necessary to inhibit microbial pathogens. Hence, stilbene synthase gene has been used to transform to synthesize resveratrol in heterologous plant species to enhance resistance against pathogens. In the present study, we investigated the antimicrobial activities of resveratrol and piceid to bacterial and fungal pathogens, which causing severe damages to rice plants. In addition, disease resistance was compared between transgenic rice varieties, Iksan 515 and Iksan 526 transformed with stlibene synthase gene and non-transgenic rice varieties, Dongjin and Nampyeong. Minimum inhibitory concentration of resveratrol for Burkolderia glumae was 437.5 ${\mu}M$, and the mycelial growth of Biplaris oryzae was slightly inhibited at concentration of 10 ${\mu}M$. However, other bacterial and fungal pathogens are not inhibited by resveratrol and piceid. The expression of the stilbene synthase gene in Iksan 515 and Iksan 526 did not significantly enhanced resistance against bacterial grain rot, bacterial leaf blight, sheath blight, and leaf blight. This study is the first report on the effect of resveratrol and piceid against pathogens of rice plant, and changes of disease resistance of transgenic rice plants transformed with stilbene synthase gene.

Effects of Pot Raising Seedling in Extremely Late Seasonal Cultivation for the Increase of Rice Production (기후변화 대응 벼 극만기 재배에서 벼 생산성 향상을 위한 폿트육묘 재배 효과)

  • This study was carried out to evaluate the productivity of rice cultivation in extremely late season in Korea and to confirm the effects of pot-raising seedling culture in this case by assessing the growth and yield performance of rice cultivars that are well adapted to late culture. Suitable cultivars for extremely late culture should not have premature heading habit when transplanted around late July and also secure appropriate growth duration before heading. Geumobyeo 1 and Manjongbyeo showed small decrease in the emerged leaves in transplanting in late July or early August and heading occurred at the same time relatively without premature heading. At transplanting from late July to early August in extremely late rice cultivation, ripening was relatively good until transplanting on August 5. At transplanting on July 30, accumulated temperature from heading to the first frost was $853^{\circ}C$ and ripening was good enough. However, heading was delayed by low temperature at transplanting on August 10 and ripening was very poor due to low accumulated temperature of below $800^{\circ}C$, causing drastic decrease of rice yield. The rice yield increased in transplanting with pot seedling, especially the effects of pot seedling was higher in Geumobyeo 1, but panicle number was not enough to secure rice yield owing to short growth duration. In order to cultivate rice in extremely late season, we should select adaptive rice cultivars that have suitable growth duration and excellent ripening in low temperature. Here, pot-seedling did a great role to secure early growth and bigger growth amounts.

Changes of Physicochemical Properties and Fatty acid Compositions of Rough Rice Stored at Different Storage Temperatures and Periods (벼 정조저장 중 저장온도 및 저장기간에 따른 쌀의 이화학적 특성 및 지방산 조성의 변화)

  • This study was conducted to investigate changes of physicochemical properties and fatty acid compositions of rough rice stored at different storage temperatures and periods. So we analyzed texture, alkali digestion value (ADV), toyo glossiness value, pasting properties, fat acidity and fatty acid compositions of five rice varieties every 4 month on the condition of which rough rice had been stored at different temperatures (ambient and low temperature condition at $15^{\circ}C$) for 2 years. Hardness of cooked rice was increased by storage periods and cohesiveness of cooked rice was not considerably different among varieties according to storage temperatures and periods. ADV was significantly different among varieties and storage periods but not different with storage temperatures. Toyo glossiness value of cooked rice was continuously decreased from 4 months after storage regardless to storage temperature. The pasting properties were considerably affected by storage temperatures and periods of rough rice. Increase in peak viscosity, final viscosity and breakdown was observed but setback was decreased by storage periods. Fat acidity of brown rice was much higher than that of milled rice during storage of rough rice and tend to increase by storage period. Oleic acid among fatty acids of brown rice except Sindongin and Hitomebore tended to be decreased by storage periods and linoleic acid among fatty acids of brown rice of Hopum was decreased by storage periods. The contents of linoleinic acid and stearic acid among fatty acids of milled rice were comparatively decreased from 4 months after storage, whereas the content of palmitic acid tended to be increased by storage periods.

Control Efficacy of Environment-friendly Agricultural Materials against Alfalfa Weevil, Hypera postica Gyllenhal at Chinese Milk Vetch Field (자운영포장에서 알팔파바구미에 대한 친환경농자재의 방제효과)

  • This study was conducted to evaluate pest control efficacy of environment-friendly agricultural materials (EFAMs) according to their spray time and frequency against alfalfa weevil, Hypera postica Gyllenhal, at Chinese milk vetch field in Miryang, Korea. Larva, pupa and adult of H. postica occurred more in year 2010 than those in 2011, and the larval population peaked 5 days faster in year 2010 than on $10^{th}$ May in 2011. Control efficacies of EFAMS sprayed on various time and in several frequencies to H. postica during April in 2010 were 73.6%, 71.9% and 66.2% at single spray in early, middle and late April, respectively and ranged from 77.1% to 78.9% when sprayed twice. With three times spray the control efficacy averaged 87.2%. Control efficacies of EFAMs with a single application in early, middle and late April in 2011 were 57.9%, 66.8% and 65.2%, respectively and ranged from 73.7% to 76.8% when sprayed twice. Control efficacy averaged with 82.7% when EFAMs were sprayed for three times. Accordingly, control efficacy of EFAMs against was increased with increasing spray frequency. Among the tested, Ungsamee(R), Wangjoongwangeco(R) and Muchungjidae(R) with a single application showed relatively higher control efficacy with a 4 to 16% lower control rate than a chemical insecticide, etofenprox 20EC. Therefore, Ungsamee(R), Wangjoongwangeco(R) and Muchungjidae(R) can be effectively used for management of H. postica when sprayed at early April for high occurring population and middle April for low occurring population with a single spray.

"Youhan", New Whole Crop Barley Cultivar of Hooded Spike and Fine Growth Ability in Spring (초기생육이 빠른 삼차망 청보리 신품종 '유한')

  • "Youhan" (Hordeum vulgare L.), a new whole-crop barley cultivar, was developed by the breeding team at National Institute of Crop Science, RDA in 2012. Youhan has the growth habit of III, a light green and mid-sized leaf, hooded and lax-type spikes. The cultivar showed 107 cm of culm length, 641 spikes per $m^2$. The heading date of Youhan was May 1, which is one day later than that of the check cultivar "Yuyeon" in upland, and 2 days earlier than that of Yuyeon in paddy field. The maturation time was similar to check cultivar Yuyeon on June 4 in upland and May 31 in paddy field. In terms of winter hardiness and resistance to lodging and disease, Youhan also performed better than the check cultivar. The average forage dry matter (DM) yield in the regional yield trial was approximately 12.6 ton $ha^{-1}$ and 12.0 ton $ha^{-1}$ in upland and paddy field, respectively, which were 6% and 5% higher than that of the check. The yield also showed 7.3% of crude protein, 26.8% of ADF (acid detergent fiber), 47.8% of NDF (neutral detergent fiber), 67.7% of TDN (total digestible nutrients), and a higher grade of silage quality for the whole-crop barley. Fall sowing cropping of Youhan is recommended only in those areas where the average daily minimum-mean temperatures in January are higher than $-8^{\circ}C$, and it should not be cultivated in mountainous areas of Korea.

Growth, Productivity and Forage Values of Winter Cereal Crops at Paddy Fields in the Southern Region of Korea (남부지역 논에서 동계 맥류의 생육특성, 생산성 및 사료가치)

  • In order to select high-quality winter cereal crops with high yield and to increase self-sufficiency rate of forage, their growth, yield, and feeding value of several cereal crops cultivated in winter were investigated in the paddy field of the southern region. Four wheat cultivars and green barley headed in early and mid-April, while oat and Italian ryegrass headed in early May. Fresh forage yields of wheats, green barley, and oat were significantly higher than that of Italian ryegrass, and dry forage yields of wheats and green barley were significantly higher than those of not only Italian ryegrass but also oat. In particular, the yield of a wheat cultivar 'Cheongwoo' was the highest. Mineral contents of wheat forages, even though low, were in the range 27.8~33.7mg·g-1 DW suitable for feeding cattle and young female cows. Crude protein content of a wheat cultivar 'Cheongwoo' was high up to 7.6%, similarly to 7.0% requiring for feeding cattle. Feeding values such as total digestible nutrients (TDN) and relative feed value (RFV) of wheats and green barley were superior to those of oat and Italian ryegrass. In addition, dry matter rates of 4 wheat cultivars and green barley were in the range 30~40%, indicating that wheat cultivars and green barley could be used for various feeding purposes such as green or dried forage, and silage. Based on these results, wheat cultivars including 'Cheongwoo' and green barley could be encouraged to be cultivated in paddy fields, as high-quality winter forage crops with high yield.
