In an effort to obtain basic data of carbohydrate metabolism during spermiogenesis of the sexually-matured Jindo dog, the glycogen distribution in the testis was investigated by light and transmission electron microscopy. Periodic acid thiocarbohydrazide silver proteinate physical development(PA-TCH-SP-PD) staining method provided better results in the detection of glycogen granules from Sertoli cells and germ cells than the periodic acid schiff(PAS) staining method did. Pre-treatment of the tissue sections with ${\alpha}$-amylase elicited a significant decrease in PA-TCH-SP-PD stained granules, which suggested that the stained granules were of glycogen origin. High concentration of the glycogen granules were observed in the Sertoli cells, especially in its column, sheet-like processes, club-like processes, and tubular processes. The glycogen granules were unevenly distributed in some Sertoli cell columns. These results strongly indicated that the Sertoli cells of Jindo dogs showed vigorous activity of carbohydrate metabolism.

Cadmium (Cd) is an inorganic toxicant of great environmental and occupational concern which was classified as a human carcinogen in 1993. Occupational cadmium exposure is associated with lung cancer in human. In the present study, we established the mechanistic basis of apoptotic cell death induced by Cd in WI38 human lung fibroblast. Cd at 20-80 ${\mu}$M decreased viablility of cells in a concentration-dependent manner. PI staining, TUNEL staining and DNA fragmentation analysis demonstrated the apoptotic cell death by Cd. Through previous studies, the central nervous system (CNS) was collected by dividing the scuttle fly into larval, pupa, and adult stages by developmental stage, and the morphological characteristics were observed. In situ hybridization (ISH) using the collected central nervous system, it was possible to confirm the location and extent of expression of the neurotransmitter corazonin (Crz) at each stage of development. In this study, paraffin specimens were prepared using central nervous system tissues of 3rd instar larval stage of scuttle fly, which had completed in situ hybridization, and general histochemical staining (hematoxylin-eosin, H-E) and special histochemical staining (luxol fast blue-cresyl violet) was performed to observe the histological and cytological morphology characteristics of corazonin neurons. As a result, a variety of nerve cell body existed between many myelin sheath. The corazonin neurons compose cortex of central nervous system with other neurons congregating in this tissue and show larger cell body relatively in neurohistochemical analysis.
목 적: 본 연구에서는 에스트로겐 수용체의 agonist로 작용할 수 있는 내분비계 교란물질인 bisphenol A (BPA)가 정소 내 세포들의 유전자 발현 및 세포자멸사에 미치는 영향을 관찰하였다. 연구방법: 그룹 I의 7주령 수컷 생쥐들에는 sesame oil에 녹인 BPA를 1 mg/kg, 10 mg/kg, 100 mg/kg를 1회 복강주사하여 급성 영향을 조사하였고, 그룹 II의 생쥐들에는 BPA 10 ${\mu}g/kg$, 1 mg/kg, 100 mg/kg를 하루에 1회씩 14일간 피하주사하여 장기적 효과를 보았다. 생쥐의 정소를 채취하여 정소의 다양한 표지유전자들의 발현을 RT-PCR로 조사하였고, 조직학적 관찰, 형광면역염색법과 TUNEL 염색을 수행하였다. 결 과: RT-PCR 결과, 정소의 표지유전자 중 특이적으로 Leydig cell의 표지유전자인 LHR가 고농도의 BPA 처리군에서 감소되어 있었다. 정원세포 표지유전자인 ERM은 장기간 BPA를 주사 받은 생쥐들의 정소에서 감소되었다. 1회의 BPA를 주사 받은 그룹에서는 조직학적 영향은 없는 것으로 보였다. 또한 세포자멸사의 표지인 Bax가 Leydig cell이 존재하는 정소의 interstitial area에서 BPA에 의해 증가된 것을 관찰하였으며 이는 TUNEL 염색이 positive하게 나타난 부분과 일치하였다. 결 론: 본 연구는 BPA가 정소에서 주로 Leydig cell에 영향을 주며 이는 Leydig cell이 에스트로겐 수용체와 에스트로겐 합성효소를 발현하는 내분비 세포라는 사실에 부합되는 것으로, 차후 그 분자적 기작을 밝히는 연구로 이어져야 할 것이다.
Chlamydia trachomatis is one of the most common cause in non-gonococcal urethritis. There are several diagnostic methods for Chlamydia trachomatis; culture method using McCoy cell, enzyme immunoassay and direct immunofluorescence staining etc. We have studied a sensitivities of culture, chlamydiazyme and direct immunofluorescence staining(DIF). 85 male patients previously conformed to non-gonococcal urethritis have been selected in this study. Three samples were concurrently collected in the same patient. First sample was used to inoculation in McCoy cell, 2'nd sample was used to Chlamydiazyme test and 3'rd sample was used to direct immunofluorescence staining method. The results are following. 1. All culture, Chlamydiazyme and DIF positive cases are 15/85(17.7%). 2. Culture and Chlamydiazyme positive but DIF negative cases absent. 3. Culture and DIF positive, but Chlamydiazyme negative cases are 2/85(2.4%). 4. Chlamydiazyme and DIF positive, but culture negative cases are 9/85(10.6%). 5. Culture positive, but Chlamydiazyme and DIF negative cases are 6/85(7.1%). 6. Chlamydiazyme positive, but culture and DIF negative cases are 7/85(8.2%). 7. DIF positive, but culture and Chlamydiazyme negative cases are 3/85(3.5%). 8. All culture, Chlamydiazyme and DIF negative cases are 43/85(50.1%). In summarized, anyone positive cases of culture, Chlamydiazymc and DIF are 42/85(49.9%).
Background: Hepatocellular carcinoma (HCC) is the fifth most prevalent cancer and thirdly leading cause of cancer-related death worldwide. The estimated risk of hepatocellular carcinoma is 15 to 20 times as high among persons infected with HCV as it is among those who are not infected, with most of the excess risk limited to those with advanced hepatic fibrosis or cirrhosis. Glypican3 (GPC3) plays a key role in relation to signaling with growth factors, regulating the proliferative activity of cancer cells. Glutamine synthetase (GS) catalyzes the synthesis of glutamine from glutamate and ammonia in the mammalian liver. GS was suggested as a specific marker for tracing cell lineage relationships during hepatocarcinogenesis. In normal liver, GS expression is seen in pericentral hepatocytes, but not by midzonal or periportal hepatocytes. In HCC, strong and diffuse GS expression in seen in tumor cells. Results: Glypican3 immunopositvity was highly specific and sensitive indicator for hepatocellular carcinoma as well as glutamine synthetase which was found to be a sensitive and specific indicator for development of hepatocellular carcinoma when compared to cirrhosis, liver cell dyspalsia and metastatic carcinomas. Statistical analysis revealed a significant association between GPC3 and GS with tumor size (P=0.003, p=0.006, respectively). Diffuse staining significantly associated with large tumor size while, focal and mixed staining was detected more with small tumor size. Studying the relation with tumor grade also revealed significant association between diffuse GPC3 and GS staining with high tumor grade. Diffuse staining was detected in 91.7% and 100% respectively of poorly differentiated specimens and only in 33.3% and 22.2% of well differentiated specimens. Conclusions: While using GPC3 and GS to screen for premalignant hepatic lesions remains controversial, our data suggest that GPC3 and GS may be a reliable diagnostic immunomarkers to distinguish HCC from benign hepatocellular lesions. However, negative immunostaining should not exclude the diagnosis of HCC.
We cytogenetically analyzed a triploid King-Nupchi strain of the olive flounder Paralichthys olivaceus to define the simplest, most rapid, and most effective method of ploidy analysis in aquaculture farms. Female triploidy of the flounder King-Nupchi strain was induced by cold shock (3 min post-fertilization at 2-4℃ for 45 min). Triploid induction was confirmed by erythrocyte measurement (nuclear volume, 29.15±2.10 μm3); flow cytometry (2.14±0.03 pg/cell); chromosome count (3N=72); Ag-NOR banding; and silver staining. Silver staining of finned cells obtained using a solid tissue technique was the most effective method of ploidy verification.
The anti-cancer effects of Angelica keiskei ethanol extract were evaluated in human breast cancer MDA-MB-231 cells. The concentrations of extract were 1, 2, 3, 4 and 5 mg/mL. Dose-dependent reductions in the number of cells with altered cell shape and pyknotic nuclei were observed at 48 h after treatments. MTT assay also exhibited a similar dose-dependent reduction in mitochondrial reductase activity (p<0.05), in particular, with a rapid reduction in the activity of the 5 mg/mL group. Analysis of cell death with propidium iodide (PI) staining revealed only a slight increase in cell death in the 5 mg/mL group. Analysis of bromodeoxyuridine (BrdU) incorporations also showed a dose-dependent reduction in cell proliferation (p<0.05). Finally, increases in total radical oxygen species (ROS) accumulation in cells, as revealed by DCF-DA staining, were observed in the treated groups in a similar dose-dependent fashion (p<0.05). These results indicate that Angelica keiskei ethanol extract exhibiting anti-cancer effects in MDA-MB-231 cells causes multiple changes in cell shape, enzyme activity, and ROS accumulation, thereby inducing cell death.

Atopic dermatitis (AD) is a chronic inflammatory skin disease with a complex etiology that encompasses immunologic responses. AD is frequently associated with elevated immunoglobulin (Ig) E levels, and common environmental factors contribute to its pathogenesis. Several recent studies have documented the role of specific lactic acid bacteria in the treatment and prevention of AD in humans and mice. In this study, the efficacy of Duolac ATP, a probiotic preparation, was determined in a mouse model with AD-like skin lesions. Alterations in the cytokine levels and histological staining suggested the alleviation of AD. The in vivo test showed that T helper (Th)2 cytokines, IgE, interleukin (IL)-4, and IL-5, were significantly downregulated, whereas Th1 cytokines, IL-12p40 and interferon (IFN)-${\gamma}$, were upregulated in all groups of mice treated with Duolac ATP compared to that observed in the group of mice treated with 1-chloro-2,4-dinitrobenzene (DNCB) alone. Moreover, the scratch score decreased in all mice treated with Duolac ATP. Staining of the dorsal area of the mice in each group with hematoxylin and eosin and toluidine blue further confirmed the alleviation of AD in mice orally treated with Duolac ATP. These results suggest that Duolac ATP inhibits the development of AD-like skin lesions in NC/Nga mice by suppressing the Th2 cell response and increasing the Th1 cell response. Thus, Duolac ATP is beneficial and effective for the treatment of AD-like skin lesions.
Background: To investigate the effects of ASMase mediated endothelial cell apoptosis in multiple hypofractionated irradiations in CT26 tumor bearing mice. Materials and Methods: Thirty-five CT26 tumor bearing mice were subjected to single ionizing radiation (IR) of 0, 3, 6, 9, 12, 15, 18 Gy. Eight hours after IR, the mice were sacrificed and tumor tissues were used for CD31 immunohistochemistry staining, TUNEL and CD31 double staining, ASMase activity assay. Then 6 and 12 Gy were chosen for multiple hypofractionated IR experiments according to the above results. Each time after IR, 5 mice were sacrificed and assayed as above. Results: The ASMase activities were increased significantly after a single IR of 12 Gy or higher which was accompanied with remarkable increased endothelial cell apoptosis and decreased MVD. For 6 Gy which was not high enough to trigger ASMase activation, after 2 or more times of IR, the ASMase activities were significantly increased accompanied with remarkable increased endothelial cell apoptosis and decreased MVD. While for 12 Gy, after 2 or more times of IR, the ASMase activities and endothelial cell apoptosis rates were maintained without remarkable increase; however, the MVD was significantly decreased. What's more, the cancer cell apoptosis rates were significantly increased after multiple IR for both 6 Gy and 12 Gy. Conclusions: ASMase mediated endothelial cell apoptosis may play an important role in the process of multiple hypofractionated IR for CT26 colorectal carcinoma.
