The changes in chemical constituents of onion in Korea and the degree of hydrolysis in onion by-drolysates by treatment of viscozyme L, celluclast 1.5L, pectinex and cereflo were investigated. Proximate compositions of onion in Korea were as follows; The contents of moisture, protein, lipid, carbohydrate and ash were 89~90%, 1.8~1.9%, 0.4~0.5%, 7.1~8.5% and 0.3~0.5%, respectively. Major free sugars were identified to be glucose, fructose and sucrose and major free amino acids were as cysteine, histidine, arginine and glutamic acid. The degree of hydrolysis by treatment of viscozyme L, celluclast 1.5L, pectinex and cereflo was 69%, 71%, 80% and 62%, respectively. During the enzymatic hydrolysis, free sugar and total sugar content increased however, pyruvic acid content decreased significantly.
Enzyme-assisted extraction is a cost-effective, safe, and efficient method to obtain bioactives from plant materials. During this study, 10 different marine algae from Sri Lanka were individually extracted by using five commercial food-grade carbohydrases. The enzymatic and water extracts of the seaweeds were analyzed for their antioxidant and anti-inflammatory activities. The highest DPPH, hydrogen peroxide ($H_2O_2$) and intracellular $H_2O_2$ scavenging abilities were observed from the Celluclast extract of Sargassum polycystum (CSp). CSp exerted protective effects against oxidative stress-induced cell death in hydrogen peroxide-induced Chang cells and in model zebrafish. The Celluclast extract of Chnoospora minima (CCm) showed the strongest anti-inflammatory activity against lipopolysaccharide (LPS)-induced NO production in RAW 264.7 macrophages ($IC_{50}=44.47{\mu}g/mL$) and in model zebrafish. CCm inhibited the levels of iNOS, COX-2, $PGE_2$, and TNF-${\alpha}$ in LPS stimulated RAW 264.7 macrophages. Hence, CSp and CCm could be utilized in developing functional ingredients for foods, and cosmeceuticals.
The potential antioxidative activity of water-soluble enzymatic hydrolyzates from a kelp, Ecklonia cava was evaluated by free radical scavenging and lipid peroxidation assays. To prepare water-soluble hydrolyzates from E. cava the seaweed was enzymatically hydrolyzed by five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase). Among all the hydrolyzates, Celluclast hydrolyzate effectively scavenged free radicals released from DPPH (1,1-diphenyl-2- pricrylhydrazyl) and recorded around 73% scavenging activity at the concentration of 4 mg ${\cdot}ml^{-1}$. This hydrolyzate was thermally stable and DPPH radical scavenging activity remained 80% or higher at heating temperatures of 40 and 60$^{\circ}C$ up to 12 h and around 80% at 100$^{\circ}C$ up to 8 h. AMG and Ultraflo hydrolyzate inhibited the lipid peroxidation of fish oil as that of $\alpha$-tocopherol. These results suggested that an enzymatic extraction will be an effective way for the production of a potential antioxidant from seaweeds.
The antioxidative activity of various enzymatic extracts from Sarcodon aspratus (S. aspratus) was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the S. aspratus were enzymatically hydrolyzed by seven carbohydrases (Viscozyme, Celluclast, Dextrozyme, AMG, Promozyme, Maltogenase, and Termamyl) and eight proteases (${\alpha}$-chymotrypsin, Alcalase, Flavourzyme, Neutrase, papain, pepsin, Protamax, and trypsin). The DPPH radical scavenging activities of Viscozyme and pepsin extracts were the highest, and the half maximal inhibitory concentration ($IC_{50}$) values were 0.896 and 0.734mg/mL, respectively. The Celluclast and trypsin extracts showed the highest scavenging activities on alkyl radical, and their $IC_{50}$ values were 0.278 and 0.575mg/mL, respectively. The Celluclast extracts was decreased cell apoptosis in PC-12 cells against $H_2O_2$-induced oxidative damage. The findings of the present study suggest that enzymatic extracts of S. aspratus exhibit antioxidative activity against oxidative stress on PC-12 cells.
리그노셀룰로오스 바이오매스를 원료로 하여 발효당을 생산하고자 할 때 최종적인 당수율은 적용하는 전처리 기술의 종류와 전처리 조건에 따라 크게 달라질 수 있다는 사실은 널리 알려져 있다. 또한 전처리물의 효소당화에서 셀룰로오스의 당전환율은 효소의 종류와 사용량에 의존적이므로 부족한 전처리 효과를 일부 보완하기도 한다. 본 연구에서는 바이오매스의 효소당화에 흔히 사용되고 있는 특정효소와 최근 공급되기 시작한 새로운 효소를 시료로 하여 해바라기 줄기의 열수전처리에 이은 효소당화에서 효소의 종류가 최종 당수율, 효소당화 시간 및 전처리효과에 미치는 영향을 측정하였다. 해바라기 줄기 분말을 $180^{\circ}C$에서 30분 동안 열수전처리하였을 때 헤미셀룰로오스의 수율이 최대가 되었으나 이 조건에서는 후속 효소당화에 의한 포도당 수율이 높지 않았다. Celluclast 1.5L 혹은 Celluclast conc BG는 전처리 물의 당화속도가 상대적으로 빠른 편이었고, 이 효소에 의한 당수율은 해바라기 줄기 셀룰로오스 함량의 80% 내외였으며, 바이오매스 1g당 효소첨가량이 3ml까지 증가함에 따라 당수율도 꾸준히 증가하는 경향을 보였다. 반면에 노보자임 스코리아로부터 분양받은 효소로서 NS22074와 NS50010의 혼합물은 Celluclast보다 약 10% 더 높은 당수율을 보여주었으나 당화는 상대적으로 느리게 진행되어 72시간 이상이 소요되었다. Endoxylanase 혹은 hemicellulase 등 다른 효소를 NS22074와 NS50010의 혼합물에 가하여도 당수율의 증대 효과는 미미하거나 거의 없었다. 시험에 사용한 효소제제에는 포도당, 소르비톨 등 여러 가지 당들이 보존제 혹은 안정화제로 함유되어 있는 것으로 나타나서 사용에 주의할 필요가 이었다.
This study investigated the effects of dietary supplementation with Sargassum horneri celluclast hydrolysate (SHCH) on the growth performance, feed utilization, non-specific immunity and antioxidant capacity of the Pacific white shrimp Penaeus vannamei. Six diets were prepared by SHCH supplemention levels of 0, 5, 10, 20, 40 and 80 g/kg (designated as Con, S5, S10, S20, S40 and S80, respectively). A total of 540 shrimp were randomly distributed in triplicate across 18 tanks and fed one of the diets for five weeks. Weight gain did not differ significantly among the groups except for S80. The feed conversion ratio was significantly lower in S10 than in S80. Phenoloxidase and nitro-blue tetrazolium activities were significantly higher in S5 than in Con. The prophenoloxidase gene was significantly upregulated in S10 compared to that in Con. The gene expression of crustin, glutathione peroxidase, lipopolysaccharide and β-1,3-glucan-binding protein were significantly upregulated in S5 compared to Con. The superoxide dismutase gene was significantly upregulated in S20 and S80 compared to Con. Supplementing shrimp diets with up to 8% of SHCH did not impair non-specific immunity or antioxidant capacity. However, based on growth results, supplementation up to 4% was considered appropriate.
A Trichoderma sp. SO-571 producing cellulase was isolated from soil, and a pilot-scale cultivation and separation of cellulase were conducted. The cellulase activity was about 14.5 unit/ml after 112 hr of cultivation in a 301 fermenter containing 3.0% cellulose, 4.0% soybean powder, 3.0% wheat bran, 0.5% ($NH_4$)$_2$$SO_4$0.2% urea, 1.0% CSL, 0.5% $KH_2$PO$_4$, and 0.2% Tween 80. The cellulase was purified over 4.6 folds in three steps with 47.86% yield. The optimum pH of cellulase was pH 5.0 and optimum temperature was $60^{\circ}C$. To investigate the effect of the cellulase-treated cellulosic fabric, the weight loss was compared. The weight loss of denim treated with cellulase from Trichoderma sp.SO-571 was 2.9% and that with Celluclast 1.5L was 2.2%. In tencel treatement with enzyme, cellulase showed 0.7% higer weight loss than that with Celluclast 1.5L.

We investigated the improvement of the antioxidative activity of black garlic with enzymatic treatment and lactic acid bacteria cultivation conditions. Celluclast, a commercially-available polysaccharide hydrolyase, was selected to obtain high total polyphenol content in a black garlic suspension. A lactic acid bacterial strain showing fast growth and high acid production in a black garlic suspension was isolated from Kimchi. This strain was identified as Lactobacillus pentosus 310-7. Enzymatically hydrolyzed black garlic was fermented using the L. pentosus 310-7 strain at $30^{\circ}C$ for 15 hr. The pH and titratable acidity achieved were 4.24 and 0.35%, respectively, after 15 hr fermentation. The viable cell population of L. pentosus 310-7 slowly increased to 7.54 log CFU/g. The polyphenolic compound content, known antioxidants, in black garlic was enhanced with Celluclast treatment and L. pentosus 310-7 cultivation. Total polyphenolic compounds were increased to approximately 60% of the initial concentration, and electron donating ability was also improved, from 39.8 to 65.9%.
In this study, we focused on natural water-soluble antioxidants from fresh water microalgae, Pediastrum duplex and Dactylococcopsis fascicularis from Jeju Island, Korea. They were prepared by enzymatic digestion using five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase), and the potential antioxidant activity of each was assessed. All enzymatic digests from P. duplex showed significant DPPH scavenging effects. Termamyl (60.6%) digest from P. duplex possessed the highest effects on hydrogen peroxide scavenging. Celluclast (58.1%) and Kojizyme digests (56.9%) from D. fascicularis possessed higher effects on superoxide anion radical scavenging. All enzymatic digests exhibited significant effects on both NO· scavenging and metal chelating. Lipid peroxidation was significantly in inhibited Viscozyme, Termamyl and Kojizyme digests from P. duplex and Ultraflo, Protamex, Kojizyme and Alcalase digests from D. fascicularis. These data suggest that enzymatic digests of the fresh water microalgae, P. duplex and D. fascicularis might be valuable sources of antioxidant which can be applied in food and pharmaceutical industry.
