A Study of Dewatering Phenomena of Potato Slice Cytorrhysed by High Molecules (고분자 용액의 세포 압착 현상에 의한 감자 절편의 탈수 현상에 관한 연구)

  • To study simultaneous water and solute transport kinetics during soaking in concentrated solution, the influence of the concentration and molecular weight of the solute(polyethylene glycol(PEG) and NaCl) in the soaking solution and the temperature on the water loss and solute gain rates were observed by using a model vegetable tissue(potato). When potato slices$(4cm{\times}4cm{\times}0.1cm)$ soaked in 60% PEG solutions, the water loss rate of the early phase decreased with increasing of the molecular weight of PEG from 200 to 6,000, while the final water loss increased with increasing the molecular weight of PEG and it reached to 80%. The cell wall of potato tissue was permeable to NaCl and PEGs of which average molecular weight is smaller than 400 but it was not permeable to PEG 600 and larger molecules. PEG which has average molecular weight below 600 induced plasmolysis and those above 600 induced cytorrhysis. The water loss rate of potato sample soaked in smaller molecular weight PEG solution was faster than those soaked in higher molecular weight PEG solution before cytorrhysis happened. The water loss rate was reversed after cytorrhysis happened. The volume change of potato within the first 60 minutes was larger in low molecular PEG solution but the final ratio of decreasing volume was larger in high molecular PEG solutions. In PEG 200 solution, the potato tissue was slightly shrinked without shape change. However, in PEG 4,000 solution, volume of potato was reduced significantly and potato tissue was twisted.

Leucaena Seeds as Protein Supplement in the Rations of Growing Sheep

  • The study was carried out to evaluate leucaena seeds as a protein replacement of mustard seed cake (MSC) in the concentrate mixture of growing lambs. Fifteen owing male lambs (Local${\times}$Corridale) with an average body weight of 16.3 kg were allocated into three dietary treatments (T1, T2, and T3) with five animals in each group. Animals were offered dry mixed grass, berseem hay and concentrate mixture to meet their nutrient requirements. In concentrate mixture of T1, (Control) MSC was used as protein source, while in T2 and T3 groups, 25 and 50% of MSC was replaced by leucaena leucocephala seeds. On completion of three months (90 days) of feeding, a digestion cum-metabolism trial was conducted to determine DMI, nutrient utilization, and nitrogen balance. Changes in body weight were recorded at 15 day internals and eating patterns were recorded for 3 consecutive days at the end of the feeding trial. MSC had higher CP contents than leucaena seeds (27.0%). Mimosine contents in leucaena seeds were 1.1 compared to 0.2 and 0.4% in concentrate mixture of T2 and T3 group, respectively. Dry matter intake varied non-significantly ($79.3{\pm}1.2$ to $83.4{\pm}1.3g/kg$ $w^{0.75}$) across the dietary treatments. Digestibility of DM and cell wall polysaccharides (NDF, ADF. Cellulose and hemicellulose) were comparable, however CP digestibility was relatively lower in leucaena luecocephala seeds based groups (T2 $45.5{\pm}1.7$ and T3 $46.7{\pm}3.5$) compared to MSC supplemented group (T1 $47.7{\pm}0.9%$). The growth rate of lambs was non-significantly higher in T1 ($79.2{\pm}5.4$) compared to T2 ($73.8{\pm}8.8$) and T3 ($73.9{\pm}7.0$), respectively. The animals were in positive nitrogen balance and N-balance varied from 1.8 to 2.9 g/d across treatment groups. The eating rate (% of total offered) of concentrate up-to 15 min was relatively higher in T1 (82.4) than T2 (74.2) and T3 (77.8%). However no effect of leucaena seeds was recorded on total DMI of animals. The results of the study revealed that the inclusion of up to 50% leucaena seeds, as protein source in concentrate mixture of lambs had no adverse effect on DMI, nutrient utilization, eating patterns, nitrogen balance and growth performance of lambs.

Nutrient Intake, Its Utilization, Rumen Fermentation Pattern and Blood Bio-Chemical Constituents of Sheep Fed Urea Treated Mustard (Brassica campestris) Straw

  • A study was conducted to compare the feeding value of urea treated and untreated mustard straw (MS) for sheep. Treated MS was prepared by adding urea-N at 1.84% and followed by packing in a pit silo for 21 days. Two groups of six empty Avikaline ewes were fed untreated (UTMS) and treated (TMS) mustard straw along with 200 g concentrate per head daily for 90 days. Untreated MS had 0.41% N and the urea treatment increased its N value to 1.58 %. The cell wall constituents were decreased in the TMS except for cellulose which remained unaffected. Dry matter intake of TMS was consistently higher than that of UTMS. Digestibility of DM, OM and fibre fractions of MS improved by the urea treatment. Ewes in both groups were in positive N balance while % N retention was lower in UTMS (26.30%) than in TMS (52.14%). The TMS fed group on average consumed 30.2 g DM, 2.9 g digestible crude protein and $0.2MJ\;DE\;per\;kg\;BW\;day^{-1}$ and maintained their weight whereas, the UTMS fed ewes lost weight. The VFA concentration in rumen liquor was higher in TMS than in UTMS. Total-N, ammonia-N and TCA-precipitable-N were also higher in TMS fed ewes. Blood glucose concentrations in the two groups were similar at initiation of the study. However the glucose concentration of UTMS fed group was significantly (p<0.01) lower than those fed UTMS at the termination of the study. Urea-N concentration was also higher in TMS fed group after 90 days of feeding period. It is concluded that urea treatment of MS improved N value of MS from 0.41% to 1.58% along with sizable improvement in nutritive value and in conjunction with 200 g concentrate, TMS can serve as maintenance ration for sheep.   • Gametogenesis and gonadal development of the sea squirt Halocythia roretzi, which is two years old were investigated by histological study. The specimens were collected in Guryong-po coastal area Kyoungsangbuk-do, Korea from May 1996 to April 1997. The sea squirt is hermaphrodite and oviparous. The ovary is located in the inner wall of the tunic year-round, but the testis can be distinguished from in June. The ovary is composed of 6∼8 gonoducts at the left side and 8∼10 ones at the right side, the testis consists of the complex gonad having irregular sacular structures. Oogonia in the ovarian sac were 11.7∼15.6 ${\mu}m$ in diameter. The early developing oocytes were 39.6∼47.6 ${\mu}m$ and nucleus 10.0∼25.0 ${\mu}m$ in diameter. Oocytes in the ovarian sacs during vitellogenesis were 158.6∼210.0 ${\mu}m$, and fully ripe oocytes which were to 210.0∼230.9 ${\mu}m$ in diameter had several test cells in the cortical parts showing a characteristic of vertebrate. The testis showed a general spermatogenesis as in the marine animals. The three-year old sea squirt occurred the first spawning between January to February under 10$^{\circ}C$

Physicochemical Properties of Wheat Bran in Different Areas Prepared by a High-pressure Homogenizer Process (고압균질처리에 따른 산지별 밀기울의 이화학적 특성)

  • In this study, the effects of high-pressure homogenizer treatment on the physicochemical properties of wheat bran from different areas were evaluated. The results showed that the high-pressure homogenizer process could effectively decrease particle size and loosen the microstructure of the wheat bran matrix. As the particle size decreased, the bulk density of wheat bran was significantly decreased (p<0.05) and the water-holding capacity, swelling capacity, oil-holding capacity, and cation-exchange capacity were substantially increased. In addition, microscopic analysis revealed the gradual disintegration of the original cell wall structure and the dissociation of bran tissues over the course of high-pressure homogenization treatment. Scanning electron micrographs showed that the process could also effectively separate out the structural components of wheat bran. These results suggest that the high-pressure homogenizer process is an effective method to modify the physicochemical properties of wheat bran and likely other cereal brans, which might provide potential fiber-rich ingredients for use in functional foods.

Foliar ultrastructure of Korean Orostachys species (한국산(韓國産) 바위솔속(屬) 엽육조직(葉肉組織)의 미세구조(微細構造))

  • Ultrastructural characteristics were examined with leaves of three species, O. japonicus A. Berger, O. malacophyllus Fisch., and O. sikokianus Owhi that probably have CAM mode. The mesophyll cells of these Orostachys possessed vacuoles with precipitates, myelin-like figures, and plasmalemmasomes, along with typical chloroplasts, microbodies and darkly stained bodies in their thin peripheral cytoplasm. Separation of the plasmalemma from the cell wall, leaving a space between them, was a common phenomenon in these species. A complex array of small to large vacuoles which contain small, membrane-bounded vesicles or vacuole-like structures were frequently found. A well-developed thylakoid system was observed in the chloroplasts and this indicates that the photosynthetic capacity of these mesophyll cells is probably active. A peculiar configuration of cytoplasm, especially around the chloroplasts, was also encountered. The variety of cytoplasmic constituents and vacuoles suggest the water-storing mesophyll cells may be complex in function. Some cellular features detected in this study strongly suggest the possible occurrence of CAM mode in Orostachys species.

Effect of Compaction Method on Induced Earth Pressure Using Dynamic Compaction Roller (진동롤러에 의한 다짐방법이 인접구조물의 다짐토압에 미치는 영향)

  • To increase the structural integrity of concrete box culvert good compaction by the dynamic compaction roller with bi9 capacity is as effective as good backfill materials. It is needed for effective compaction that a compaction roller closes to concrete structure with high frequency. However structural distress of the culvert could be occur due to the excessive earth pressure by great dynamic compaction load. To investigate the characteristics of Induced stress by compaction, a box culvert was constructed with changing cushion materials and compaction methods. Two types of cushion material such as tire rubber chip and EPS(Expanded Polystyrene) were used as cushion panels and they are set on the culverts before backfill construction. Laboratory test result of cushion material says that the value of dynamic elastic modulus of rubber is lesser than that of EPS. On the other hand, material damping of rubber material is greater than that of EPS. In most case, dynamic compaction rollers with 10.5 ton weights were used and vibration frequency was applied 30Hz for the great compaction energy. This paper presents the main results on the characteristics of dynamic earth pressures during compaction. The amounts of induced dynamic pressures$(\Delta\sigma\;h)$ by compaction are affected with construction condition such as compaction frequency, depth of pressure cell, distance between roller and the wall of culvert and roller direction. Based on the measured values dynamic lateral pressure on the culverts, it could be said that orthogonal direction of roller to the length of culvert is more effective to compaction efficiency than parallel direction.

  • The pine wilt disease is one of the most serious forest diseases that kill the pine trees, and the study on the invasion and movement of the pine wood nematode within the tree is very important for understanding the inhabitation of pine wood nematode. In this relation, the microscopic observation was carried out to study the place of inhabitation and movement of pine wood nematode within the infested wood. In result, the rays were mainly infested by pine wood nematode and showed dark discoloration due to their necrosis in cross, radial and tangential surface. Also, the intensive damage was found in the resin canals. On the other hand, some traumatic resin canals in tangential band were identified in the sapwood near the cambium. In the ray, the pine wood nematode occurred more commonly in the ray parenchyma cell and fusiform ray with horizontal resin canal than in the ray tracheid and uniseriate ray without horizontal resin canal, respectively. The pine wood nematode was thought to move from ray to tracheid through the large natural opening, window-like pit, in the cross-field, neither through the small natural opening, bordered pit, in the tracheid nor through the tracheid wall by creating a bore hole.

  • Malignant mesenchymoma is a very rare tumor presented during the embryonic and infant period and malignant mesenchymoma in the adult is extremely rare. Tumor is composed of two or more unrelated mesenchymal derivatives apart from fibrous tissue. These tumors are thought to be originated from embryonic mesenchyme capable of differentiating into any type of connective tissue. A 61 years old man with complaints of cough and copious sputum of onset of two months was admitted after initial examinations, showing a very huge mass over the right upper lobe. Right pneumonectomy with partial rib resection of 3rd, 4th, and 5th ribs was performed due to the initial diagnostic impression of squamous cell carcinoma by the fine needle aspiration biopsy. The operative field presented a mass locating across the interlobal fissure with severe adhesions to the chest wall. Postoperatively, the patient received 5,000 rads of radiotherapy and presently, 6 months later, has shown no signs of recurrence.

THE EFFECT OF THE CITRIC ACID ON THE REPAIR OF THE DENUDED ROOTS TRANSPLANTED IN PERIODONTALLY INVOLVED EXTRACTION SOCKETS IN DOGS (성견 치주질환 발치와에 이식된 구연산 처리 치근의 치유에 대한 연구)

  • The author transplanted periodontally-diseased teeth which had been treated with citric acid into a clinically healthy extraction sockets and periodontally-affected extraction sockets, and compared with the healing processes within these tissues. Recipient sites were prepared by surgically removing a part of alveolar bone of premolars of adults dogs, placing elastic orthodontic ligatures for 8weeks, thereby inducing periodontal disease. The diseased roots were extracted and transplanted into healthy extraction sockets, and these were designated as control group 1. Diseased roots transplanted into diseased sockets were designated as control group 2. Diseased roots which had been root planed, treated with citric acid and transplanted into healthy sockets were designated as experimental group 1, while identically treated roots which had been transplanted into diseased sockets were designated as experimental group 2. Observations were made at weeks 2, 8 and 12, with following results. 1. At week 2, experimental group 2 showed some inflammatory cell infiltration in the connective tissue above the extraction sockets, while control groups showed less inflammatory or foreign body reactions throughout the experiment. 2. In both control groups, root surface resorption was observed throughout the experiment, while experimental groups showed a little resorption. 3. Control group 1 & 2 showed ankylosis by newly-formed bone ground the resorbed root surfaces, while experimental group 1 & 2 displayed collagen fibers which are not functionally-arranged, with random, loose arrangement or parallel orientation to root surfaces, and newly-formed bone outside of them. 4. In both control groups & experimental groups which had been transplanted into a clinically healthy extraction sockets & periodontally affected extraction sockets groups, histological differences were not significant. 5. Root resorption or ankylosis in control group 1 & 2 had increased quantitatively as experiment progressed. 6. New bone formation developed from the base and lateral wall of extraction sockets. In both control groups & experimental groups, root surfaces lying next to the upper portion of extraction sockets showed little alveolar bone formation and surrounded by connective tissue fiber at weeks 2 & 8, while at weeks 12, they did show alveolar bone formation. 7. At week 12, experimental group 2 showed numerous cells which appeared to be periodontal ligament cells, with functionally arranged connective tissue fibers between the roots and alveolar bone.
