한국인의 대장암 세포주에서 p53 돌연변이의 발견과 발현에 관한 연구 (Study on the expression and detection of the p53 mutation in Korean colon cancer cell lines)

  • Background: Inactivation in p53 tumor suppressor gene through a point mutation and deletion is one of the most frequent genetic changes found in human cancer, with 50% of an incidence. This high rate of mutation mostly suggests that the gene plays a central role in the development of cancer and the mutations detected so far were found in exons 5 to 8. Mutation of p53 locus produced accumulation of abnormal p53 protein, and negative regulation of cell proliferation and transcriptional activation as a suppressor of transformation were lost. In addition, inhibition of its normal cellular function of wild-type by mutant is an important step in tumorigenesis. Method: 4 colon cancer cell lines (SNU C1, C2A, C4, C5) were examined for mutation in exons 5 to 8 of the p53 tumor suppressor gene by PCR-SSCP analysis and expression pattern by western blotting and immunoprecipitation. p53-mediated transactivation ability were examined by CAT assay and base substitution of p53 in SNU C2A cell were detected by DNA sequencing. Results: 1) SNU C2A cell and SNU C5 cell were detected mobility shifts each in exon 5 and exon 7 of p53 gene by the PCR-SSCP method, implicating being of p53 mutation. 2) 3 colon cancer cell lines (SNU C1, SNU C2A, SNU C5) expressed wild type and mutant type p53 protein. 3) In northern blot experiment, SNU C2A and SNU C5 cell expressed high level of p53 mRNA. 4) Results of p53-mediated transactivation in colon cancer cell lines by CAT assay represented only SNU C2A cell has transcriptional activity. 5) DNA sequencing in SNU C2A cell showed missense mutation in codon 179 of one allele, histidine to arginine and wild type p53 in the other allele. Conclusion: Colon cancer cell lines showed correlation with mutation in p53 gene and accumulation of abnormal p53 protein. Colon cancer cell SNU C2A retained p53-mediated transactivation as heterozygous p53 with one mutant allele in 179 codon and the other wild-type allele.

  • The development of routine techniques for the isolation and in vitro maintenance of conducting airway epithelial cells in a differentiated state provides an ideal model to study the factors involved in the regulation of the expression of mucocilicary differentiation. Several key factors and conditions have been identified. These factors and conditions include the use of biphasic culture technique to achieve mucociliary differentiation and the use of such stimulators, the thickness of collagen gel substratum, the calcium level, and vitamin A, and such inhibitors, the growth factors EGF and insulin, and steroid hormones, for mucous cell differentiation. Using the defined culture medium, the life cycle of the mucous cell population in vitro was investigated. It was demonstrated that the majority of the mucous cell population in primary cultures is not involved in DNA replication. However, the mucous cell type is capable of self-renewal in culture and this reproduction is vitamin A dependent. furthermore, differentiation from non-mucous cell type to mucous cell type can be demonstrated by adding back a positive regulator such as vitamin A to the “starved” culture. Cell kinetics data suggest that vitamin A-dependent mucous cell differentiation in culture is a DNA replication-independent process and the process is inhibited by TGF-${\beta}$1.

로봇용 자연급기형 연료전지의 최적구조에 관한 연구 (Study on optimum structure of free-breathing Polymer Electrolyte Membrane Fuel Cell (PEMFC) for robotic application)

  • The performance of free-breathing polymer electrolyte membrane fuel cell (PEMFC) was studied experimentally and the effect of cathode separator structure on the cell performance was investigated. Cathode separators were used for the cell with $18cm^2$ active area. In channel type, the contact resistance is low, and the nature convection. which is strongly affected by the cross-sectional shape of cathode separator channel, is dominant in a cell performance. The maximum power density with $18cm^2$ active area is $105mW/cm^2$ using the 10mm depth and 2mm width channel. A computational analysis was optimum structure of free-breathing channel type PEMFC for robotic application.

동양달팽이 (Nesiohelix samarangae) 타액선의 조직화학적 및 미세구조적 연구 (Histochemical and Ultrastructural Studies on the Salivary Gland of a Land Snail, Nesiohelix samarangae)

  • Histochemical and ultrastructural studies on the salivary gland and salivary duct of a land snail Nesiohelix samarangae were conducted to observe structural characteristics and function. The salivary gland consisted of one type of epithelial cell, one type of supporting cell, and six types of gland cells. Four out of six gland cell types were histochemically identified on these secretions. The one secreted acid mucopolysaccharide and the other three secreted neutral mucopolysaccharide. The salivary duct epithelium had only one type of columnar cell with microvilli on its luminal surface. The basal protoplasmic membranes of the epithelial cells were deeply infolded so many times all along the cell bases.

Structural Transition of A-Type Zeolite: Molecular Dynamics Study

  • Molecular dynamics (MD) calculations were carried out in order to investigate the effect of MD cell size to predict the melting phenomena of A-type zeolite. We studied two model systems: a pseudocell of $(T_2O_4Na)_n$, (L= 12.264 $^{\AA}$, N= 84) and a true-cell of (SiAlO$_4Na)_n$. (L= 24.528 $^{\AA}$, N= 672), where T is Si or Al. The radial and bond angle distribution functions of T(Si, Al)-O-T(Si, Al) and diffusion coefficients of T and O were reported at various temperatures. For the true-cell model, the melting temperature is below 1500 K and probably around 1000 K, which is about 600-700 K lower than the pseudocell model. Although it took more time (about 30 times longer) to obtain the molecular trajectories of the true-cell model than those of the pseudocell model, the true-cell model gave more realistic structural transition for the A-type zeolite, which agrees with experiment.

  • 본 논문에서는 네트워크가 많은 endpoint를 가질지라도 낮은 비트율의 데이터를 효율적으로 전송할 수 있는 AAL 유형 2 스위치를 포함하는 ATM스위치 구조를 제안한다. 컴퓨터 프로그램으로 모델링한 ATM스위치는 AAL 유형 1, AAL 유형 2, AAL 유형 3/4 및 AAL 유형 5 셀로 이루어진 모든 유형의 AAL 셀에 대해 셀 스위칭을 지원하고 있다. 우리는 2가지 방식의 스위치를 제안하고 있는데, 하나는 개별적인 입력 포트마다 AAL 유형 2 셀 처리를 지원하는 스위치 패브릭이고, 다른 하나는 모든 입력 포트에 대한 전체적인 AAL 셀 처리를 지원하는 스위치 패브릭이다. 시뮬레이션 결과는 후자의 방식이 전자의 방식보다 우수한 것으로 나타났다. 그러나, 전자의 방식이 구현이 용이하고, 확장성에 대한 장점을 가지고 있다. 따라서 본 논문에서는 전자의 방식을 채용한 AAL 유형 2 스위치 모듈을 VHDL 언어를 사용하여 설계하였으며, 이를 FPGA로 구현하였다. 설계된 칩은 52MHz에서 동작하였다. 본 논문의 ATM 스위치 패브릭은 범용의 ATM 스위치 패브릭으로서 뿐만 아니라 ATM 네트워크상으로 모바일 통신, 협대역 서비스 및 무선 ATM등에 폭넓게 응용될 것으로 사료된다.

  • Transient current (TC) on bulk-state liquid crystal mixtures was measured. We found that TC is very sensitive to impurities and the features of TC curves depend on the type of contamination, from which the quality of materials can be definitely evaluated and the type of impurities can also be revealed.

Nanoscale imaging of rat atrial myocytes by scanning ion conductance microscopy reveals heterogeneity of T-tubule openings and ultrastructure of the cell membrane

  • In contrast to ventricular myocytes, the structural and functional importance of atrial transverse tubules (T-tubules) is not fully understood. Therefore, we investigated the ultrastructure of T-tubules of living rat atrial myocytes in comparison with ventricular myocytes. Nanoscale cell surface imaging by scanning ion conductance microscopy (SICM) was accompanied by confocal imaging of intracellular T-tubule network, and the effect of removal of T-tubules on atrial excitation-contraction coupling (EC-coupling) was observed. By SICM imaging, we classified atrial cell surface into 4 subtypes. About 38% of atrial myocytes had smooth cell surface with no clear T-tubule openings and intracellular T-tubules (smooth-type). In 33% of cells, we found a novel membrane nanostructure running in the direction of cell length and named it 'longitudinal fissures' (LFs-type). Interestingly, T-tubule openings were often found inside the LFs. About 17% of atrial cells resembled ventricular myocytes, but they had smaller T-tubule openings and a lower Z-groove ratio than the ventricle (ventricular-type). The remaining 12% of cells showed a mixed structure of each subtype (mixed-type). The LFs-, ventricular-, and mixed-type had an appreciable amount of reticular form of intracellular T-tubules. Formamide-induced detubulation effectively removed atrial T-tubules, which was confirmed by both confocal images and decreased cell capacitance. However, the LFs remained intact after detubulation. Detubulation reduced action potential duration and L-type Ca2+ channel (LTCC) density, and prolonged relaxation time of the myocytes. Taken together, we observed heterogeneity of rat atrial T-tubules and membranous ultrastructure, and the alteration of atrial EC-coupling by disruption of T-tubules.

가리비, Patinopecten yessoensis 혈구의 형태학적 동정과 기능 (Identification of Blood Cells and Their Physiological Functions in the Scallop, Patinopecten yessoensis)

  • 일본 북해도의 망주만에서 방류양식된 가리비, Patinopecten yessoensis를 재료로 하여, 전자현미경적 및 조직화학적 관찰을 토대로 혈구의 미세구조에 따른 형태학적 동정 및 생리학적 기능을 연구한 결과는 다음과 같다. 가리비의 체내에 분포하는 혈구는 3종류로 구분되었다. I형 : 세포의 외형은 타원형으로 세포질의 전자밀도가 비교적 낮다. 타원형의 핵은 가끔 2핵으로 분엽하며 그 주변부에 소관상의 골면소포체와 액포가 발달하나, 유리 리보솜의 양은 적다. II형 : 세포의 외형은 장타원형으로 세포질은 전자밀도가 높다. 핵은 타원형으로 분엽하지 않으며 그 주변부에 소양상의 골면소포체와 유리 리보솜이 발달하나, 액포는 존재하지않는다. III형 : 세포의 외형은 원형으로 전자밀도는 3종의 혈구중 가장 높다. 핵은 차축상으로 염색질결절이 많으며 세포질에는 조면소포체가 현저하게 발달하나, 액포는 존재하지 않는다. 이들 혈구에 대한 미세구조 및 식작용에 관한 조직화학적 실험을 통하여 그 기능을 검토한 결과, I형은 주로 세균 등의 외래 물질 및 내인성 노화세포의 식작용에, II형은 영양물질의 운반에 관여하며, 특히, III형혈구는 체액성 방어기구과 관련한 모종의 단백질 생산분비 기능을 가지는 것으로 판단되었다

한국산 낙지(Octopus minor) 흡반(Sucker)내 분비 및 감각수용세포 II (Secretory and Sensory Receptor Cells in the Sucker of Korean Octopus minor II)

  • 두족강 낙지류(Octopus minor)의 흡반 누두부 상피조직에서 관찰된 감각세포들은 A1, A2, B, C그리고 D형세포 등 5종류가 관찰되었다. A형세포는 A1과 A2형 등 두 종류가 확인되었다. A1형은 주로 흡반의 누두부 상피조직에서 B형세포와 나란히 연접되어 있었으며 핵은 불규칙한 형태였고 핵질은 전자밀도가 높아서 어둡게 관찰되었다. 세포질 속에는 대, 소형의 많은 공포들(직경 $0.4\sim0.04{\mu}m$)로 가득차 있었으며 이들은 세포의 상단쪽으로 이동하면서 당피막을 통해 분비되었다. A2형은 주로 상피조직의 하단부에 존재하였으며, 핵의 형태는 A1형과 같았다. 세포질은 사상형 또는 망상형으로 전자밀도가 높게 나타났다. B형세포는 난원형의 핵을 소지하고 있었으며, 세포질속에는 소포체 모양의 많은 공포들과 전자밀도가 높은 다양한 크기$(0.6\sim0.25{\mu}m)$의 둥근 과립들이 있었는데 이들은 당피막을 통해 세포의 자유면으로 분비되었다. C형세포와 D형세포는 주로 흡반상피의 주름진 부위에서 단층 또는 다층 구조 형태로 관찰되었다. C형세포는 전자밀도가 낮은 긴 타원형의 핵을 소지한데 비해, D형은 이질염색질이 발달된 불규칙한 핵을 소지하고 있었다.
