Nuclear factor kappa-B- and activator protein-1-mediated immunostimulatory activity of compound K in monocytes and macrophages

  • Background: Compound K (CK) is a bioactive derivative of ginsenoside Rb1 in Panax ginseng (Korean ginseng). Its biological and pharmacological activities have been studied in various disease conditions, although its immunomodulatory role in innate immunity mediated by monocytes/macrophages has been poorly understood. In this study, we aimed to elucidate the regulatory role of CK on cellular events mediated by monocytes and macrophages in innate immune responses. Methods: The immunomodulatory role of CK was explored by various immunoassays including cell-cell adhesion, fibronectin adhesion, cell migration, phagocytic uptake, costimulatory molecules, reactive oxygen species production, luciferase activity, and by the measurement of mRNA levels of proinflammatory genes. Results: Compound K induced cell cluster formation through cell-cell adhesion, cell migration, and phagocytic activity, but it suppressed cell-tissue interactions in U937 and RAW264.7 cells. Compound K also upregulated the surface expression of the cell adhesion molecule cluster of differentiation (CD) 43 (CD43) and costimulatory molecules CD69, CD80, and CD86, but it downregulated the expression of monocyte differentiation marker CD82 in RAW264.7 cells. Moreover, CK induced the release of reactive oxygen species and induced messenger RNA expression of proinflammatory genes, inducible nitric oxide synthase, and tumor necrosis factor-alpha by enhancing the nuclear translocation and transcriptional activities of nuclear factor kappa-B and activator protein-1. Conclusion: Our results suggest that CK has an immunomodulatory role in innate immune responses through regulating various cellular events mediated by monocytes and macrophages.

Effect of Inosiplex on the Cell-mediated Immune Response of Patients with Lepromatous Leprosy (Inosiplex가 나종형 나환자의 세포성 면역반응에 미치는 영향)

  • This study was attempted to investigate the effect of inosiplex on the cell-mediated immunity in the patients with lepromatous leprosy. Fourteen patients received inosiplex (6gm/day) for 1 month. About 20% of the patients (3 of 14) showed conversion of the lepromin reaction and bacteriological index was significantly decreased in 3 of 7 patients. However, inosiplex administration had no effect on the lymphocyte blastogenesis to M. leprae. The clinical evolution showed a favorable activity of the drug on cutaneous lesions in some patients. The tolerance of the drug was excellent. No side effects were observed. These results suggest that inosiplex may have a moderate immunopotential value in lepromatous leprosy.

  • As a mood-altering drug, long-term alcohol consumption have significant harmful effects on the human body and people's mental functioning. This study observed that the suppression of cell mediated immunity induced in spleen of ICR mouse by long-term alcohol administration. After 8% alcohol voluntary administered for 120 days, the splenic tissue irnmunohistochemically stained by following ABC method that used monoclonal antibody including L3T4(CD4), Ly-2(CD8), IL-2 receptor(CD25R) and NK-1.1(CD56) after embedding with paraffin. The results were as follows. 1. The size of marginal zone in splenic white pulp was diminished and the number of macrophage in marginal zone was decreased in test group than control group. 2. After alcohol administration, the number of Helper T lymphocyte, cytotoxic T lymphocyte, and IL-2 receptor were decreased in periarterial lymphatic sheaths of white pulp and penicilla artery of red pulp and the degree of CD4, CD8, and CD25R positive reaction were soften. 3. In test group, the number of NK cell were decreased. These results indicated that the secretion of lymphokine as IL-2 was inhibited by long-term alcohol administration and subsequently prevent to activate and proliferate splenic T lymphocytes and NK cells as cell mediated immunity component.

The Effects of Saenghyuldan(shengxiedan) on Bone Marrow Failure (생혈단의 골수부전 치료효과에 대한 실험적 연구)

  • Objectives: This experimental study was carried out to prove the effect of Saenghyuldan(SHD; shengxiedan) on bone marrow failure induced by cyclophosphamide(CY) and irradiation in mice. Methods: The following were performed; immunopathology, histopathlogical findings of bone marrow and in the smear of myelocyte. hematopoietic cytokine(IL-3, GM-CSF, TPO), hematopoietic stem cell colony assay, humoral immunity(LPS mitogen response), cell-mediated immunity (Con A mitogen response) and nonspecific immunity(macrophage adherence & phagocytosis) in vitro or vivo. Results: SHD showed a protective effect on bone marrow failure induced by cyclophosphamide(CY) and irradiation in mice. SHD increased lymphoproliferative responses to LPS and Con A, and activated macrophage adherence and phagocytosis to SRBC. Conclusions: We expect that SHD can be used to treat bone marrow failure and immune suppression induced by the chemotherapy or radiation.

  • This study was designed to investigate age-related changes in body composition and cell mediated immunity. Male Sprague-Dawley rats were fed stock diet ad libitum and 12 rats were sacrified at 1, 4, 6, 12 months of age. Body weight Increased sharply from 1 to 4 months. and Increased steadily thereafter. The weights of liver, epididymal fat pads, and kidney increased in similar pattern as body weight, but their relative ratio to body weight decreased with age. The ratio of epididymal fat pads to body weight increased with age. The weight of extensor digitorium longus, coleus and plantaris increased from 1 to 4 months, but it decreased at 6, 12 months. The protein content of muscles decreased or increased through- out 12 months. The T cell proliferation response to Con A stimulation was significantly lower at 6 months than 1 month and lower at 12 months than 6 months.

  • In order to evaluate the effects of the cell-mediated immunity of the animal in the chronic diseases the guinea pigs and rabbits were inoculated with Mycobacterium bovis. After 6 weeks these animals were sensitized and challenged with 2,4-dinitrochlorobenzene. The cutaneous reactions observed in these animal species were similar each other. Macroscopic and microscopic responses in the animal experimentally infected with M. bovis were markedly reduced compared to those in the control animals. The results indicated that the cell-mediated immunity of the animals was depressed by infection of M. bovis.

  • Mancozeb is a protective fungicide on plants and a polymeric complex of ethylene bisdithiocarbamate manganese with zinc salt. It is reported to induce teratogenic and carcinogenic effect in laboratory animals. But the immunomodulating effects of Mancozeb exposure have not been systemically evaluated. The purpose of this study was to investigate the effects of Mancozeb on cell-mediated immunity in mice. For ex vivo assessment, mice were orally exposed to Mancozeb dissolved in distilled water as concentrations of 2,500, 5,000, 10,000 mg/kg for single occasion (acute exposure) or 250, 1,000, 1,500 mg/kg/day 5 days a week for 30days(subacute). Immunohistochemical Observation of Plasma Cell Granuloma in Intraoral Chronic Inflammatory Lesions (구강내 만성염증병소에서 보이는 형질세포육아종의 면역조직화학적 관찰)

  • Purpose: Chronic inflammatory gingival lesions occur as pyogenic granulomas or non-specific chronic suppurative lesions. Methods: Of the 59 chronic inflammatory gingival lesions examined, plasma cell granuloma (n=14), which showed an intense antibody-mediated immune reaction with the increased infiltration of plasma cells, was observed as a pseudotumor-like gingival overgrowth and myofibroblastic or fibrohistiocytitc proliferation of stromal cells with a heavy collection of plasma cells. The levels of CD3, CD20, CD31, CD68, RANKL, cathepsin G, cathepsin K, lysozyme, TNF${\alpha}$, MMP-2, and MMP-9 in the 14 cases of gingival plasma cell granuloma with immunohistochemical detection were measured to determine the pathogenetic progresses of the plasma cell granuloma compared to the common pyogenic granuloma (n=45) in the gingiva. Results: The gingival lesions of the plasma cell granuloma could be divided into three histological types, plasma cell predominant type (PPT, n=8), mixed inflammatory cell type (MICT, n=2), and sclerosed fibrosis type (SFT, n=4). The PPT showed a condensed infiltration of plasma cells into the perivascular spaces of the granulomatous lesion with frequent formation of Russel's body in their cytoplasm. The MICT showed the concomitant infiltration of many macrophages together with plasma cells, resulting in the diffuse destruction of stromal fibrous tissue. The SFT showed granulomatous lesions replaced gradually by thick collagenous fibrous tissue, resembling an inflammatory pseudotumor. The SFT expressed strongly the lymphocytic markers, CD3 and CD20, and the macrophage/monocyte markers, CD31 and CD68, but showed reduced expression of common inflammatory markers, TNF${\alpha}$, cathepsin G, lysozyme, MMP-2, and MMP-9, as well as the reduced expression of osteoclastogenic markers, RANKL and cathepsin K. Conclusion: These results suggest that a gingival plasma cell granuloma shows variable gene expression for cell-mediated immunity and stromal tissue degeneration, undergoing sclerotic fibrosis with a persistent inflammatory reaction.

Effects of Vinblastine and Vincristine on the Primary and Secondary Cell-mediated Immunity (Vinblastine과 Vincristine이 1차(次) 및 2차(次) 세포성(細胞性) 면역반응(免疫反應)에 미치는 영향(影響))

  • Effects of vinblastine(VLB) and vincristine(VCR) on cell-mediated immunity(CMI) were studied with the microcytotoxicity test(MCT) after normal or pre-sensitized Balb/c mice had been treated in vivo with a combination of two different doses of VLB or VCR(single dose of 20% and 60% $LD_{50}$, i.p.) at different times (from day -6 to day +4) plus allo-transplantation antigen(allo-TA, cells from C3H mice at day 0). The results were that $LD_{50}$ of VLB for female Balb/c mouse was 7.3mg/kg body weight (i.p.) and $LD_{50}$ of VCR was 4.3mg/kg body weight and that VLB and VCR acted as immunosuppressive agents on the primary CMI when administered after allo-TA(antigen-drug-phase), but showed no effect when administered prior to allo-TA(drug-antigen-phase). Change of doses of VLB and VCR(20% $LD_{50}$, 60% $LD_{50}$) caused quantitative or qualitative variations in the immunomodulating effects of these two drugs. Neither VLB nor VCR had any immunomodulating effect on the secondary CMI. Lastly, the results support that the four parameters (type of drug, sensitization status, time of drug treatment in relation to antigen injection, and drug dosis) are significant for the effects of the VLB and VCR on the CMI, and that VLB and VCR may inhibit the proliferation of antigen-stimulated T effector lymphocytes but not memory-cytotoxic T lymphocytes.

A comparative study of the gamma-interferon assay and the single intradermal tuberculin test for the diagnosis of bovine tuberculosis under field conditions (소 결핵병 검사에 사용되는 감마인터페론법과 단일피내접종법의 상관관계 비교분석)

  • Bovine tuberculosis (bTB) is a wide-spread zoonotic disease in cattle, which is casued by Mycobacterium bovis that is a part of Mycobacterium tuberculosis complex (MTC). This study describes a field trial conducted in 42 herds with the history of prevalence bovine tuberculosis. Two cell-mediated immunity tests, the gamma-interferon (${\gamma}-IFN$) assay and the single intradermal tuberculin test (SIT) were applied for the diagnosis of bovine tuberculosis in 5,289 animals. The ${\gamma}-IFN$ assay presented 144 (2.7%) head of cattle with the positive result, and 112 (2.1%) head of cattle were shown to be bTB-positive by the SIT. The positive concordance was 45.5%, and the negative concordance was 98.2%. The ${\gamma}-IFN$ assay showed more positive results in younger cattle, especially between 12 and 23 months of age. It is shown that the strategic combination of both cell-mediated immunity test methods is more efficient for the detection of bTB to reduce the number of false positive individuals which are being slaughtered.
