Genome Sequence of Spinach Cryptic Virus 1, a New Member of the Genus Alphapartitivirus (Family Partitiviridae), Identified in Spinach

  • A distinct double-stranded RNA (dsRNA) cryptic virus, named spinach cryptic virus 1 (SpCV1), was identified from spinach transcriptome datasets. The SpCV1 genome has two dsRNA genome segments. The larger dsRNA1 has an open reading frame for a conserved RNA-dependent RNA polymerase (RdRp). The smaller dsRNA2 encodes a putative coat protein (CP). The sequence identity of SpCV1 RdRp and CP to the closest cryptic virus is 81% and 60%, respectively. Phylogenetic analysis indicates that SpCV1 is a novel member of the genus Alphapartitivirus (family Partitiviridae).

  • Recent attempt to enhance the safety against collision reshaped the simple shell structures into the integrated complex shell structures. Moreover, due to various regulations continuously tightened for environment protection, weight reduction of automobiles becomes an increasingly important issue. Auto parts lightening is mainly accomplished by more reasonable design, adoption of lighter materials and miniaturization of the auto bodies. Focusing on the locally enhanced design approach among the above three ways, we here attempt to develop a patching optimization method, and also to determine the thicknesses of an integrated shell structure, both bringing a specified amount of stress relaxation. We first select a cross member as a patching optimization model. Based on the finite element stress calculations, we relieve the stress of cross member by patching in two ways-nonuniform thickness patching and optimized uniform thickness patching, the latter of which is more effective in a practical point of view for the preset amount of stress relaxation. Selecting a box type subframe as another finite element analysis model, we then determine the thickness of each part by axiomatic design approach for a preset amount of stress relaxation. The patching methodology and the axiomatic approach adopted in this work can be applied to the other complex shell structures such as center member and lower control arm.

A Study on the Optimum Design of the Automotive Side Member to Maximize the Crash Energy Absorption Efficiency (충돌에너지 흡수효율 최대화를 위한 자동차 사이드 멤버 최적 설계에 관한 연구)

  • In this study, the design optimization of the automotive side member is performed to maximize the crash energy absorption efficiency per unit weight. Design parameters which seriously influence on the frontal crash performance are selected through the sensitivity analysis using the Plackett-Burman design method. And also the design variables, which are determined from the sensitivity analysis, are optimized by two methods. One is conventional approximate optimization method which uses the statistical design of experiments (DOE) and response surface method (RSM). The other is a methodology derived from previous work by the authors, which is called sequential design of experiments (SDOE), to reduce a trial and error procedure and to find an appropriate condition for using micro-genetic algorithm. The proposed optimization technique shows that the automotive side member structure can be designed considering the frontal crash performance.

  • In this study, the collapse characteristic of CFRP/Al compound square tube was investigated experimentally. The conclusions are as follows; The impact collapse characteristic of CFRP/Al compound square tube was found to be the most superior stacking conditions $[90^{\circ}]_8$. It showed that a very stable collapse mode was crushing. In the member with $[0_2{^{\circ}}/90_2{^{\circ}}]_s$ and $[90_2{^{\circ}}/0_2{^{\circ}}]_s$, stacking conditions, $0^{\circ}$ fibers were splayed to the external by laminar bending, while the $90^{\circ}$ fibers were held between the folds of the aluminum member by laminar bending, local buckling and transverse crack. In the member with $[45_2{^{\circ}}/45_2{^{\circ}}]_s$ stacking conditions, fibers were held between the folds of the aluminum member by local buckling and transverse crack.

  • Fundamental equations of a member to analysis the elasto-plastic buckling analysis based on the deflection method are derived in this paper, and its validity and accuracy are shown by the numerical examples. The model discussing in the present paper has three elasto-plastic springs which are located at the both ends and center of a member and two elastic beam elements between them. The elasto-plastic springs represent the elasto-plastic behavior of the member and elastic beam element represents buckling behavior of the member. Numerical example shows the validity of this formulation.

The Role of Nuclear Receptor Subfamily 1 Group H Member 4 (NR1H4) in Colon Cancer Cell Survival through the Regulation of c-Myc Stability

  • Nuclear receptor subfamily group H member 4 (NR1H4), also known as farnesoid X receptor, has been implicated in several cellular processes in the liver and intestine. Preclinical and clinical studies have suggested a role of NR1H4 in colon cancer development; however, how NR1H4 regulates colon cancer cell growth and survival remains unclear. We generated NR1H4 knockout (KO) colon cancer cells using clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR-associated protein-9 nuclease (CAS9) technology and explored the effects of NR1H4 KO in colon cancer cell proliferation, survival, and apoptosis. Interestingly, NR1H4 KO cells showed impaired cell proliferation, reduced colony formation, and increased apoptotic cell death compared to control colon cancer cells. We identified MYC as an important mediator of the signaling pathway alterations induced by NR1H4 KO. NR1H4 silencing in colon cancer cells resulted in reduced MYC protein levels, while NR1H4 activation using an NR1H4 ligand, chenodeoxycholic acid, resulted in time- and dose-dependent MYC induction. Moreover, NR1H4 KO enhanced the anti-cancer effects of doxorubicin and cisplatin, supporting the role of MYC in the enhanced apoptosis observed in NR1H4 KO cells. Taken together, our findings suggest that modulating NR1H4 activity in colon cancer cells might be a promising alternative approach to treat cancer using MYC-targeting agents.

A New Member of Human TSA/AhpC as Thioredoxin-dependent Thiol Peroxidase

  • A new type of the human TSA homologous gene was cloned from a HeLa cell cDNA and characterized. The gene product consists of 161 amino acids with a molecular mass of 16,900. The TSA homologous protein, as a new 6th member of the human TSA (hTSA VI), exerted a thioldependent peroxidase activity with the use of thioredoxin system as a physiological electron donor. The values of $V_{max}/K_m$ of hTSA VI for $H_2O_2$ and t-butyl hydroperoxide (t-BOOH) were calculated as $5.53{\times}10^{-2}$ and $3.70{\times}10^{-2}$, respectively. This implies that hTSA VI is a peroxidase, which reduces $H_2O_2$ and t-BOOH. The mutation of $Cys^{47}$ to serine resulted in a complete loss of the peroxidase activity. This suggests that $Cys^{47}$ acts as a primary site of catalysis. The analysis of the tryptic digest derived from hTSA VI revealed that the $Cys^{47}$ exists as a free thiol form. Taken together, these results suggest that the TSA homologous protein is a new type of the human family, which exerts thioredoxin-linked peroxidase activity toward $H_2O_2$ and alkyl hydroperoxide.

Dissemination of Advanced Mouse Resources and Technologies at RIKEN BioResource Center

  • RIKEN BioResource Center (BRC) has collected, preserved, conducted quality control of, and distributed mouse resources since 2002 as the core facility of the National BioResource Project by the Ministry of Education, Culture, Sports, Science and Technology (MEXT), Japan. Our mouse resources include over 5,000 strains such as humanized disease models, fluorescent reporters, and knockout mice. We have developed novel mouse strains such as tissue-specific Cre-drivers and optogenetic strains that are in high demand by the research community. We have removed all our specified pathogens from the deposited mice and used our quality control tests to examine their genetic modifications and backgrounds. RIKEN BRC is a founding member of the Federation of International Mouse Resources and the Asian Mouse Mutagenesis and Resource Association, and provides mouse resources to the one-stop International Mouse Strain Resource database. RIKEN BRC also participates in the International Gene Trap Consortium, having registered 713 gene-trap clones and their sequences in a public library, and is an advisory member of the CREATE (Coordination of resources for conditional expression of mutated mouse alleles) consortium which represents major European and international mouse database holders for the integration and dissemination of Cre-driver strains. RIKEN BRC provides training courses in the use of advanced technologies for the quality control and cryopreservation of mouse strains to promote the effective use of mouse resources worldwide.

  • A statistical annoyance model to railway noise at platform was proposed by jury evaluation test performed in hearing laboratory. ITX-Saemaeul and Mugunghwa were chosen as the noise sources of the test, and announcement sound was included to simulate real situation. Logistic regression analysis produced %HALAB curve. Hosmer-Lemeshow test and classification accuracy test were used to verify the model's statistical significance. It was shown that the model which was generated from relatively small number of samples is statistically significant.
