Xyloglucan in the Differentiating Xylem of the Populus deltoides M.

  • The chemical composition of the differentiating xylem of Populus deltoides M. was investigated and compared with that of sapwood. The cell wall polysaccharides were extracted sequentially from adifferentiating xylem and fractionated with gel chromatography. The sugar composition of each fraction was analyzed with G..C and H.P.L.C. The cell wall of the differentiating xylem is rich with the pectin substance and hemicellulose compared with that of sapwood. The water-extracted polysaccharides from the differentiating xylem were composed mainly of xylose and glucose residues. The sugar composition of some of the fractions in the gel filtration of purified $H_{2}O$ polysaccharide suggest that xyloglucan was extracted with $H_{2}O$ from differentiating xylem. Also, we can supposed that the purified $H_{2}O$ polysaccharide might be xyloglucan from the spectrometric data(IR and NMR) of purified $H_{2}O$ polysaccharide.

Detection of Bacillus Cereus Using Bioluminescence Assay with Cell Wall-binding Domain Conjugated Magnetic Nanoparticles

  • Bacillus cereus can cause blood infections (i.e., sepsis). Its early detection is very important for treating patients. However, an antibody with high binding affinity to B. cereus is not currently available. Bacteriophage cell wall-binding domain (CBD) has strong and specific binding affinity to B. cereus. Here, we report the improvement in the sensitivity of an ATP bioluminescence assay for B. cereus detection using CBD-conjugated magnetic nanoparticles (CBD-MNPs). The assay was able to detect as few as 10 colony forming units (CFU) per mL and $10^3CFU\;per\;mL$ in buffer and blood. CBD-MNPs did not show any cross-reactivity with other microorganisms. These results demonstrate the feasibility of the ATP assay for the detection of B. cereus.

대두 Protoplast의 세포벽 합성과 세포분열에 대한 Cytokinin의 영향 (Effects of Cytokinin on Cell Wall Regeneration and Cell Division of Soybean Protoplasts)

  • 대두(Glycine max var. Acme)의 자엽조직에서 유래된 callus의 부유배양세포로부터 protoplast를 분리하고, 이들 protoplast의 viability, 세포벽재생 및 세포분열에 대한 benzyladenine(BA)의 영향과 protoplast에 의한 BA의 흡수특성을 조사하였다. Protoplast의 viability는 BA처리에 의하여 증가되었고, 세포벽재생과 세포분열 그리고 callue의 생장도 BA처리에 의하여 촉진되었다. Protoplast에 의하여 흡수된 BA의 양은 BA 처리후 약 20시간에 최대에 이르렀고, 이중에서 약 2/3가 6시간 이내에 흡수되었다.

Microscopic Patterns of Decay caused by Tyromyces palustris and Gloeophyllum trabeum in Korean Red Pine and Radiata Pine Woods

  • The objective of this study was to elucidate the microscopic patterns of decay caused by brown-rot fungi of Tyromyces palustri and Gloeophyllum trabeum in Korean red pine (Pinus densiflora) and radiata pine (Pinus radiata) woods through light and electron microscopies. The ultrastructural changes of cell walls attacked by the two brown-rot fungi were compared in this respect. Macroscopically, radiata pine showed more ring and radial checks than Korean red pine. Microscopically, with the progress of decay, spiral checks associated with cross-field pits and bore holes in the cell wall were more remarkably numerous in the radiata pine than in the Korean red pine. In the radiata pine, G. trabeum produced more spiral checks in the cell wall than T. palustris. In the advanced stages of decay by G. trabeum, the erosions of ray cell walls were identified both in the Korean red pine and radiata pine but $S_3$ layers of tracheid walls were eroded only in the Korean red pine.

Analysis of the Formation of Protoplasts and Regeneration of Cells in Phycomyces blakesleeanus

  • It is possible ot prepare protoplasts of the zygomycete fungus, Phycomyces blakesleeanus, by digesting the cell wall of spore germlings with commercially available chitinase and chitosanase. However, the cells without any cell walls immediately form large aggregates, and thus, it is difficult to isolate the individually separated protoplasts. Inherent problem with the formation of aggregates in preparing protoplasts could be solved by the use of bovine serum albumin (BSA). As a result, we were able to prepare a large number of single protoplsts quickly and easily. We took time-lapse photomicrographs of the formation of protoplasts, and found that there were certain regions of the cell wall of spore germlings that were sensitive to chitinase and chitosanase, although the cell wall of the original spores is known to be insensitive to these enzymes. There are two kinds of cell walls on a spore germling; one with a bound wheat germ agglutinin (WGA), and the other a bound concanavalin A (ConA). Furthermore, only cells with walls which had bound WGA were able to regenerate, while those with walls with bound ConA were not able to regenerate.

낙엽송 열처리재의 세포벽 미세구조 및 항팽윤효율 평가 (Evaluation of Cell-Wall Microstructure and Anti-Swelling Effectiveness of Heat-Treated Larch Wood)

  • 본 연구에서는 낙엽송 열처리재의 세포벽 미세구조와 항팽윤효율을 평가하고, 이들 간의 상호관계를 분석하고자 하였다. 이를 위하여 낙엽송 판재를 190℃와 220℃의 온도 조건에서 12시간, 18시간, 24시간 동안 열처리하였다. 열처리된 낙엽송재의 횡단면을 1,000배 확대하여 관찰한 결과, 열처리재의 경우 세포가 원래의 형태를 유지하지 못 했으며, 세포의 단면이 거칠게 절단되었고, 세포 간 결합이 끊어지면서 세포간극이 늘어났음을 확인할 수 있었다. 또한, 처리 조건 별 팽윤 실험 결과, 열처리 온도와 시간이 증가함에 따라 수분 흡수량과 팽윤량이 감소하였으며, 항팽윤효율은 증가하는 경향을 보였다. 수분 흡수량의 감소는 열처리에 의한 세포벽의 화학적 변화에 영향을 받은 것으로 보이며, 팽윤량의 감소 및 항팽윤효율의 증가는 화학적 변화 뿐만 아니라 세포벽의 구조적인 변화와 같은 물리적인 변화가 복합적으로 영향을 미쳤기 때문인 것으로 생각된다.

  • This paper describes a new method to decide the margin for the sustain voltage of AC PDPs based on the wall-charge distribution. We model the discharge cell and measure the wall-charge when sustain pulses are applied to the AC PDP. The measured wall-charge distribution informs us of the voltage forming the maximum wall-charge which should be chosen as the sustain voltage.

Enterococcus faecalis 2B4-1 세포벽 성분 중 Polysaccharide Fraction의 종양세포 증식억제 효과 (Growth Inhibition of Polysaccharide Fraction in Cell Wall Components from Enterococcus faecalis 2B4-1 against Tumor Cell Lines)

  • This study was developed to evaluate the growth inhibition effects of cell wall components of Enterococcus faecalis 2B4-1 obtained from feces of neonates against tumor cell lines. Polysaccharide fraction (PS) shown sensitive growth inhibition effect in the cell wall components was isolated and characterized. In growth inhibition effects, residue fractin of whole cell was shown sensitive level of percent survival about 30% when administrated at ehe concentration of 100${\mu}$g/ml, and that was more effective than that of supernatant fraction against the tumor cell lines, SNU-1, 3LL, FARROW and HEC-1-B. Sensitive growth inhibition effects against SNU-1, FARROW and HEC-1-B were performed by whole cell (WC) fraction from Ent. faecalis 2B4-1. Cytoplasm fractin (CP) of WC was shown non-inhibition effect, however, the other part of WC, precipitate of disrupted cell (PD), was sensitive against the tumor cell line mentioned above. Followed by separation to peptidoglycan fraction (PG) and polysaccharide fraction (PS) were all sensitive which the latter was shown more sensitive percent survival than the former. Composed sugars of polysaccharide fraction were determined to D-glucose, L-rhamnose and D-glucosamine, and the rate fo composition was calculated to about 1:1:1 by the data of elemental analysis, IR, TLC and HPLC.

경피적 폐생검술후 흉벽에 전이된 폐암;1례 보고 (Chest Wall Implantation of Lung Cancer after Percutaneous Fine Needle Aspiration - Report of one case -)

  • Percutaneous needle aspiration has been widely used in the diagnosis of pulmonary lesions, because it is a fairly simple procedure with good diagnostic accuracy and low complication rate. Among its complications, the spread of malignant cells along the needle tract is rare but serious one. We report a case of chest wall implantation of lung cancer after the percutaneous fine needle aspiration biopsy. A 57-year-old man had undergone a right upper lobectomy for squamous cell carcinoma [T2N0M0] of the lung, 3 months after the operation, a growing mass, located far from the previous thoracotomy incision, developed on the right anterior chest wall where the diagnostic thin needle biopsy had been performed before the lobectomy. A wide excision of the chest wall mass was performed, and permanent histology showed squamous cell carcinoma as noted before.

애기장대 MYB7 유전자의 리그닌 생합성 억제 조절 (AtMYB7 Acts as a repressor of lignin biosynthesis in Arabidopsis)

  • 식물의 이차생장의 결과로 생산되는 바이오매스의 대부분은 식물의 이차세포벽에 축적된다. 식물의 이차세포벽은 크게 셀룰로스, 헤미셀룰로스, 리그닌이라는 3가지 물질로 구성되며 이들 중 페놀성 복합 화학 물질인 리그닌은 셀룰로스나 헤미셀룰로스와 달리 알코올 발효과정의 저해 물질로이다. 따라서, 식물의 이차세포벽 생합성의 과정을 조절함으로 전체 바이오매스의 생산량과 조성을 바이오 에너지 생산을 위해 최적 조건으로 조절함으로써 바이오 에탄올 생산을 최대화할 수 있을 것이다. 본 연구에서는 애기장대 유래 MYB7 유전자를 CaMV35S 프로모터 조절 하에서 과별현되게 한 식물체와 식물의 전사조절에서 하위 특정 유전자의 발현을 저해시키는 것으로 잘 알려진 SRDX 융합 단백질 즉, MYB7-SRDX 과발현체를 제작하여 그 특성을 조사하였다. 그 결과 MYB7 전사조절 인자가 리그닌의 생합성을 억제 조절한다는 결과를 관찰하였다. 이는 MYB7 전사조절인자를 이용하면 바이오 에탄올 발효과정에 저해 물질로 작용하는 리그닌을 경감시킬 수 있는 기초 자료로 사용할 수 있을 것이다.
