This study demonstrated anti-oxidative and neuroprotective effects of Rhei Rhizoma. Anti-oxidative effects were studied on BV-2 microglia cells damaged by $H_2O_2$ and nitric oxide. Neuroprotective effects were studied by using oxygen/glucose deprivation of the organotypic hippocampal slice cultures. The results obtained are as follows; The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of neuronal cell death area and cell death area percentages in CA1 region of ischemic damaged hippocampus cultures during whole 48 hours of the experiment. The group treated with 50 mg/ml of Puerariae Radix demonstrated decreases of neuronal cell death area and cell death area percentages in CA1 region, but these were not significant statistically. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of neuronal cell death area and cell death area percentages in dentate gyrus of ischemic damaged hippocampus cultures during whole 48 hours of the experiment. The group treated with 50 mg/ml of Puerariae Radix demonstrated decreases of neuronal cell death area and cell death area percentages in dentate gyrus, but these were not significant statistically. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of TUNEL-positive cells in both CA1 region and dentate gyrus of ischemic damaged hippocampus cultures. The group treated with 50 mg/ml of Puerariae Radix demonstrated significant decrease of TUNEL-positive cells in CA1 region, but not in dentate gyrus of ischemic damaged hippocampus. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of LDH concentrations in culture media of ischemic damaged hippocampus cultures. The group treated with 50 mg/ml of Puerariae Radix demonstrated decrease of LDH concentrations in culture media, but it was not significant statistically. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant increases of cell viabilities of BV-2 microglia cells damaged by $H_2O_2$. The group treated with 50 mg/ml of Puerariae Radix demonstrated increase of cell viability of BV-2 microglia cells, but it was not significant statistically. The group treated with 0.5 mg/ml of Puerariae Radix revealed significant increase of cell viability of BV-2 microglia cells damaged by nitric oxide. The groups treated with 5 and 50 mg/ml of Puerariae Radix demonstrated increases of cell viabilities of BV-2 microglia cells, but these were not significant statistically. These results suggested that Puerariae Radix revealed neuroprotective effects through the control effect of apoptosis and oxidative damages.
A strong oxidative stress-inducible peroxidase promoter (referred to as SWPA2 promoter) was cloned from tell cultures of sweetpotato (Ipomoea batatas) and characterized in transgenic tobacco cultured cells in terms of biotechnological applications. Employing a transient expression assay in tobacco protoplasts, with five different 5'-deletion mutants of the SWPA2 promoter fused to the $\beta$-glucuronidase (GUS) reporter gene, the 1314 bp deletion mutant showed approximately 30 times higher GUS expression than the CaMV 35S promoter. The expression of GUS activity in suspension cultures of transgenic cells derived from transgenic tobacco leaves containing the -1314 bp SWPA2 promoter-GUS fusion was strongly expressed following 15 days of subculture compared to other deletion mutants, suggesting that the 1314 bp SWPA2 promoter will be biotechnologically useful for the development of transgenic cell lines engineered to produce key pharmaceutical proteins. In this respect, we developed transgenic cell lines such as tobacco (Nicotiana tabacum L. BY-2), ginseng (Panax ginseng) and Siberian ginseng (Acanthopanax senticosus) using a SWPA2 promoter to produce a human lactoferrin (hLf) and characterized the hLf production in cultured cells. The hLf production monitored by ELISA analysis in transgenic BY-2 cells was directly increased proportional to cell growth and reached a maximal level (up to 4.3% of total soluble protein) at the stationary phase in suspension cultures. The SWPA2 promoter should result in higher productivity and increased applications of plant cultured cells for the production of high-value recombinant proteins.
The physiological characteristics of cultures of very high cell mass (e.g. 10g cell mass/L), termed“ultrahigh cell density cultures”is reviewed. A close relationship was found between the length of the optical path (OP) in flat-plate reactors and the optimal cell density of the culture as well as its areal (g m$\^$-2/ day$\^$-1/) productivity. Cell-growth inhibition (GI) unfolds as culture density surpasses a certain threshold. If it is constantly relieved, a 1.0cm OP reactor could produce ca. 50% more than reactors with longer OP, e.g. 5 or 10cm. This unique effect, discovered by Hu et al. [3], is explained in terms of the relationships between the frequency of the light-dark cycle (L-D cycle), cells undergo in their travel between the light and dark volumes in the reactor, and the turnover time of the photosynthetic center (PC). In long OP reactors (5cm and above) the L-D cycle time may be orders of magnitude longer than the PC turnover time, resulting in a light regime in which the cells are exposed along the L-D cycle, to long, wasteful dark periods. In contrast, in reactors with an OP of ca. 1.0 cm, the L-D cycle frequency approaches the PC turnover time resulting in a significant reduction of the wasteful dark exposure time, thereby inducing a surge in photosynthetic efficiency. Presently, the major difficulty in mass cultivation of ultrahigh-density culture (UHDC) concerns cell growth inhibition in the culture, the exact nature of which is awaiting detailed investigation.
본 연구에서는 Phellinus linteus 균사체의 액상배양을 통한 면역증강 생리활성 효능의 단백다당체 생산공정을 개발하기 위한 시도로서, 우선 생산균체의 원형질체 형성을 통한 고생산성 균주를 개발하고자 하였으며, 발효기 액상배양 시 최적 배양형태의 유도를 통해 균사체와 단백다당체의 생산성을 극대화하고자 하였다. 본 연구실에서 채취한 생산 균주를 ITS rDNA sequencing 방법과 blast search 방법에 의해 조사한 결과 다양한 Phellinus linteus 종들과 99.67% 이상의 유사성 확인되어, 이 균주를 Phellinus linteus라고 최종적으로 동정할 수 있었다. 이 동정된 균주로부터 균주 개량을 시도하기 위해 Phellinus linteus 균사체로부터 대량의 원형질체 형성 및 재생에 의한 단일 콜로니 획득 방법을 개발함으로써 균주를 신속하게 개량할 수 있었다. Sorbitol을 이용한 banding filtration 방법을 이용하여 원형질체를 회수한 결과 $10^5{\sim}10^6\;protoplasts/ml$를 얻을 수 있었으며, 원형질체 재생률은 $10^{-2}{\sim}10^{-3}$로 나타났다. 균주개량을 위해 원형질체 재생배지와 고체배양배지에서 고성장성 및 고안정성을 보이는 균주들을 지속적으로 대량 선별하여, 액상 생산배양을 수행하였다. 그 결과 균사체량은 13~15 g/L로 대부분 비슷하게 자랐으며, 조단백다당체의 함량 또한 5.8~6.4%로 거의 비슷하게 분포하는 것으로 나타났는데, 이로부터 고체배양배지에서 빠른 성장속도를 보여주는 균주들이 대부분 액상 생산배양에서도 고생산성 및 고안정성을 보여주는 것을 확인할 수 있었다. 한편 Phellinus linteus 균사체의 경우 조단백다당체의 함량이 세포 무게당 거의 일정한 양을 함유하고 있는 것으로 확인되었으므로, 조단백다당체의 생산성을 증가시키기 위해서는 최종 생산배양에서의 균체량 증가가 가장 중요한 것으로 판단되어, 균사형성 고등균류의 균사체 배양 시 균체량 증가에 가장 중요한 요인 중의 하나인 생산균주의 배양형태적 특성에 대해 집중적으로 조사하였다. 균주개량 실험을 통해 고생산성 균주로 최종 결정된 AR147 균주를 이용해서 다양한 배양조건에서 발효조 배양을 수행한 결과, 최종 생산발효조로의 접종원이 고농도의 균사모양인 경우에 생산균주의 배양형태가 매우 작은 compact한 펠렛 모양(대부분 직경 0.5 mm 이하)을 유지하는, 이상적인 균사체 액상배양 공정이 이루어지는 것으로 확인되었다. 즉 생산 발효조배양에서 직경 0.5 mm 이하의 compact한 펠렛 모양의 배양형태가 유도되었을 경우, lag phase 시간의 획기적 감소와 1.5배 이상의 높은 세포비성장속도로 인해, 최종 균사체생산성이 다른 배양형태를 유도한 경우에 비해 약 3.3배 더 높은 주목할 만한 배양결과를 얻을 수 있었다. 이로부터 균사 형성 Phellinus linteus의 산업용 발효조 배양 시, 각 배양단계에서의 생산균체의 배양형태가 최종 균체생산성, 궁극적으로는 최종 단백다당체의 생산성에 심각한 영향을 미친다는 것을 알 수 있었다.
Guggulsterone, a hypolipidemic natural agent, is produced in resin canals of the plant Commiphora wightii. In this study, the stimulatory effects of growth retardants [ALAR (N,N-dimethylaminosuccinamic acid) and CCC (chlormequat chloride)] and fungal elicitor on guggulsterone accumulation in cell cultures of C. wightii are reported. CCC at $1\;mg\;l^{-1}$ enhanced guggulsterone content (${\sim}123\;{\mu}g\;l^{-1})$ when added on the fifth day after inoculation, while ALAR at $2.5\;{\mu}g\;l^{-1}$ increased guggulsterone content (${\sim}116\;{\mu}g\;l^{-1}$) when added on the tenth day. In a two-stage fed-batch process, combined treatment with fungal elicitor and growth retardant caused a significant increase (${\sim}353\;{\mu}g\;l^{-1}$) in guggulsterone content in cell cultures after 17 days of growth. This represents an approximately fivefold increase over the guggulsterone contents in initial cultures of this plant.
Effects of various environmental factors on cell size index(FCW/DCW) in Thalictrum rugosum. Lithospermum erythrorhizon and Taxus cuspidata plant cell suspension cultures were investigated. Time course change of cell size index were also observed. In batch cultures, FCW/DCW increased according to the decrease of sugar concentration. For short-term experiment within 24 hr, FCW/DCW value could be reduced significantly by increasing sugar concentration. When an osmoticum such as mannitol was added, FCW/DCW converged to a low value. Therefore, it was confirmed that osmolality of the medium was important in determining cell size or water content of the cells. Inorganic salts or treatment with organic solvent also exhibited some effect on the cell size index. However, pH and centrifugal force did not show any influences. On the other hand, it was found that the addition of Pluronic F-68 reduced FCW/DCW. By combining these results effectively, it may be possible to increase the cell concentration in high density culture to a higher extent.
In the present study, the protective effects of Bcl-2 over-expression in a suspension culture (without any adaptation) and spent medium (low nutrient and high toxic metabolite conditions) were investigated. In the suspension culture without prior adaptation, the viability of the control cell line fall to 0% by day 7, whereas the Bcl-2 cell line had a viability of 65%. The difference in the viability and viable cell density between the Bcl-2 and control cell lines was more apparent in the suspension culture than the static culture, and became even more apparent on day 6. Fluorescence microscopic counting revealed that the major mechanism of cell death in the control cell line in both the static and suspension cultures was apoptosis. For the Bcl-2 cell lines, necrosis was the major mode of cell death in the static culture, but apoptosis became equally important in the suspension culture. When the NS0 6A1 cell line was cultured in spent medium taken from a 14 day batch culture, the control cell line almost completely lost its viability by day 5, whereas, the Bcl-2 still had a viability of 73%. The viable cell density and viability of the Bcl-2 cell line cultivated in fresh medium were 2.2 and 2.7 fold higher, respectively, than those of the control cultures. However, the viable cell density and viability of the Bcl-2 cultivated in the spent medium were 8.7 and 7.8 fold higher, respectively, than those of the control cultures. Most of the dead cells in the control cell line were apoptotic; whereas, the major cell death mechanisms in the Bcl-2 cell line were necrotic.
Cell cultures of Commiphora wightii (Arnott.) Bhandari were grown in shake flasks and a bioreactor and an increase in guggulsterone accumulation up to $18{\mu}g\;l^{-1}$ was recorded in cells grown in the production medium containing a combination of sucrose:glucose (4% total), precursors (phenylalanine, pyruvic acid, xylose, and sodium acetate), morphactin, and 2iP. A yield of $10g\;l^{-1}$ biomass and ${\sim}200{\mu}g\;l^{-1}$ guggulsterone was recorded in a 3-l flask and in a 2-l stirred tank bioreactor compared with 6.6 g biomass and $67{\mu}g\;l^{-1}$ guggulsterone in 250-ml flasks. Increased vessel size was correlated with increased biomass and guggulsterone accumulation. 2iP alone was not effective for biomass and guggulsterone accumulation in cell cultures of C. wightii.
Cell cultures of Cayratia trifolia (Vitaceae), a tropical lianas, were maintained in Murashige and Skoog's medium containing $0.25mg\;1^{-1}$ NAA, $0.2mg\;1^{-1}$ kinetin and casein hydrolysate $250mg\;1^{-1}$. Cell suspension cultures of C. trifolia accumulate stilbenes (piceid, resveratrol, viniferin, ampelopsin), which on elicitation by any of $500{\mu}M$ salicylic acid, $100{\mu}M$ methyl jasmonate, $500{\mu}M$ ethrel and $500mg\;1^{-1}$ yeast extract, added on the 7th day, were enhanced by 3- to 6-fold ($5-11mg\;1^{-1}$) by the 15th day.
Thalicrtrum rugosum 식물세포 현탁배양에 의한 berberine 생산에 미치는 여러 가지 종류의 elicitor들의 영향에 관해 연구 하였다. 효모 유래의 elicitor, 15가지의 서로 다른 종류의 무생물 종류 elicitor, 3가지 곰팡이로부터 얻은 elicitor 들을 처리하여 세포의 생장 및 berberine 생산에 미치는 영향을 비교 분석해 본 결과, 이들 elicitor들은 T. rugosum에 의한 berberine 생산을 크게 증대시키지 못함을 확인하였다.
